1.The research progress of Lynch syndrome-associated endometrial cancers

Lynch syndrome.(LS) is an autosomal dominant condition caused by a mutation in the mismatch repair genes (MMR).Endometrial cancer (EC) is the most common extra-colonic cancers of LS Ⅱ type.Among the female members of LS family with the MMR gene mutation,EC has an overall lifetime risk more than 35%.LS-associated EC is more likely present as the clinical pathological characteristics of early age of onset,endometrioid adenocarcinoma,lower grade,lower uterine segment involvement,and better prognosis.LS patients with family history and early age of onset should be performed with a combined screening and genetic testing,while high-risk patients should be taken clinical monitoring methods as endometrial biopsy.Oral contraceptives mav be reasonable chemopreventive agents in the patients with LS.Although prophylactic hysterectomy with bilateral salpingo-oophorectomy is an effective strategy to prevent EC in women with LS,preoperative counseling should address the trade-offs between the reduction in the risk of cancer,and the risks and side effects of surgery.The patients need hormone replacement therapy after surgery.
2.Survival analysis of HIV/AIDS patients with antiretroviral therapy among drug users in Yili Prefecture from 2005 to 2019

Objective:
To understand the survival status and influencing factors of HIV/AIDS patients with highly active antiretroviral therapy ( HAART ) among drug users in Yili Prefecture, Xinjiang from 2005 to 2019, so as to provide references for reducing AIDS mortality.
Methods :
The demographic information, clinical stage, baseline CD4+T lymphocyte ( CD4 ) level and treatment status of HIV/AIDS patients with HAART in Yili Prefecture from 2005 to 2019 were collected through AIDS Antiretroviral Therapy Information System. The survival rate was calculated by the life table method. The influencing factors for survival time were analyzed by Cox proportional hazard regression model.
Results:
Totally 1 935 patients were recruited, the median age receiving HAART was 37 years old and the median CD4 counts was 293/μL. The cumulative survival rates at 1, 5, 7 and 10 years were 97%, 78%, 73%, and 66%, respectively. The multivariate Cox proportional hazards regression analysis showed that the patients with body mass index of 18.5-<28.0 kg/m2 ( HR: 0.391-0.656, 95%CI: 0.234-0.958 ), baseline CD4>200/μL ( HR: 0.354-0.667, 95%CI: 0.232-0.841 ) , or missed medication in the last 7 days ( HR=0.009, 95%CI: 0.001-0.061 ) had lower risk of death; the patients with WHO clinical stage of Ⅱ-Ⅳ ( HR: 1.479-2.311, 95%CI: 1.004-3.288 ) or treatment delay ≥1 years ( HR: 1.287-1.388, 95%CI: 1.029-1.826 ) had higher risk of death.
Conclusions
The 5-year cumulative survival rate of HIV/AIDS patients with HAART in Yili Prefecture is 78%. Body mass index, baseline CD4 level, WHO clinical stage, treatment delay and missed medication in last 7 days were the influencing factors for survival time.
3.The comparison of clinical efficacy of SOX and XELOX in the neoadjuvant chemotherapy for advanced gastric carcinoma

Objective To investigate the clinical efficacy of SOX and XELOX in neoadjuvant chemo-therapy for advanced gastric carcinoma .Methods Seventy-five cases with advanced gastric carcinoma were se-lected from our hospital from Feb 1,2011 to Oct 1,2015.SOX and XELOX were used to these patients treatment . The relationship between SOX and XELOX with tumor stage ,clinical curative effect evaluation ,operation ,postop-erative pathology ,adverse reaction ,survival analysis with advanced gastric carcinoma had been analyzed retrospec -tively.Results The efficacy of SOX and XELOX were 47.22%,41.03%.Compared with the two groups ,the CT curative effect evaluation,effective rate,disease control rate had no significant differences (P>0.05).The median survival time of DFS and OS were significant between the two groups (P<0.05).Conclusion The SOX and XELOX for neoadjuvant chemotherapy of advanced gastric carcinoma treatment is safety and effective , and the XELOX is better than SOX in terms of long -term effect,prognosis and clinical benefits .
4.Effect of Xianxiong decoction on acute lung injury mice induced by lipopolysaccharide.

