1.Research advances of antibody-dependent cell-mediated cytotoxicity in the resistance of rituximab

Rituximab,a chimeric monoclonal antibody targeted against the pan-B-cell marker CD20,comparing with chemotherapy has significantly improved the treatment outcome of all subgroups of B-cell lymphoma patients.However,not all patients respond to rituximab plus chemotherapy or relapse in a short time after response,which suggests the existence of resistance mechanisms.Antibody-dependent cell-mediated cytotoxicity (ADCC) is one of the main mechanisms of rituximab,so any abnormalities in ADCC pathway may affect the efficacy of rituximab.With the widely application of rituximab,its resistance mechanisms are attracting more attention.This review will summarize the factors within ADCC pathway that may affect the antitumor effect of rituximab.
2.CT perfusion imaging of lower extremity skeletal muscle for diabetic peripheral arterial disease

Objective To investigate the feature of CT perfusion of the lower limb skeletal muscle in patients with diabetic peripheral arterial disease (PAD).Methods It was a prospective study that collected out-patients and in-patients of interventional vascular surgery from January 2008 to January 2013.The patients were divided into three groups,including 27 patients (54 limbs) in diabetic PAD group,27 patients (54 limbs) in common PAD group and 9 patients (18 limbs) in control group.Patients in diabetic PAD group were selected when PAD were diagnosed through CTA and patient had the exact history of diabetes.There were 54 limbs in diabetic PAD group,including 10 limbs in Fontaine Ⅰ class,21 limbs in Fontaine Ⅱ class,14 limbs in Fontaine Ⅲ class and 9 limbs in Fontaine Ⅳ class.Patients in common PAD group were selected when PAD were diagnosed through CTA,patients those had the exact history of diabetes or coronary heart disease were exclusion.Patients in control group were collected with negative CTA results.CT perfusion scans of lower extremities were performed in all patients.Time density curve (TDC),Blood flow (BF),blood volume (BV),mean transit time (MMT) and permeability surface (PS) of skeletal muscle in different pathologic stage were obtained with perfusion software.The differences of perfusion parameters among diabetic PAD group,common PAD group and control group were compared by analysis of variance; and the differences of perfusion parameters among control group and all Fontaine classes in diabetic PAD group were also compared.Results TDC of normal skeletal muscle in control group continued to rise,and raise rapidly at an early stage; TDC of diabetic PAD group and common PAD group continued to rise,but raise slowly; the TDC of ischemia skeletal muscle has smaller slope and lower peak value than that of control group.The values of BF were (10.41±0.92) ml · 100 ml-1 · min-1 in control group,(13.37± 1.15) ml · 100 ml-1 · min-1 in diabetic PAD group and (17.12 ±0.81)ml· 100 ml-1· min-1in common PAD group.The values of BV were (1.04± 0.13)ml/L in control group,(1.23 ±0.16)ml/L in diabetic PAD group and (1.84 ±0.23) ml/L in common PAD group.The values of MTT were (11.63 ± 1.45) s in control group,(7.92±0.35) s in diabetic PAD group and (7.82±0.38) s in common PAD group.The values of PS were (3.46±0.84) ml· 100 ml 1· min-1 in control group,(9.84±0.87) ml· 100 ml-1· min-1 in diabetic PAD group and (12.11 ±0.70)ml· 100 ml-1· min-1in common PAD group.The differences of the values of BF,BV,MTT and PS among the three groups were statistically significant (F values were 10.892,4.492,27.543 and 20.506,P all<0.05).The differences of the values of BF,BV among control group and all Fontaine classes in diabetic PAD group were not statistically significant(P all>0.05),the differences of the values of MTT,PS were statistically significant.And the differences of the values of MTT,PS between control group and each Fontaine class in diabetic PAD were statistically significant (P all<0.05).Conclusions CT perfusion imaging of Lower extremity skeletal muscle can prompt the existence of early skeletal muscle ischemia for diabetic PAD.The method can be valuable for clinical early diagnosis.
3.Construction of mcpr1gene vector and expression of mcpr1 in escherichia coli

Objective: To construct mcpr1prokaryoti c expression vector and to express MCPR1 protein.Methods:PCR was used to obtain coding region of mcpr1. Construction of a high-level fusion protein expression vector pGEX-4T-mcpr1 was conducted by inserting the fra gment of coding region of mcpr1into a fusion protein expression vector pGEX -4T-1. Then the recombinant plasmid was transferred into E. colito prepar e the MCPR1/GST fusion protein. DNA sequencing and endonucleases digesting were used to check the coding region. Results:pGEX-4T- mcpr1 wa s constructed successfully and the coding region was inserted into the vector co rrectly. A new protein band of 36 000 was observed by SDS-PAGE analysis after i nduction by IPTG. The 36 000 protein amounted to 39 percent of the total prote in and existed mostly in precipitation of broken bacteria. Conclusion: MCPR1 protein can be expressed in E. coliexpression system and purif ied initially.
4.Clinical effect of neovascular glaucoma treated by vitrectomy and cyclophotocoagulation

