1.Role of centrosome in tumorigenesis and oncotherapy

The centrosome plays a crucial role in the maintaining of cell conformation,mitosis and chromosome seg-regation.It responds to the DNA damage and keeps the genome stability via cross-talking with the intranuclear DNA damage repair system.The apoptosis of tumor cells can be induced through inhibition of the duplication of centro-some.So the inhibitors of centrosomal proteins will be a potential anti-tumor therapy.
2.Clinical effect of combined nickel-titanium instruments and ultrasonic technique and warm vertical technique used in one-visit endodontic treatment for infected root canals

Objective To evaluate the effect of one-visit endodontic treatment for infected root canals with combination of nickel-titanium instruments and ultrasonic technique and warm vertical technique.Methods According to treatment,90 teeth were randomly divided into experimental and control groups.Two groups were used nickel titarlium root canal preparation.Experimental group:51 teeth for ultrasonic root canal irrigation,and warm vertical technique was employed to fill the root canal.Control group:39 teeth for rinse syringes,and warm vertical technique was employed to fill the root canal.Washing fluid was 2% sodium hypochlorite.The root canal filling was observed after a week and one year of postoperative pain response.Results A week later,two groups of post-operative pain showed that there was significant difference in responses (x2 =15.525,P < 0.05).One year later,the two groups showed no significant difference in efficacy(x2 =0.085,P > 0.05).Conclusion The combination of nickel-titanium instruments and ultrasonic irrigation technique and warm vertical obturation,for the treatment of infected root canals was a method to complete the root canal filling,could reduce both the number of patients subsquent visits and the postoperative pain response,while it could improve the quality of root canal filling.
3.Monitoring results and epidemic analysis of gerbil plague in northern Ningxia in 2009

Objective To analyze gerbil plague in northern Ningxia and the monitoring results,to master the plague epidemic dynamics,and to provide the basis for developmenting countermeasures.Methods Monitoring data of gerbil plague focus in northern Ningxia were collect in 2009,counted main host density,rate of dye fleas,flea body index and bacteriology,serology detect and analyzed the epidemic situation.Results An average density of main host was 1.74/hm2,the average rate of infected fleas was 28.60%,and the average rat body flea index was 0.76.Monitoring found 4 plagues from rat plague epidemic,plague bacteria were found in 16 strains,of which gerbils inspection bacteria 10 strains,Meriones unguiculatus 2 strains,the same type cheopis 2 strains,and bald disease fleas 2 strains.Indirect hemagglutination (IHA) was used to test 529 copies of samples,2 copies were found positive,and hemagglutination-positive rate was 0.38%; eight copies were examined by reverse indirect hemagglutination(RIHA),7 material were found positive,and hemagglutination-positive rate was 87.50%.Conclusions In recent years,the disease is active among animals in gerbil plague focus Ningxia.The density of rodents is higher,and local plague epidemic is found in rats.Monitoring efforts should be strengthened and scope of monitoring should be expanded.We should pay close attention to the epidemic dynamics,control the prevalence and spread of animal disease,and prevent the occurrence of human plague.
4.Proteomic analysis of differentially expressed proteins involved in primary focus of human colorectal carcinomas and normal colonic mucosa

Objective To study the differentially expressed proteins and their biological behavior in colorectal carcinoma tissues and the normal colonic mucosa by proteomics and molecular biology techniques. Methods The technique of fluorescence two dimension differential gel electrophoresis (2-D DIGE) was used to analyze the expression of differential proteins in normal colorectal mucosa and primary cancer foci. Liquid chromatography with electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS) was used to identify the differential proteins. Transfection experiment of colorectal cancer cells was performed with the differential protein cDNA, and the changes in cytobiological behavior were observed. Results Significant differences of protein expression levels were found by two-dimension electrophoresis. Eight differential protein spots were analyzed and identified. Human carbonic anhydrase Ⅱ and protein disulfide isomerase were detected in normal colorectal mucosa, but not in primary cancer foci. Phosphoglycerate kinase 1, fumarate hydratase and aldolase A were expressed in primary cancer. After transfection with human carbonic anhydrase Ⅱ cDNA, the abilities of Lovo cells were obviously reduced in invasiveness, chemotaxy motor and drug resistance. Conclusions Differences on protein expression levels are found between normal colorectal mucosa and primary cancer foci by 2-DE DIGE. The pathogenesis of colorectal carcinoma is related to the reduced expressions of carbonic anhydrase II and protein disulfide isomerase and enhanced expression of aldolase A. The technique of differential proteomics is useful in reaching a indepth understanding of the pathogenesis mechanisms of human colorectal cancer.
5.Long term follow up of carotid cavernous fistula patients treated with carotid occlusion

