1.Controlled release by novel lysostaphin-loaded hydroxyapatite/chitosan composites.

Lysostaphin is highly effective on eliminating methicillin resistant Staphylococcus aureus (MRSA). In order to achieve controlled release of lysostaphin, a biocompatible drug carrier is needed. Hydroxyapatite/chitosan (HA/CS) composites were chosen to carry lysostaphin and sample composites with different weight ratios of HA to CS, including 80/20, 70/30, 60/40, and 40/60, were prepared. Multiple analyses were performed to determine the structural and physicochemical properties of the composites, including scanning electron microscopy, X-ray diffraction and Fourier transform infrared spectroscopy. We immersed HA/CS composites loaded with 1 wt% lysostaphin to test in vitro release activity and cultured MC3T3-E1 cells to carry out biocompatibility test. The result of the release behavior of the composites revealed that the controlled release of lysostaphin from 60/40 HA/CS composites was the highest release rate of (87.4 ± 2.8)%, which lasted for 120 hours. In biocompatibility testing, MC3T3-E1 cells were able to proliferate on the surface of these composites, and the extract liquid from the composites could increase the growth of the cells. These results demonstrate the controlled release of lysostaphin from HA/CS composites and their biocompatibility, suggesting the potential application of these composites to bone injury and infection applications.

2.Determination of Amino Acids inQihong Maitong Injection through Pre-column Derivatization HPLC

In this article, an HPLC method for the contents determination of amino acids in Qihong Maitong injection was reported. In detailed, OPA-Fmoc pre-column derivatization was adopted, and related HPLC methods to determine the contents of amino acids was established. Linear relationship was well constructed for 17 amino acids through the method mentioned above. Briefly speaking, the optimized method was accurate and reproducible, and suitable for the determination of amino acids in Qihong Maitong injection and corresponding quality control.
3.Evaluation of cystatin C with two kinds of detection systems

Objective To evaluate the performance of Cys C results among two detection system.Methods The particle-enhanced immunonephrometic assay was used in Dade Behring BNII. Immunoturbic assay was used in Hitachi 7170 to evaluate the JING' YUAN reagents.We compared the precison,linearity,interference,correlation,and calibrators agreement with Dade Behring BNII.Results The total CV of the samples that contain 0.6-5.0 mg/L was less than 10%.The Dade Behring and JING'YUAN method showed good linearity.Haemoglobin(10 g/L),Bilirubin(300 mg/L), Vitamin C(5 g/L)in the tested sample had no significant interference in the assay(interference 0.05) between JING' YUAN and Dade Behring reagents.Values were slightly lower than that from the Dade Behring BNII method,the mean bias was-0.16.The bias range was 1.1%-23% between JING'YUAN and Dade Behring for one sample.Conclusions The precision,linearity and interference test were suitable for routine Cys C measurement on automated biochemistry analyzer,but results has bias.
4.Development of new radial artery puncture and fixation device

Objective To develop a new radial artery puncture fixing device to conquer the deficiencies of traditional method. Methods The device was composed of an arm support,a hand support,a wrist part,an arm fixing belt,a palm fixing belt and a tourniquet.The arm support was concave-shaped,and was connected with the hand support with the wrist part.The wrist part had a raised block which was gifted with an inflatable bag.The balloon linked with an inflation balloon with an inflation tube.There was a valve between the inflation tube and balloon.The fixing belt had one end directly connected with the arm support, hand support and wrist part, and the other end linked with the other ends of the above components with Velcro. There was a compression hemostatic balloon at the internal surface of the tourniquet. Results The device behaved well in puncture time and success rate,and decreased the incidence rates of errhysis and hematoma after withdrawing the puncture needle while increased the satisfaction of medical staffs and patients. Conclusion The device gains advantages in simple structure,convenience,practicability and safety,and meets the desired requirements.[Chinese Medical Equipment Jour-nal,2018,39(5):44-46]
5.Comparison of serum creatinine,Cystatin C and estimated glomerular filtration rate on evaluation of glomerular filtration function in chronic kidney disease patients

Objective To compare the coherence of serum creatinine,creatinine clearance(Ccr), Cystatin C,and estimated glomerular filtration rate(eGFR)in each stage of chronic kidney disease(CKD) patients.Methods Creatinine in serum and urine were determined by Jaffe method;serum Cystatin C was measured by particle enhanced turbidimetric method,while eGFR was calculated using the abbreviated Modification of Diet in Renal Disease(MDRD)equation which was mainly based on the serum creatinine concentration.According to the American national kidney foundation-Kidney Disease Outcome Quality Initiative(NKF-K/DOQI)guideline,all cases were grouped by eGFR into 5 stages.Results In these 228 cases,as eGFR decreased gradually,the average levels of creatinine and Cystatin C increased,while Ccr decreased.The level of each items showed a statistic difference among each stage(P0.05);in eGFR 60-89 ml/min group,the average level of creatinine was 83.3 ?mol/L,the abnormal rate was only 6.8%,it was not a sensitive marker to detect the slightly damaged GFR,the levels of Ccr and Cystatin C showed a marginal decrease and increase,with an abnormal rates of 70% and 86%,there was a statistic difference among the three abnormal rates(P
6.Development of disposable anticoagulant negative pressure syringe

