1.Hair follicle stem cells promote the healing of skin wound

BACKGROUND:Hair fol icle stem cel s derived from skin and hair have the most readily available sources, and cannot lead to serious complications and immunogenicity after col ection, which can be used for autologous transplantation. OBJECTIVE:To provide effective experiment basis for hair fol icle stem cel s in the repair of skin wound through reviewing the role of hair fol icle stem cel s in promote the repair of skin wound. METHODS:Articles related to hair fol icle stem cel s for the repair of skin wound were retrieved in CNKI and PubMed databases (1999-01/2014-12). The key words were“hair fol icle stem cel , skin repairing, regenerative medicine, tissue engineering”in Chinese and English, respectively. A total of 45 articles based on inclusion criteria and exclusion criteria were obtained for the review. RESULTS AND CONCLUSION:Hair fol icle stem cel s belong to adult stem cel s, which are easy to access, have a significant number, and possess differentiation potential and safety. Hair fol icle stem cel s can promote the repair of skin wound and provides good seed cel s for regenerative medicine and tissue engineering research through early vascularization of the skin, epidermal and appendage regeneration, signaling pathways, and transcription factors.
2.Study on microsatemte instability of p14,p15,p16 gene in gastric carcinoma

Objective To detect the microsatellite instability (MSI) of D9S166,D9S171,D9S941, D9S942 and IFNA located at chromosome 9p21 in gastric carcinoma with high-grade intraepithelial neoplasia and normal tissue,and investigate the correlation of p14,p15,p16 gene and the gastric carcinogenesis. Objective To investigate the clinical significance of soluble CD40 ligand (sCD40L ) and lipoprotein associated phospholipase A2 (Lp-PLA2 ) in the assessment of coronary artery severity and risk classification in patients with non-ST-segment elevation acute coronary syndrome (NSTE-ACS).Methods Of the 9 6 patients with coronary heart disease diagnosed by coronary angiography,2 8 patients had stable angina pectoris (SAP),38 patients unstable angina pectoris (UAP)and 30 patients acute non-ST-segment elevation myocardial infarction (NSTEMI).Another 30 patients with non-coronary heart disease (NC)served as controls.The sCD40L and Lp-PLA2 levels were determined by enzyme-linked immune sorbent assay (ELISA)method.The Gensini score was used to assess the severity of coronary artery and analyze the correlation with sCD40L and Lp-PLA2.The correlation of sCD40L and Lp-PLA2 with GRACE risk score was analyzed too.Results ① sCD40L was significantly higher in NSTEMI and UAP groups than in SAP and NC groups (P<0 .0 5 ),but there was no significant difference between NSTEMI and UAP groups (P>0 .0 5 )or SAP and NC groups (P>0 .0 5 ).Lp-PLA2 was significantly higher in NSTEMI group than in UAP,SAP and NC groups (P<0.05).Lp-PLA2 was significantly higher in UAP group than in SAP and NC groups (P<0.05).② We found that sCD40L had obvious correlation with Lp-PLA2 (r=0.284, P<0.01),Gensini score (r=0.213,P<0.05),and GRACE (r=0.224,P<0.05).Lp-PLA2 was significantly correlated with Gensini score (r=0.270,P<0.05),and GRACE (r=0.323,P<0.01).③ Multivariate logistic regression analysis showed that Lp-PLA2 was independently associated with NSTE-ACS (P<0.05).Conclusion The sCD40L and Lp-PLA2 which were significantly elevated in NSTE-ACS are correlated with the severity of coronary artery disease.The two indexes indicate the instability of atherosclerotic plaque;thus they can be used as predictors of risk assessment in coronary heart disease.
5.Relationship of CD31+/CD42b- and CD144+ endothelial micropaticles with intima-media thickness of carotid artery

Objective To detect the levels of CD31+/CD42b- and CD144+ endothelial microparticles (EMPs)in plasma,and analyze the relationship with carotid intima-media thickness (CIMT)and the risk factors of atherosclerosis.Methods Totally 122 outpatients and inpatients in our hospital were selected,and carotid artery ultrasound examination was performed.The results were divided into a thickening group (62 cases)and a control group (60 cases).Flow cytometry was used to detect the levels of CD31+/CD42b- and CD144+ EMPs in the blood to analyze the relationship between the level of endothelial cells and CIMT,and to investigate the relationship between the level of plasma EMPs and the risk factors of atherosclerosis.Results Compared with the control group,the levels of EMPs of CD31+/CD42b- and CD144+ were significantly higher.In the correlation analysis, plasma CD31+/CD42b- and CD144+EMPs levels were positively correlated with CIMT;in multi-factor Logistic regression analysis,CD31+/CD42b- EMPs levels were correlated positively with age,TC and Cys C;plasma CD144+ EMPs levels were correlated with UA and BMI.Conclusion The levels of CD31+/CD42b-,CD144+EMPs in plasma were correlated with CIMT,guiding early prevention of cardiovascular diseases.

7.Enhancement of immune responses to hepatitis B DNA vaccine by superantigen SEA in mice.

