Aim The nasal absorption of furosemide and promotion action of l_menthol was examined. Methods In situ nasal recirculation methods was used to study the does_effect relationship of nasal absorption of furosemide and the effect of l_menthol on nasal absorption of furosemide in rats. Results When the concentration of furosemide in circulation liquid was respectively(4.66?0.33) ?(9.76?0.45) and (18.83?1.89) ?g?ml-1,the absorption rate constant k(min-1) of nasal in situ was correspondingly 0.0067?0.0011,0.0134?0.0024 and 0.0163?0.0037;and urine liquid appearing time was (73.33?1.53)?(52.67?7.02) and (47.00?4.58) min.But the larger the concentration of l_menthol, the little the absorption rate constant k. Conclusion Furosemide can be absorbed through the nasal mucous, and l_menthol can promote its absorption.

Objective To investigate the effects of hydroxyethyl starch 130/0.4 (HES) used as priming fluid for cardiopulmonary bypass (CPB) on the plasma colloid osmotic pressure (COP) and lactic acid (LAC) concentration in infants undergoing cardiac surgery.Methods Forty infants of either sex with age ≤6 yr undergoing cardiac surgery with CPB were randomly divided into 2 groups (n =20 each): HES group and control group. The left radial artery and the right internal jugular vein were cannulated for blood pressure (BP) and the central venous pressure (CVP) monitoring. Arterial blood gases, blood LAC concentration, hemoglobin (Hb), hematocrit (Hct), mean arterial pressure (MAP) and nasopharyngeal temperature were measured and recorded immediately before and 5 min after aortic cross-clamping, at the end of CPB and operation. Plasma COP was measured before induction of anesthesia (T1), at 5 and 30 min of CPB (T2 and T3, respectively), before routine ultra-filtration (T4), at the end of CPB (T5) and2 h in ICU (T6).Results The plasma LAC concentration was significantly lower and the COP significantly higher in HES group than in control group (P＜0.05 or 0.01).The plasma LAC concentration increased after aortic cross-clamping, reached the peak at the end of CPB and then declined at the end of operation, but was still higher than that before aortic cross-clamping in both groups. Plasma COP was significantly decreased during CPB as compared with the baseline at T1, but increased at T6 in both groups.Conclusion Using HES 130/0.4 as pdming fluid for CPB can effectively improve plasma COP and reduce blood LAC level in infants undergoing cardiac surgery with CPB.

Objective To develop a stable and sensitive microemulsion liquid chromatography method by using oil-in-water microemulsion mobile phase;To simultaneously determine five major active lignans, namely, schizandrin, schisandrol B, schisantherin A, deoxyschizandrin, and schisandrin B, in Schisandrae Sphenantherae Fructus and Schisandrae Chinensis Fructus. Methods The quantitative determination was conducted by using Kromasil C18 column (250 mm × 4.6 mm, 5μm). The influence of the composition of the microemulsion system on separation effect and retention time was studied. The method was optimized to sodium dodecyl sulfonate 3.3%(SDS, W/V), n-octanol 1.2% (V/V) and n-butanol 6.6% (V/V) in 20 mmol/L phosphate buffer (adjusted to pH 7.0 with orthophosphoric acid and sodium hydroxide). The flow rate was set at 0.7 mL/min. The compound peaks were detected by UV at 254 nm. The column temperature was 25 ℃. Results Under the optimized conditions, baseline separation of 5 lignans was achieved in 20 min, and the separation was satisfying. Schizandrin, schisandrol B, schisantherin A, deoxyschizandrin, and schisandrin B were in linear relation within the ranges of 2.0-155.0, 8.3-166.7, 8.8-175.8, 8.3-166.7, 8.9-177.1μg/mL (r>0.999). The limit of detection ranged from 0.06μg/mL to 0.18μg/mL, and the limit of quantitation ranged from 0.17μg/mL to 0.55μg/mL. The average recovery values were between 97.22%and 100.8%. Conclusion This method is simple, accurate and reliable, and can be successfully applied to simultaneous determination of five major active lignans in 13 batches of Schisandrae Sphenantherae Fructus and Schisandrae Chinensis Fructus from different producing areas in China.

Objective To evaluate the effect of polysaccharide nucleic acid fraction of bacillus Calmette-Guerin (BCG-PSN) on peripheral blood CD4+CD25+Foxp3+ regulatory T (Treg) cells in patients with condyloma acuminatum (CA).Methods Forty-two patients with first onset of CA were randomly assigned to receive either injection of BCG-PSN (0.35 mg every other day for 3 months) after fulguration (combination group,26 patients),or fulguration only (fulguration group,16 patients).Venous blood samples were obtained from all the patients at the initial visit and three months after the beginning of treatment,as well as from 30 healthy checkup examinees.The percentage of peripheral Treg cells in CD4+ T lymphocytes was determined by flow cytometry.The recurrence of CA was evaluated during the three months after the beginning of treatment.Results The percentage of peripheral Treg cells in CD4+ T lymphocytes was significantly higher in patients with CA than in the controls (8.31％ ± 1.24％ vs.5.15％ ± 0.72％,P ＜ 0.01),and in patients with clinical recurrence of CA than in those without (9.34％ ± 0.72％ vs.7.45％ ± 0.85％,P ＜ 0.01).The recurrence rate was significantly lower in the combination group than in the fulguration group (30.77％ vs.68.75％,P ＜ 0.05).After three months of treatment,the combination group showed lower percentage of Treg cells in CD4+ T cells compared with the fulguration group (5.87％ ± 1.05％ vs.6.60％ ± 0.75％,P ＜ 0.05).Conclusions The percentage of Treg cells has a close relationship with the progression of CA,i.e.,the higher the percentage,the more frequent the relapse.BCG-PSN may enhance the antiviral immune response in patients with CA and improve their prognosis by reducing the number of Treg cells.

OBJECTIVE:To establish a method for the determination of residuals of petroleum ether,ethanol,xylene and ace-tic acid in ecabet sodium crude drug. METHODS:Capillary GC was performed on the column of PGE-20M capillary column at the flow rate of 1.7 ml/min,detector was hydrogen flame ionization detector,carrier gas was nitrogen with high purity,column temper-ature was 45 ℃,maintaining 4 min,it increased to 80 ℃ with rate of 10 ℃/min,then increased to 135 ℃ with rate of 30 ℃/min,maintaining 3 min,the injection mode was direct injection,inlet temperature was 250 ℃,and the volume injection was 1.0 μl. RESULTS:The mass concentration was 0.050-1.952 g/L for petroleum ether,0.050-1.941 g/L for ethanol,0.024-0.948 g/L for xy-lene and 0.050-1.947g/L for acetic acid(r=0.999 1-0.999 7);RSDs of precision,stability and reproducibility tests were no more than 1.7%;recoveries were 99.3%-101.0%(RSD=0.7%,n=9),102.3%-103.7%(RSD=0.4%,n=9),101.2%-102.1%(RSD=0.3%,n=9) and 100.3%-102.2%(RSD=0.6%,n=9),respectively. CONCLUSIONS:The method is simple and accurate,and can be used for the control of residual of organic solvents in ecabet sodium crude drug.
