Objective To investigate mRNA expression levels of cathepsin B,cathepsin L and uPA in non-Hodgkin lymphoma (NHL) samples, and analyze the relationship between three genes and clinicopathological factors. Methods Real-time fluorescence quantitative RT-PCR was used to quantify the relative expression levels of cathepsin B,cathepsin L and uPA mRNA in 39 NHL patients without chemotherapy or radiotherapy and 15 patients with lymph node reactive hyperplasia.Relationship between their expression and clinico-pathological factors was analyzed. Results mRNA expression levels of cathepsin B,cathepsin L and uPA in NHL group were significantly higher than those in the control group (0.403 vs 0.128,0.209 vs 0.057,0.459 vs 0.031,all P < 0.05).The mRNA expression levels of cathepsin B,cathepsin L and uPA were correlated with each other in NHL.The mRNA expression levels of cathepsin B,cathepsin L and uPA in advanced staged NHL were markedly higher than those in early staged NHL (0.453 vs 0.341,0.204 vs 0.085,0.372 vs 0.196,all P < 0.05).mRNA levels of cathepsin B,cathepsin L and uPA were higher in NHL patients sensitive to chemotherapy than in those resistant to chemotherapy(0.401 vs 0.556,0.085 vs 0.205,0.316 vs 0.499,all P < 0.05).Conclusions The high expression of cathepsin B,cathepsin L and uPA could be detected in NHL,and their expression has a close relationship with the biological behavior characters of NHL.The dynamic examination of cathepsin B,cathepsin L and uPA may be used as clinical auxiliary diagnosis and prognosis of NHL.
4.Study on relationship between efflux pumps and Candida albicans resistance mechanism
Chinese Journal of Infectious Diseases 1997;0(04):-
Objective To investigate whether efflux mechanism is involved in azole resistant Candida albicans strains isolated in China. Methods We compared rhodamine 6G uptake and glucose induced efflux between azole sensitive and azole resistant strains to elucidate resistance to azole due to efflux pump, then measured mRNA level of efflux gene cdr1 and cdr2 by reverse transcriptase polymerase chain reaction (RT PCR). Results Rhodamine 6G moved from extra into the intracellular compartment in both azole resistant and azole sensitive strains quickly when in glucose free incubation condition. However, efflux of rhodanmine 6G was enhanced significantly in azole resistant strains and decreased in azole sensitive strains while adding glucose in the media. The mRNA level of efflux gene cdr1 and cdr2 was higher in clinical azole resistant strains and fluconazole induced resistant strains than strains reverted from resistance to sensitivity. However, all such strains had higher efflux gene expression than azole sensitive strains. Conclusions Efflux mechanism is associated with the resistance to azole in Candida albicans strains isolated in China. Fluconazole can induce efflux pumps expression leading to resistance to azole but the resistance to azoles can be reversed after withdrawing fluconazole.

MicroRNA is a type of endogenous noncoding small molecular RNA with a length of 19-25 nucleotides. MicroRNAs are involved in almost all key biological processes, such as cell proliferation, differentiation, apoptosis, and hematopoiesis, via repres-sion of target gene expression. Acute lymphoblastic leukemia (ALL) is the most common type of pediatric cancer. It is a heterogeneous disease with different clinical signs, morphology, immunophenotype, and cytogenetics. Currently, several microRNAs have implicated aberrant expression in ALL and play an important role in cancer initiation and development. MicroRNA expression pattern is associated with clinical feature, prognosis, and treatment. Therefore, microRNAs could act as a novel marker to help scientists learn about the pathogenesis of ALL and to provide more effective treatment options.

Chinese Journal of Medical Instrumentation 2014;38(3):168-185
Heart disease is the major disease that threaten human health. ECG is an important tool for the diagnosis of cardiac disease. As traditional ECG measurement devices have many disadvantages, a non-contact ECG measurement system was designed. With the non-contact electrode based on capacitive coupling, the signals were collected and then they were amplified and filtered. The conditioned analog signal was converted to digital data which was sent to the mobile terminal through bluetooth. Finally, the ECG data was analyzed to extract the key ECG parameters. The results showed that the precise ECG signals can be got with the non-contact electrode and the key ECG parameters can be acquired accurately.

Progress of guidelines for quantifying iron overload in the 56th American Society of Hematology (ASH) annual meetings was reviewed. This article reviewed the use of historical data, serological measures, and MRI to estimate somatic iron burden. Before chelation therapy utilization, transfusional volume was an accurate method for estimating liver iron burden, whereas transferrin saturation reflected the risk of extrahepatic iron deposition.Liver biopsy was invasive and plagued by sampling variability. In the current study,we recommended annual liver iron concentration to be measured by MRI for all patients on chronic transfusion therapy. And it was important to measure cardiac T2* by MRI every 6-24 months depending on the clinical risk of cardiac iron deposition.Recent validation data for pancreas and pituitary iron assessments was also presented, while the further confirmatory data was suggested before these techniques could be recommended for routine clinical use.

Journal of Leukemia & Lymphoma 2015;24(3):132-134
New progresses of timing of allogeneic hematopoietic stem cell transplantation (allo-HSCT) for myelodysplastic syndrome (MDS) and aplastic anemia in the 56th ASH annual meetings were reviewed.Allo-HSCT for MDS was a potentially curative procedure, but it was associated with a significant risk of morbidity and mortality. With the recent approval of disease-modifying agents, the appropriate timing of alloHSCT needed to be addressed. For low and intermediate-1 IPSS risk groups, the decision to delay HSCT from the time of diagnosis maximized overall survival. For patients with intermediate-2 and high-risk disease, immediate HSCT at the time of diagnosis was associated with a greater number of life-years than HSCT at a delayed time point. The methods that underwent HSCT were after azacitidine,leukemia-type induction chemotherapy,or both.for severe aplastic anemia (SAA),HSCT was a proven cure, but HLA-matched sibling donors were found in fewer than 25 % of newly diagnosed patients. The use of early unrelated donor HSCT was an evolving concept that will became more accepted as improvements in HSCT outcomes continued.Moving forward,HLA-matched related and unrelated donor HSCT will likely become the treatment of choice for most patients with higher-risk MDS and newly diagnosed SAA.

Journal of Leukemia & Lymphoma 2015;24(4):211-213
New progress of guidelines for novel targets to iron overload in the 56th American Society of Hematology (ASH) annual meeting was reviewed.β-thalassemia and hereditary hemochromatosis disorders, and inappropriately low levels of the liver hormone hepcidin are responsible for the increased iron absorption, leading to toxic iron accumulation in many organs.Several studies have shown that targeting iron absorption could be beneficial to reducing or preventing iron overload in these 2 disorders.New approaches target Tmprss6.Additional strategies in β-thalassemia are showing beneficial effects in ameliorating ineffective erythropoiesis and anemia. The goal of this review is to discuss the major factors controlling iron metabolism and erythropoiesis and to discuss potential novel therapeutic approaches to reduce or prevent iron overload in these 2 disorders and ameliorate anemia in β-thalassemia.