To investigate the adjuvant effect of plasmid DNA encoding superantigen SEA (D227A) (pmSEA) on immune responses induced by HBV DNA vaccine containing HBV preS2 and S antigen in BABL/c (H-2d). BALB/c mice were immunized intramuscular injection with HBV DNA vaccine (pHBVS2S) mixed with or without pmSEA plasmid. Antibodies againat HBV PreS2 and S antigen in the sera were accessed by Anti-HBs ELISA, and the HBsAg specific cytotoxic T lymphocytes (CTLs) activity was determined by 5 Chromium Release Assay. The HBs peptide-specific IFN-gamma secreting T cells were detected by ELISPOT. Anti-HBs antibody titers and CTLs activity in mice immunized with pmSEA + pHBVS2S group were significant higher (P < 0.05) than pHBVS2S DNA vaccine group. The ratio of IgG1/IgG2a (0.282) was apparently different from the group immunized with peptide (10). Mice immunized with HBV DNA vaccine plus adjuvant produce higher titer of IgG1 and IgG2a antibodies against HBV S antigen 1.36 and 1.73 time higher than that without adjuvant respectively. HBs peptide--specific IFN-gamma secreting T cells increased 2 - 3 times by the pmSEA adjuvant, compared to DNA vaccine group. HBV DNA vaccine (pHBVS2S) induces humoral and cellular immuno-responses in BALB/c mice, and the responses could be significantly boasted by the plasmid encoding mSEA. Therefore the pmSEA was a potential adjuvant for DNA vaccines.

8.Clinical value of (18)F-FDG positron emission tomography-computed tomography in local liver neoplasm ablation.

OBJECTIVETo assess the value of (18)F-fluoro-2-deoxy-D-glucose positron emission tomography-computed tomography ((18)F-FDG PET-CT) in ultrasound-guided local ablation of malignant liver tumors.
METHODSThirteen patients with 35 local residual tumor foci following previous tumor ablation underwent (18)F-FDG PET-CT and ultrasound-guided local ablation with intratumoral alcohol injection.
RESULTSAfter the second local ablation guided by (18)F-FDG PET-CT and ultrasound, radioactive defects were detected in the corresponding location in 31 of the 35 residual foci, and after the third local ablation, the other 4 foci also showed radioactive defects.
CONCLUSION(18)F-FDG PET-CT can sensitively and accurately identify tissue necrosis and residual tumors, and serves as an excellent approach for ultrasound-guided local ablation of local residual tumors.

9.Allogeneic stem cell transplantation for 8 patients with malignant infantile osteopetrosis in China.

OBJECTIVEOsteopetrosis is a rare genetic disorder and the malignant infantile osteopetrosis (MIOP) is the worst subtype of this disease. Seventy percent of patients die in six years of life without proper treatment. Hematopoietic stem cell transplantation (HSCT) offers the only chance of cure for MIOP.
METHODRetrospective analysis was performed on 8 patients with MIOP who underwent HSCT in Beijing Children's Hospital during the period from 2006 to 2011.
RESULTEight cases (4 male and 4 female, mean age at HSCT 13.5 months) were diagnosed as malignant infantile osteopetrosis. Conditioning regimen included fludarabine, busulfan and cyclophosphamide. All patients received cyclosporin for prophylaxis of graft vs. host disease (GvHD). A UMD recipient underwent CD34(+) cell selection. ATG/ALG, mycophenolate mofetil (MMF) and methotrexate (MTX) used for recipients with unrelated cord donor (2) and recipients with haplo-identical donors (5). Average time for neutrophil engraftment was 15.7 day (9 - 36), platelet engraftment was 43.3 day (10 - 68). The patients were followed up from 47 days to 5 years, 1 patient died of post-transplant complications. Seven cases presented better in clinical manifestation. Acute GvHD I°-II° was observed in 6 patients, III°-IV° in 2 patients. It was controlled by anti-GvHD therapy.
CONCLUSIONNon-allogenic stem cell transplantation treatment of infantile MIOP showed high survival rate and restoration of hematopoiesis in haploid transplant patients, therefore, non-allogenic HSCT may be an option to treat MIOP in children.

10.Effect of hypertension on cerebral aneurysms and model production of cerebral aneurysm in a rat

Objective To establish a rat model of cerebral aneurysms and explore the impact of hypertension on it. Methods Forty-five rats were randomly divided into hypertensive, normotensive, and normal control groups (n=15). The origin of the right external carotid artery was digested with porcine pancreatic elastase, ligated and cut at 1.5 mm from the carotid bifurcation. Cerebral aneurysm models were established in this stump in the hypertensive and normotensive groups, and then, stenosis of the bilateral renal artery was induced surgically in the hypertensive group. The systolic blood pressures in the 3 groups were measured before and at 6 and 12 weeks after the operation. At 12 weeks after the operation, the aneurysms, after measurement of the size, were fixed by formalin perfusion. The vascular tissues in the aneurysms were isolated for pathological examination using HE staining, Van-Geson staining and Verhoeff staining. Results A significant increase in the blood pressure occurred in the hypertensive group to the level of 197.48±15.44 mm Hg, and the aneurysm was obviously enlarged measuring 2.38±0.31 mm in length and 1.89±0.35 mm in width;no significant changes occurred in the normotensive and normal control groups. Pathological examinations demonstrated the absence of the intima and rupture of the elastin layer in the hypertensive group;the normotensive group retained part of the elastic fibers, and no vascular damage was found in the normal control group. Conclusion This aneurysm model can be prepared conveniently with good stability, and can well simulate the structure and pathology of human cerebral aneurysm. Hypertension significantly affects the elastin and collagen in the blood vessels to contribute to aneurysm enlargement.
