Objective To determine quercetin content in Polygonum chinense L.from different growing areas.Methods RP-HPLC was used to determine quercetin content.Chromatographic conditions were as follows:chromatographic column of Agilent TC-C18 (4.6 mm?250 mm,5 ?m),mobile phase consisting of methanol-0.6 %phosphoric acid (52:48),with a velocity of 1 mL?min-1,column temperature at (25?1)℃,detection wavelength being 365 nm,elution time of 20min,and an injection volume of 20 ?L.Results Standard curve regression equation of quercetin is:Y=85 355X-48.16(r=0.999 3).A good linearity was showed in the range of 0.004～0.012 mg?mL-1,with average recovery being 100.3 %,and RSD of 1.44 %.The content of quercetin in the samples from different growing areas is in the range of 183.0～848.1 ?g?g-1.Conclusions The method is simple and reliable,and can be used to control the quality of Polygonum chinense L.The existence of some differences of quercetin content between medicinal materials from different growing areas indicates that we should make a quality standard to ensure the safety and effectiveness of Polygonum chinense L.in clinic.

Objective To explore status quo of Shanghai community health services based on the investigation of rural community doctors in Shanghai suburb,and to propose measures on training rural doctors. Methods Retrospective investigation was carried out in 114 rural community health service centers and 1 563 clinics.Primary investigation included the registration,distribution,age,academic titles and education background. Results The results showed insufficient number of human resources and inadequate investment in human capitals in the primary medical services of Shanghai suburbs,and there was a serious shortage of general practitioners.The other problems included older age,insufficient academic qualifications,and without professional titles. Conclusion We should improve the status and treatment of rural doctors.Job training and well-trained professional students should be encouraged for the primary health services.More and more doctors should be attracted to work in community health service centers.General practitioners' personnal information network and general medical training network should be established,and the general medical education system gets improvement.

Object To investigate the effects of Panax notoginseng saponins (PNS) on myocardial Gs? mRNA expression of seriously scalded rat. Methods A 30% skin full thickness scald model was produced by immersing rat in 95 ℃ water for 10 s. The effects of PNS on myocardial Gs? mRNA level were observed with dot blotting hybridization and in situ hybridization technique; effects on cAMP and adenyl cyclase (AC) activities were determined with radioimmunoassay. Results Myocardial Gs? mRNA, AC activity and cAMP content were reduced significantly 3 h after scalding. PNS (100, 200 mg/kg) could markedly increase the level of myocardial Gs? mRNA expression. Objective To construct the prokaryotic expression vector of mouse CD25 extracellular domain and to express it in E coli.Methods Total RNA was isolated from splenocytes of Balb/c mice.The CD25 extracellular domain gene was amplified by RT-PCR and cloned into the PET-32a vector.A positive recombinant,PET-32a-CD25e,was identified by enzyme cleaving and sequencing before its expression in E.coli,and transferred into E.coli BL21(DE3) plysS for its expression.After purification with Ni+ resin and renaturation in vitro,a relative molecular mass(Mr) of the interesting protein was detected by SDS-PAGE and Wes-tern blotting.Effect of the purified interesting protein on the proliferation of splenocytes from T cell vaccine-immunized syngeneic mice was detected by MTT assay.Results The cloned CD25 extracellular domain gene was identified to be functional by sequencing and expression.The purified interesting protein could significantly induce the proliferation and IL-4 secretion of splenocytes from T cell vaccine-immunized mice in vitro.Conclusion Mouse CD25 extracellular domain gene can be successfully expressed in prokaryotic cells with biological activity,which lays a foundation for further relative studies.
