1.Effect of recombined human epidermal growth factor combined with alprostadil on Wnt/β-catenin signal pathway in rats with diabetic ulcer

Objective To explore the effect of recombined Human epidermal growth factor (rhEG) combined with alprostadilon Wnt/β-Catenin signal pathway in rats with diabetic ulcer. Methods Rats with diabetic ulcer were randomly divided into four groups :control group ,rhEG group ,epidermal group and combined treatment group. Ulcer skin was smeared by normalsaline in control group ,and by rhEG in rhEG group. Epidermal was administered from caudal vein in epidermal group.Combined treatment group was treated by rhEG and epidermal at the same time. The healing status were observed. The proteinand mRNA expression of Wnt-1 ,β-Catenin and GSK-3β were measured 14 days after treatment. Results The healing rates in combined treatment group were (9.76 ± 2.37 )% ,(35.74 ± 3.65 )% ,(51.37 ± 4.16)% and (84.42 ± 5.35 )% respectivelyin 6th , 10th and 14th day after treatment , which were significantly higher than in rhEG group and epidermal group (P<0.05). The mRNA levels of Wnt-1 ,β-Catenin and GSK-3βin combined treatment group were (1.42 ± 0.19) ,(1.56 ± 0.21) and (0.95 ± 0.15) after treatment for 14 days ,which were significantly higher than in rhEG group and epidermal group (P<0.05). Protein levels of Wnt-1 ,β-Catenin and GSK-3βin combined treatment group were (1.17 ± 0.16) , (1.38 ± 0.18 ) and (0.81 ± 0.13 ) after treatment for 14 days ,which were significantly higher than in rhEG group and epidermal group ( P < 0.05 ). Conclusion rhEG combined with epidermal can significantly accelerate the healing of diabetic ulcer ,and can regulatethe Wnt/β-Catenin signal pathway by increasing the expression of Wnt-1 ,β-Catenin and GSK-3β.

3.Clinical significance of detecting the NKT cell content and its surface receptors NKG2A and NKG2D in the peripheral blood of patients with esophageal carcinoma

Objective NKT cells are very important as a kind of non-specific immune cells. Much attention in antitumor significance has been received in the study of its effect on malignant diseases. The aim of this study was to detect the expression of NKT cells and its CD+8 NKT subsets in peripheral blood of esophageal patients and normal person, and to analyze the changes in the expression of NKG2D and NKG2A receptorsand its clinical pathological factors. Methods By flow cytometric analysis, 53 patients with esophageal carcinoma and 39 normal controls were analysed for peripheral blood of NKT cells and CD+8 NKT subsets, and the expression of NKT cells NKG2A and expression of NKG2D receptor. The clinical pathological factors were collected for the comparative analysis. Results Compared with the normal control group, the expression of NKT cells in peripheral blood of esophageal patients increased [(4.32±0.73) %, (5.97±1.29) %] (t =3.562, P <0.01), and the expression level of its surface receptor NKG2D reduced [(17.56±5.92) %, (15.12±1.56) %] (t =3.892, P <0.05), but the express levels of NKG2A [(4.02±1.41) %, (5.99±4.59) %] in creased (r = 4.015, P <0.05), those expression change with the development of the esophageal cancer. Conclusion The increased expression of NKT cells and CD+8 NKT subsets in the peripheral blood of patients with esophageal carcinoma refletcs that the immune feedback of patients' antineoplastic effect is strengthened. The decresed expression of the active receptor NKG2D and the increased expression of the inhibitory receptor NKG2A on NKT cells might be one of mechanisms leading to the reduction of NKT cell activity and immune escape of patients with esophageal carcinoma. The changes of surface receptors of NKT cells may be associated with the development of the esophageal cancer.
4.Numerical simulation of chemico-biological particles transportation inside human micropassage

Objective To investigate the transportation of chemico-biological particles(CBP) through the micropassage inside the human body in order to improve chemico-biological protection.Methods Dissipative particle dynamics (DPD) method was used to study CBP transportation through micropassages inside the human body.Results The Poiseuille flow could be ensured by imposing boundary conditions including pressure gradient and no-slip.The axial velocity between fluid particles and CBPs was well matched except the area close to the passage wall.However, CBPs tended to accumulate and the density of CBPs slightly increased, leading to the jam effect and producing particle accumulation.Conclusion The characteristic of CBP transportation is better understood,which can help develop some chemico-biological protection devices according to movement of CBPs and improve the performance of CBPs during chemico-biological protection.

