Objective:To explore the mechanism of growth factor autocrine stimulation in human pituitary tumors. Method: Immunohistochemical analyses of EGF、TGF-αand EGFR were studied on paraffin-embeded sections of 30 pituitary tumors. Results: Expression of EGFR and its ligand EGF、TGF-αhas a variable stained cells intensity,density and type. EGFR and the ligands expressed in majority of pituitary tumors.Conclusion:The EGF autocrine stimulation exists in pituitary tumors,and tyrosine kinases inhibitors may be useful for pituitary tumors treatment.

Objective To investigate the effect of formoononetin on endostatin ( ES ) , vascular endothelial growth factor ( VEGF ) , matrix metalloproteinases 2(MMP-2), basic fibroblast growth factor (bFGF) in hydrothorax and serum, and tumor biomarkers of patients with elderly advanced lung cancer.Methods 67 cases elder with advanced lung cancer were selected and randomly divided into two groups.33 cases in control group were treated with conventional therapy of NP chemotherapy regimen, 34 cases in experiment group were combined with formononetin.The changes of ES, VEGF, MMP-2, bFGF, tumor marker levels in hydrothorax and serum and life quality evaluation were compared.ResuIts Compared with control group, the contents of ES, VEGF, MMP-2, bFGF in hydrothorax and serum of experiment group significantly decreased (P<0.05); the carcino embryonie antigen (CEA), neuron specific enolase (NSE), cytokeratin-19-fragments (CYFRA21-1), and carbohydrate antigen 125 (CA125) in serum of experiment group significantly decreased ( P<0.05 ); the life quality evaluation improved better of experiment group (χ2 =4.96, P<0.05 ). ConcIusion Formononetin has better clinical effect in treatment of elderly patients with advanced lung cancer patients.It could effectively improve the quality of life, reduce ES, VEGF, MMP-2, bFGF and other indicators in hydrothorax and serum, which has the vital significance to the clinical therapy.

Recombinant human epidermal growth factor (rhEGF, 20 ng/ml) and Genistein (740 μmol/ml) were added to the cells from 12 cases of human pituitary tumors and co-cultured in vitro. The 3H-thymidine uptake of tumor cells was measured by scintillation counter and tyrosine kinases activity was detected by using immunocytochemical method. The results showed that 3H-thymidine uptake and tyrosine kinases activity were increased in 10 out of 12 cases of human pituitary tumor by EGF and both declined by Genistein. It was suggested that EGF had a strong mitogenic effect on majority of pituitary tumors. Genistein could abolish the effects of EGF promoting the proliferation of the tumor cells, inhibit the growth of the tumor and might be useful in pituitary tumor treatment.

OBJECTIVE: To compare the difference in bond strength between titanium alloy with different surface roughness and dentin. METHODS: Thirty titanium alloy cylindrical specimens were fabricated and randomized to 6 groups. All groups of specimens were rubbed separately with 60, 80,100,150, 240, and 320 grit silicon carbide (Si-C) sand paper. The surface roughness of each group was measured. The occlusal surfaces of 30 human molars freshly extracted within 1 month were flattened to expose an adequate area of lower dentin. The dentin bonding surface was produced using 320, 400, and 600 grit Si-C sand paper, respectively. Titanium alloy and dentin specimens were bonded with Ketac~(TM) Cem Easymix ionomers. All bonded specimens were stored for 24 hours at 37 ℃. The microtensile bond strength was measured through the use of universal testing machine. The fractured surface of titanium alloy was observed under scanning electron microscopy.RESULTS: The titanium alloy group rubbed with 80 grit Si-C sand paper yielded the strongest bond strength, 6.65 MPa, compared with other groups (P < 0.05). The surface roughness of titanium alloy in this group was (2.69±0.32) μm. Scanning electron microscopy results revealed that all fractured surfaces appeared at the interface of dentin and ionomer cement. CONCLUSION: The surface roughness could influence the bond strength between titanium alloy and dentin. Titanium alloy with (2.69±0.32) μm surface roughness bonding to dentin showed the maximum bond strength.

