Journal of Leukemia & Lymphoma 2014;23(7):401-404
Objective To investigate mRNA expression levels of cathepsin B,cathepsin L and uPA in non-Hodgkin lymphoma (NHL) samples, and analyze the relationship between three genes and clinicopathological factors. Methods Real-time fluorescence quantitative RT-PCR was used to quantify the relative expression levels of cathepsin B,cathepsin L and uPA mRNA in 39 NHL patients without chemotherapy or radiotherapy and 15 patients with lymph node reactive hyperplasia.Relationship between their expression and clinico-pathological factors was analyzed. Results mRNA expression levels of cathepsin B,cathepsin L and uPA in NHL group were significantly higher than those in the control group (0.403 vs 0.128,0.209 vs 0.057,0.459 vs 0.031,all P < 0.05).The mRNA expression levels of cathepsin B,cathepsin L and uPA were correlated with each other in NHL.The mRNA expression levels of cathepsin B,cathepsin L and uPA in advanced staged NHL were markedly higher than those in early staged NHL (0.453 vs 0.341,0.204 vs 0.085,0.372 vs 0.196,all P < 0.05).mRNA levels of cathepsin B,cathepsin L and uPA were higher in NHL patients sensitive to chemotherapy than in those resistant to chemotherapy(0.401 vs 0.556,0.085 vs 0.205,0.316 vs 0.499,all P < 0.05).Conclusions The high expression of cathepsin B,cathepsin L and uPA could be detected in NHL,and their expression has a close relationship with the biological behavior characters of NHL.The dynamic examination of cathepsin B,cathepsin L and uPA may be used as clinical auxiliary diagnosis and prognosis of NHL.
4.Study on relationship between efflux pumps and Candida albicans resistance mechanism
Chinese Journal of Infectious Diseases 1997;0(04):-
Objective To investigate whether efflux mechanism is involved in azole resistant Candida albicans strains isolated in China. Methods We compared rhodamine 6G uptake and glucose induced efflux between azole sensitive and azole resistant strains to elucidate resistance to azole due to efflux pump, then measured mRNA level of efflux gene cdr1 and cdr2 by reverse transcriptase polymerase chain reaction (RT PCR). Results Rhodamine 6G moved from extra into the intracellular compartment in both azole resistant and azole sensitive strains quickly when in glucose free incubation condition. However, efflux of rhodanmine 6G was enhanced significantly in azole resistant strains and decreased in azole sensitive strains while adding glucose in the media. The mRNA level of efflux gene cdr1 and cdr2 was higher in clinical azole resistant strains and fluconazole induced resistant strains than strains reverted from resistance to sensitivity. However, all such strains had higher efflux gene expression than azole sensitive strains. Conclusions Efflux mechanism is associated with the resistance to azole in Candida albicans strains isolated in China. Fluconazole can induce efflux pumps expression leading to resistance to azole but the resistance to azoles can be reversed after withdrawing fluconazole.

MicroRNA is a type of endogenous noncoding small molecular RNA with a length of 19-25 nucleotides. MicroRNAs are involved in almost all key biological processes, such as cell proliferation, differentiation, apoptosis, and hematopoiesis, via repres-sion of target gene expression. Acute lymphoblastic leukemia (ALL) is the most common type of pediatric cancer. It is a heterogeneous disease with different clinical signs, morphology, immunophenotype, and cytogenetics. Currently, several microRNAs have implicated aberrant expression in ALL and play an important role in cancer initiation and development. MicroRNA expression pattern is associated with clinical feature, prognosis, and treatment. Therefore, microRNAs could act as a novel marker to help scientists learn about the pathogenesis of ALL and to provide more effective treatment options.

Heart disease is the major disease that threaten human health. ECG is an important tool for the diagnosis of cardiac disease. As traditional ECG measurement devices have many disadvantages, a non-contact ECG measurement system was designed. With the non-contact electrode based on capacitive coupling, the signals were collected and then they were amplified and filtered. The conditioned analog signal was converted to digital data which was sent to the mobile terminal through bluetooth. Finally, the ECG data was analyzed to extract the key ECG parameters. The results showed that the precise ECG signals can be got with the non-contact electrode and the key ECG parameters can be acquired accurately.

