1.The effect of Rad001 and ATO on activation of the excessive autophagy and apoptotic pathways in SKOV3 ovarian cancer

Objective To investigate the synergistic anticancer effect of ATO combined with Rad001 on human ovarian cancer cells in vitro and the underlying mechanisms. Methods SKOV3 cells were treated with Rad001, ATO and a combination of Rad001 and ATO, respectively. Cell relative luminescence units, viability, and combination index were tested. The apoptosis was quantified by Flow cytometry. Autophagy and apoptosis associated proteins were measured with immunoblotting. Results Significant decrease in live cell number were observed in cells treated with combination of Rad001 and ATO , compared with single compound treatment (P <0.05). Moreover, a higher rates of autophagy as well as apoptosis were observed in the combination treatment compared with single compound treatment (P < 0.05). Conclusions combination of Rad001 and ATO can result in synergistic cytotoxicity through activation of the excessive autophagy and apoptotic pathway.
2.Outcome analysis of endemic fluorosis control in Guangxi in 2006

Objective To investigate the distribution of water fluoride and the status of water-improving defluoridation projects,thus to explore the condition of endemic fluorosis in Guangxi Province.Methods According to"The National Technical Scheme for Endemic Disease Contml in 2005",the fuorine content in water Was determined by F-ion selective electrode,children's dental fluorosis was checked by Dean method.and the skeletal fluorosis was checked by the standard of clinical scale of skeletal fluorosis.Results 305 water samples in 61 villages were examined,among which 71 waters were exceeded the standard,accounting for 23.28%(71/305).The projects of defluoriding drinking water were running well except one was discarded.The prevalence of dental fluorosis was 13.55%(356/2627),the prevalence of skeletal fluorosis was 4.02%(65/1615).Conclusions The situation of endemic fluorosis control is not optimistic in Guangxi,which needs fuaher prevention and controls.
3.Lipoprotein lipase expression in the hippocampus and its effects on vitamin E levels in rats with epilepsy.

OBJECTIVETo investigate the dynamic changes of lipoprotein lipase (LPL) expression in the hippocampus of epileptic rats and to study its effect on vitamin E levels in rats following status epilepticus (SE).
METHODSRat model of SE was induced by intraperitoneal injection of pilocarpine. The rats receiving an injection of normal saline were used as a control group. The expression of LPL in the hippocampal tissue was determined using immunofluorescent methods and the level of vitamin E was examined by the colormeric method 12 hrs, 24 hrs, 3 days, 7 days and 14 days after SE.
RESULTSLPL was expressed in the control and SE groups. In the SE group, the LPL expression began to increase 24 hr after SE (P<0.05), reached a peak 3 days after SE (P<0.01), and kept at a high level 7 days after SE (P<0.01). By 14 days, the LPL expression was reduced to the level similar to the control group. The level of vitamin E began to decline 12 hrs after SE (P<0.01), and decreased to a nadir 24 hrs after SE (P<0.01). At 3 and 7 days after SE, the levels of vitamin E were still significantly lower than the controls (P<0.05). By 14 days, the vitamin E level increased to the level similar to the control group.
CONCLUSIONSThe over-expression of LPL in the hippocampus may play an important role in the oxidative stress mechanisms following SE by regulating the uptake of vitamin E.

4.Clinical study of topography-guided off-flap Epi - LASlK in treating traumatic corneal astigmatism

To evaluate the visual performance of the patients with traumatic corneal astigmatism, after the treatment of topography guided off-flap epipolis laser in situ keratomileusi (off-flap Epi-LASlK).
●METHODS: This prospective clinical study was comprised of 21 eyes of 21 patients with irregular corneal astigmatism caused by trauma, they were treated by off-flap Epi - LASlK from July 2012 to December 2013. The data included uncorrected visual acuity ( UCVA), best spectacle - corrected visual acuity ( BSCVA ), contrast sensitivity 1, 6mo before and after surgery; the healing area percentage of corneal epithelia, the healing time of corneal epithelia and pain score at 3d after surgery.
●RESULTS: Postoperative 1mo both UCVA and BSCVA were improved significantly than that before surgery (t =15. 703, 4. 351, P< 0. 05); Compared with the 1mo after surgery, UCVA at 6mo after surgery raised significantly (t= 6. 867, P <0. 05). There was no statistical significance between 6 and 1mo after surgery about BSCVA (t= 1. 497, P = 0. 140 ). After surgery, mean spherical equivalent (SE) was reduced from -2. 43±3. 02D to -0. 23±0. 49D (P<0. 05), and the mean cylinder was reduced from -1. 86± 2. 23D to - 0. 46 ± 1. 03D (P< 0. 05). Postoperative 1mo,4 kinds of spatial frequency and contrast sensitivity had no significant difference compared with the preoperative (P>0. 05 ). Postoperative 6mo except the 3c/ d spatial frequency, the remaining 3 spatial frequency contrast sensitivity compared with those before operation were significantly improved ( P < 0. 05 ). The healing area percentage of corneal epithelia was 92. 46% ±8. 24% (80% -100%) at 3d after surgery; The healing time of corneal epithelia was 3. 50 ± 1. 56d; Pain scores at 3 and 7d after surgery was 1. 54±1. 32 and 0. 04±0. 64, respectively.
●CONCLUSlON: Topography-guided off-flap Epi-LASlK is safe and effective in treating the patients with traumatic corneal irregular astigmatism. The operation can improve both the contrast sensitivity and the visual performance.
5.Phenolic components from Rhodiola dumulosa.

