1.Alkaloidal constituents of Nauclea officinalis

Objective To investigate the alkaloidal constituents of Nauclea officinalis, an anti-inflammatory and antibacterial traditional Chinese herb. Methods The alkaloids were isolated and purified by silica gel, Sephadex LH20, and C-18 ODS column chromatography repeatedly, and their structures were identified by spectral analysis. Results Eleven alkaloids were isolated from the stems of N. officinalis and their structures were identified as angustoline (Ⅰ), 19-O-ethylangustoline (Ⅱ), 3-S-3, 4-dihydroangustoline (Ⅲ), 3-R-3, 4-dihydroangustoline (Ⅳ), naucleamide A (Ⅴ), strictosamide (Ⅵ), vincosamide (Ⅶ), 6′-acetyl-strictosamide (Ⅷ), 2′-acetyl-strictosamide (Ⅸ), pumiloside (Ⅹ), 3-epi-pumiloside (Ⅺ). Conclusion Compounds Ⅰ-Ⅺ are isolated from this plant for the first time.
2.Progress on pain control during the perioperative period of shoulder arthroscopy.

Successful pain management of perioperative shoulder arthroscopy may allow patients to go home earlier, improve the quality of life in perioperative period, and facilitate rehabilitation. A comprehensive method to perioperative pain control has three stages including preoperative, intraoperative and postoperative phase. Successful pain reduction should begin preoperatively because of an excellent communication between patient and physician, moreover, preoperative analgesia also should be administered. Intraoperative efforts should include local wound infiltration and the administration of anesthetic medication intra-articularly. Postoperative management should include oral analgesics, constant infusion devices, Patient Controlled Analgesia (PCA), sedative-hypnotic drug, continuous cryotherapy and vicarious treatment.

3.Antitumor effects of extract from Ligia exotica(Roux)

Objective To study the antitumor activities of extract from Ligia exotica(Roux).Methods The dried powder of total Ligia exotica(Roux) was extracted by 37 ℃ water.The solution was concentrated in vacuum,and then was freeze-dried to afford crude extract.The inhibitory effect of the extract on tumor cells proliferation was assayed by MTT method,and transplant tumor model of sarcoma 180(S180) was used.Results The extract from Ligia exotica displayed obvious proliferation inhibitory effect on HeLa,7901,NCI cells,and no growth inhibitory effect on 929 cells in vitro.After administration at the doses of 0.25,0.50,1.00 g?kg-1,ip,for 7d in tumor-bearing mice with S180,The extract caused 26.9 %,45.3 %,64.6 % inhibition rates,respectively.Conclusion The extract from Ligia exotica showed significant antitumor activity.
4.Determination of adenosine in Ligia exotica with ultraviolet spectrophotometry

Objective To establish a UV method of determination of adenosine in Ligia exotica. Methods Adenosine was extracted with 37℃ water and separated by TLC silica gel, the absorbance was detected at 260nm with UV spectrophotometry.Results The cabibration curve for adenosion was linear within the range of 1.0~20.0?g?mL -1, r=0.9998 (n=6), the average recovery was 99.3%,RSD=1.9% (n=3). Conclusion The method was simple, convenient and accurate for determination of adenosine in Ligia exotica.
5.A new indole alkaloid glycoside from stems of Nauclea officinalis.

AIMTo investigate the chemical constituents of water-soluble part of stem of Nauclea officinalis.
METHODSThe compounds were isolated and purified by resins, silica gel, and Sephadex LH20 column chromatography repeatedly, and their structures were identified by spectral analysis.
RESULTSSeven compounds were isolated and identified as: naucleamide A-10-O-beta-D-glucopyranoside (I), 5-beta-carboxystrictosidin (II), sweroside (III), loganin (IV), 3,4-dimethoxyphenyl-beta-D-glucopyranoside (V), 3, 4, 5-trimethoxyphenyl-beta-D-glucopyranoside (VI), 2-phenethylrutinoside (VII).
CONCLUSIONCompound I is a new indole alkaloid glucoside, compounds I-VII were isolated from this plant for the first time.

6.Development of transgenic maize with anti-rough dwarf virus artificial miRNA vector and their disease resistance.

Maize is one of the most important food crops. Rice black-streaked dwarf virus is a maize rough dwarf disease pathogen. The occurrence and transmission of maize rough dwarf disease brings great damage to maize production. The technology of using artificial miRNA to build antiviral plant has been proven effective in a variety of plants. However, such trials in maize have not been reported. We designed primers based on the sequence of maize zea-miR159a precursor and sequence of function protein genes and silencing RBSDV coding genes in RBSDV genome. We constructed amiRNA (artificial miRNA) gene for silencing RBSDV coding gene and gene silencing suppressor. We constructed pCAMBIA3301-121-amiRNA plant expression vector for transforming maize inbred lines Z31 by using agrobacterium mediated method. After molecular analysis of transgenic maize, homozygous lines with high miRNA expression were selected by molecular detection for a subsequent natural infection experiment. We studied the severity of maize rough dwarf disease according to a grading standard (grade 0 to 4). The experiment results showed that the disease resistance of transgenic homozygous maize with the anti-rough dwarf virus amiRNA vector was better than that of wild type. Among the transgenic maize, S6-miR159 transgenic maize had high disease resistance. It is feasible to create new maize variety by the use of artificial miRNA.

7.Investigation on occupational exposure to 5-fluorouracil in pharmacy intravenous admixture service of a hospital.

