1.Effect of hydrogen peroxide on human leukocyte antigen-G expression in placental trophoblasts in pre-eclampsia

Objective To explore the effect of hydrogen peroxide (H2O2) on the expression of human leukocyte antigen-G ( HLA-G) in placental trophoblasts in pregnant women with pre-eclampsia.Methods Forty pregnant women,delivered through cesarean section in the Department of Obstetrics of and Gynecology,the First Affiliated Hospital of Nanjing Medical University from October 2008 to October 2009,were enrolled,including 20 women with pre-eclampsia and 20 healthy gravidas (control group).Colorimetry and western blot were applied,respectively,to determine the level of H2O2 and the expression of HLA-G protein in placental tissues and the correlation between them were analyzed.After 24 hours of seeding,JEG-3 cells (the HLA-G positive cell line of choriocarcinoma) were divided into two groups:intervention group (exposure to 175 μmol/L H2O2) and control group (without H2O2).Immunofluorescence and western blot were used to investigate the expression of HLA-G protein in JEG-3 cells at 24 hours and 48 hours after incubation.Results (1 )The level of H2O2 in placenta in the pre-eclampsia group was significantly higher than that in control group[(105 ±13) nmol·mg-1·prot-1 vs (62 ± 18) nmol·mg-1 ·prot-1,P < 0.05].(2) The expression of HLA-G protein in placenta of the pre-eclampsia group was reduced by88%compared with that of the control (0.20 ± 0.08 vs 1.67 ± 0.65,P < 0.05).( 3) Negative correlation was found between HLA-G level and H2O2 expression in the placenta in both groups(r =-0.895,P =0.000).(4) Compared with the control group,the expression of HLA-G protein in JEG-3 cells,after 24 hours and 48 hours exposure to H2O2,reduced by 39% and 80%,respectively,(3.21 ±0.33 vs 1.95 ±0.25 and 0.65 ±0.08,P <0.05,respectively) and 67% reduction was detected from 24 hours to 48 hours of H2O2 exposure (P <0.05).Fluorescence microscope observed reduced expression of HLA-G in JEG-E cells in the intervention group at 48 hours compared to the control group (P<0.05).Conclusion High level of H2O2 could down-regulate HLA-G expression in the placental trophoblasts in pre-eclampsia which may be involved in the pathogenesis of pre-eclampsia.
2. Effects of protocatechuic acid on expression of D2DR, iNOS, and TH in striatum and midbrain of Parkinson's disease model mice

The aim of this paper is to investigate the effects of protocatechuic acid (PCA) on the expression of D2DR, iNOS, and TH in striatum and midbrain of Parkinson's disease (PD) mice induced by MPTP. C57BL brown mice were used as experimental animals and were injected with MPTP (25 mg/kg. d) for 7 d to build PD model mice. PCA (10 mg/kg. d) and Madopar (125 mg/kg. d) had been ip administered to the PD mice for the precaution and treatment at the first and lasted for 7 d. The expression of D2DR, iNOS, and TH in the striatum and midbrain of PD mice induced by MPTP was detected by Western blotting method. The results showed that iNOS expression increased while D2DR and TH expression decreased in the striatum and midbrain of MPTP-induced PD mice. However, PCA significantly decreased the expression of iNOS while obviously increased the expression of D2DR and TH in the striatum and midbrain of MPTP-induced PD mice. PCA has a neuroprotective effect on the PD mice induced by MPTP, and its protective effects on mice brain are achieved at least partly by increasing TH and D2DR expression while decreasing iNOS expression.
3. Expression changes of alpha adrenergic receptors in heart and mesenteric artery in autonomic dysreflexia rats after spinal cord transection