ObjectiveTo explore efficacy,durability and possible impacts on life quality of carotid occlusion treatment to carotid cavernous fistula (CCF) patients.MethodsCCF patients since 2001 were retrospectively analyzed,the clinical features,2 weeks post procedure mRS score and ratio of carotid occlusion were recorded.Headache impact test (HIT-6) and Short form health survey(SF-36) were used to assess impact of sequelae in patients' daily life,by phone call,questionnaire and clinic recheck.Results Total 96 cases were studied composed of 81 direct CCF and 15 dural AVF.Thirty-two direct CCF cases underwent carotid occlusion during procedure and many ophthalmologic signs but visual impairment got recovery after 2 weeks,the mRS score less than 2 were revealed.The one year post operation HIT-6 score more than 50 was more likely found in carotid occlusion cases comparing with those preserved carotid artery while the 3 year SF-36 scores of carotid occlusion cases revealed inferior to those with patent artery,especially in body pain,general health and vitality subscales.ConclusionCarotid occlusion seems to be a feasible,effective and durable alternative for CCF treatment,but which could play a negative role on quality of patients' life in the long run.
6.Comparison of proliferative and adipogenetic potentials between three human mesenchymal stem cells

Objective To isolate and culture mesenchymal stem cells (MSCs)from human term umbilical cord and placenta,and to study the proliferative and adipogenic capacities of human fetus tissue MSCs (FT-MSCs ), umbilical cord MSCs (UC-MSCs)and placenta MSCs (PL-MSCs), and to provide an experimental basis for its clinical use.Methods The human term umbilical cord and placenta were obtained in sterile condition. The UC-MSCs and PL-MSCs were isolated by digestion using collagenaseⅠ,the FT-MSCs were provided by Zhongke Bio-engineering Co.,Ltd.The 3th or 4th passages of MSCs were selected in this study.The cell morphology was observed by inverted microscope.The proliferative ability was detected by living cell number counting method.The cell cycle and the positive expression rates of the surface markers of three kinds of MSCs were detected by flow cytometry (FCM).The 3th generation of UC-MSCs,PL-MSCs,and FT-MSCs were induced to differentiate into adipogenic cells.18 d after being induced,the differentiated cells were stained with Oil red O and the positive cells were counted. The adipogenic differentiation abilities of MSCs from the different tissues were assessed. Results The MSCs from different tissues were spindle-shaped adherent cells.The FCM results showed the surface marker of MSCs were positive for CD44,CD73,CD90,and CD105,and negative for CD14,CD34,and CD45. The FT-MSCs displayed higher proliferative ability than UC-MSCs and PL-MSCs (P<0.01).The proliferative index (PI)of FT-MSCs (36.66%±1.30%)was significantly higher than those of UC-MSCs (18.23%±1.10%) and PL-MSCs (10.03%±1.20%) (P<0.01).The lipid droplets were found after induction,and the results of Oil red O staining were positive in all kinds of MSCs. However the adipogenic capacity of UC-MSCs was significantly higher than those of PL-MSCs and FT-MSCs (P<0.01).Conclusion UC-MSCs,PL-MSCs and FT-MSCs have similar biological characteristics and cell surface markers. But FT-MSCs have the strongest proliferative ability and activity,UC-MSCs have the strongest adipogenic capacity invitro.
7.Effects of inhalation of different concentrations of sevoflurane combined anesthesia on somatosensory evoked potentials in adolescent patients undergoing scoliosis surgery

Objective To investigate the effects of inhalation of different concentrations of sevoflurane combined anesthesia on somatosensory evoked potentials (SEPs) in the adolescent patients undergoing scoliosis surgery.Methods Forty-five adolescent patients of both sexes,aged 12-17 yr,with body mass index of 15-23 kg/m2,undergoing scoliosis surgery under general anesthesia,were randomly divided into 3 groups ( n =15 each):low concentration sevoflurane group ( group S1 ),moderate concentration sevoflurane group ( group S2 ) and high concentration sevoflurane group (group S3 ).Anesthesia was induced with fentanyl 4 μg/kg,midazolam 0.03 mg/kg,propofol 2.0 mg/kg,and rocuronium 0.6 mg/kg.The patients were endotracheal intubated and mechanically ventilated.Anesthesia was maintained with propofol infusion at 8 mg· kg-1 · h-1 and remifentanil infusion at 0.25 mg· kg-1 ·min-1.After beginning of the surgery,1.8%,4.0% and 6.0% sevoflurane were inhaled for 10 min in groups S1-3 respectively,SEPs were measured by stimulating the posterior tibial nerve and significant SEP suppression was detected and recorded.The wave P40 latency,wave P40-N50 amplitude,and time for P40 waveform changes were recorded before sevoflurane inhalation (baseline) and at 10 min of sevoflurane inhalation,and the time for recovery of P40 waveform was recorded after sevoflurane inhalation was stopped.The prolonged percentage of P40 latency and decreased percentage of P40-N50 amplitude were calculated.Results The rates of significant SEP suppression were 80%,100% and 100% in groups S1,S2 and S3 respectively ( P > 0.05).Compared with group S1,the prolonged percentage of P40 latency and decreased percentage of P40-N50 amplitude were significantly increased,the time for P40 waveform changes was significantly shortened,and the time for recovery of P40 waveform was significantly prolonged in groups S2 and group S3 ( P < 0.05).Compared with group S2,the decreased percentage of P40-N50 amplitude was significantly increased,the time for P40 waveform changes was significantly shortened,and the time for recovery of P40 waveform was significantly prolonged in group S3 ( P < 0.05).Conclusion 1.8%,4.0% and 6.0% sevofiurane combined anesthesia can suppress SEPs in the adolescent patients undergoing scoliosis surgery,and they are not suitable for the surgeries requiring SEP monitoring.
8.Preliminary experience on early mechanical recanalization of middle cerebral artery for acute ischemic stroke and literature review

