2. A new phenolic glycoside from rhizomes of Sinacalia tangutica

Objective: To study the chemical constituents from the rhizomes of Sinacalia tangutica. Methods: The compounds were separated and purified by D101 macroporous resin, silica gel, and ODS column chromatography. Their structures were elucidated by extensive spectroscopic methods including 1H-NMR, 13C-NMR, DEPT, HSQC, HMBC, and HR-ESI-MS. Results: One new phenolic glycoside was isolated from S. tangutica, and identified as 5-hydroxyeugenol-4-O-β-D-xylopyranosyl-(1→6)-O-β-D-glucopyranoside (1). Conclusion: Compound 1 is a new compound named singuticoside.
3. Two new monoterpene glucosides from roots of Aruncus sylvester

Objective To study the chemical constituents from the roots of Aruncus sylvester. Methods The compounds were separated and purified by D101 macroporous resin, silica gel and ODS column chromatography. Their structures were elucidated by extensive spectroscopic methods including 1D and 2D NMR, HR-ESI-MS and ECD spectrum. Results Two new monoterpene glucosides were isolated from A. sylvester, and identified as (2E,4S,5R)-4-hydroxy-3-[2-(β-D-glucopyranosyloxy)ethylidene]- 5-(2-methylprop-1-en-1-yl) dihydrofuran-2(3H)-one (1) and (2Z,4S,5R)-4-hydroxy-3-[2-(β-D-glucopyranosyloxy) ethylidene]-5- (2-methylprop-1-en-1-yl) dihydrofuran-2 (3H)-one (2). Conclusion Compounds 1 and 2 are new compounds, and named as sylvesteroside A and sylvesteroside B, respectively.
4.Effect of NR2E1 on the division and proliferation of neuroblastoma cells.

OBJECTIVE:
To explore the effects of nuclear protein-like transcription factor nuclear receptor subfamily 2 group E member 1 (NR2E1) on the growth, division, and proliferation of neuroblastoma cell line IMR32.
METHODS:
A NR2E1 shiRNA plasmid vector was constructed and transfected into neuroblastoma cell line IMR32 using lipofedamine™2000. Subsequent cell growth was measured by cell counting and the protein expression of somatic nuclear division was examined by immunofluorescent staining.
RESULTS:
At 48 h after the neuroblastoma cells IMR32 were transfected with NR2E1-shiRNA vector, the related nuclear division protein and the proliferation of the transfected cells IMR32 were remarkably depressed.
CONCLUSION
Cells division and proliferation of neuroblastoma cell line IMR32 is inhibited through transfection with the NR2E1-shiRNA plasmid vector.

5.Association of serotonin and norepinephrine transporter gene polymorphisms with the susceptibility to depression.

OBJECTIVETo determine whether the serotonin transporter (5-HTT) and norepinephrine transporter (NET) gene polymorphisms were associated with the susceptibility to depression.
METHODSFive hundred and seventy-nine patients with depression, evaluated using a 17-item Hamilton Depression Rating Scale for Depression (HAMD), and 437 healthy controls, all of Chinese Han origin, were genotyped by polymerase chain reaction.
RESULTSBoth genotype distributions (P=0.033) and allele frequencies (P=0.023, OR = 1.250, 95% CI = 1.031-1.517) of NET-T182C were significantly different between patients and controls, where the T allele was associated with the onset of depression. Both NET-T182C T- and 5- HTTLPR L-carriers had higher baseline HAMD scores (P=0.032 and 0.023, respectively). There was an interaction between NET-T182C and 5-HTTLPR, where the combined genotype distributions were associated with both onset of depression (P=0.006) and the baseline HAMD scores (P=0.007).
CONCLUSIONThis study suggested a positive relationship between the NET-T182C polymorphism and the susceptibility to depression, and a positive relationship between NET-T182C/5-HTTLPR polymorphisms and the severity of depression.

