Objective:To observe the effect of He-Ne laser combined with ammonium glycyrrhetate in the treatmnet of 62 patients.　Methods:62 cases of patients with psoriasis vulgar were divided into two groups. 36 cases (A group) received He-Ne laser combined with ammonium glycyrrhetate intravenous infusion, 26 cases(B group) received only ammonium glycyrrhetate. The therapy course of both groups were 30 days.　Results:The total effective rate of two groups was 82.2%. The effective rate of A group and B group were 88.9% and 73.1%, respectively. There was siginfinant difference by the analysis(ridit test)between two groups(U＝2.76，P＜0.01).　Conclusion:The effect of He-Ne laser combined with ammonium glycyrrhetate in the treatment of patients with psoriasis vulglar than that of ammonium glycyrrhetatealone.

Objective To explore the application of diffusion tensor imaging in the assessment of ischemic muscle in hind-limb ischemia model of rabbit. Methods Excision of femoral artery in unilateral hind limb was done in 14 New Zealand white rabbits and ischemic model were established in 12 rabbits. Three (12 rabbits),10(10 rabbits) ,28(7 rabbits),56(5 rabbits) days after the model establishment, DTI scan was performed on bilateral hind limbs in each of the models, respectively and, λ_1,λ_2,λ_3, ADC and FA values were measured. Histological analysis was also performed at these time points. Pared t test was used to compare the differences of these indexes in bilateral hind limbs. Results Following femoral artery excision, a rapid ascending of ADC,λ_2,λ_3 values with sharply reduced FA value was observed in ligated hind-limb, which reached maximal on 3 days post-excision (ADC_(ligated)= 1.72±0.16, ADC_(unligated)= 1.53±0.16, t = 6.48, P < 0.01 ; λ_(2 ligated)= 1.70±0.15, λ_(2 unligated)= 1.51±0.06, t=10.87, P < 0.01 ; λ_(3 ligated_ =1.17±0.12, λ_(3 unligated)= 0.88±0.12, t=6.67, P < 0.01 ; FA_(ligated)= 0.24±0.04, FA_(unligated) =0.39±0.03, t = -10.61 ,P <0.01) and histologic analysis revealed the severest muscle damag at that time. Ischemic muscle recovered very slowly during the first 10 day post-excision accompanied with reduction of ADC , λ_2, λ_3 values, however there was also difference of ADC,λ_2, λ_3 values between ligated and nonligated limbs except λ_1 ( ADC_(ligated) = 1.65±0. 16, ADC_(unligated)= 1.50±0.12, t =6.42, P <0.01 ; λ_(2 ligated) = 1.62±0.32, λ_(2 unligated) =1.48±0.31, t=5.09, P < 0.01) ; λ_(3 ligated)= 1.11±0.13, λ_(3 unligated)= 0.85±0.09, t=6.26, P <0. 01;λ_(1 ligated)=2.20±0.21, λ_(1 unligated) =2.18+0.20, t=0.87, P=0.40). After 28 days, ADC and λ_3 returned to normal (ADC_(ligated)= 1.51±0. 16, ADC_(unligated)= 1.55±0.14, t=-1.35, P=0.23 ; λ_(3 ligated) =0.95±0. 10, λ_(3 unligated)= 0.92±0.06, t=1.70, P=0.14), but λ_2 and FA of ligated limb were still different from those of nonligated limb (λ_(2 ligated)= 1.45±0.23, λ_(2 unligated)= 1.52±0. 95, t=-3.56, P=0.012; FA_(ligated)=0.35±0.02, FA_(unligated)=0.40±0.03, t=-3.83, P<0.01). After 56 days, all parameters retuned to normal(ADC_(ligated) =1.57±0.18, ADC_(unligated)=1.58±0.23, t=-0.71, P=0.52; λ_(1 ligated) =2.18±0.18, λ_(1 unligated)=2.24±0.14, t=-0.22, P=0.10; λ_(2 ligated)=1.64±0.13, λ_(2 unligated)=1.59±0.15, t=0.89, P=0.42; λ_(3 ligated)=0.89±0.1,λ_(3 unligated)=0.91±0.07, t=- 1.64,P= 0.18; FA_(ligated)= 0.39±0. 03, FA_(unligated)= 0.41±0.02, t=-0.83, P=0.47). Conclusion DTI is a quantitative and relatively accurate technique to assess time-dependent changes of ischemic muscle in hindlimb ischemia model of rabbit.

Objective To investigate the clinical and prognosis characteristics of SLE patients whose initial clinical presentations were Sj(o)gren's syndrome (SS).Methods Medical charts of 41 consecutive SS/SLE inpatients admitted to Peking Union Medical College Hospital (PUMCH) from February 1998 to February 2008 were systematically reviewed,including demographic data,clinical features,laboratory findings,treatment as well as prognosis.Two hundred and fourteen cases were randomly selected as controls from 2331 non SS-onset lupus (NSs/SLE) inpatients weated in PUMCH at the same time period.Results There were significant differences between SS/SLE and NSS/SLE patients in the following aspects (P＜0.05):①gender composition:(F/M) (41/0 vs 184/30),age at the diagnosis of SLE [(43±41) yrs vs (32±31) yrs)],disease duration L(114±84) mollths vs (45±18) months];②clinical features:xerostomia (85.3% vs 6.1%),xerophthalmia (75.6% vs 2.3%),faeial rash (9.8% vs 46.3%),renal tubular acidosis (21.9% vs 0),nephrotic syndrome (7.3% vs 31.3%),central nervous system invoivement (4.9% vs 19.6%),interstitial lung disease (12.2% vs 2.8%);③labratory findings:ESR[(65±75) mm/1 h vs (46±34) mm/1 h)],patients with elevated IgG level (56.4% vs 29.9%) and IgA level (38.5% vs 20.4%),RF,prevalence of anti-SSA and anti-SSB antibodies (70.8% vs 20.3%,82.9% vs 43.4% and 39.0% vs 7.9%);④SLEDAI score (8±8 vs 10±10),glucocorticosteroid treatment (methylprednisolone bolus/1～2 mg·kg-1·d-1 prednisone/1 mg·kg-1·d-1 prednisone)(8/26/7 vs 91/102/21),and rate of death and/or severe irreversible organ failure (2.4% vs 14.9%).SS/SLE patients were followed up for (33±34) months,40 cases remained stable and only one patient died of acute pulmonary embolism.Conclusion Compared to NSS/SLE,SS/SLE patients ale older and have more insidious disease course.They have higher prevalence rate of anti-SSA and anti-SSB antibodies,renal tubular acidosis and interstitial lung disease,but less severe neuropsychiatric and renal involvements and much better prognosis.

