1.Effect of Matrine on the expression of X-linked inhibitor of apoptosis protein in human rhabdomyosarcoma RD cells

Objective To investigate the effect of Matrine on the expression of X-linked inhibitor of apoptosis protein (XIAP) in human rhabdomyosarcoma RD cells in vitro.Methods Cultured human rbabdomyosarcoma RD cells were divided into Matrine intervention groups (0.5 g/L,1.0 g/L and 1.5 g/L) and a control group.The proliferation-inhibition rates in RD cells treated with different concentrations of matrine were detected by methylthiazolyl blue colorimetric assay.Flow cytometry analysis was performed for the apoptosis rates of RD cells.Reverse transcription-polymerase chain reaction analysis was used to measure the XIAP mRNA expression.Results There was a significant difference in the proliferation-inhibition rates [0.5 g/L Matrine group:(15.84 ± 2.58)%,1.0 g/L Matrine group:(23.13 ±4.19)%,1.5 g/L Matrine group:(30.32 ±3.02)%,and the control group:(8.92 ± 1.23)%];apoptotic rates [0.5 g/L Matrine group:(12.33 ± 1.15)%,1.0 g/L Matrine group:(16.67 ± 0.99)%,1.5 g/L Matrine group:(25.33 ± 1.91)%,and the control group:(9.47 ± 0.96)%];XIAP mRNA expression(0.5 g/L Matrine group:0.633 ± 0.046,1.0 g/L Matrine group:0.441 ± 0.055,1.5 g/L Matrine group:0.326 ± 0.065,control group:0.794 ±0.029)in RD cells among 0.5 g/L,1.0 g/L,1.5 g/L Matrine groups and the control group (F =14.15,83.37,50.57,all P < 0.05).The proliferation-inhibition and apoptotic rates in RD cells were gradually increased with the increasing Matrine concentration.The expression of XIAP mRNA was significantly decreased in different Matrine groups compared with the control group,exhibiting a dose-dependent manner.Conclusions Matrine can inhibit the proliferation of RD cells and induce the apoptosis in a dose-dependent manner,which may be related to the down-regulated XIAP mRNA.

3.Protective effects of the specific cyclooxygenase-2 inhibitor on microglia activation-mediated neuronal damage

Objective To investigate the protective effect of NS398,a specific cyclooxygenase-2 inhibitor,on microglial activation-mediated neuronal damage.Methods The microglia of neonatal SD rat were isolated and cultured in the medium containing 1 μg/ml of LPS.The morphological changes of microglial were observed.The IL-1β and TNF-α levels were detected by Western blot in LPS group (group L) and control group (group C).The hippocampal neurons of neonatal SD rats were cultured in the control medium (group C),LPS-activated microglial conditioned medium (group L) and LPS-activated microglial conditioned medium with NS398 (group N),respectively.The survival rate of neurons were detected by MTT.The respiration of hippocampus neurons was determined by detecting the ratio of lactic acid/pyruvic acid.Results LPS-induced microglial activation was characterized by the morphological change and increased secretion of IL-1β and TNF-α.The survival rate of neurons cultured by microglia activated conditioned medium was 64.37%,while the group N was 80.25% (P<0.01).In group L,the ratio of lactic acid/pyruvic acid (27.34±8.53) was significantly higher than that of group N (20.32±6.05,P<0.01) and group C (14.95±4.72,P<0.01).Conclusion NS398,a specific cyclooxygenase-2 inhibitor,has a protective effect on rat hippocampus neuron damaged by activated microglia.
4.Clinical observation of locally advanced pancreatic cancer treated by compound matrine injections combined with three-dimensional conformal intensity modulated radiotherapy

Objective To study the recent clinical efficacy of compound matrine injections combined with three-dimensional conformal intensity modulated radiotherapy in the treatment of locally advanced pancreatic cancer.Methods 48 cases of untreated locally advanced pancreatic cancer were randomly divided into treatment group (compound matrine injections combined with three-dimensional conformal intensity modulated radiotherapy) and control group (three-dimensional conformal intensity modulated radiationtherapy),there were 24 cases in each group.Three-dimensional conformal intensity modulated radiation therapy used 95 % isodose coverage PTV.Using 6 MV X-ray irradiation,2 Gy/day,5 times/week,the total radiation dose was 50 Gy/25 times/5 weeks to complete.Treatment group:one week before treatment infusion of compound matrine injections 20 ml/time,l/d,21-day was one cycle.Control group:simple intensity modulated radiotherapy.Results After treatment,the clinical benefit response rate of the treatment group was 87.5 % (21/24),the control group was 70.83 % (17/24),the difference was statistically significant (P =0.045).Term efficacy rate of the treatment group was 79.17 % (19/24),the control group was 50.00 % (12/24),the difference was statistically significant (P =0.035).Adverse reactions were hematologic toxicity,leukopenia occurred in the treatment group was 16.67 % (4/24),but the control group was 45.83 % (11/24),the difference was statistically significant (P =0.029).Conclusion The effects of compound matrine injections combined with three-dimensional conformal intensity modulated radiotherapy for locally advanced pancreatic cancer are good,they can relieve pain,improve short-term efficacy,reduc side effects,and improve the quality of life.
5.Changes of enzyme histochemistry and ultrastructure in mitochondrial myopathy and encephalomyopathy