Objective To evaluate the feasibility,efficacy and complication of early middle cerebral artery(MCA) mechanical recanalization(MER) for treatment of acute ischemic stroke.Methods Seven cases undergone MER of MCA for the treatment of acute cerebral infarct were retrospectively reviewed and analyzed,including the etiology,mechanism,Qureshi grading scale,location and size of infarcts,NIHSS score of pre and post procedure,endovascular technique and complications.Referring to the literature,the indications of MCA recanalization were further identified.Results A total of 7 cases with mean age of 48 yrs were reviewed,which included 3 cases of atherosclerotic thrombosis and 4 embolic cases with pre NIHSS score ranging from 3 to 22.Mechanical recanalization succeeded in 6 cases,but 2 cases of cardiogenic embolism died of intrac ranial hemorrhage postoperatively.Favorable clinical outcomes were achieved in 4 cases whereas 1 deteriorated.Overall complications seemed to be consistent with literatures reviewed.Conclusions Early MER of MCA may benefit to a certain subset of acute ischemia stroke patients,however,embolic cases,elder patients and those with severe neurologic deficits are often accompanied by higher complications and unfavorable outcome.
9.Effects of different dose tranexamic acid in fibrinolysis during liver transplantation

Objective To investigate the effects of different dose of tranexamic acid in fibrinolysis during liver transplantation. Methods Sixty ASA Ⅱ~ Ⅳ liver transplant recipients, were randomly, double-blind assigned to one of 3 groups (n = 20): group control (group C), group tranexamic acid 1 (group T1) and group tranexamic acid 2 (group T2). The patients in group C received a loading dose of normal saline 10 mL, then continued infuse normal saline at 20 mL/h until neohepatic phase 120 min, while in other two groups, patients received a loading dose of tranexamic acid 1 g, totally 10 mL, followed by continuous infusion at 10 mg/(kg·h) in group T1 or 20 mg/(kg·h) in group T2 until neohepatic phase 120 min. Prothrombin time, fibrinogen, fibrin degradation product and D-dimers were measured before operation (T0), 120 min after the skin incision (T1), nonhepatic phase 30 min (T2), neohepatic phase 120 min (T3). Blood loss, fresh frozen plasma dosage, fibrinogen dosage and thromboembolic events were recorded. Results The plasma concentration of fibrin degradation product and plasma concentration of D-dimers were different in the 3 groups, group T2< group T1
10.Different Effects of Simvastatin on the Expression of Lectin-like Oxidized Low-Density Lipoprotein Receptor-1 (LOX-1) Induced by Oxidized Low-Density Lipoprotein (ox-LDL) or Glucose

Objective To investigate the effects of simvastatin on the expression of lectin-like oxidized low-density lipoprotein receptor-1 (LOX-1) induced by oxidized low-density lipoprotein (ox-LDL) or Glucose in U937 macrophages, and explore the role of NF-κB in modulating of LOX-1 expression. Methods U937 macrophages were treated with PMA to induce differentiation, which were co-cultured with 50 mg/L ox-LDL or/and 25 mmol/L glucose. Pyrrolidine dithiocarbamate (PDTC) and simvastatin (1 μmol/L or 10 μmol/L) were used to treat cells. The expression of LOX-1 protein and NF-κB ac- tivity were detected by enzyme-linked immunosorbent assay technology. The expression of LOX-1 mRNA was measured by RT-PCR. Results The expression of LOX-1 was up regulated by ox-LDL, glucose and combination of both. The inhibitor of NF-κB PDTC suppressed this up-regulation. Simvastatin suppressed the expression of LOX-1 induced by ox-LDL, and showed a significant effect in the higher concentration. There was no significant effect of simvastatin on the expression of LOX-1 induced by glucose. The variation of NF-κB activity was similar to that of LOX-1 expression. Conclusion Simvas- tatin suppressed the expression of LOX-1 induced by ox-LDL, while no significant effect on the expression of LOX-1 in- duced by glucose. The expression and regulation of LOX-1 were related with NF-κB pathway.