OBJECTIVETo investigate the level of occupational exposure to 5-fluorouracil (5-Fu) in the pharmacy intravenous admixture service (PIVAS) of a hospital, and identify the sources of 5-Fu contamination.
METHODSThe 5-Fu concentrations in air, on the surface of different areas in PIVAS and personal protective equipments were detected using UV-vis spectrophotometry.
RESULTSThe 5-Fu in air could not be detected. The 5-Fu concentrations on five different surfaces of biological safety cabinets were (22.00 +/- 6.35), (13.99 +/- 2.46), (14.13 +/- 0.72), (7.25 +/- 1.19) and (9.87 +/- 1.23) ng/cm2, respectively, which were significantly higher than those [(3.14 +/- 0.04), (5.43 +/- 0.65), (2.26 +/- 0.17), (2.26 +/- 0.17) and (3.63 +/- 0.46) ng/cm2] of corresponding controls (P < 0.05 or P < 0.01). The 5-Fu concentrations of the floor under cabinets [(18.19 +/- 5.22) ng/cm2], the floor in front of cabinets [(10.25 +/- 2.57)ng/cm2], the office floor [(11.64 +/- 2.53) ng/cm2], the terrace floor [(99.89 +/- 14.06 ) ng/cm2], the floor beside trash can in dressing room [(24.54 +/- 0.23) ng/cm2] were significantly higher than those of control [(3.36 +/- 0.11 ) ng/cm2] (P < 0.05 or P < 0.01). The 5-Fu concentrations of the tables in preparation room [(7.22 +/- l.04) ng/cm2] and the tables in office [(11.81 +/- 1.18) ng/cm2] were significantly higher than those of control [(5.56 +/- 0.14) ng/cm2] (P < 0.05 or P < 0.01). The 5-Fu concentrations of the indoor handle in preparation room were significantly higher than those of controls (P < 0.05 or P < 0.01). 5-Fu concentrations on the surfaces of outdoor handle and floor beside door in preparation room were not significantly increased compared with controls (P > 0.05). The 5-Fu concentrations on the surfaces of infusion bags, transfer box, transfer trays were significantly higher than those of controls (P < 0.05). The differences of 5-Fu concentrations between outer and inner masks and controls were not significant (P > 0.05). The 5-Fu concentrations of gloves of preparing and checking staffs were significantly higher than those of controls (P < 0.05 or P < 0.01).
CONCLUSIONThe preparing and checking process of 5-Fu and the treatment of medical wastes are major sources of 5-Fu contamination.

8.Studies on chemical constituents and volatile oil of Xiaochengqi decoction.

OBJECTIVETo study the chemical constituents and volatile oil of Xiaochengqi decoction.
METHODThe constituents in decoction were separated by means of column chromatography and their structures were identified by spectral data and compared with literature data. As well as the volatile oil of Xiaochengqi Fang were analyzed and identified by GC-MS.
RESULTEleven compounds were isolated and identified as chrysophanol (1), physcion (2), magnolol (3), beta-sitosterol (4), sitosterol trans-cinnamic acid (5), emodin (6), aloe emodin (7), rhein (8), gallic acid (9), chrysophanol-8-O-beta-D-glucopyranoside (10), aurantiamarin (11). The volatile oils extracted with steam distillation from Xiaochengqi were identified 67 components, and the main components are including.
CONCLUSIONAll of the eleven compounds were isolated from Xiaochengqi decoction for the first time and the study on their activities in Chinese prescription is being carried out. D-limonene (42.61%), p-cymene (16.43%), and 8-terpinene (14.46%).

9. The sonographic characteristics of papillary thyroid microcarcinoma and its risk evaluation of cervical lymphatic metastasis

Objective:
To investigate the sonographic characteristics of papillary thyroid microcarcinoma(PTMC) and the preoperative prediction value for cervical lymphatic metastasis of PTMC.
Methods:
Three hundred and fifty-eight PTMC patients who were confirmed by pathology and accepted partly or totally thyroidectomy and neck dissection were collected to analyze the sonographic characteristics, all patients were classified according to multifocal carcinoma nodules and solitary carcinoma nodules; cervical lymphatic metastasis or not; the maximum diameter of carcinoma nodules. 10.Expression of free fatty acid receptor GPR40 in adult monkey hippocampus after ischemia

Objective To investigate the expression of free fatty acid receptor GPR40 and evaluate the possible function of GPR40 in the adult monkey hippocampus after ischemia. Methods According to the post-ischemic adult monkey model of Yamashima, a total of 24 adult monkeys were randomly divided into 4 groups: sham-operated group (control) and post-ischemic day 4, 9, 15 (d4, d9,d15). The expression of free fatty acid receptor GPR40 was detected at the protein level by immunoblotting and immunohistochemistry in the adult monkey hippocampus. Results Immunoblotting analysis showed the expression of GPR40 was decreased in CA1 and increased in DG after ischemia (posfischemic group vs control, P>0.05). Immunohistochemistry data revealed that the double stained cells of GPR40 and NeuN were also decreased by 50% (d15) in CA1 and had no significant changes in DG after ischemia. Interestingly, the co-labeled cells of GPR40 and GFAP were increased 2.5 folds (d4) in post-ischemic SGZ. Conclusions There is the different expression of GPR40 in adult monkey hippocampal CA1 and DG regions after ischemia. Co-labeled cells of GPR40 and GFAP are increased in post-ischemic SGZ, which indicates that polyunsaturated free fatty acid such as DHA, a ligand of GPR40, may alleviate neuronal injury in post-ischemic hippocampus.