OBJECTIVE: To survey the immediate bond strength of the two glass ionomer cements with different pressure times.METHODS: Totally 70 teeth were prepared to expose the dentin, and then divided into the Ketac~(TM)Cem Easymix GIC glass ionomer cement group (3M group), and the TOKUSO glass ionomer cement (TOKUSO group). Each group was divided into 7 subgroups according to pressure times. The immediate tensile strengths of all specimens were measured after pressed for 5,10,15, 20, 25, 30 and 35 minutes.RESULTS: The immediate bond strengths of the two glass ionomer cements with different pressure time showed obviously significant differences (P < 0.01), which exhibited the greatest immediate bond strengths at 25 minutes after pressure. The bond strength of 25 minutes subgroup of 3M was higher than that of TOKUSO group, but the difference had no significance.CONCLUSION: Both Ketac~(TM) Cem Easymix and the TOKUSO glass ionomer cement can harvest the maximum bond strength after 25 minutes pressure.

Objective To discuss the uhrasonography and hemodynamic characteristics of carotid atherosclerosis in elderly patients with coronary heart disease (CHD).Methods The 548 people were divided into CHD group (n = 373) and control group (n = 175) by the coronary angiography.And the patients in CHD group were sorted into myocardial infarction (MI) group (n= 160) and non-MI group (n= 213).All patients were above 60 years of age, and underwent carotid artery ultrasound examination.The data of carotid sonography and hemodynamics were classified and analyzed using SPSS 10.0 software.Results The carotid intima-media thickness (IMT), plaque thickness and score were much more in CHD group than in control group [(0.96±0.24) mm vs.(0.78±0.11) mm, (1.90±0.36) mm vs.(1.66±0.13)mm, (2.2±0.7) vs.(1.9±0.4), t= 14.81, 27.89 and 27.83, all P=0.00].A total of 161 (43.2%) patients had atherosclerotie plaques in CHD group, and there were 7 (4.0%) people with plaques in healthy control group(χ~2 =337.18, P =0.00).And there were 81.3% of patients with plaques in MI group, it was more than non-MI group, in which there were 12.9% of patients with plaques (χ~2 =60.88, P=0.00).The carotid peak systolic velocity (PSV) and resistance index (RI) were both higher in CHD group than in control group [(77.0±11.9) cm/s vs.(65.2±3.1) cm/s and 0.77±0.06 vs.0.67±0.08, (t= 12.26 and 50.43, P=0.00)].Conclusions Carotid ultrasonography could evaluate not only CHD, but also CHD severity in the elderly.

In this study,the loop-mediated isothermal amplification(LAMP)method was used to develop a rapid and simple detection system for porcine circovirus type 2(PCV2).According to the PCV2 sequences published in GenBank,multiple LAMP primers were designed targeting conserved sequences of PCV2.Using the DNA extracted from PCV2 isolates HUN-09 and SD-09 as the template,LAMP reactions in a PCV2 LAMP system was performed,the amplification products were detected by adding SYBR Green I and could be observed directly by the naked eye.The results showed highly-efficient and specific amplification in 30 min at 63℃ with a LAMP real-time turbidimeter.Furthermore,PCV2 DNA templates,with a detection limit of 5.5×10-5ng of nucleic acid,indicated that this assay was highly sensitive.The results obtained with the naked eye after SYBR Green I staining were consistent with those detected by the real-time turbidimeter,showing the potential simplicity of interpretation of the assay results.The LAMP assay appeared to have greater accuracy than PCR and virus isolation for the analysis of 18 clinical samples.In addition it offers higher specificity and sensitivity,shorter reaction times and simpler procedures than the currently available methods of PCV2 detection.It is therefore a promising tool for the effective and efficient detection of PCV2.