OBJECTIVETo investigate the effect of Xianxiong decoction on the mice with acute lung injury induced by lipopolysaccharide.
METHODEighty female ICR mice were randomly divided into 8 groups: model group, Xianxiong decoction group, Daxianxiong decoction group, Xianxiong decoction group without Kansui Radix group, Xianxiong decoction group without Glycyrrhizae Radix et Rhizoma group, Glycyrrhizae Radix et Rhizoma and Kansui Radix group, normal group and control group. Animals of each group, except normal group, were undertaken intraperitoneal injection and intranasal inhalation of lipopolysaccharide (LPS) on day 1, 2, 3 to establish acute lung injury (ALI) model. 30 min after modeling, 0.2 mL corresponding drugs were administrated to each mice, dexam ethasone and normal saline were given to the mice of control group and normal group respectively. White blood cell in blood, neutrophil percentage of blood and bronchoalveolar lavage fluid (BALF) supernatant, the ratio of wet and dry lung tissue ( W/D), histopathological changes of lung tissue were estimated. Sixty ICR mice were randomly divided into normal, model, control, high, middle and low dose Xianxiong decoction groups and were modeled in the same way. ELISA was applied to detect the level of NF-kappaB, TNF-alpha and IL-6 in BALF, PCR for NF-kappaB and TNF-alpha mRNA in lung tissue, and Western blot for NF-kappaB and TNF-alpha. Half of 20 ICR mice were administrated with Xianxiong decoction of its maximum tolerant normal saline.
RESULTCompared with model group, the number of WBC in blood of Xianxiong decoction group mice decreased (P < 0.01), percentage of neutrophils in both blood and BALF decreased as well (P < 0.01, P < 0.05); it also significantly reduced the ratio of W/D (P < 0.01); and found the alveolar wall, the number of inflammatory cells infiltrating improved, compared with model group. Xianxiong decoction reduced the level of NF-kappaB, TNF-alpha and IL-6 in BALF (P < 0.01, P < 0.01, P < 0.05); its high and low dose groups only found TNF-alpha level declined. Five mice died 24 h after administration of Xianxiong decoction which indicated its toxicity when other influential factors were considered.
CONCLUSIONXianxiong decoction is effective on the ALI mice induced by LPS, but it is of toxicity at 3 g x mL(-1).

5.A Canine Portal Hypertension Model Induced by Intra-portal Administration of Polyurethane-Tetrahydrofuran Solutions.

This study was to build a canine portal hypertension model by intra-portal administration of high polymer material polyurethane and organic solvent tetrahydrofuran mixed solutions in order to evaluate the effectiveness of the model. Twelve local crossbreed dogs were selected randomly, with intra-portal administration of 8% (weight/volume) polyurethane- tetrahydrofuran solutions through an incision in the upper abdomen to build the portal hypertension model. We measured the portal vein pressure before modeling, during modeling, and four-, eight-, and twelve- weeks after modeling, respectively. Then we evaluated the effectiveness of the model comparing values of data with those data obtained before modeling started, which were regarded as the normal values. The results showed that the portal vein pressure rose by 2. 5 times after the solution administrated instantly as much as that before modeling, and maintained at 1. 5 times after 4 weeks. This method presents an easy operation, low animal mortality and reliable model of portal hypertension. Its less abdominal adhesions and its ability in keeping normal anatomic structure specially make it suit for surgical research of portal hypertension.