Objective To investigate the effects of CD40-P227A SNP at human B cell signaling and function. Methods Human Ramos B cells were transfected with plasmids of CFP fusion wild type CD40 and mutant type CD40-P227A, together with plasmids of luciferase fusion NF-κB and AP-1, as well as plasmids of GFP fusion NF-κB. Activation of NF-κB and AP-1 pathway were detected by luciferase assay and flow cytometry, degradation of IκBα and nuclear translocation of NF-κB subunits were tested by Western blot and transfactor ELISA assay. Results Compared with wild type CD40, CD40-P227A SNP induced more degradation of IκBα, increased nuclear translocation of p65, p50 and c-Rel. As well as higher activity of NF-κB, as shown by increased NF-κB luciferase and GFP-NF-κB expressing B cells. Moreover, CD40-P227A SNP induced more activation of AP-1 pathway than CD40. CD40-P227A enhanced B cells function by inducing more excretion of IL-6 and TNF-α. Conclusion Our data indicate that CD40-P227A is a gain-of-function phenotype which induces activation of NF-κB and AP-1 signaling pathways and may contribute to the pathogenesis of the SLE autoimmune disease.

OBJECTIVE To investigate the distribution and the resistance variance tendency of the pathogenic bacteria isolated from the blood cultureing specimens collected during the period of 2002-2005.METHODS A retrospective analysis was made to the blood cultureing results during the period of 2002-2005 with WHONET 5.1 software.RESULTS Gram-negative rods were the predominant bacteria which caused sepsicemia,the isolated rates of Escherichia coli,Klebsiella pneumoniae and Staphylococcus aureus were the most high during the period of 2002-2004 but the S.epidermidis and S.hominis were also the important pathogenic ones which caused blood stream infection.Vancomycin and the teicoplanin were the most effective to the Gram-positive bacteria,and the imipenem and the cefepime were the most effective to the Gram-negative ones.CONCLUSIONS It′s important to strengthen the blood cultureing for blood stream infection patient.

Objective To study the influence of quedcetin on the collagen synthesis of cultured fibroblasts and to explore the mechanism.Methods The inhibitory effect of quercetin and radiation on fibroblast proliferation was assayed using MTT assay.Collagen synthesis was detected by hydroxyproline colorimetric analysis.Immunocytochemical staining method was used to investigate the expression of collagen Ⅰ and Ⅲ.The mRNA expression of typeⅠ,type Ⅲ collagens and TGF?-1 were assayed by reverse transcriptionpolymerase chain reaction(RT-PCR) and real-time PCR technique.Results Keloid fibroblast cell proliferation and collagen synthesis of fibroblasts were inhibited by quercetin in a dose-dependent manner.Significant inhibition was observed by the treatment with quercetin and radiation together.Immunocytochemical staining indicated the IOD of type I and Ⅲ collagen protein was down-regulated by quercetin and radiation.Both collagen Ⅰ and collagen Ⅲ gene in the quercetin groups showed a significantly decreased mRNA expression compared with that in the untreated group,especially in the group treated with both quercetin and X-ray.Procollagen gene expression was inhibited and then decreased type Ⅰ and Ⅲ protein syntheses of fibroblsts,particularly type Ⅰ procollagen gene(P

Objective To explore the chromosome features of pancreatic cancer cell lines from Chinese patients. Methods G-band Chromosome karyotyping was performed on pancreatic cancer cell lines of PC1, PC2, PC3, PC4 and PC7 that were established in our laboratory. The results of cytogenetic analysis were confirmed by fluorescence in situ hybridization using Chromosome 3, 13 18 and 20 paint probes. Results All the 5 cell lines were hypotriploid and showed a modal number of 58 for PC1, 56 for PC2, 61 for PC3, 53 for PC4 and 54 for PC7 with different proportion of complex chromosome rearrangements including dicentric chromosomes, double-minute chromosomes, ring chromosomes, acentric fragments or complex chromosome translocation, etc. Conclusion Hapotriploid accompanied with complex numerical and structural chromosomal rearrangements is the main cytogenetic marker of chromosomal instability in pancreatic cancer cell lines. Molecular cytogenetic techniques have to be used beside conventional G-band karyotyping for accurate identifying abnormal chromosomes of pancreatic cancer cell lines.

Objective To establish a UV method of determination of adenosine in Ligia exotica. Methods Adenosine was extracted with 37℃ water and separated by TLC silica gel, the absorbance was detected at 260nm with UV spectrophotometry.Results The cabibration curve for adenosion was linear within the range of 1.0~20.0?g?mL -1, r=0.9998 (n=6), the average recovery was 99.3%,RSD=1.9% (n=3). Conclusion The method was simple, convenient and accurate for determination of adenosine in Ligia exotica.
