1.Change and Significance of Soluble Intercellular Adhesion Molecule-1 in Cerebro spinal Fluid of Bacterial Meningitis Induced by Escherichia Coli in Rabbits

Objective To investigate the changes and significance of concentration of soluble intercellular adhesion molecule-1(sICAM-1) in cerebrospinal fluid (CSF) of bacterial meningitis(BM) in rabbits. Methods A total of 36 rabbits were randomly divided into m eningitis group, meningitis cefotaxime-treated group and control group. BM indu ced by escherichia coli(Ec) via cerebellomedullary cistern inoculated. Normal sa line was injected in control group. CSF was sampled in different time. The conce ntration of CSF sICAM-1 was detected by ELISA.Results 1.There was a low concentration of sICAM-1 in CSF in 85 percent of normal rabbi ts.2.In meningitis group, there was a sharp rise in the concentration of CSF sIC AM-1 at 6 hours after Ec was inoculated, reached a peak level at 12 hours, and t here was higher concentration of CSF sICAM-1 between 6 and 24 hour than that at 0 hour.3.In meningitis cefotaxime-treated group, the concentration of CSF sICAM -1 at 6 hour and 12 hours was similar to meningitis group, the time that get pe ak level was at 24 hours that at 12 hours after making use of antimicrobial agen t.The peak level was higher than meningitis group. The concentration of CSF sICA M-1 decreased markedly at 48 hours that made use of antimicrobial agent 36 hour s later,but the concentration was still higher than the peak level of meningitis group.Afterwards, with the time of making use of antimicrobial agent lengthened ,the concentration of CSF sICAM-1 decreased gradually.4.Experimental results in dicated by correlating analysis to these data that there was positive correlatio n in the concentration of CSF sICAM-1 with the brain water content.Conclusions sICAM-1 participates in the pathological process of BM, and contributes to the damage of blood brain barrier and the formation of brain edema.There is importan t significance that drugs which can resist the discharge of sICAM-1 will be impl ied. 3.Antitumor effect of Tanshinone Ⅱ A submicron emulsion and effect on its reversal of multidrug resistance in SMMC-7721/VCR tumor

Objective:To explore the anti-tumor effect of Tanshinone Ⅱ A microemulsion and to research its reversal effect of multidrug resistance on SMMC-7721/VCR tumor.Methods The human hepatocellular carcinoma cell line SMMC-7721 and human hepatocellar carcinoma drug-resisitant cell line SMMC-7721/VCR were cultured in vitro.Each cell was divided three groups:experimental group (cell suspension + 3 kinds of anti-tumor drugs),negative control group (cell suspension + culture) and blank control group (culture).Tanshinone Ⅱ A microemulsion and Tanshinone Ⅱ A in five concentrations (0.5,1.0,2.0,4.0,8.0 μg · mL-1) intervention SMMC-7721 cells.The inhibition effect of Tanshinone Ⅱ A microemulsion of 0.5,1,2,4,8 μg · mL-1 in 24 h and 48 h on SMMC-7721 cell proliferation was tested by MTT assay.The IC50 of VCR,CDDP,5-fluorouracil(5-Fu) for the cell of SMMC-7721,SMMC-7721/VCR were determined by MTT assay.The IC50 of VCR,CDDP,5-Fu for the SMMC-7721/VCR after intervention with Tanshinone Ⅱ A microemulsion were determined also by MTT assay.The sensitivity change and drug reversal effect of Tanshinone Ⅱ A microemulsion before and after intervention on the SMMC-7721/VCR cell line was investigated by MTT assay.Results The inhibition rate after intervention with Tanshinone Ⅱ A microemulsion on SMMC-7721 cells for 48 h were 47.93%,55.92%,66.34%,95.61%.Accordingly,the inhibition rate of Tanshinone Ⅱ A were 38.83%,49.31%,58.24%,82.46%.The half inhibitory concentration were (0.67 ±0.32),(1.91 ±0.53) μg · mL-1,respectively.The difference had statistical significance (all P < 0.01).With the increase of concentration,the inhibition rate increased of Tanshinone Ⅱ A microemulsion on SMMC-7721 cell with dose dependence and time dependence.When the concentration of Tanshinone ⅡA and its microemulsion for 0.5 ug · mL-1,on SMMC-7721 cells and SMMC-7721/VCR cell growth inhibition rate were 9.53%,7.74%,both less than 10% and no obvious toxic effect,so concentration of 0.5 μg · mL-1 was choosed as liver cancer drug resistant safety experiment dose.When 0.5 ug · mL-1 Tanshinone Ⅱ A microemulsion was intervention on SMMC-7721/VCR cell,VCR to SMMC-7721/VCR half inhibitory concentration was from (12.27 ±0.84) ug · mL-1 to (3.25 ±0.14) ug · mL-1.Reverse ratio is 3.78.Half inhibitory concentration of CDDP on SMMC-7721/VCR cell is from (20.42 ± 0.18) μg · mL-1 to (6.98 ±0.99) μg · mL-1,reverse ratio is 2.93.Half inhibitory concentration of 5-FU on SMMC-7721/ VCR cell is from (35.21 ± 0.68) μg · mL-1 to (18.27 ± 2.18) ug · mL-1;reverse ratio is 1.93.Compared with the effect on SMMC-7721 ceils,VCR,CDDP,5-FU effect on SMMC-7721/VCR cells had significant difference (P <0.01).Compared with the effect on SMMC-7721/VCR cell which was dealed by Tanshinone Ⅱ A microemulsion,VCR,CDDP,5-FU effect on SMMC-7721/VCR cells had significant difference (P < 0.01).Conclusion The Tanshinone Ⅱ A submicroemulsion can inhibit the growth of SMMC-7721 cells and SMMC-7721/VCR cells.The Tanshinone Ⅱ A submicroemulsion can partially reverse the multidrug resistance of SMMC-7721/VCR in vitro.

