2.Purification of L-amino acid oxidase from Naja atra venom and its effects on endothelial cells

Aim To purify L-amino acid oxidase(LAAO) from the venom of Naja atra and study its effect on endothelial cells.Methods The NAV-LAAO was purified by ion-exchange chromatography and affinity chromatography.The MTT assay and Western blot were used to detect the viability and apoptosis of HUVEC.The tubule-forming was used to study the angiogenesis of cells.Results The NAV-LAAO was purified successfully from the venom of Naja atra.The molecular weight of NAV-LAAO was determined to be 58 ku by SDS-PAGE.NAV-LAAO effectively inhibited the growth and tubule-forming of HUVEC,and the 50% inhibitory concentration(IC50) was 21.42 mg?L-1.Compared with control,the levels of caspase-3 and caspase-8 increased in HUVEC treated with NAV-LAAO.Conclusions The NAV-LAAO is purified successfully from Naja atra venom by ion-exchange chromatography and affinity chromatography.The NAV-LAAO inhibits the growth and tubule-forming capacity of HUVEC in a dose-dependent manner.
3.Case-control study of factors associated with aortic dissection DeBakey type Ⅲ

Objectives To investigate the recurrence risk factors and the protective factors of aortic dissection (AD) DeBakey type Ⅲ. Methods 43 patients with AD DeBakey type Ⅲ who were in Guangdong General Hospital from May 2014 to September 2014, were enrolled as the case group, while 27 volunteers exclude AD as the control group. Blood chemistries and other information obtained immediately after admissions , χ2 test or T test was used for univariate analysis of independent samples. Multivariate Logistic regression analysis was used to screen patients with recurrence risk factors or protective factors. Results The prevalence of hypertension (93.02%vs. 18.52%, P = 0.000) and proportion of smokers (34.88% vs. 11.11%, P = 0.027) were significantly higher in case group than control group. Logistic regression analysis showed that hypertension (OR=5.148, 95%CI= [2.209~13.058], P=0.001) and albumin level (OR=0.709, 95%CI = [0.541~0.929], P=0.013) were significantly associated with recurrence of aortic dissection DeBakey type Ⅲ. Conclusion Hypertension is an independent risk factor for recurrence of aortic dissection DeBakey type Ⅲ, and albumin level is a protective factor.
4.Advances in the research of pharmacogenomics of cyclophosphamide.

Cyclophosphamide (CPA) is the most common alkylating antineoplastic agent, as well as a strong immunosuppressant that is frequently applied to autoimmune diseases and organ transplantation. It is metabolized by cytochrome P450 oxidases (CYPs) to its active metabolite which played a critical role in therapy. CPA has serious and even fatal side effects, and its efficacy and adverse reactions are significantly varied among individuals. In this review, the association of the genetic polymorphisms in the metabolic enzymes and transporters involved in the disposition of CPA with the efficacy and adverse effects of CPA were summarized, thereby providing fundamental reference for further pharmacogenomic study of CPA.

5.Preventive Effect on Nosocomial Infection with Herbal Medicine Atractylodes chinensis Disinfectant for Air Sterilization in Cardiothoracic Surgery

OBJECTIVE To observe the preventive effect on nosocomial infection by the herbal medicine Atractylodes chinensis disinfectant for air sterilization in cardiothoracic surgery.METHODS A.chinensis disinfectant and ultraviolet irradiation were used to disinfect the patients′ rooms of our cardiothoracic surgery department.The total number of germ and fungus in the indoor air before and after using the two methods were collected.The nosocomial infection rates of the same period were also studied.RESULTS A.chinensis disinfectant had strong effect on sterilization of indoor germs and fungi,and the sterilization rate was 92.7%.The comparison between A.chinensis disinfectant and ultraviolet irradiation showed significant difference in their disinfection effects,while no significant difference in their nosocomial infection rates.CONCLUSIONS The herbal medicine A.chinensis disinfectant can achieve good sterilization effect and prevent nosocomial infection.
6.Mini-puncture hematoma scavenging technique, traditional craniotomy operation, or medicine expectant treatment for patients with hypertensive intracerebral hemorrhage

Two hundred and five patients with hypertensive intracerehral hemorrhage (HICH) received mini-puncture hematoma scavenging technique (MPST; n=80), traditional craniotomy operation (TCO; n=78), or medicine expectant treatment (MET; n=47), respectively. Clinical data demostrated that consciousness level and Glasgow Coma Scale (GCS) was more obviously improved in the MPST group,while postoperative complication rate was relatively lower. The MPST group and TCO group saw no significant difference in over-all matality, although both were significantly lower than the MET group. Severe disability rate in the MPST group was significantly decreased. In comparison with the TCO or MET group, 1 to 3 class activity of daily life score, language function recovery and response rate in the MPST group significantly improved (all P<0.05). We suggest that because of lower severe disability rate, less postoperative complications, and improved quality of life, MPST should be a better treatment of choice for HICH patients.