Objective To develop a disposable negative pressure syringe to solve the problems in bone marrow aspiration. Methods The syringe was composed of an injector and a self-contained puncture needle.The injector consisted of a barrel,a plunger and a tip.The plunger had its front end placed in the barrel and connected with the tip.The tip clung to the inner wall and could slide along it,and the upper end of the cylinder was connected closely with a negative-pressure sealing cap.A negative-pressure cavity was formed between the sealing cap and the tip,and an anticoagulation layer existed at the cylinder inner wall in the cavity.The blood taking needle was made up of a bone puncture needle and a hub.Results The developed syringe gained stable negative pressure,low time and workload consumption during bone marrow aspiration when compared with the conventional tools. Conclusion The syringe increases the quality of bone marrow aspiration while decreases the workload of medical staffs,and thus is worthy promoting practically.
7.Effect of interventional chemotherapy on angiogenesis and apoptotic protein expression in pathological tissues of patients with lung cancer

Objective To explore the effect of interventional chemotherapy on angiogenesis and apoptotic protein expression in pathological tissues of patients with lung cancer.Methods Ninety-four lung cancer patients were divided into control group and treatment group,47 cases in each group.The patients in two groups were treated with lung cancer resection.In control group,no chemotherapy was given preoperatively.The treatment group was given adriamycin combined with cisplatin before chemotherapy.The expression of angiogenesis related factors and microvessel density (MVD) were compared between the two groups.The expressions of survivin and p53 in two groups were detected.Results The positive rates of vascular endothelial growth factor (VEGF) expression in control group and treatment group were 51.06% and 23.40%,the positive rates of Sema3 protein were 4.26%,25.53% and microvessel density were 23.17 ± 2.45 and 10.16 ± 2.05,all with significant difference (all P < 0.01).The relative expression of puma protein in control group and treament group were 0.87± 0.01 and 1.11 ± 0.01,and the relative expression of survivin protein were 1.55 ± 0.02 and 0.56 ± 0.01,all with significant difference (all P < 0.01).Conclusion The mechanism of preoperative chemotherapy may be related to inhibition of angiogenesis,up-regulation of the expression of proapoptotic protein puma and down-regulation of survivin expression of anti-apoptotic protein.
8.The Detoxification Technology and Common Detoxification Mechanism of Toxic Chinese Medicine

Chinese medicine processing is a kind of traditional pharmaceutical technology,which can reduce toxicity,enhance the curative effect,change the properties and facilitate dispensing preparations in clinical use.The technology of detoxification is a great pioneering work for the safe application of toxic Chinese medicine,which can reduce the side effects of certain drugs,ensure the clinical safe use and expand the scope of clinical application through processing.In this paper,we searched the toxic Chinese medicine and the processing technology in ancient literatures of TCM,and then we summarized and analyzed their lat-est studies at home and abroad.Thus,we make a summary of toxic Chinese medicine including its classification,site of action,the toxic components,detoxification and its clinical application.Meanwhile,we proposed an opinion that toxic Chinese medi-cines with the same processing technology had common detoxification rules.Therefore,we hope this work will provide new perspectives for the research on the processing of toxic Chinese medicine and the mechanism of detoxification.
9.Exploration of differential expressed genes involved in the development and progression of hepatocellular carcinoma using oligo microarray.

OBJECTIVESTo analyze the differential expression genes (DEGs) among hepatocellular carcinoma (HCC), para-cancerous tissue (PCT) and normal liver tissue (NLT) and explore the target genes related to the development and progression of HCC.
METHODSThe total RNAs of matched HCC, PCT and NLT of HCC patients were isolated using one step Trizol method. Matched RNAs were qualified using 10 g/L agarose gel electrophoresis and lab-on-chip. cRNAs were synthesized, fluorescence labeled and purified after total RNAs were purified. The RNAs of HCC and NLT, HCC and PCT were hybridized with Agilent oligo microarray (21,074 probes). The fluorescence intensity features were detected by Agilent scanner and quantified by feature extraction software. The selected candidate genes were confirmed by SYBR Green I stained real time RT-PCR.
RESULTS(1) The total RNA, reverse transcription product and fluorescence labeled cRNA were all of high quality; (2) There were 420 up-regulated genes and 552 down-regulated genes among 2-fold DEGs, including DKK1 (dickkopf homolog 1) which was 5-fold up-regulated; (3) The results of real time RT-PCR, using beta-actin as an internal control, showed that the 2-Delta Ct values of DKK1 in HCC, PCT and NLT were 0.089 504, 0.007,65 and 0.000,631 respectively.
CONCLUSION(1) The high throughput and effective Agilent oligo microarray can screen novel therapy targeted genes by analyzing the DEGs in development and progression of HCC; (2) The development and progression of HCC is a complicated process involving multigenes and multiprocedures; (3) DKK1, as a novel gene, is involved in the development and progression of HCC and may be a new therapy target.

10.The correlation analysis between the employed nurse' emotional intelligence and the strategies of emotional work

Objective To investigate the relationship between the employed nurse' emotional intelligence and the strategies of emotional work and to provide effective measures for the nurses' managers. Methods 286 employed nurses were investigated by using emotional intelligence scale and emotional work questionnaire. Results Dealing with the emotional work,most of the employed nurse used natural acting(3.741 ±0.632),the second was deep acting(3.436 ± 0. 634), and the least was surface acting ( 2. 634 ± 0. 548); the natural acting score and the deep acting score were significantly different for the nurses on different levels of emotional intelligence(F = 23. 658, P < 0. 001; F = 12. 705, P < 0. 001 ) ; Correlation analysis showed that: emotional intelligence had positive relationship with natural acting and deep acting, while had no obvious relationship with surface acting. Conclusions To improve the employed nurse' emotional intelligence can increase the using of natural acting and deep acting.