6.Evaluation of left ventricular systolic hemodynamics in patients with hypertrophic cardiomyopathy using vector flow mapping

Objective To investigate the left ventricular(LV) systolic hemodynamics and the distribution of blood flow in patients with hypertrophic cardiomyopathy(HCM) by vector flow mapping(VFM).MethodsThirty-five random non-obstructive HCM patients(case group) and forty healthy volunteers (control group) were enrolled.Peak systolic velocity (Vs),peak systolic flow (Fs) and total systolic negative flow (SQ-) of two groups were compared.Velocity vector,streamline and vortex distribution of two groups were observed.Results Vs,Fs,SQ- of the control group and Vs,Fs of the case group all showed a decreasing trend from the basal segments to the apical segment(P<0.05).SQ- of the case group lost this decreasing tendency.Vs,Fs of all segments and SQ- of the middle and apical segment of the case group were higher than those of the control group.SQ- of the basal segment of the case group were significantly lower than that of the control group(P<0.01).Distribution of blood flow:The velocity of blood flow was faster in the case group than that in the control group and the direction was disordered.The streamlines of the case group were discontinuous,and vortexes in the case group were more than that in the control group.Conclusions VFM technology could quantitatively evaluate LV systolic hemodynamics in patients with HCM,and can clearly show the distribution of blood flow.
7.Changes in cerebral hemodynamics in different body positions in healthy volunteers

Objective To investigate the changes in cerebral hemodynamics in different body positions in healthy volunteers.Methods Sixty right-handed healthy male volunteers,aged 22-26,height 167-178 cm,weighing 51-67 kg,were randomly divided into 4 groups ( n =15 each): 15 degrees head-down tilt group (group Ⅰ ),30 degrees head-down tilt group (group Ⅱ ),15 degrees head-up tilt group (group Ⅲ) and 30 degrees head-up tilt group (group Ⅳ ).Blood flow signals of right middle cerebral artery (MCA) and extracranial internal carotid artery (EICA) were detected by transcranial Doppler and systolic blood flow velocity (Vs),diastolic blood flow velocity (Vd ),mean blood flow velocity (Vm),pulsatility index (PI) and resistance index (RI) were recorded at supine position (baseline),immediately,10 and 30 min after body position change(T1-3 ).Lindegaard index was calculated.Results Compared with the baseline,in group [Ⅱ Lindegaard index was increased at T2,while Vs and Vm of MCA were decreased at T2,3,in group Ⅳ Vs of MCA and PI of EICA at T2,Vd and Vm of MCA at T2,3 were decreased ( P < 0.05),while there was no significant difference in the variables mentioned above in the other two groups ( P > 0.05 ).Conclusion In healthy adults,cerebral blood flow velocity decreases in 30 degrees headdown and head-up tilt positions,however there is no change in cerebral blood flow velocity in 15 degrees headdown and head-up tilt positions.

Objective To investigate the effect of adenovirus-bone morphogenic protein 9 ( Ad-BMP9 ) on osteogenic differentiation of immortalized calvarial mesenchymal progenitor cells ( iCALs ) .Methods iCALs were infected with adenoviral vectors encoding BMP-9 or green fluorescent protein ( GFP) and the early osteogenic differentiation was assessed by detecting alkaline phosphatase (ALP) activity after being cultured for 3, 5 and 7 days.14 days after infection, alizarin red S staining was performed to study the formation of osteogenic calcium nodules .The expression of osteogenic marker genes Runx2 and OCN was assessed by quantitative real-time ( RT )-PCR and Western blotting .Results Significant increases in ALP activity and in the expressions of Runx 2 and OCN were detected in BMP-9 treated iCALs compared with GFP-treated cells(P<0.05).Meanwhile, alizarin red S staining showed that more mineralized nodules were found in the BMP-9 induced group .Conclusion BMP-9 can promote the osteogenic differentiation of iCALs .

10. Comparison on therapeutic effect of stent implantation and drug treatment for young cerebral infarction combined with middle cerebral artery stenosis

Objective: To compare the effect of stent implantation and drug treatment for young cerebral infarction combined with middle cerebral artery stenosis. Methods: A total of 71 young cerebral infarction patients (aged 30-44 years) with severe stenosis of middle cerebral artery (stenosis rate >70%) were enrolled and divided into drug group (n=39, received conventional oral medication) and stent group (n=32, underwent stent implantation). The therapeutic effects and follow-up outcomes during 24 months were compared between 2 groups. Results: The success rate of stent group was 100%, and 32 stents were implanted. Significant differences were found between the scores at enrollment and those at 3, 6, 12 and 24 months of modified Rankin scale (mRS) and National Institutes of Health Stroke Scale (NIHSS) within both the two groups during the follow-up (all P<0.05), while there was no statistically significant difference of mRS nor NIHSS scores at any comparison between 2 groups (all P>0.05). Within 24 months' follow-up, the incidence of end-point event was 20. 51% (8/39), the stroke recurrence rate was 20.51% (8/39), and the disability rate of recurrence stroke (mRS score ≥ 2 points) was 20. 51% (8/39) in drug group, while in stent group was 6.25% (2/32), 3.13% (1/32) and 3.13% (1/32), respectively. There was no statistically significant difference of the incidence of endpoint events between 2 groups (P=0.17), while the recurrence rate of stroke (P=0.04) and the disability rate of recurrent stroke (P=0.04) in stent group were both lower than those in drug group. Conclusion: Compared with drug therapy, stent implantation can reduce stroke recurrence rate and disability rate of in young cerebral infarction with middle cerebral artery stenosis.
