OBJECTIVE To develop the strategies for preventing nosocomial infection in the department of(laboratory).METHODS The current situation in nosocomial infection management in department of laboratory was analyzed.RESULTS There were some problems in training knowledge of nosocomial infection,preparing(equipment) and supplies for prevention,implementing prevention measures,and cultivating good work habit.(CONCLUSIONS) It is very important to strengthen nosocomial infection management for preventing efficiently(nosocomial) infection in the department of laboratory.
4.Prevention of Nosocomial Infections in Laboratory Departments of Large Scale Hospitals:Current Situation and Measures

OBJECTIVE To analyze the reasons of nosocomial infection in laboratory departments, and then advance corresponding measures to overcome them. METHODS The current situations in laboratory department between domestic large scale hospitals and overseas hospitals were compared, especially paying attentions to those parts involving in management system and precautionary measures. RESULTS There were a lot of shortcomings existed in the supervision of nosocomial infections in laboratory departments; many measures should be taken to increase the management level. CONCLUSIONS To reform and improve the system of nosocomial infection control and prevention, and establish an effective and systematic alerting and prevention system will benefit all kinds of the hospitals.
5.The effect of NaAsO2 on the binding of methyl CpG binding protein 2, DNA methyltransferase 1 and histone deacetylase 1 to the promoter of MGMT gene in HaCaT cells

Objective To investigate the effect of NaAsO2 on the binding levels of methyl CpG binding protein 2(MeCP2),DNA methyltransferase 1 (DNMT1) and histone deacetylase 1(HDAC1) to the hypermethylation promoter region of MGMT gene in HaCaT cells,in order to provide a basis to deepen the interpretation of the role of arsenic poisoning mechanism.Methods HaCaT cells were treated repeatedly and interval with different concentrations of NaAsO2(3.13,6.25,12.50,25.00 μnol/L,respectively) for 72 h.Untreated HaCaT was used as blank control group and human epidermal squamous carcinoma cell line(A431 cells) as positive control group.The binding levels to the two transcription regulatory regions(ChIP1,ChIP2) and to the coding region(ChIP3) of MGMT 8ene were detected by chromatin immuno-precipitation combined with quantitative PCR.Results The differences of binding levels of MeCP2,DNMT1 and HDAC1 to ChIP1 and ChIP2 in each group were significant (F=7.387,84.634,78.442 and 19.263,69.649,26.546,all P < 0.05).The binding levels of MeCP2,DNMT1 and HDAC1 to ChIP1 and ChIP2 in each NaAsO2 exposed group[3.13 μmol/L NaAsO2 exposed group:(136.00 ±16.97)%,(145.00 ± 2.83)%,(88.50 ± 19.09)% and (106.50 ± 37.48)%,(112.34 ± 8.73)%,(59.71 ± 8.49)%;6.25 μmol/L NaAsO2 exposed group:(130.00 ± 42.43)%,(154.50 ± 4.95)%,(101.00 ± 1.27)% and (88.50 ±3.54)%,(134.32 ± 2.82)%,(102.75 ± 19.91)% ; 12.50 μmol/L NaAsO2 exposed group:(141.50 ± 23.33)%,(161.50 ± 7.78)%,(125.00 ± 11.31)% and (119.50 ± 24.75)%,(171.59 ± 3.54)%,(167.61 ± 10.61)%; 25.00μmol/L NaAsO2 exposed group:(134.50 ± 43.13)%,(472.50+ 50.20)%,(383.50 ± 30.41)% and (180.09 ±12.73)%,(348.50 ± 27.58)%,(158.45 ± 12.02)%] were higher than that in the blank control group[(51.50 ±9.19)%,(82.00 ± 12.73)%,(25.03 ± 2.91)% and (37.02 ± 4.24)%,(91.56 ± 26.16)%,(19.09 ± 2.90)%,all P < 0.05].The differences of binding levels of MeCP2 to ChIP3 in each group were not significant(F =1.670,P >0.05),but the differences of binding levels of DNMT1 and HDAC1 to ChIP3 were significant (F =4.404,9.863,all P < 0.05),and only the binding levels in the 25.00 μmol/L NaAsO2 exposed group [(615.85 ± 29.63)%,(306.09 ± 59.40)%] were higher than that in the blank control group[(99.70 ± 12.02)%,(92.45 ± 48.79)%,all P < 0.05].Conclusions MeCP2 can bind to the methylated MGMT gene transcriptional regulatory regions which are induced by arsenic and leads to histone deacetylation by the recruitment of DNMT1 and HDAC1 and,meanwhile,DNMT1 can bind to the coding region of MGMT gene to recruit HDAC1 in a methyl DNA binding protein(MBD) independence manner and media MGMT gene silencing through the chromatin remodeling way,which might be the early molecular events of arsenic poisoning.
6.Analysis of 32 patients with colon cancers co-existing with appendicitis