Objective To investigate the tissue distribution character of cinobufagin in in vivo in rats,in order to provide some references for rational drug use in the clinic.Methods Two kinds of solutions of cinobufagin were given to rats(0.5 and 1 mg/kg) by sublingual iv injection.The cinobufagin concentration in rat tissues corresponding to different time-point was determinated by HPLC.The biological samples of the different tissuses were performed by liquid-liquid extraction method.Results Cinobufagin distributed rapidly to tissue after sublingual iv injection.At 30 min cinobufagin in eight tissues except fat was detected.At 60 min the concentration in a majority of tissue achieved the peak value and liver′s was tiptop.When the concentration of cinobufagin increased by gemination,the time of reaching peak value had no change.Only the concentration of cinobufagin increased slightly corresponding to the time-point.Conclusion The distribution of cinobufagin in tissues of rats is extensive.Both the absorption and elimination of cinbufagin are quick and up to the top concentration in brain tissue.

Objective To compare the clinical value of three-dimensional dynamic contrast enhanced magnetic resonance angiography (3D DCE MRA) and digital subtraction angiography (DSA) in diagnosing inferior vena cava diseases in suspected case of Budd-Chiari syndrome (BCS).Methods Radiological findings of 91 suspected BCS cases obtained from 3D DCE MRA and DSA in the Affiliated Hospital of Xuzhou Medical University were retrospectively analyzed.DSA test was considered as golden standard,which assess the capacity of 3D DCE MRA in diagnosing inferior vena cava diseases,including sensitivity,specificity and accuracy.Kappa test was utilized to compare the coincidence ratio of 3D DCE MRA and DSA in diagnosing inferior vena cava diseases.Results Among 91 suspected BCS cases with 3D DCE MRA,a total of 17 cases without inferior vena cava diseases were misdiagnosed as inferior vena cava stenosis,two patients with inferior vena caval obstruction was misdiagnosed as falsely negative.Seventy-two patients with 3D DCE MRA were confirmed via DSA in diagnosing inferior vena cava diseases,sensitivity was up to 97.7％ (58/60),false positivity 54.8％ (17/31),specificity 45.2％ (14/31),respectively.Fair coincidence ratio of 3D DCE MRA and DSA in diagnosing inferior vena cava diseases (Kappa =0.474,P ＜ 0.05).Conclusions There could be clinical value of 3D DCE MRA for its high sensitivity and low specificity in diagnosing inferior vena cava diseases,and favorable coincidence ratio was discovered between 3D DCE MRA and DSA.Comprehensive consideration is needed for suspected cases of inferior vena cava stenosis detected by 3D DCE MRA,and further analysis may figure out potential causes of misdiagnosis and decline false positive events.

Objective To explore the application value of the real-time three-dimensional ultrasound Bi plane technology in evaluation of the foramen ovale blood in fetus.Methods The coronal plane of inferior vena cava entering the right atrium in 186 normal fetuses were obtained.The dimensions of foramen ovale (DFO) and inferior vena cava at the entrance of right atrium (DIVC) were measured in the coronal plane.The ratio of DFO and DIVC (DFO/DIVC) was calculated.The correlation be tween DFO/DIVC and gestational age was analyzed.Results There was no correlation between DFo/DIvc and gestational age (r=0.228,P＞0.05).There were 155 cases during second trimester (21 28 weeks),and the DFO was (4.46±0.57)mm,DIVC was (4.55±1.22)mm,DFO/DIVC was 1.03±0.22.There were 31 cases during late pregnancy (29-36 weeks),and the DFO was (5.20±0.43)mm,DIVC was (5.90±1.33)mm,DFO/DIVC was 0.92±0.17.The mean 95％ confidence interval of DFO/DIVC was (0.98,1.04),the 95％ normal reference range was (0.80,1.44).Conclusion The coronal plane of inferior vena cava entering the right atrium can be obtained with real-time three-dimensional ultrasound Bi-plane technology,the flow from the inferior vena cava enters right atrium which is divided by the upper atrial septum can be observed,which is important in evaluating the foramen ovale blood.
