1.Epidemiological investigation of host animals infected with zoonotic parasitic worms in Nanbai Town of Zunyi County, Guizhou Province

Objective A field investigation of worm eggs in dog feces,classification of crabs and metacercaria infection were carried out in Nanbai Town of Zunyi County,Guizhou Province.Methods One or two villages were selected as the survey sites in Nanbai Town of Zunyi County and the local dogs and crabs were chosen as survey objects.Worm eggs in dog feces were detected using natural precipitation method.On the base of holotype,the classification of crabs was identified.Then,crabs were mashed and metacercaria were detected.Results From November 2014 to September 2015,through this survey in Xiejia Village and Longquan Administration Village,fifty-three fresh feces of dogs were collected and three worm eggs were detected,such as Dipylidium caninum egg,Toxocara canis egg and Spirometra mansoni egg.Forty-two crabs were caught,including fourteen female adults (two larvaes) and twenty-eight male adults (nine larvaes).All these crabs were identified as Sinopotamonzunyiense by morphological method and metacercaria was not detected.Conclusion Nanbai Town of Zunyi County only has Sinopotamonzunyiense;metacercaria is not detected,which means that this local area is not a Paragonimus epidemiological region;only three worm eggs are detected in dog feces.
2.Study on clinical therapeutic effect of activating brain and regaining consciousness needling method on poststroke depression and the mechanism.

OBJECTIVETo probe into an effective method for treatment of poststroke depression.
METHODSTwo hundred and fifty-six cases of poststroke depression were randomly divided into a control group (n = 76) treated by oral administration of amitilin and an acupuncture group (n = 180) treated by activating brain and regaining consciousness needling method. Their clinical therapeutic effects and changes of relative factors of poststroke depression and plasma contents of monoamine neurotransmitters were observed.
RESULTSIn the acupuncture group, the clinical symptoms and the relative indexes in the depression scales improved and plasma contents of the monoamine neurotransmitters increased significanty as compared with the control group (P < 0.05).
CONCLUSIONThe needling method for activating brain and regaining consciousness can improve relative factors to cure poststroke depression.

4.Primary study on the significance of ascites microRNA in the differential diagnosis of tuberculous and tumor

Objective To explore the value of microRNA in differential diagnosis of tuberculous ascites and cancerous ascites.Methods From January 2011 to October 2013,31 patients with ascites were enrolled in this study,19 cases of whom had cancerous ascites (two cases of ovarian cancer,three cases of liver cancer,one case of bile duct carcinoma,five cases of gastric cancer,three cases of pancreatic cancer,four cases of colon cancer and one case of peritoneal mesothelioma) and 12 cases had tuberculous ascites.Ascites was collected for microRNA microarray detection,and the possible differential expressed microRNA was screened.The results of microarray were confirmed by TaqMan stem-loop real-time polymerase chain reaction (PCR) method.The t test,receiver operating characteristic (ROC) curve and area under the ROC curve (AUC) were performed for statistical analysis.Results The results of microRNA expression profiles indicated that there were differences between tuberculous ascites and cancerous ascites.The findings of TaqMan real-time PCR showed the expression of microRNA-21 in cancerous ascites was 39.3±11.6,which was much higher than that of tuberculous ascites (12.6 ±4.1),and the difference was statistically significant (t=4.921,P<0.05).The expression of microRNA-134 in cancerous ascites was 68.2±20.4,which was lower than that of tuberculous ascites (210.2±37.2),and the difference was statistically significant (t =3.430,P < 0.05).The AUC of microRNA-21 and microRNA-134 in differential diagnosis of tuberculous ascites and cancerous ascites was 0.882 (95 % CI 0.816-0.917) and 0.877 (95% CI 0.782-0.901).The AUC of combined microRNA-21 and microRNA-134 in differential diagnosis of tuberculous ascites and cancerous ascites was 0.915 (95% CI 0.863-0.967).Conclusions There are differences in microRNA expression profiles between tuberculous ascites and cancerous ascites.The detection of microRNA-21 and microRNA-134 expression in ascites is of great importance in differential diagnosis.
5.Comparison of major bioactive components from leaves of Chrysanthemum morifolium.

