1.Effect of 2?-(3-hydroxypropoxy)-calcitriol on proliferation and differentiation of osteoblast-like cells

Objective To study the effect of 2?-(3-hydroxypropoxy)-Calcitriol (ED-71) on the cell growth and differentiation of human osteoblast-like cell line OS-732. Methods Osteoblast-like cells were cultured with various concentration of ED-71.The effects of ED-71 on proliferation and differentiation were detected by (MTT), alkaline phosphatase activity and level of osteocalcin (bone gla protein,BGP). Results The proliferation rate of osteoblast-like cells cultured for 4 days was significantly inhibited by 10~ -7mol/L concentration of ED-71. ED-71 in 10~ -8, 10~ -9mol/L concentration can significantly increase BGP level, but ED-71 in 10~ -7mol/L concentration may decrease BGP level.Alkaline phosphatase activity compared with control was not changed significantly. Conclusions ED-71 can inhibit cell growth in high concentration and improve cell differentiation in lower concentration.
2.The role of NgR - Rhoa - Rock signal pathway in retinal ganglion cell apoptosis of early diabetic rats

To study the function and mechanism of the NgR-Rhoa- Rock signal pathways which exists in the retinal ganglion cells apoptosis in diabetes mellitus (DM) rats.
● METHODS: Some healthy SD rats were operated by means of single intraperitoneal injection of 1%streptozotocin based on the standard of 50mg/ kg wight, after that the blood sugar value was greater than 16. 7mmol/ L as DM model, then randomly divided into 3 groups, each group was 10 rats. ln addition to take 10 healthy SD rats as control group. Four groups of rats were bilaterally eyeball intravitreal injection in turn with NgR-siRNA virus 10μ L (siRNA group), NgR-siRNA virus diluted 10μ L ( DM group), NgR - siRNA virus - negative -control solution 10μ L (siRNA blank group), NgR- siRNA virus diluted 10μ L ( normal control group ), and fed normally. During that time, some life indexes like blood glucose, body mass, etc. were measured and recorded. After 12wk, the expression of NgR and Rhoa, HE staining, and TUNNEL staining were detected by Western blot analysis.
● RESULTS: Western blot analysis: compared with normal control group, the expression of NgR and Rhoa in DM group and siRNA blank group increased significantly (P<0. 01), while siRNA group was no significant change (P > 0. 05); compared with DM group and siRNA blank group, the expression of those proteins significantly lowered in siRNA group. HE staining: compared with normal control group, some extent ganglion cells arranged disorder, irregular shape, spacing not consistent were all found in three groups of model rats;compared with DM group and siRNA blank group, there was some improvement in siRNA group of ganglion cells about the order and shape size. TUNEL staining:compared with normal control group, there were retinal ganglion cells apoptosis in all of three groups of model rats. Compared with DM group and siRNA blank group, the number of retinal ganglion cells apoptotic cells was less, and the shape of cells had improved significantly in siRNA group.
●CONCLUSlON: ln the DM phase, the expression of NgR and Rhoa were up - regulation, the condition of diabetic retinal ganglion cell apoptosis was improved after that the NgR-Rhoa-Rock signal pathways had been inhibited.
3.Effect of psychological first aid training on medical students' cognitive appraisal about sudden events

Objective To explore the influence of psychological first aid training on cognitive appraisal of sudden events in medical students.Methods A medical university undergraduate students were selected and divided into the training group(n=42) and the control group(n=53).The former was treated with 20 hours short-term targeted psychological first aid,and the latter was treated as blank control.The SAM and LMSQ were adopted to measure students' cognitive appraisal before and after the training.Results After the training,the training group gained significant differences (P< 0.05) in threat (11.95 ±3.17) vs (10.45±2.48)(t=2.426,P=0.020),challenge (12.64t2.84) vs (14.40±1.95)(t=-3.304,P=0.002),controllable-by-self (13.05±3.15) vs (14.93±2.00) (t=-3.240,P=0.002),controllable-by-others (12.40±3.10) vs (14.52±2.52) (t=-3.553,P=0.001),stressfulness (13.26±2.69) vs (12.00±1.96) (t=2.592,P=0.013) of SAM,and Physical threat ((23.88±7.59) vs (19.76±4.82),t=3.057,P=0.004) of LMSQ compared with before training.Other factors had no significant difference(P>0.05).Compared with two groups,the training group gained significant increase(P<0.05) on the score differences for SAM factors (challenge (t=-2.466,P=0.016),controllable-by-self (t=-2.557,P=0.012),controllable-by-others (t=-2.800,P=0.006)) than the control group,and significant reduction (P<0.05) in physical threat (t=-2.308,P=0.023) of LMSQ.Conclusion Short-term targeted psychological first aid training has certain effects on changing cognitive appraisal about sudden events for medical students.
4.Identification of related substances in nicergoline by HPLC-MS.