AlM: To observe the postoperative intraocular pressure ( lOP) and operation safety in the eyes of the neovascular glaucoma pateints treated by intraocular cyclophotocoagulation which needed vitrectomy at the same time.
METHODS: A total of 12 neovascular glaucoma cases ( 14 eyes ) secondary to diabetic retinopathy, retinal detachment surgery and trauma were reviewed in our study. This procedure mainly used intraocular photocoagulation catheter to highlight the ciliary processes until the ciliary became white atrophy or plosion after vitreous surgery treatment. The intraocular photocoagulation catheter was performed at a power of 300-500mW, for a duration of 0. 1-0. 2ms. Postoperative follow-up was at least for 6mo. The observation of 14 postoperative neovascular glaucoma was performed at 1wk, 1, 6mo observing the lOP and complications.
RESULTS:lOP of the 11 eyes was significantly declined and controlled in normal. After cyclophotocoagulation, average lOP at 1wk was 16. 7±14. 4mmHg, 15. 7±8. 8mmHg at 1mo and 12. 9±4. 5mmHg at 6mo, which compared with untreatment ( 39. 6 ± 10. 0mmHg ) was statistically significant different (P<0. 01). ln follow up time 3 cases were relapsed which were supplied with transscleral or endoscope cyclophotocoagulation. During the follow-up period no endophthalmitis and complications such as eyeball atrophy were found.
CONCLUSlON: The intraocular cyclophotocoagulation and vitrectomy simultaneously can deal with the primary disease and secondary neovascular glaucoma. The operation can be accurately performed under direct cyclophotocoagulation and it is a safe and effective way for neovascular glaucoma which needs vitreous surgery.
5.Genetic Toxicity Induced by Carbon Tetrachloride in Male Mice

Objective To study the genetic toxicity of carbon tetrachloride for male mice and assess the potential genetic toxic effect on mankind. Methods Micronucleus test and sperm malformation test were used respectively to determine the change of micronucleus cell frequencies (MNCF) and the rate of sperm deformity of germ cell induced by carbon tetrachloride in male mice. Carbon tetrachloride was given through intraperitoneal injection at does of 5,15,25 mg/kg respectively for 24 h and 48 h, the rates of micronucleus in the bone marrow cell and sperm deformity of germ cell of male mice were investigated by counting the number of micronucleus cells per 5 000 polychromatic erythrocyte (PCE) in bone marrow and sperm deformity cells per 5 000 germ cells from male mice. To investigate the clinical effect of tonifying the kidney and promoting blood circulation to promote oocyte decoction in the treatment of anovulatory infertility caused by polycystic ovary syndrome. Sixty cases were selected from the out-patient department of Xiyuan hospital of China academy of Chinese medical sciences and the Chinese academy of traditional Chinese medicine, Chinese medicine out-patient department. Sixty patients with PCOS patients were randomly divided into the treatment group and the control group, with 30 cases and 30 cases respectively. The treatment group was given decoction of the reinforcing kidney, activating blood circulation and ovarian stimulation compound recipe. The control group was treated with clomiphene. Through the treatment of 1-2 courses, in the treatment group the pregnancy rate was 56.67%, the ovulation rate 61%; in control group of clomiphene citrate ovulation ratepregnancy rate was 30% , 72.84% of ovulation rate. The difference was significant between two groups (P < 0.05), the pregnancy rate in the treatment group was higher than the control group. The treatment group has regulatory effect on FSH, LH and their ratio, and increase E2 level, decrease T, PRL, INS and other hormone levels, contributing to the mature development of the follicles and endometrium growth, increase the ovulation rate and pregnancy rate. The control group on FSH, E2 increased, LH, T, PRL and INS showed no obvious effect.

8.Bases Selecting and Quality Analysis of Sihuang Gao

Objective To select the optimum bases for Sihuang Gao and study the method for quality control.Method Ultra-fine powders of Radix Scutellariae,Cortex Phellodendri,Radix et Rhizoma Rhei and Rhizoma Coptidis were used to prepare Sihuang Gao with different formulations,the bacteriostatic action of which were compared and stability were inspected.The best prescription was identified by TLC,and the skin irritation was observed.Result Sihuang Gao made of water-soluble base had a strong antibacterial activity and stable quality,and it was proved no irritation,the contents of active component could be detected by TLC.Conclusion The quality was controllable,and the improvement prescription can be used in the clinic.
9.Study on Optimum Preparation Process of Xiaochuan Tiegao by Orthogonal Design

Objective To optimize the preparation process of Xiaochuan Tiegao. Methods The effective compound imporatorin of Dahurian Angelica Root was determined by HPLC. The best condition of preparation process for the ethanol-extraction method was investigated by L9(34) orthogonal design. Results The optimum technical condition:adding 2 times ethanol, cooking 1 hour and 2 times with 90% ethanol. Conclusion The method is reliable, accurate and easy to operate.
10.Determination of Sinapine Thiocyanate in Semen Sinapis Albae of Different Processed Products

Objective To develop an HPLC method for the assay of sinapine thiocyanate in Semen Sinapis Albae of different processed products. Methods It was assayed by RP-HPLC. The column was Hypersil BDS Phenyl (5 ?m, 250 mm?4.6 mm), the mobile phase was acetonitrile-0.08 mol/L KH2PO4 (15∶85). The UV detection wavelength was 240 nm. The flow rate was 1.0 mL/min. Results Good linearity of sinapine thiocyanate was showed within the range of 0.8~4.0 ?g (r=0.999 7). The average recovery was 100.98% (RSD=3.51%). Conclusion The method is simple, accurate and good reproducible. It can be used for quality control of Semen Sinapis Albae of different processed products.
