1.Inspiratory and expiratory high resolution CT lung findings in normal adult subjects

Objective To evaluate inspiratory and expiratory high resolution CT (HRCT) lung findings in normal adults and set up inspiratory and expiratory HRCT criterion for further clinical diagnosis and differentiating diagnosis. Methods Sixty-four healthy adults underwent inspiratory and expiratory lung HRCT scanning. The lung density and slice area in upper, middle and lower fields were measured in inspiratory and expiratory phases respectively. Results The lung density in inspiratory phase was (-795?58)HU, the lung density in expiratory phase was (-706?59)HU, the inspiratory expiratory value difference was (90?43)HU, and the expiratory inspiratory area ratio was 0.72? 0.14. Conclusion HRCT will be a new method for lung function testing in future.
2.MRI Signal Measurement of Hippocampal Formation

Objective To define the range of the normal values of MRI signals of hippocampal formation(HPF) for the diagnosis of early stage Alzheimer's disease and hippocampal sclerosis.Methods MRI signals of 254 normal adults were measured on the transverse section.Results HPF signal intensities were:T1 relaxation times,629±73 ms;T2 relaxation times,83±5.5 ms;and proton density value,5978±651.The T2 value range(χ±2S)was 72~94 ms,and mean 83 ms.T2 value greater than 104 ms will be associated with evidence of hippocampal pathologic changes.The mixed signal intensity of T2WI(30,60,120)were:3907±407,2657±347,1288±174.The signal intensity ratios of T2WI were:1.02,1.07,1.13.Conclusion As for the histological features of temporal lobe cortex and HPF,T1relaxation times and proton density values are matched but T2 relaxation times of HPF are relatively longer.
3.Anti-tumor effects of p53N15-based fusion peptide in the transfected H1299 lung cancer cells

ObjectiveLoss-of-function mutation of p53,a tumor suppressor gene,is an important mechanism for the development of human cancers.In this study we tried to transfect p53N15-based fusion peptide into H1299,a lung cancer cell line,and evaluate the anti-tumor effects of the fusion peptide.MethodsAdeno-associated virus ( AAV) vectors were used for transfecting p53N15 fusion peptide into p53-null lung adenocarcinoma H1299 cells.The anti-replication effects of p53N15 fusion peptide were evaluated with inverted microscopy,MTT test for cell viability and flow cytometry.ResultsFusion peptides in H1299 cells was highly expressed and had detectable suppressive effects on cell proliferation.A large amount of dead cells were seen under microscope after the transfection of recombinant viruses for 72 hours.Cells activity was reduced significantly in the virus-transfecting groups as demonstrated by MTT test.The flow cytometry showed that a large number of dead cells were present in the virus-transfecting groups.ConclusionThe growth of H1299 lung adenocarcinoma cells could be inhibited in vitro by being transfected with p53N15 fusion peptide,which may be a potential gene therapy alone or as an adjuvant option in the treatment of lung cancer.
4.ADENO-ASSOCIATED VIRUS INTRODUCED INTEGRATION AND EXPRESSION OF FOREIGN GENES IN PC12 CELLS

Objective To investigate integration and expression of adeno-associated virus (AAV) vectors in neuronal PC12 cells,the result of which can be applied in further gene therapy of diseases of the central nervous system. Methods Human neurotrophin-3(hNT3)genes were inserted into AAV vectors. Then the recombinat AAV plasmids were encapsidated as recombinant virions. PC12 cells were transfected with the virions and the positive cells were selected by G418. The transfection positive (hNT3 modified)PC12 cells were cultured for several generations and the cellular genomic DNA and total RNA were extracted. We investigated the integration locus of AAV vectors by Southern blot and transcript situation of foreign genes by dot blot. Results The hybridization tests showed that AAV introduced foreign genes were stably integrated, but at random locus, and robustly transcribed in hNT3 modified PC12cells. Conclusion AAV vectors can serve as high efficiency vectors of target genes in neuronal PC12 cells.
5.Influences of different flow rates of high pressure injector on hysterosalpingography

Objective To analyze the contrast agent back flow,balloon ejection and pain which appeared during the examination process,and define the advantages and disadvantages of different rates of high pressure injector to take hysterosalpingography (HSG) examination.Methods 229 patients who took HSG examination in our department were divided into two groups randomly.The rate of conventional group was 0.3 mL/s,and low rate group was 0.15 mL/s.The contrast agent back flow,balloon ejection and pain which appeared on the examined patients of two groups were recorded.T test was adopted to compare the age difference of two groups.χ2 test was used to compare the differences of diagnosis results,contrast agent back flow and balloon ej ection of two groups.Wilcoxon rank sum test was used to compare the pain difference of two groups.Results Two groups had no significant differences on age, diagnosis results and contract agent back flow (t=1.237,χ2=0.004,χ2=1.521,P>0.05).Balloon ejection of two groups had significant difference(χ2=17.319,P<0.05).The appeared pain of two groups had significant difference (U=-2.337,P<0.05).Conclusion Using high pressure injector to inject contrast agent with low flow rate is better for HSG.
6.Analysis of multi spiral CT features of acute blunt or penetrating intestinal injury

Objective To analyze the CT signs of acute blunt or penetrating intestinal injury,and to improve the diagnostic accuracy of multi spiral CT for intestinal injury.Methods The CT and clinical data of 63 patients with intestinal injury confirmed by clinical surgical exploration who underwent emergency CT scan were collected,and the CT findings and surgical findings were comparatively analyzed.Results There were 63 cases of intestinal injury,of which 26 cases were complicated with mesenteric injury.The direct CT signs of intestinal injury included intestinal wall thickening sign and intestinal discontinuity sign,which accounted for 64%(40/63)and 17%(10/63),respectively.The indirect CT signs of intestinal injury included intraperitoneal/retroperitoneal gas sign,intraperitoneal/retroperitoneal effusion sign,intramural air,and portal venous gas,which accounted for 72%(45/63),88%(55/63),7%(5/63)and 5%(3/63),respectively.Conclusion Recognizing the CT signs of intestinal injury,such as intestinal wall thickening sign,intestinal discontinuity sign,intraperitoneal/retroperitoneal gas sign,intraperitoneal/retroperitoneal effusion sign,intramural air,and portal venous gas can help to make the early and correct diagnosis of intestinal injury if combined with clinical practice.