6.Effect of cerebral ischemic preconditioning on endogenous injury after cerebral ischemic reperfusion in rats

Objective To study effects of cerebral ischemic preconditioning on the changes of Fas,FasL,Caspase-3 proteins in infarction tissue after cerebral ischemia reperfusion of rats and to explore its mechanism.Methods The rats were randomly divided into three groups as follows:model Ⅰ group,model Ⅱ group and the sham operated group.Each group had thirty rats.The rats in model Ⅰ were prepared by Zea-Longa'middle cerebral artery occlusion.The right internal carotid artery of rats in model Ⅱ group was blocked transiently 10 minutes at one time.After 3 days,the middle cerebral artery occlusion of rat in model Ⅱ group was used to establish focal cerebral ischemia reperfusion injury model according to the model Ⅰ group.The sham operated group only isolated carotid artery.The neural behavior of rats was evaluated and the volume of cerebral infarction was detected after operation at 6,12,24,48,72 h(each group had six rats).The expressions of Fas,FasL and Caspase-3 protein of rats at different time points after operation were observed by immunohistochemical method.The apoptosis rate of nerve cells were detected by TdT -mediated dUTP nick-end labeling (TUNEL)method.Results At the time of 24 h after operated,the neural function defect point in model Ⅰ group,model Ⅱ group and sham operated group of rats were (2.8 ± 0.8),(1.8 ± 0.8),0 point.The expression of Fas protein positive cells in rats' brain tissue were (35.67 ± 2.16) %,(22.83 ± 1.71) %,(14.17 ± 2.32) % in that three groups.The expression of FasL protein positive cells in rats' brain tissue were(36.50 ±1.38)%,(22.67 ±2.50)%,(13.33 ±1.21)% in that three groups.The expression of Caspase-3 protein positive cells in rats' brain tissue were(37.33 ± 1.03)%,(23.17 ± 1.47)%,(11.33 ± 1.37)%.The apoptotic rate in brain tissue of rats were(40.58 ± 1.72)%,(23.25 ± 1.13)%,(11.75 ± 1.37)% in that three groups.Compared with model Ⅰ group,the neural function defect point of rats or the expression rat of Fas,FasL,Caspase-3 protein and the number of apoptotic nerve cells in model Ⅱ group at the same time all decreased;but the two groups were significantly higher than the sham operated group,(all P < 0.05).Conclusion Cerebral ischemic preconditioning can reduce the expression of Fas,FasL and Caspase-3 proteins.Negative regulation of Fas and FasL pathway after ischemic preconditioning may be an important endogenous mechanism of the resistance to damage.
7.Role of P2X4 receptor,a subtype of ionotropic purinoceptors, in central nervous system and its potential role as a drug target for treating neurological disorders

Adenosine 5-triphosphate is not only the source of cellular energy,but also the extracel-lular signal molecule that binds two different classes of ATP receptors:ionotropic P2X receptors and G-protein-coupled P2Y receptors.P2X4 receptor is one of the ionotropic P2X receptors widely expressed in the central nervous system(CNS).This subtype of the ionotropic P2X receptor is implicated in many physiological and pathological functions in the CNS, including chronic pain, alcohol abuse, seizures, Alzheimer′s and Parkinson′s diseases,and stroke.Therefore,P2X4 receptor has attracted attention as a promising drug target.Here,we review the findings of research or the physiological and pathological functions of P2X4 in the CNS. Recent progress in the development of new pharmacological agents of P2X4,such as selective antagonists,is also presented here.
8.The Predictive value of umbilical cord blood regulatory T cells for bronchopulmonary dysplasia in preterm infants