Eight phenolic compounds (1-7) were isolated from the methanol extract of the root of Rhodiola dumulosa. Their structures were identified as kaemperol (1), Quercetin (2), Gallic acid (3), (+) -Isolariciresinol-3alpha-O-beta-D-glucopyranoside (4), (-)-Isolariciresinol-3alpha-O-beta-D-glucopyra-noside (5), kaemperol-3-O-beta-D-glucopyranoside-7-alpha-O-L-rhamnoside (6), rutin (7) respectively on the basis of chemical and spectroscopic evidence. The compounds 3-7 were isolated from R. dumulosa for the first time.

6.Study on steroids of Cacalia tangutica.

Seven steroids and two cumarins were isolated from the petroleum ether extract of the specie Cacalia tangutica of the family Compositae which were collected in Minhe county, Qinhai province of China. The structures were identified as Stigmast4-en-3beta, 6beta-diol (1), 24-ethyl-5alpha-cholestane-3beta, 5, 6beta-triol (2), 7beta- methoxy-stigmast-5-en-3beta-ol (3), Schleicherastatin 1 (4), Stigmast-5-en-3beta, 7alpha-diol (5), umbelliferone (6) and hydrangetin (7) by the means of chemical and modern spectroscopic analysis (MS, 1H-NMR, 13C-NMR, DEPT). The compounds 1-5 were isolated from Cacalia tangutica for the first time.

7.Experimental Study of Adrenomedullin in Autoimmune Myocarditis Induced by Immunization of Mice with Lactobacillus Casei Cell Wall Element

Objective To study adrenomedullin (AM) mRNA and protein expression level in myocardium of autoimmune myocarditis animal models induced by immunization of mice with lactobacillus casei cell wall element(LCWE). Methods Forty-five Balb/c male mice were randomly divided into experimental group (n = 30) and control group (n = 15), which were intraperitoneally injected with LCWE and phosphate buffered solution(PBS) at day 0,3,5 and 10,respectively. Sera and myocardium samples were gained 14,21 and 28 days after the first immunization. AM expression levels were determined by semiquantitative reverse transcriptase-polymerase chain reaction(RT- PCR) and immunchistochemistry,and mycardial histopathological lesions were observed. The anti- myosin antibodies in different stages were examined by an ELISA. Results There were myocardial necrosis or inflammatory infiltration in the experimental group, but myocardial lesions were not found in the control group. Anti - myosin antibodies were detected in sera of experimental mice,but not in control group. 9.Effects of Fructose,Glucose and Exogenous Nitric Oxide Donor Sodium Nitroprusside on Germination of Psychotria rubra(Lour.)Poir. Seeds Under Drought Stress

Objective To observe the effect of fructose(Fru),glucose (Glc)and exogenous nitric oxide donor sodium nitroprusside(SNP)on the germination of Psychotria rubra(Lour.)Poir. seeds under drought stress,and to explore the optimal concentration for clearing the drought stress. Methods We established the drought stress model of Psychotria rubra(Lour.)Poir. seeds with 15%(volume fraction)PEG6000 and 0.4 mol/L D-mannitol, measured the germination rate and germination index of Psychotria rubra (Lour.)Poir. seeds under drought stress combined treatment with different concentrations of Fru (0,300,500 μmol/L),Glc (0,300,500 μmol/L) and SNP(0 , 100 , 200 μmol/L). Results The germination process of Psychotria rubra(Lour .) Poir . seeds was significantly suppressed by drought stress. Fru,Glc and SNP could significantly promote the germination of Psychotria rubra (Lour.) Poir. seeds under drought stress. The optimal condition for clearing drought stress induced by 15% PEG6000 was Fru 300 μmol/L, Glc 500 μmol/Land SNP 200 μmol/L, and the germination rate was 95% under the optimal condition . The optimal condition for clearing drought stress induced by D-Mannitol was Fru 500 μmol/L, Glc 500 μmol/L and SNP 100 μmol/L, and the germination rate arrived to100%. Conclusion Fru,Glc and SNP can significantly alleviate the negative impact induced by drought stress for the Psychotria rubra(Lour.)Poir. seed germination,improve the drought-resistant ability of the seeds.
10.Research on inorganic elements of Asari Radix et Rhizoma from different localities and species.

OBJECTIVETo explore the character of inorganic elements in Asari Radix et Rhizoma (Xixin).
METHODThe contents of 53 inorganic elements in Xixin samples from different localities and species were determined by ICP-AES and ICP-MS. The statistical data were made using SAS.
RESULTThe result demonstrated that Xixin has the high contents of Fe, Cr, Li. It has been observed that the content of Cu and Pb of the samples are much higher than the standard level. The results of hierarchical cluster analysis revealed two groups which correspond with the species of the samples. No correlations between the contents of the inorganic elements and the localities of the samples were found. Some characteristic elements were displayed in some specific areas. The difference of the contents of the 53 inorganic elements between root and rhizome of Xixin was reported for the first time. The primary form of inorganic elements in Xixin has been studied for the first time. The result demonstrated that the extraction rate between different elements varied, with the average extraction rate of (22.25 +/- 24.96)%.
CONCLUSIONThe inorganic elements analysis of Xixin can provide evidence of its identification, cultivation and application.