Leaves of Chrysanthemum morifolium were potential medicinal resource. The present study aims to estimate the main bioactive components: total flavonoids (TF), galuteolin (GA), quercitrin (QU), chlorogenic acid (CA) and 3 ,5-O-caffeoylquinic acid ( CQ), which were considered to be the main effective components, in leaves of C. morfolium cultivars in China. The TF content was estimated hy UV-VIS spectrophotometry, while GA, QU, CA, and CQ were quantitatively determined by HPLC. The highest TF content (7. 13% w/w) was found in cultivar Wan Cong (Shexian county). Cultivar Da Bo ( Bozhou county) had the highest GA content (33. 45 mg - g-1); Cultivar Hong Xin (Sheyang county) contained the highest QU content (29.25 mg · g(-1)); Cultivar Chang Ban (Sheyang county) had the highest CA content (13.14 mg ·(-1)). The maximum CQ content (7.35 mg · g(-1)) was observed in culti- r Da Yang ( Tongxiang county). Different cultivars of C. morfolium had significant difference in components, but the leaf and capitulum of C. morifolium. were found to possess similar chemical compositions. The high content of bioactive components in several cultivars suggested the potential utilization of C. morifolium leaves.

6.Initial study of acoustic radiation force impulse(ARFI) elastography quantification for evaluation of solid liver tumors

ObjectiveTo evaluate the potential value of acoustic radiation force impulse (ARFI)elastography in the characterization of solid liver tumors.MethodsForty-three patients with 56 liver tumors were evaluated with ARFI,which included 21 patients with hepatocellular carcinoma (HCC),8 patients with metastase,5 patients with cholangiocarcinoma(CCC),and 9 patients with hemangioma.The shear wave velocity of the tumor and background liver parenchyma were calculated,and results were compared with 30 healthy subjects.Statistical analysis was performed on the shear wave velocity for differentiation of normal liver,background liver parenchyma,and tumors.ResultsHCC and CCC had greater stiffness than metastase (P <0.05),there were no statistical differences between HCC and CCC (P = 0.179).Malignant liver tumors had significantly greater stiffness than hemangioma and normal liver (P = 0.000).34.5% (9/26) HCC and 33.3% (4/12) hemangioma appeared softer than the background liver.With a cut-off value of 1.5 m/s for the shear wave velocity,the sensitivity,specificity,positive predictive value and negative predictive value for malignancies were 79.5%,83.3%,94.5% and 52.6%,respectively.ConclusionsARFI elastography quantification is a promising noninvasive technique for assessing solid liver tumors.Use of ARFI elastography quantification may lead to new quantitative tissue characterization parameters for differentiating hemangioma and malignant liver tumors.
7.Effect of triptolide on reversing hypermethylation of apc gene in Jurkat cells and its possible mechanisms.

This study was aimed to investigate the effect of traditional Chinese medicine, Triptolide (TPL) on reversing hypermethylation of antioncogene (apc gene) in acute lymphoblastic leukemia cell line Jurkat in vitro and to explore its mechanisms. The effects of TPL on cell growth, proliferation and cell cycle were detected by growth curve, MTT assay, colony formation test and flow cytometry, respectively. The effect of TPL on apc gene methylation of Jurkat cells was analyzed by nested methylation specific PCR; the expressions of apc gene, dnnt3a, dnmt3b mRNA were measured by RT-PCR; the protein expression of apc gene was detected by Western blot. The results showed that as compared with untreated control cells, the TPL of different concentrations could significantly inhibit growth and proliferation of Jurkat cells in dose-and time-dependent manners with IC₅₀ 19.7 ng/ml at 48 hours. All cytosines in CpG dinucleotides in untreated Jurkat cells had no changed, while all cytosines in Jurkat cells treated with TPL had been converted to thymidine suggesting the methylation of apc gene in Jurkat cells. The TPL could reverse hypermethylation of apc gene and induced the mRNA and protein expression of apc gene in dose-dependent manner. It is concluded that the small dose of TPL can obviously suppress the proliferation of Jurkat cells, activate and up-regulate the expression of apc gene through demethylation of apc gene resulting from DNMT and/or direct action, thereby inhibit the proliferation rate of Jurkat cells.