Chinese Journal of Urology 2001;0(07):-
Objective To construct radiation inducible-specific hytk gene therapy vector using radiation inducible gene Egr-1 promoter, and to determine its feasibility in bladder carcinoma gene therapy. Methods A radiation inducible-specific hygromycin phosphotransferase-thymidine kinase fusion gene(hytk)vector was constructed by subcloning hytk gene into pCIneo/Egr-1 vector, which was carrying Egr-1 promoter. Then the vector was transferred into bladder carcinoma cells lines EJ with FuGENE TM 6.Antitumor effects were observed after irradiation with 60 Co-rays as well as gancyclovir(GCV) treatment. Results The new cell line,EJ/Egr-1-hytk,incorporated and expressed the hytk gene, which was proved by RT-PCR and Western blotting.Egr-1 promoter actively drived hytk gene expression in the presence of ?-rays in the EJ cells, while lower hytk expression was detected without ?-rays.Antitumor effects were observed after ?-rays irradiation with 60 Co rays and GCV treatment. Conclusions Hytk-based vector harboring Egr-1 promoter is a novel ideal candidate vector for bladder carcinoma gene therapy.

Chinese Journal of Hospital Administration 1996;0(04):-
Pharmacy intravenous admixture service was built in accordance with the Australian criteria for intravenous drug admixture and managed in light of the ideas contained in the Accreditation Standards for Hospital released by the US Joint Commission International. Cytoxic drugs, total parenter-al nutrition solutions and intravenous infusions were centrally admixted in the service. Aseptic operation was stressed in the admixture of intravenous drugs while the appropriateness of drug use and the rationality of drug formulations were stressed in the review of doctors' orders. In practice, priority was given to environment and equipment management, qualification and certification of personnel and continuous education, the process was standardized, regulations for standard post operation and the rational use of drugs were formulated, and criteria for assessing doctors' orders were established All this has led to satisfactory results.

Objective To summarize the experience of pterional craniotomy microsurgical resection of giant tuberculum sellae meningiomas. Methods A total of 18 patients with giant tuberculum sellae meningiomas were treated microsurgically in this hospital from February 2000 to November 2004. The patients were operated on by use of pterional craniotomy on the side of worse vision. The basal part of the tumor was firstly dissected to control the blood supply of the lesion. Through the cerebral cisterns at the sellar region and the interfaces between the tumor and the adjacent structures, the tumor was removed to the greatest possible extent with minimal invasion to the neighbouring structures. Results[WTBZ] A total resection was conducted in 16 patients (88.9%), and a subtotal resection, in 2 patients (11.1%). No surgery related death was observed. Follow-up examinations in 14 patients for 3 months ~ 4 years (mean, 2.5 years) found no recurrence of meningiomas. Conclusions[WTBZ] The pterional approach provides excellent exposure of the middle fossa and the parasellar area. Pterional approach microsurgical technique can improve the rate of total resection of giant tuberculum sellae meningiomas.

Chinese Journal of Minimally Invasive Surgery 2005;0(11):-
Objective To evaluate the curative effect of endoscopic sphincterotomy (EST) combined with laparoscopic cholecystectomy (LC) in the treatment of gallstones complicated with common bile duct stones. Methods A total of 36 cases of gallstones complicated with common bile duct stones were enclosed. First EST was performed to remove the bile duct stones. Then LC was applied to resect the gallbladder. Results EST was successfully performed in 34 cases (94.4%) and failed in 2 cases because of the terminal stricture of the bile duct that was afterwards cured by an open choledochojejunostomy. LC was performed following EST in 34 cases, 32 of which were successful (94.1%) and 2 of which required a conversion to open surgery. Conclusions EST combined with LC is the best way for treating gallstones complicated with common bile duct stones. The biliary stricture mainly accounts for the failure of EST and should be treated by choledochojejunostomy as the first choice.