6.Research progress on the role of pyroptosis in the occurrence and development of peritoneal fibrosis

Peritoneal dialysis is a recognized renal replacement therapy. Long term peritoneal dialysis will lead to changes in the morphology and function of the peritoneum, that is, peritoneal fibrosis, which is a known cause of the loss of peritoneal ultrafiltration capacity. Pyroptosis is a special type of soluble programmed cell death, characterized by cell swelling, rupture, secretion of cell contents and significant proinflammatory effect. The pyroptosis can be divided into typical and atypical pathways, and the inflammatory body of NOD like receptor heat protein domain related protein 3 (NLRP3) is the most important initiator. Current evidence shows that high glucose peritoneal dialysis fluid can induce peritoneal Mesothelium to scorch, and the inflammation and cell damage caused by it can aggravate the progress of peritoneal fibrosis. Different signal pathways have been proved to regulate the occurrence of pyroptosis. The latest research has proved that some potential targeted methods to inhibit pyroptosis can effectively inhibit the inflammation of peritoneal mesothelium and alleviate peritoneal fibrosis. This article mainly discusses the molecular mechanism of pyroptosis and the relationship between pyroptosis and peritoneal fibrosis.
7.Analysis of the effectiveness and safety of early radiotherapy intervention in oligometastatic non-small cell lung cancer

Objective:To investigate the prognostic factors of oligometastatic (OM) non-small cell lung cancer (NSCLC) patients and the safety and effectiveness of early radiotherapy intervention.Methods:A retrospective analysis was conducted, including 159 OM NSCLC cases (metastatic sites≤5, metastasis organs≤3) admitted to Department of Radiation Oncology in First Affiliated Hospital of Soochow University from January 2015 to December 2018. Among 159 cases, there were 107 males and 52 females, with the median age of 63 years. 137 cases were administrated via early radiotherapy intervention, and 22 cases via delayed radiotherapy intervention. The receiver operating characteristic curve (ROC) was used to determine the progression-free survival time (PFS)/overall survival time (OS) to ascertain the best cut-off value for local control and prognosis. Survival analysis was calculated by Kaplan-Meier curves, and Log rank test was used for comparison of these curves. Cox proportional hazards regression model was used for multivariate survival analysis.Results:The median follow-up time of 159 cases was 28.2 months. During the follow-up period, there were 16 cases with complete remission (10.1%), 53 cases with partial remission (33.3%), 27 cases with stable disease (17.0%), and 63 cases with progressed disease(39.6%). The local control rates at 3, 6 and 12 months were 83.9%, 59.7% and 41.0%, respectively. The median progression-free survival (PFS) of 159 patients was 8.0 months, the median survival time (OS) was 35.0 months, and 1, 2, and 3-year survival rates were 77.3%, 63.0% and 45.1%, respectively. Adverse reactions related to radiotherapy were relatively mild, mostly grade 1 and 2. PFS/OS= 0.3 is the best cut-off value for determining the patient′s local control and prognosis. The result of univariate analysis showed that gender, number of OM organs, T staging, radiotherapy intervention mode, tumor target volume absorbed dose (DT-GTVnx), PFS/OS were significantly related to median PFS ( χ2=4.175, 16.508, 4.408, 10.300, 6.842, 38.175, P<0.05); gender, pathological type, number of OM organs, initial diagnosis stage, T stage, N stage, lobectomy, radiotherapy intervention mode, tumor target volume (V-GTVnx), tumor load, local control status were significantly related to median OS ( χ2=6.672, 8.330, 21.299, 5.398, 6.874, 6.893, 5.611, 115.206, 4.017, 5.110, 21.299, P< 0.05). The result of multivariate analysis showed that delayed radiotherapy intervention ( HR=3.728, 95% CI 2.099-6.622, P<0.001) was an independent risk factor for PFS in patients with OM NSCLC, and PFS/OS>0.3 ( HR=0.123, 95% CI 0.062-0.246, P<0.001) was an independent protective factor for PFS in patients with OM NSCLC; male ( HR=1.665, 95% CI 1.024-3.043, P=0.033), high tumor burden ( HR=2.113, 95% CI 1.088-4.107, P=0.027), delayed radiotherapy interventions ( HR=15.076, 95% CI 7.925-28.680, P<0.001) were independent risk factors for OS in patients with OM NSCLC. Conclusions:OS of patients with OM NSCLC is significantly prolonged in female, low tumor burden and early radiotherapy intervention. Early radiotherapy intervention significantly improved the prognosis, and radiotherapy-related adverse reactions could be tolerated. These might suggest that local radiotherapy is safe and effective in the treatment of OM NSCLC patients.
8.Effects of a recombinant adenovirus expressing human hypoxia-inducible factor la double-mutant on the in vitro differentiation of bone marrow mesenchymal stem cells to cardiomyocytes

