1.Clinical application of INRatio coagulometer for fast measuring international normalized ratio

ObjectiveTo compare the correlation and agreement of international normalized ratio (INR) measured by INRatio coagulometer and standard laboratory venous blood measurement in order to judge the clinical application value of measuring INR by INRatio coagulometer.MethodsOne hundred who received warfarin were chosen randomly.Venous blood and finger capillary blood were taken simultaneously for measurement of INR by laboratory coagulation analyzer SYSMEX CA-7000 and INRatio coagulometer respectively.The correlation and agreement of INR results tested by the two methods were analyzed.Results When INR ≤5,there was a good correlation between INRatio coagulometer and laboratory coagulation analyzer SYSMEX CA-7000 (r =0.898,P =0.000).The mean bias of the two methods was 0.36 and INR measured by INRatio coagulometer were higher than those tested by laboratory coagulation analyzer SYSMEX CA-7000.By Bland-Altman,it showed a good agreement for 91.2% (62/68) of the difference INR located 95% the limited of agreement when the mean INR 1.60 - 3.50,and a bad agreement for only 50.0%(1/2) of the difference INR located 95% the limited of agreement when the mean INR > 3.50.ConclusionsIn the therapeutic range,INR measured by INRatio coagulometer shows a good correlation and agreement with INR measured by the established laboratory method.INRatio coagulometer can be used to monitor INR in the patients received oral anticoagulant therapy.

4.Effects of Traditional Chinese Medicine Treatment from Lung on mRNA Expression of iNOS in Lung Tissues of Rats with Ulcerative Colitis

This study was aimed to observe effects of traditional Chinese medicine (TCM) treatment from lung on mRNA expressions of iNOS in lung tissues of rats with ulcerative colitis (UC). A total of 52 rats were used to estab-lish the UC rat model by using rabbit intestinal mucosal tissue allergenic model and TNBS-ethanol model (with the model success rate of 78%). Eight rats, which were randomly selected from 40 successfully modeled rats and the normal group, were used as the model group and the normal group before intervention (time point of week zero). The rest 32 successfully modeled rats were randomly divided into the model group, the western medicine (salazosulfapyri-dine) group, treatment from lung (Huang-Qi Jie-Geng, HQJG decoction) group, and treatment from intestine (Huang-Qi Huang-Lian, HQHL decoction) group, with 8 rats in each group. Another 8 normal rats were used as the control group. The intervention was given for 4 weeks. And the mRNA expression of iNOS was detected in week ze-ro, and four weeks after the treatment using real time-PCR. The results showed that in the acute phase of week zero, the mRNA level of iNOS in lung tissues of model group was significantly increased compared with that in normal group (P < 0.05); after 4-week treatment, compared with the normal group, the mRNA level of iNOS in the model group was significantly increased (P< 0.05). Compared with the model group, the mRNA level of iNOS in the treat-ment from lung group was significantly decreased (P < 0.01). It was concluded that NO which was catalyzed and synthesized by iNOS played an important role in pulmonary diseases. It can cause airway inflammation, edema, lung injury, and etc. Treatment from lung can alleviate the lung injury by inhibiting abnormal activation of iNOS.
5.Progress in the mechanisms of keratoacanthoma regression

Keratoacanthoma ( KA) is an epithelial neoplasm occurring in sun -exposed skin of the elder-ly.Two striking features of KA are its clinical behavior with spontaneous regression after rapid growth and its nos -ological position on the border between benignity and malignancy .The regression of KA may be associated with the hair follicle cycle,apoptosis and immune function of organism .This article summarizes physiological mecha-nisms such as Wnt/retinoic acid signalling cross -talk,the function of p53,bcl-2,bcl-xL and bak in apopto-sis,and the mechanism of action of several immune -related cells,that drives KA regression .It can provide theo-retical basis for us to select the method as well as time of the treatment in KA .
6.Protective effects and its mechanism on neural cells after folic acid intervention in preeclampsia rat model