Objective: To investigate the changes of alpha adrenergic receptors (α-AR) mRNA expression in heart and mesenteric artery in autonomic dysreflexia (AD) rats after spinal cord transection, so as to explore the possible mechanism of AD. Methods: The spinal cord of rats was exposed and the fourth thoracic spinal cord was transected; 4 weeks later, rats' rectum was stimulated by self-made catheter and those with a mean arterial blood pressure increased by more than 15 mmHg (1 mmHg=0.133 kPa) were chosen as AD group (n=16). Heart and mesenteric arteries along with their branches were harvested. mRNA expression of α1A-, α1B-, α1D-, α2A-, α2B- and α2C-AR was quantified by real time PCR and the result was compared with that in sham-operated group (the fourth thoracic spinal cord was exposed but not transected). Results: Compared with sham-opera ted group, rats in AD group had a lower expression of α1A-AR mRNA (P<0.05) and α1B-AR mRNA (P<0.01) in their heart, but the expression of α1D-AR mRNA had no obvious change. Also, rats in AD group had a higer expression of α1A-, α1D-, α2B-AR mRNA (all P<0.01) and α2C-AR mRNA (P<0.05) in their mesentric artery, but α1B- and α2A-AR mRNA expression had no obvious change. Conclusion: AD may occur in the chronic stage of spinal cord transection in rats. The abnormal expression of α1A- and α1B-AR mRNA in the heart and α1A-, α1D-, α2B- and α2C-AR mRNA in the mesenteric artery may be related to AD.
4.Clinical application of INRatio coagulometer for fast measuring international normalized ratio

ObjectiveTo compare the correlation and agreement of international normalized ratio (INR) measured by INRatio coagulometer and standard laboratory venous blood measurement in order to judge the clinical application value of measuring INR by INRatio coagulometer.MethodsOne hundred who received warfarin were chosen randomly.Venous blood and finger capillary blood were taken simultaneously for measurement of INR by laboratory coagulation analyzer SYSMEX CA-7000 and INRatio coagulometer respectively.The correlation and agreement of INR results tested by the two methods were analyzed.Results When INR ≤5,there was a good correlation between INRatio coagulometer and laboratory coagulation analyzer SYSMEX CA-7000 (r =0.898,P =0.000).The mean bias of the two methods was 0.36 and INR measured by INRatio coagulometer were higher than those tested by laboratory coagulation analyzer SYSMEX CA-7000.By Bland-Altman,it showed a good agreement for 91.2% (62/68) of the difference INR located 95% the limited of agreement when the mean INR 1.60 - 3.50,and a bad agreement for only 50.0%(1/2) of the difference INR located 95% the limited of agreement when the mean INR > 3.50.ConclusionsIn the therapeutic range,INR measured by INRatio coagulometer shows a good correlation and agreement with INR measured by the established laboratory method.INRatio coagulometer can be used to monitor INR in the patients received oral anticoagulant therapy.
5.Management strategy of implantable medical device in clinical department

7.Relationship among hardiness, perceived organizational support and work engagement of clinical nurses

Objective To know about the current status of the work engagement of the clinical nurses,and discuss the relatioaships among hardiness,perceived organizational support and work engagement so as to provide basis for improving the level of the work engagement of the clinical nurses.Methods The work engagement scale,hardiness scale and perceived organizational support scale were used to investigate a total of 630 clinical nurses.Results The general score of work engagement,hardiness and perceived organizational support were showed respectively as following:(52.54±8.08),(70.09±14.44) and(82.41±24.98).Every two items of hardiness,perceived organizational support and work engagement were in significant positive correlation.Regression analysis showed after the variable of demographic was controlled,the hardiness and perceived organizational support respectively had significant positive predictive function on work engagement as respective independent variables.Conclusions Hospital managers should create a good working environment for clinical nurses,measures should be taken from individual and organizational aspects so as to improve the hardiness and perceived organizational support and the work positivity of the clinical nurses,in order to make the nurses more positively devoted to their work.It had important practical significance to improve the quality of their services and stabilize nursing group.
8. Protective effects of different solvent fractions of Boschniakia rossica on acute liver injury induced by acetaminophen in mice