8.Evaluation the value of number of metastatic perigastric lymph nodes on prognosis in patients with gastric cancer.

OBJECTIVETo find out a simple and reproductive prognostic index in gastric cancer patients,which can be used as a comparable parameter among different regions of China.
METHODSThe perigastric metastatic lymph nodes(PGMLN) with long-term survival data were retrospectively evaluated in 148 gastric cancer patients, undergone potentially curative resections.
RESULTSThe 3-year cumulative survival rate was 62.8% and the survival rate decreased significantly with the increase of PGMLN, especially when the PGMLN was more than 6. The PGMLN had a clear linear relationship with total positive lymph nodes(r=0.94, P<0.01), while it had little correlation with total resected lymph nodes(r=0.18,P=0.2).
CONCLUSIONSPGMLN is a convenient and reliable prognostic factor in gastric cancer patients, which may be a good candidate of comparable parameters among different regions of China.

9.Association of transcriptional repressor Snail with Lauren classification of gastric cancer.

OBJECTIVETo explore the association of Snail expression and Lauren classification of gastric cancer.
METHODSThe protein levels of Snail and E-cadherin were detected by Western blot in N87 (intestinal-type gastric cancer cell line) and AGS(diffuse-type gastric cancer cell line) cell lines and those after transfection of GSK-3β plasmid. The study included a total of 77 patients with primary gastric cancer who underwent curative gastrectomy in the Zhongshan Hospital from February 2000 to December 2005 without any chemotherapy or radiation therapy before surgery. Tissues of gastric cancer specimens were stained using immunohistochemistry to determine Snail expression.
RESULTSSnail expression was low in N78 and high in AGS. E-cadherin expression showed reverse expression pattern. After transfection with GSK-3β, the expression of Snail was significantly suppressed and that of E-cadherin elevated (P<0.01). Different concentrations of GSK-3β inhibitor lithion chloride were used to treat the cell lines and Snail expression was significantly up-regulated in a dose-dependent manner (P<0.01). Snail expression was elevated in 16 out of 21 N78 cell lines, and in 21 out of 56 AGS cell lines, and the difference was statistically significant (P<0.01).
CONCLUSIONThe expression of Snail is closely associated with the Lauren classification of gastric cancer, and it may be a potential marker of the gastric cancer classification.

10.Oridonin inhibits proliferation of Jurkat cells via the down-regulation of Brg1.

OBJECTIVETo investigate the effect of oridonin on the human acute lymphocytic leukemia cell line Jurkat and its mechanism.
METHODSJurkat cells were cultured in vitro and treated with various concentrations (0, 1.25, 2.5, 5, and 10 μmol/L) of oridonin for different lengths of time (24, 48, and 72 hours). The proliferation of Jurkat cells was analyzed by MTT assay. The changes in nuclear morphology were evaluated by fluorescence microscopy at 12 hours after treatment with various concentrations of oridonin. The expression levels of Brg1, P53, and C-myc were determined by semi-quantitative Western blot in Jurkat cells treated with various concentrations of oridonin for 24 hours or 5 μmol/L oridonin for various lengths of time (0, 2, 6, 12, and 24 hours). The expression levels of P53 and C-myc and proliferation of Jurkat cells were evaluated after Brg1 expression was knocked down by Brg1-specific siRNA.
RESULTSCompared with the control group, the proliferation of oridonin-treated Jurkat cells was significantly inhibited in a concentration- and time-dependent manner (P<0.05). According to the florescence microscopic analysis, oridonin treatment led to nuclear pyknosis in Jurkat cells. Compared with the control group, Jurkat cells treated with 5 μmol/L oridonin had reduced expression of Brg1 and C-myc but elevated expression of P53. Brg1 knock-down led to a significant reduction in proliferation of Jurkat cells (P<0.05), up-regulated expression of P53, and down-regulated expression of C-myc.
CONCLUSIONSOridonin can inhibit the proliferation of Jurkat cells, probably via the Brg1 signaling pathway.