Objective To assess the safety and efficacy of domestic sirolimus-eluting stent(SES)compared with bare metal stent(BMS)in the primary percutaneous coronary intervention(PCI)for patients with ST-segment elevation AMI in a real-world scenario.Method From January 2005 to March 2006,a total of 143 patient with ST-segment elevation AMI were enrolled in this study,and all of them underwent primary percutaneous coronary intervention(PCI).Among the 143 patients,74 were treated with domestic SESs(Firebird stent)and 69 with BMSs.The incidence of major adverse cardiovascular events(MACE:death,reinfarction,and target vessel revascularization[TVR])was evaluated at 30 days and 180 days.Continuous variables were compared using Student's unpaired t test.Categorical variables were compared using Fisher's test.Cox proportional hazard survival models were used to assess risk reduction of adverse events.P value

OBJECTIVETo constitute HPLC fingerprint of the methanol extract from Swertia mussotii grown in Sichuan Province.
METHODRP-HPLC, methanol and water including 0.02% acid as mobile phase, gradient elution, flow rate 1.0 mL x min(-1), the detection wavelength was 260 nm, temperature was 30 degrees C.
RESULTThe RSD values of peak area and retention time of common peaks in precision, repeatability and stability were lower than 5.0%, respectively, similarity was over 0.805 in S. mussotii collected from 10 different habitats.
CONCLUSIONAll results above exhibit that this method is simple, practicable, and reliable as a standard method in controlling the quality of S. mussotii.

6.Clinical study of transcrestal maxillary sinus floor elevation with the disk-up sinus reamer.

OBJECTIVETo evaluate the clinical effect of the disk-up sinus reamer (DSR) applied to transcrestal maxillary sinus floor elevation with simultaneous placement of implants.
METHODSThirty-seven patients underwent transcrestal maxillary sinus floor elevation with fifty-one implants placed simultaneously using the DSR. The residual bone height (RBH) was 3 to 8 mm, (5.61 ± 1.61) mm on average. The safety of this technique and the pain index during the operation was evaluated. The final prostheses were restored in 3-6 months postoperatively. The follow-up period was 3 to 24 months. The stability and osseointegration of the implants were clinically evaluated, and the endo-sinus bone gain around the implants were measured.
RESULTSThe elevation height ranged from 2 to 8 mm, with an average of (4.75 ± 1.55) mm. There was no detectable sinus membrane perforation, no serious suffering or uncomfortable subjective sensation in any patients during operation with a pain index of (2.22 ± 0.98). During the follow-up period, no sinus complication was observed. Favorable osseointegration was obtained. There were no implants or prostheses which were loose or lost. The survival rate was 100%. The radiographic results demonstrated that the endo-sinus bone gain tended to reach stabilization after 6 months and the marginal bone loss was (1.20 ± 0.72) mm after 12 months.
CONCLUSIONSTranscrestal maxillary sinus floor elevation with simultaneous implant placement by DSR is a safe, invasive and handy technique, with higher elevation height, fewer clinical complications and less pain. It shows satisfactory clinical results in short term and a long-term observation is still needed.