To study the related substances in nicergoline, electrospray positive ionization high resolution TOF/MS was used for the determination of the accurate mass and elemental composition of the related substances. Triple quadrupoles tandem MS/MS was employed for the determination of the fragmentations of the parent ions. 16 related substances were detected and identified to be eight synthetic by-products and eight degradation products, by using impurity references matching, product mass spectra fragmentations elucidation, and verified further according to synthetic processes and stress testing results. The results obtained are valuable for nicergoline manufacturing process control and quality assurance.

5.The effects of lactoferrin on the proliferation,migration and osteogenic differentiation of human periodontal ligament cells

Objective:To observe the effects of lactoferrin(LF)on the proliferation,migration and osteogenic differentiation of human periodontal ligament cells(HPDLCs)in vitro.Methods:HPDLCs were cultured and identified.The proliferation and migration of HP-DLCs cultured with 0,10 and 20 μg/ml lactoferrin respectively were tested by MTT assay,Transwell assay and scratch test.The os-teogenic differentiation of the cells was evaluated using alizarin red staining and real-time PCR.Results:Lactoferrin at 10 and 20μg/ml increased the proliferation(P <0.05),increased the quantities of mineralized nodules and the expression of alkaline phospha-tase(ALP),osteocalcin(OCN)and osteopontin(OPN)(P <0.05).Conclusion:Lactoferrin promotes the proliferation,migration and osteogenic differentiation of HPDLCs.
6.Effects of olanzapine on glucose and lipidmetabolism, liver function and prolactin in childhood onset schizophrenia

Objective To study the effects of olanzapine on glucolipid metabolism,liver function and prolactin level in childhood onset schizophrenia(COS) to provide reference for clinical medication.Methods Thirty-eight patients with COS aged 13-17 years old were treated with olanzapine for at least two weeks.The changes of body mass,alanine aminotransferase(ALT),aspartate aminotransferase (AST),total cholesterol (TC),triglyceride (TG),glucose (GLU) and prolactin (PRL) were detected and compared between before and after treatment.Results The body mass after medication in children patients was significantly increased,average increase by (3.50-t-1.90)kg (P<0.01).The levels of ALT,AST,TC,TG and PRL after treatment were higher than before treatment (P<0.05).However,there was no statistically significant difference in blood GLU level between before and after treatment(P=0.598).The body mass change before and after treatment was positively correlated with ALT and AST levels(r=0.366,0.377,P<0.05);whereas the PRL level before and after treatment was negatively correlated with the body mass change (r=-0.432,P<0.01).Conclusion Olanzapine can lead to the body weight gain and increase of ALT,AST,TC,TG and PRL levels in COS patients.
7.Influence of looming cognitive style and fear of negative evaluation on anxiety in college students

Objective To explore the effect of looming cognitive style and negative evaluation on anxiety in college students.Methods A total of 300 college students were assessed with looming maladaptive style questionnaire(LMSQ),Beck anxiety inventory(BAI) and fear of negative evaluation scale (FNE).Results (1)There was significant gender difference in fear of negative evaluation in college students (90.16±11.69 vs 97.63± 11.69,t=-2.770,P<0.05),and there was no significant gender difference in looming cognitive style and anxiety(50.49± 11.59 vs 53.67± 11.65,t=-2.770,P>0.05;29.82±7.89 vs 28.84±7.22,t=0.813,P>0.05).(2) LCS were positively correlated with fear of negative evaluation and anxiety (P< 0.01).(3) Looming cognitive style could forecast anxiety in college students (β=0.257).Looming cognitive style and fear of negative evaluation could forecast anxiety in college students together (β1 =0.137,β 2 =0.247).(4) The meditation effects of fear of negative evaluation in total effect were 0.468.Conclusion Looming cognitive style and fear of negative evaluation are important factors influencing the anxiety of college students,fear of negative evaluation plays an important mediation role between looming cognitive style and anxiety of college students.
8.The mediating effect of coping style on the relationship between looming cognitive style and anxiety and depression of university students