Objective To explore the reason of colon cancer with appendicitis and reduce the rate of misdiagnosis. Methods We car-ried out analysis retrospectively to analyze 1 094 hospitalized patients with colon cancer in the First Affiliated Hospital of Harbin Medical Uni-versity from January 2006 to March 2013. Results Among 1 094 colon cancer patients,31 patients who firstly diagnosed with acute appendi-citis received appendectomy,1 patient with appendiceal abscess received non-surgical treatment. Among 31 patients with acute appendicitis,6 patients were found to be with ileocecal tumors during surgeries;24 patients were diagnosed with colon cancer within one year;incision of 1 patient did not heal, with continuous drainage of brown liquid;the appendicitis of 1 patient who received non-surgical treatment was recur-rent. Conclusion As lack of typical symptoms,colon cancer is prone to misdiagnose and diagnose incorrectly when the cancer co-exists with appendicitis and symptoms of appendicitis were showed firstly. Therefore,clinicians should be vigilant for patients with appendicitis. In order to induce the rate of misdiagnosis,it is essential that requesting disease history carefully,timely and relevant inspection and appropriate ex-ploratory surgery for the patients with appendicitis.
7.Value of the combining detection of p53, p27 and bcl-2 in early diagnosis and the implementation of the intervention for non-small cell lung cancer

BACKGROUND: The quality of life(QOL) of patients with lung cancer is ffected due to depression, reduced lung function, subjective reduced body force, fatigue, and poor stamina, etc., and the survival of the patientswould be affected by complications or advanced stage cachexia.OBJECTIVE: To investigate the relationship between three gene proteins including p53, p27 and bcl-2 and the pathological characters of non-small cell lung cancer (NSCLC).DESIGN: An experimental trial by employing pathological specimens as subjects.SETTING: Department of respiration of a university affiliated hospital and the Central laboratory of a university.PARTICIPANTS: Totally 76 specimens of NSCLC after surgical resection between June 1997 and December 2002, which were all primary lung cancer without any other therapy.METHODS: The expression of three gene proteins in 76 NSCLC specimens was detected by SP immunohistochemical analysis.MAIN OUTCOME MEASURES: Positive expression of p53, p27 and bcl-2.RESULTS: Among 76 specimens, 28 cases(37%, 28/76) with excessive expression of p53, 34 cases(45%, 34/76) with excessive expression of p27, 37 cases(49%, 37/76) with excessive expression of bcl-2, and 7 cases with excessive expression in all three proteins. The positive expression of p53 elevated with the reduced gradation in differentiation; bcl-2 and p27positive expressions reduced with the reduced gradation in differentiation and there was significant difference between high-differentiation group and low-differentiation group( P < 0. 05) . However, there was no significant relationship between the positive expressions of three proteins and the histological classification, lymph node metastasis, and pathological aging of lung cancer( P > 0.05).CONCLUSION: The excessive expression of p53, p27 and bcl-2 genes might be related with the occurrence and development of NSCLC.
8.Traditional Chinese medicine to prevent environmental pollutant-induced epigenetic changes of sperm DNA.

In recent years, the incidences of male infertility and recurrent abortion are increasing. The causes of the conditions are varied and complex, but the affected stability of the genetic material in the sperm is an important etiological factor. Environmental pollutants may invade the body through respiration, diet and other channels, resulting in infertility and abortion or even affecting the reproductive system development of the offspring by changing the epigenetics of sperm DNA. In this paper, we propose the idea of using traditional Chinese medicine to prevent environmental pollutant-induced epigenetic changes of sperm DNA, hoping to provide a new insight into the protection against genetic material damage, male infertility, and recurrent abortion caused by environmental pollutants.