Objective To investigate protective effects and mechanism of folic acid on brain neural cells in preeclampsia rat model.Methods Adult pregnant Wistar rats were randonly divided into 4 groups (n = 10 in each group).Rats in model group were injected intraperitoneally with homocysteine (Hcy,200 mg · kg-1 · d-1) daily and were injected subcutaneously every other day with monosodium glutamate (MSG,1 g · kg-1 · 48 h-1) from the 10th day of pregnancy to establish the model of preeclampsia. Lowdose folic acid (low dose group 10 ng · kg-1· d-1) and high-dose folic acid (high dose group 20 mg · kg-1 · d-1 ) were given intragastric administration with folic acid tablets dissolved in saline daily at the same time of establishing model.Rats in control group were injected or intragastric administration with the same dose of saline as above up to the 20th day of pregnancy.Brain tissue was fixed on the 20th day of pregnancy, so was that plasma folic acid was measured with automatic electro-chemiluminescence.Rats' immunohistochemical staining.bcl-2 mRNA and protein expression changes were observed by using reverse transcription(RT) -PCR and western blot.Results ( 1 ) Plasma folate concentrations were ( 39.5 ± 3.4 )nmol/L in low dose group and (40.1 ±5.4) nmol/L in high dose group, which were all significantly higher than (26.9 ± 6.7 ) nmol/L in model group( P < 0.01 ).Plasma folate in low dose and high dose group did not show significant difference( P > 0.05 ); ( 2 ) Apoptosis cell were 48.2 ± 9.1 in low dose group and 44.7 ±8.3 in high dose group, which were significantly lower than 75.8 ± 10.1 in model group (P<0.01).However, apoptosis cell in low dose and high dose group did not show significant difference( P >0.05 ) ;(3 )significant difference( P > 0.05 ); (4) bcl-2 mRNA and protein expression were 0.98 ± 0.49 and 0.89 ±0.52 in low dose group and 0.95 ± 0.38 and 0.92 ± 0.47 in high dose group which was significantly higher than 0.62 ± 0.20 and 0.45 ± 0.37 in model group ( P < 0.01 ); bcl-2 expression in low dose and high dose group showed no significant difference ( P > 0.05 ).Conclusions Folic acid has a protective role on neural activation and promoting bcl-2 gene and protein expression.
7.Value of virtual touch tissues quantization in the stages of type 2 diabetic nephropathy

Objective To investigate the application value of virtual touch tissues quantization(VTQ)in the stages of type 2 diabetic nephropathy.Methods 120 cases of type 2 diabetes were divided into 3 groups according to urinary albumin excretion rate (UAER):diabetes without urinary albumin,diabetes with trace albuminuria and diabetes with mass albuminuria.The control group consisted 30 normal cases.VTQ was used to measure the renal shear wave velocity (SWV),then compare were made between the 4 groups.Besides,correlation analysis was made between SWV and UAER in diabetic nephropathy group.Results When compared SWV,there was no significant difference between diabetes without urinary albumin and control group (P =0.144),but there was significant difference between each 2 group of diabetes with trace albuminuria,diabetics with mass albuminuria group and control group (P < 0.05).There was also significant difference between each 2 group of diabetes (P < 0.05).The correlation coefficients for SWV and UAER in diabetes were 0.697(P <0.05).ROC analyses showed the area under curves for SWV diagnosis for diabetic nephropathy was 0.93.The best cutoff point,sensitivity and specificity were 2.43 m/s,85.7% and 84.5 %,respectively.Conclusions The renal SWV increases as the renal injury progresses,which also plays an important role in diabetic nephropathy stages.

9.Role of cAMP-PKA signal transduction pathway in the ischemic preconditioning-induced attenuation of ischemia-reperfusion injury In isolated rat hearts