OBJECTIVE: To investigate the protective effect of different solvent fractions of Boschniakia rossica on acute liver injury induced by acetaminophen (APAP) in mice. METHODS: The mice were randomly assigned to the normal control, model control, bifendate (positive control), as well as groups of different solvent fractions of Boschniakia rossica. Animals were treated once daily for 7d. APAP were given intraperitoneally to the mice of groups, then the alanine aminotransferase (ALT), aspartate aminotransferase (AST), albumin (ALB), reactive oxygen species (ROS), lipid hydroperoxide (LPO), malondialdehyde (MDA), glutathione (GSH), catalase (CAT), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), Na+-K+-ATPase, Ca2+-Mg2+-ATPase nitric oxide (NO) and inducible nitric oxide synthase (iNOS) were detected by the colorimetric method. RESULTS: The administration with butanol soluble fraction and aqueous fraction oi Boschniakia rossica reduced the serum ALT and AST activities, increased the scrum ALB level, decreased the hepatic ROS, LPO and MDA levels, increased the CAT, GPx, SOD activities and GSH level, increased the Na+-K+-ATPase and Ca2+-Mg2+-ATPase activities of liver mitochondria, and decreased hepatic iNOS activity and NO level of liver in mice with acute liver injury. CONCLUSION: The different solvent fractions of Boschniakia rossica have protective effects on acute liver injury induced by APAP in mice, probably via anti-oxidative and anti-inflammatory activities.
9. Effect of Yaotongning Capsules combined with Imrecoxib Tablets in 60 cases of ankylosing spondylitis with cold-dampness and blood-stasis syndrome

Objective: To observe the effect of Yaotongning Capsules combined with Imrecoxib Tablets in the treatment of ankylosing spondylitis with cold-dampness and blood-stasis syndrome. Methods: A total of 120 cases with ankylosing spondylitis were randomly divided into two groups: treatment group and control group, with 60 patients in each group. The control group was given Imrecoxib Tablets, and the treatment group was given Imrecoxib Tablets and Yaotongning Capsules. The levels of ESR, CRP, and the scores of BASDAI, BASFI, TCM syndrome were compared before and after treatment, the treatment effect of two groups were evaluated. Results: There was no significant difference in the levels of ESR, CRP and the scores of BASDAI, BASFI, TCM syndrome between two groups before treatment. After treatment, the levels of ESR, CRP and the scores of BASDAI, BASFI, TCM syndrome of the two groups were decreased (P < 0.05), and all the indexes in treatment group were lower than those in control group (P < 0.05), the total effective rate in treatment group (86.67%) was better than that in control group (65.00%) (P < 0.01). Conclusion: Yaotongning Capsules can effectively improve TCM syndrome and functional activity indexes, reduce inflammatory indexes and disease activity, has definite clinical effect on ankylosing spondylitis.
10.Correlation Analysis of HPLC Fingerprint of Medicinal Pieces of Yupingfeng Dec oction and Yupingfeng Decoction

Objective To establish the method of fingerprint analysis on Yupingf eng Decoction,and to study the correlation of HPLC fingerprint in Radix Astraga li,Radix Saposhnikovlae,Rhizoma Atractylodis Macrocephalae and Yupingfeng Deco ction. Methods HPLC with Hypersil ODS was used,acetonitrile -water(gradient el ution) as a mobile phase and detection wavelength at 220 nm,flow rate was 1 mL ?min-1,and column temperature was 30 ℃. Results There were 9,8 and 7 common peaks separated from 10 batches of medicinal material of Radix Astragali,Radix Saposhnikovlae,and Rhizoma Atractylodis Macrocephalae,respectively;11 common peaks were separated from 10 batches of Yupingfeng Decoction,of which 6 peaks were shared by Radix Astragali,4 peaks by Radix Saposhnikoviae and 1 peak by Rh izoma Atractylodis Macrocephalae. Conclusion There exists a correlation of Yupin gfeng decoction with the medicinal pieces of Radix Astragali and Radix Saposhnik oviae. The major characteristic fingerprint peaks of Yupingfeng decoction belong to those of the isoflavones from Radix Astragali and chromones from Radix Sapos hnikoviae. This will provide a reference for the rules of the compatibility and component research of Yupingfeng Decoction.