Objective To investigate the pathological features and clinical characteristics of primary mitochondrial myopathy and encephalomyopathy. Methods 11 cases of mitochondrial myopathy and encephalomyopathy were analyzed on clinical features, histochemical changes and ultrastructure observations of muscles. Results RRFs were found in all 11 cases by muscle staining of MGT under light microscope. 7 cases were only afflicated with muscle weakness, 4 cases also having impairment of central nervous system. The proportion of RRFs was 6.4% to 10.3%. We observed these cases under electronic microscope. In 9 cases,the ultrastructural alterations included subsarcolemmal accumulation of mitochondria,increases of the number,abnormal shape,disarrangement of cristae and paracrystaline inclusion bodies. In 2 cases the increase of mitochondrial number is only the alteration. Conclusions Typical RRF is valuable for diagnoses of mitochondrial myopathy and encephalomyopathy under light microscope. The accumulation of abnormal mitochondrials beneath sarcolemal,especially the paracrystaline inclusion bodies are significant in making a definite diagnosis of this disease under electronic microscope.
6.Killing effects of cytosine deaminase gene mediated by adenovirus vector on human pancreatic cancer cell lines in vitro

Objective To evaluate the killing effects of cytosine deaminase (CD) gene mediated by adeno virus vector on human pancreatic cancer cell lines in vitro. Methods After CD gene was cloned into pAdTrack CMV CD, pAdTrack CMV CD and pAdEasy 1 were recombinated in bacteria. The newly recombinated Ad CD containing green fluorescent protein (GFP) was propagated in 293 cells and purified by cesium chloride gradient centrifugation. Human pancreatic cancer cell lines Patu 8988 and SW 1990 were infected with this virus, then 5 FC was added, XTT assay was used to estimate relative numbers of viable cell. Results The positive clones were selected by using endonuclease to digest the combinatants and the concentration of viral liquids containing CD gene was 2?10 11 PFU/ml. It was found that significant cytotoxic activities were possessed by 5 FC for CD gene transduced pancreatic cell lines, but little effects on the nontransduced pancreatic carcinoma cells. Conclusions CD gene mediated by adenovirus has a high infectivity and is efficient for gene therapy of pancreatic carcinoma cell lines. These data demonstrate the therapeutic efficacy of an enzyme as a prodrug strategy in experimental pancreatic cancer.
7.The Application of MR 3D-FLASH Dynamic Enhancement in the Breast

Objective To investigate the application of MR 3D-FLASH dynamic enhanced scanning in the breast diseases. Methods 108 women (35~76 years old,mean,47 years) with breast diseases confirmed by pathology were examined by using Siemens 1.0T superconducting MRI and double breast circles.Plain MR imaging performed first,then contrast-enhanced MR scans were followed with 3D FLASH sequence,and the images were subtracted.Results Among 108 cases,62 cases were benign and 46 cases were breast cancer,the diagnosing accurate rate of MR imaging was 81.5% compared with pathology pre-operation.Conclusion 3D-FLASH enhanced MR scan is the most valuable sequence in diagnosing breast lesions.

The purpose of this study was to evaluate the killing effects of CD/5-FC suicide gene system mediated by adenovirus vector on human pancreatic carcinoma. Recombinant adenoviruses containing cytosine deaminase(CD) gene were constructed by the homologous recombination method in bacteria. The newly recombinated Ad-CD containing green fluorescent protein (GFP) was propagated in 293 cells and purified by cesium chloride gradient centrifugatioa Human pancreatic carcinoma cell line was infected with this virus, then 5-FC was added, and XTT assay was used to estimate relative numbers of viable cells. A heterotopic human pancreatic adenocarcinoma was successfully established in the flank of Balb/c nude mice. Adenoviral liquid containing CD gene was directly injected into xenografts following administration of 5-FC to observe the antitumor effects. Positive clones were selected by using endonuclease to digest the recombinants and the concentration of viral liquid containing CD gene was 2 1011 pfu/ml. It was found that significant cytotoxic activities were possessed by 5-FC for CD gene-transduced pancreatic carcinoma cells, but there was little effect on nontransduced ones. The anticancer effect was seen in vivo in xenografts of nude mice with in situ CD gene transduction. Our conclusian is that CD gene mediated by adenovirus has a high infectivity and is efficient for gene therapy of pancreatic carcinoma.