Objective To investigate the role of cAMP-PKA signal transduction pathway in the ischemie pleeonditioning-induced attenuation of ischemia.1eperfusion(ITR)injury in isohted rat hearts.Methods Fifty healthy adult SD rats weighing 300-350 g were anesthetized with intraperitoneal(IP)pentobarbital 50 meg/kg.Their he.arts were excised and perfused in a Langendorff apparatus with 37℃ oxygenated(95%O2-5%CO2)modified K-H solution at a constant pressure of 70 cm H2O,and randomly divided into 5 groups(n=10 each):group Ⅰ I/R;group Ⅱ ischemic preconditioning(IPC);groupⅢH89(PKA inhibitor);group Ⅳ PDTC(NF-κB inhibitor)and group Ⅴ db-cAMP.The experiment started after 10 min stabilization.The isolated hearts were tinct perfuzed for 30 min.followed by 60 min ischemia and 30 min leperfusion in group I/R(Ⅰ).Group IPC(Ⅱ)w88 subjected to 3 episodes of 5 min ischemia at 5 min intervals before I/R.Group Ⅲ and Ⅳ received 5 min perfnsion withH89 10 μmol/L and PDTC 100 μmol/L 3 times at 5 min intervals respectively before I/R.Group Ⅴ was peffnsed with db-cAMP 200 μmol/L for 30 min before I/R.Left ventricular developed pressure(LVDP)and ± dp/dtu were measured at stabilization period and 10, 20, 30 rain of reperfusion. Coronary flow (CF) was measured at stabilization period and 30 min of reperfusion and activities of LDH and creafinc kinase (CK) in the coronary effluent were determined. The myocardial specimens were obtained at 30 rain of reperfusion for determination of NF-κB-DNA binding activity (by EMSA) and expression of TNF-κ mBNA (by RT-PCR ) and p-CBEB (Ser133) (by Western blot). Results Compared with 1/R group, NF-κB-DNA binding activity and TNF-α mRNA expression were significandy decreased, ± dp/dt and CF were significandy increased, CK and LHD activities in the coronary effluent were significantly decreased in group IPC and db-cAMP (group Ⅱ , Ⅴ ) and p-CREB (Ser133) expression was significantly increased in group IPC, PDTC and db-cAMP (group Ⅱ , Ⅳ,Ⅴ ). Compared with IPC group, NF-κB-DNA binding activity and TNF-α mBNA expression were significantly increased, ± dp/dtmax, LVDP and CF were significantly decreased, CK and LDH activities were significantly increased in group H89 and PDTC (group Ⅲ, Ⅳ ) and p-CREB (Ser133) expression was significantly decreased in group H89(group Ⅲ ). Conclusion lschemic preconditioning can attenuate I/R injury in isolated hearts by inhibition of NF-κB-DNA binding activity via cAMP-PKA signal transduction pathway which reduces gene transcription of inflammatory cytokine.
10.Experimental study on effect of low dose radiation on autologous CIK cell immunity functions of tumor patients

Objective To observe the effects of low dose irradiation (LDI) on autoiogous CIK cell proliferation, phenotype and killing activity in tumor patients, and to provide the evidence for clinical application of adoptive immunotherapy with CIK cells. Methods Peripheral blood mononuclear cells (PBMC) were separated from 10 patients with malignant tumor, and CIK cells were cultured with different cytokines. (1) After 10 d culture, C1K cells were irradiated with different doses as 30, 50, 80, 100 and 200 Gy of X-rays was also detected. The CIK cell proliferation and killing activity were measured with 3H-TdR incorporation assay and 3H-TdR release assay, respectively and the percentage variation of CD3 +CD56 + were measured with flow cytometry after 24 h. ( 1 ) Autologous CIK cells were irradiated with 80 mGy X-rays. At different culture time ( 12, 24, 48, 72 h) after irradiation, the killing activity was measured with 3H-TdR release assay. (3) The effect of 3d low dose irradiation of 80 mGy X-rays on thekilling activity of CIK cells was also detected. Results After the CIK cells were irradiated with different doses as 50, 80, 100, 200 mGy of X-rays, the CPM values were 20 048.6 ± 2332. 2 ( t = 2.2, P <0.05), 21 832.2 ±2975.9 (t=3.5, P<0.01), 21 000.3 ±2451.1 (t=3.3, P<0.01), 19908.1 ±2051.0 ( t = 2.2, P < 0.05 ), respectively and the proliferation of CIK cells were significantly higher than that of control group. The CD3 + CD56 + cell percentage of 50, 80, 100 mGy groups were ( 30.3 ±3.8)% (t=2.3, P<0.05), (32.3±3.4)% (t=4.2, P<0.01), (29.742.9)% (t = 2.4, P<0.05 ), respectively. The killing activity of CIK cells of 80, 100 mGy groups were 55.2 ± 5.0 ( t = 3.3, P < 0.01 ), 52.8 ± 4.1 ( t = 2.3, P < 0.05 ), respectively. The killing activity of CIK cells up-regulated significantly at 24 h, dropped to normal levels at 48 h and 72 h. After 80 mGy X-ray irradiation for 3 consecutive days, the killing activity of CIK cells at different time points were 55.2 ± 5.3 (t = 2.6, P <0.05),61.9 ± 4.4 (t = 4.7, P <0.01), 67.2 ±5.7 (t = 5.7, P <0.01) for 24, 48, 72 h,respectively. Conclusions LDI might have the hormesis effect on CIK cells.