Objective To study the predictive value of umbilical cord blood regulatory T cells (Treg) for bronchopulmonary dysplasia (BPD) in preterm infants. Method From June 2017 to December 2018, premature infants with gestational age less than 32 weeks admitted to NICU of our hospiatal were prospectively selected. The umbilical cord blood was collected at birth to examine the Treg amount. The infants were assigned into BPD group and non-BPD group according to the diagnosis at discharge. The differences of Treg amount between the two groups and different degrees of BPD were analysed. Result A total of 124 premature infants (GA<32 weeks) were admitted, including 41 cases in BPD group (mild, n=18;moderate, n=14; severe, n=9) and 83 cases in the non-BPD group. The BPD group had GA of (29.6 ± 1.1) weeks and birth weight (BW) of (1128 ± 135) g. The non-BPD group had GA of (29.8 ± 1.1) weeks and BW of (1316 ± 180) g. The birth weight, 1min and 5min Apgar scores in BPD group were lower than the non-BPD group (P<0.001). The BPD group had higher incidence of respiratory distress syndrome, longer duration of mechanical ventilation (MV) and oxygen inhalation(P<0.001) than the non-BPD group. The MV duration and oxygen inhalation duration in the severe BPD group were significantly longer than the moderate and mild BPD groups, and the duration in the moderate group was longer than the mild group (P<0.001). The number of Treg in cord blood in the BPD group [(1.43 ± 0.06) × 105 cells/ml] was significantly lower than the non-BPD group [(2.57 ± 0.09) × 105 cells/ml], and the difference was statistically significant (P<0.001). Multivariate Logistic regression analysis showed that a significant decrease in the number of Treg was a risk factor for BPD in premature infants (OR=0.000, 95%CI 0.000 ~ 0.012, P=0.009). The number of Treg in umbilical cord blood was negatively correlated with the severity of BPD. The area under the ROC curve showed that the cut-off value was 1.95 × 105 cells/ml, with Youden index 0.613, sensitivity 85.4% and specificity 75.9%. Conclusion The number of cord blood Treg cells may be a useful biomarker for predicting BPD in premature infants.
9.Chemical constituents from flowers of Scabiosa tschilliensis.

Twenty-two compounds were isolated from the flowers of Scabiosa tschilliensis. Their structures were identified by spectroscopic methods as octacosanol (1), stearic acid (2), β-sitosterol (3), oleanolic acid (4), apigenin (5), luteolin (6), daucosterol (7), kaempferol-3-O-β-D-6-O-(p-hydroxycinnamoyl) -glucopyranoside (8), kaempferol-3-O-β-D- (3, 6-di-p-(hydroxycinnamoyl) -glucopyranoside (9), apigenin-7-O-β-D-glucopyranoside (10), luteolin-4'-O-β-D-glucopyranoside (11), apigenin-7-O-rutinoside (12), luteolin-7-O-β-D-glucopyranoside (13), apigenin-4'-O-β-D-glucopyranoside (14), caffeic acid methyl ester (15), loganin (16), adenosine (17), luteolin-6-C-β-D-glycopyranosyl (18), sweroside (19), sylvestrosides I (20), sylvestrosides II (21), urceolide (22). Among them, compounds 1, 2, 7-9, 12, 15, 17-18, 20-22 were isolated from the genus Scabiosa for the first time, and compounds 1-4, 6-9, 11-12, 14-22 were isolated from this plant for the first time. 13C-NMR data of 22 were reported for the first time.

10.Effect of micronutrients on the immune status of human immunodeficiency virus-positive individuals.

OBJECTIVETo evaluate the effect of micronutrients on the immune status of human immunodeficiency virus (HIV)-positive individuals.
METHODSTotally 102 HIV-positive individuals were randomly divided into supplementation group (received micronutrients supplement) and control group (received placebo). Physical examinations were performed at baseline and at the end of the trial. Immune status were determined in both two groups.
RESULTSAge, height, weight, and sex ratio were not significantly different between two groups (all P>0.05). Baseline CD3+, CD4+, and CD8+ T lymphocytes and IgA, IgG, IgM, C3 levels were not significantly different between two groups (all P>0.05), while the levels of CD3+, CD4+, and CD8+ T lymphocytes and IgA, IgG, IgM, C3 were significantly higher in supplementation group than in control group(all P0.05).
CONCLUSIONSupplementation of micronutrients can improve the immune status of HIV-positives individuals.