Background and purpose:It was reported that chemokine receptor CXCR4 and its ligand stromal cell-derived factor 1(SDF-1)were involved in the proliferation,differentiation,and metastasis of tumor.This study was designed to observe the expression of chemokine receptor CXCR4 in hypopharyngeal squamous cell carcinoma tissue and study the relationship between the expression of chemokine receptor CXCR4 and different clinicopathlogical characteristics,and further to explore the clinical significance.Methods:For the detection of the expression of chemokine receptor CXCR4,43 primary hypopharyngeal squamous cell carcinoma tissues,27 normal hypopharyngeal tissues,34 lymph node metastastatic lesions and 9 normal lymph node lesions were detected by immunohistochemical method using rabbit anti-human CXCR4 polyclonal antibody.Results:The positive expression rates of CXCR4 in 43 hypopharyngeal carcinoma tissues and normal tissues were 95.3% and 22.2%,respectively(P
8.Comparison clinical efficacy of 3%hypertonic saline solution with 20%mannitol in treatment of intracranial hypertension in patients with aneurysmal subarachnoid hemorrhage

Objective:To compare the efficacy of 3%hypertonic saline solution with 20% mannitol in treatment of intracranial hypertension in patients with aneurysmal subarachnoid hemorrhage.Methods: An alternating treatment protocol was used to compare the efficacy of 160 mL 3% hypertonic saline solution ( HSS ) with 150 mL 20%mannitol for episodes of increased intracranial pressure ( ICP ) in patients with aneurysmal subarachnoid hemorrhage.The dependent variables were the extent and duration of reduction of increased ICP after each event.Results: Both 3% HSS and 20% mannitol rapidly decreased the ICP in patients with aneurysmal subarachnoid hemorrhage ( P <0.01 ) .No difference between two medications in the extent of duration of ICP and reduction of action ( P>0.05) .Conclusion:3%HSS should be considered as the first-line osmotic drug in treatment of intracranial hypertension in patients with aneurysmal subarachnoid hemorrhage.
9.The prevalence of metabolic syndrome in adolescents according to 3 different diagnostic criteria

Objective To evaluate the differences and consistency of different diagnostic criteria for metabolic syndrome ( MetS ) in adolescents. Methods This cross-sectional study enrolled 920 healthy middle school students (aged11-16years,53.2% males). All participants underwentant hropometric and biochemical examinations,andwere diagnosed as MetS separately using criterion of the Pediatric Academy of Chinese Medical Association ( abbreviated as Chinese criterion) , 2007 International Diabetes Federation criterion ( IDF criterion) , and Cook criterion based on the National Cholesterol Education Program-Adult Treatment Panel in 2003 ( Cook criterion) . Statistic analysis was used to compare the prevalence of MetS diagnosed and the individual MetS components detected by the different diagnostic criteria, and to evaluate the differences and consistency between the three criteria. Results The prevalence of MetS diagnosed with the Cook criterion (16. 2%) was significantly higher than that by the Chinese criterion(11. 0%) and the IDF criterion. The most frequently detected components were low high-density with the Chinese criterion and IDF criterion, hypertension with the Cook criterion. The least frequently detected component was hyperglycemia with each criterion. The consistency was found stronger between the Chinese and IDF criteria ( Kappa=0. 798-0. 829, P<0.01), but weak between the Cook and the other two criteria (Kappa=0. 465-0. 789,P<0. 01). The adolescents solely diagnosed as MetS patients by the Cook criterion but not the other two criteria had more central obesity, higher plasma glucose and lipid levels and HOMA insulin resistance index (all P<0. 05). Conclusion The prevalence of MetS and components diagnosed with the three criteria was evidently different. The Cook criterion may diagnose more adolescents with higher cardiovascular risks and insulin resistance.
10.The Effect of Combination Treatment of Fosinopril and Losartan on Microalbuminuria in Patients with Essential Hypertension

Objective To evaluate the the effect of microalbuminuria of combined treatment with fosinopril and losartan,or fosinopril,losartan monotherapy in patients with hypertension.Methods In this double-blind, intention to treat study,136 patients with hypertension were randomly assigned to receive fosinopril 10 mg/d(n= 50),losartan 50 mg/d(n=41),or a combination of fosinopril 5 mg and losartan 25 mg (n=45) qd for 4 weeks, followed by titrating to the maximum recommended doses for another 4 weeks.The primary endpoint was the difference of mean sitting blood pressure and microalbuminuria excretion at baseline and week 8.Results At week 8,the combination of fosinopril and losartan therapy lowered mean mieroalbuminuria from baseline by 26.1?10~(-8) mol/L,significantly more than either monotherapy approaches (fosinopril 20 mg,18.3?10~(-8)mol/L,P