Objective To analyze the mediating effect of coping style on the relationship between Looming Cognitive Style (LCS) and anxiety and depression of university students.Methods 281 university students were investigated with looming maladaptive style questionnaire (LMSQ),simplified coping style questionnaire (SCSQ),Beck anxiety inventory (BAI) and Beck depression inventory (BDI).Results ① The differences between positive group and negative group in anxiety were significant in LCS ((62.67 ± 16.42) vs (52.56±11.23)) and physical threat of LCS((32.58±8.90) vs (26.14±6.40)) (t=2.97,P< 0.05;t=3.34,P<0.05);and the differences were not significant in social threat of LCS(t=1.86,P>0.05).The differences of the people in different degree of depression were significant in LCS and physical threat and social threat of LCS (F=11.635,P<0.05;F=10.357,P<0.05;F=4.774,P<0.05).(②)LCS were positively correlated with anxiety and depression(r=0.257,P<0.01;r=0.343,P<0.01).Negative coping style was positively correlated with anxiety and depression(r=0.286,P<0.01;r=0.299,P<0.01),and positive coping style was negatively correlated with anxiety and depression(r=-0.176,P<0.01;r=-0.248,P<0.01).③ Positive and negative coping style partly mediated the relationship between LCS and anxiety and depression,and the proportions of mediating effects in total effects were 9.5%-21.8%,the variation for mediation explained the variance of the dependent variable were 9.5%-23.0%.Conclusion Coping style mediates LCS and anxiety and depression,and coping style are needed to be considered during intervening anxiety and depression of university students.
9.Application of group feedback and formative assessment in the teaching of endotracheal intubation

Objective To investigate the effect of group feedback and formative assessment respectively on teaching effectiveness of endotracheal intubation (ETI).Methods After atheoretical lecturing and theoretical test,eighty three undergraduates were randomized into concurrent-feedback group and groupfeedback group.ETIsimulation training was given respectively by providing concurrent or group feedback in the two groups,and then undergraduates received a skill assessment.After atheoretical lecturing and theoretical test,other 91 undergraduates were randomized into final-assessment group and formative-assessment group.ETIsimulationtraining and skill assessment were given in final-assessment group.Before training,first skill assessment was performed in formative-assessment group,and then feedback was delivered based on the assessment.After training,second skill assessment was given again.Skill assessment was evaluated using the criterion-based task specific checklist combined with global rating scale.t-test was used for comparison between groups.Results There was no statistical difference (P=0.212) in theoretical scores between group-feedback group,and concurrent-feedback group.Skill assessment score of group-feedback group (39.4 ± 4.9) was higher than that in concurrent-feedback group (35.4 ± 4.7),with statistical difference (P=0.000).There was no statistical difference (P=0.395) in theoretical scores between formative-assessment group and final-assessment group.Score of formative-assessment group (39.3 ± 6.2) was higher than that in final-assessment group (32.2 ± 2.4),with statistical difference (P=0.000).Conclusion Terminal feedback or formative assessment could enhance teaching effectiveness of endotracheal intubation among medical students.
10.Clinical observation of Ahmed glaucoma valve implantation for refractory glaucoma by directly puncturing the sclerotic tunnel

To observe the clinic effects and complication of Ahmed glaucoma valve(AGV) implantation in refractory glaucoma by using the 23G syringe needle direct puncture the sclerotic tunnel.METHODS: Forty-four cases ( 44 eyes ) of refractory glaucoma underwent AGV implantation by useing the 23G syringe needle direct puncture the sclerotic tunnel. The intraocular pressure ( lOP ) , visual acuity, and complication of post - operation were contrasted with those of pre-operation.RESULTS:The success rate was 84. 1%, the mean preoperative lOP in research group was 52. 1±10. 1mmHg, and the last follow up mean lOP was 15. 6 ± 6. 9mmHg. Compared with the preoperative visual acuity, 11 eyes increased, 27 eyes had no changes and 6 eyes decreased. The main post-operative complications included shallow anterior chamber ( 4 eyes ) , choroidal detachment ( 3 eyes), drainage tube shift (1 eye), hyphema (6 eyes), drainage tube blockage ( 1 eye ) , expulsive choroidal hemorrhage (1 eye), and fiber wrap of drainage tray (5 eyes) .CONCLUSlON:AGV implantation by direct puncture the sclerotic tunnel is feasible and easy. lt avoids of making sclerotic petal and the xenogenic sclera transplanting, simplified the operation technique, prevent the leakage of around tube. The shallow anterior chamber rate is lower. lt is an effective procedure for refractory glaucoma.