7.Numerical simulation of microcarrier motion in a rotating wall vessel bioreactor.

OBJECTIVETo analyze the forces of rotational wall vessel (RWV) bioreactor on small tissue pieces or microcarrier particles and to determine the tracks of microcarrier particles in RWV bioreactor.
METHODSThe motion of the microcarrier in the rotating wall vessel (RWV) bioreactor with both the inner and outer cylinders rotating was modeled by numerical simulation.
RESULTSThe continuous trajectory of microcarrier particles, including the possible collision with the wall was obtained. An expression between the minimum rotational speed difference of the inner and outer cylinders and the microcarrier particle or aggregate radius could avoid collisions with either wall. The range of microcarrier radius or tissue size, which could be safely cultured in the RWV bioreactor, in terms of shear stress level, was determined.
CONCLUSIONThe model works well in describing the trajectory of a heavier microcarrier particle in rotating wall vessel.

8.A Retrospective Study of Pulmonary Actinomycosis in a Single Institution in China.

BACKGROUNDActinomycosis is a rare indolent infectious disease caused by Actinomyces. Although pulmonary actinomycosis is thought to be more prevalent in developing countries, data from developing countries are scanty. This study was to reveal the current situation of pulmonary actinomycosis in developing countries and the difference from that in developed countries.
METHODSPatients fulfilling the inclusion criteria for pulmonary actinomycosis from Peking Union Medical College Hospital in China between January 2003 and December 2014 were retrospectively analyzed. Baseline characteristics, clinical symptoms, underlying diseases, diagnostic methods, pulmonary function test results, chest computed tomography (CT) tests, fluoro-2-deoxy-D-glucose positron emission tomography (FDG-PET) tests, initial diagnosis, treatment and prognosis were retrieved from medical records and analyzed.
RESULTSTwenty-six patients were included in this study (mean age 52.0 + 13.1 years). The ratio of male to female was 1.17:1. Most common clinical symptoms were cough (15/26), sputum (12/26) and hemoptysis (12/26). Chest CT findings presented as masses (13/26), nodules (10/26) and infiltrates (3/26). FDG-PET had an increased standardized uptake value and 4/6 patients were misdiagnosed as malignancy. Many kinds of antibiotics were used in the treatment of pulmonary actonomycosis and all got favorable results. Five patients receiving complete resection of the lesion were cured without postoperative use of antibiotic.
CONCLUSIONSPulmonary actinomycosis is a rare disease even in developing countries, and both misdiagnosis and missed diagnosis are common. FDG-PET seems useless in the differential diagnosis, and complete resection of the pulmonary lesion without postoperative antibiotic therapy might be enough to achieve cure.

9.Expression of annexin V in decidua tissues of preeclampsia patients.

OBJECTIVETo investigate the expression of annexin V in the decidua tissues of preeclampsia patients and explore its clinical significance.
METHODSReal-time PCR, Western blotting and immunohistochemistry were employed to detect the mRNA and protein expressions of annexin V in the deciduas from 35 normal pregnant women at full term, 38 early onset severe preeclampsia patients and 33 late onset severe preeclampsia patients.
RESULTSAnnexin V was found on the cell membrane and in the cytoplasm of the decidual cells and stroma. Both the mRNA and protein of annexin V expressions in the decidua tissues were significantly different between normal pregnancy group and early or late onset severe preeclampsia group (P<0.05), being the highest in normal pregnancy group and the lowest in early onset severe preeclampsia group.
CONCLUSIONThe low expression of annexin V in the deciduas might participate in the hypercoagulability state in preeclampsia patients.

10.Clinical analysis of thoracoscopic surgery combined with intraoperative autologous blood transfusion in the treatment of traumatic hemothorax.

From January 2013 to January 2015, 19 patients of traumatic hemothorax with hemorrhagic shock were treated in our department by thoracoscopic surgery combined with autologous blood transfusion. This study retrospectively analyzed the therapeutic effect and shared our experience. The average amount of blood transfused back was 662.41 ml ± 269.15 ml. None of the patients developed transfusion reaction and were all discharged uneventfully. Thoracoscopic surgery combined with autologous blood trans- fusion is effective in the rescue of patients with progressive hemothorax and hemorrhagic shock. When corresponding indications are well managed, treatment for these patients is quicker, safer, and more effective.