Objective To observe the effects of mutant hypoxia-inducible factor-1α ( HIF-1α) adenovirus ( Adeno-HIF-1α-Ala402-Ala564 ) on cardiomyocytes ( CMCs ) differentiation from the mesenchymal stem cells ( MSCs) co-cultured with CMCs. Methods Following groups were studied: HIF-1α group ( MSCs + CMCs + Ad-HIF-1α), LacZ group ( MSCs + CMCs + Ad-LacZ) , Sham group (MSCs + CMCs + PBS) and MSC + HIF-la Group ( MSCs + Ad-HIF-1α). MSCs were co-cultured with myocardial cells in proportion of MSCs: CMCs 1:2, after 24 hours, cells were infect with virus (MOI = 100) or treated with PBS, cardiac troponin (cTnT) expression in MSCs was detected 7 days post infection by immunochemical analysis, mRNA expression of HIF-la, TGF-B,, Smad4, NKx2.5, GATA-4 was also detected by RT-PCR. Results HIF-la increased MSCs differentiation to myocardial cells (differentiation rate 32. 68%±6. 52% vs. 8. 28% ± 0.09% in the LacZ group and 10. 25 % ±2. 20% in the Sham group and 0.32% ±0.05% in the MSC group (all P< 0.05 vs. HIF-la group). mRNA expression of HIF, TGF-β1, Smad4, NKx2. 5 and GATA-4 was also significantly upregulated in HIF-1α group all P <0. 05 vs. Sham group). Conclusion HIF-1α promoted MSCs, co-cultured with myocardial cells, differentiating to cardiomyocytes via upregulating TGF-β1/Smad4 signaling pathway.
9.Monitoring and analysis of arsenic pollution in Yangzonghai lake and the influence of arsenic to the surrounding drinking water and local food in Chengjiang of Yunnan province in 2008 and 2009

Objective To find out the arsenic pollution in Yangzonghai lake and its influence to the surrounding drinking water source and the local food and to provide a scientific basis for arsenic pollution control,drinking water and food safety. Methods Arsenic monitoring were carried out in 10 points of Yangzonghai lake,45 water sources within 5 km of Yangzonghai lake, and locally produced food. Results From September 2008 to December 2009, the water arsenic concentrations of Yangzonghai lake ranged 0.06 - 0.18 mg/L, the average arsenic concentrations were 0.12 mg/L and 0.10 mg/L for 2008 and 2009, respectively, the value of 2009 was higher, and the difference was statistically significant(t = 3.284, P < 0.05). There were no significant difference in the average of arsenic concentration of Yangzonghai lake between the dry and rainy season in 2009(t = 0.905, P >0.05). The arsenic concentrations from the No. 3 spring water ranged from 0.12 - 30.36 mg/L. After a peak value early in October 2008, the No. 3 spring water showed a downward trend month by month. In 2008 the average arsenic concentration was 23.92 mg/L, 2009 was 2.41 mg/L, down 89.92% over the previous year, the difference was statistically significant(t = 9.582, P < 0.05 ). In January and February 2009, the average arsenic concentration (11.96 mg/L) was significantly higher than those of March-December(0.50 mg/L, t = 57.759, P< 0.05). In Yangzong town, the arsenic concentration in the drinking water and river met the national drinking water health standards. Among 78 samples monitored of the locally produced food in Yangzong town, 77 passed, with a pass rate of 98.72%. Conclusions Arsenic contamination has been effectively controlled. The drinking water and local food production has not been contaminated by arsenic except aquatic products in Yangzonghai Lake. It is recommended to take effective measures to prevent water contamination.
