2.Gene polymorphism of lipoprotein lipase in patients with acute hypedipidemic pancreatitis

Objective To investigate the expression and polymorphism of lipoprotein lipase (LPL) gene and their association with acute hyperlipidemic pancreatitis(HLP). Methods A total of 120 patients Were assigned to HLP group (n=20), acute pancreatitis (AP) group (n=50) and control group (n= 50). Serum levels of triglyceride (TG), cholesterol (Ch), free fatty acid (FFA), lipoprotein and apolipoprotein and serum LPL/HL activities were determined. The mRNA expressions of LPL/HL and LPL gene intron 8 polymorphisms were detected by RT-PCR and PCR-RFLP, respectively. Results The serum levels of TG, Ch, FFA and ApoE were significantly higher in HLP group than those in AP group and control group (P<0.05). The serum level of HDL was lower in HLP group than that in AP group and control group(P<0.05). The serum LPL/HL activities were significantly higher in HLP group than that in AP and control groups. The expression of LPL mRNA was up-regulated and intron 8 Hind Ⅲ H2 allele frequency was significantly inereased in the HLP group compared to control group(0.90/0.72, P<0.05). H1 allele frequency was significantly decreased in the HLP and AP groups compared to control group(0.10/0.28 and 0.14/0.28, respectively). Conclusions The high allele frequency of LPL gene intron 8 Hind H2 result in the increase of activities and expression of LPL mRNA, which exacerbate the development of HLP through changing TG metabolism such as FFA accumulation.

This paper outlines the major animal epidemic emergency management system in the United States, involving the responsibilities of United States Department of Agriculture (USDA), Department of Homeland Security (DHS), and Department of Health and Human Services ( DHHS ) .Besides the main commanding and coordinating departments, the system also involves procedures in surveillance, early warning, preparation, response and ending in case of serious animal emergency.We have also summarized and analyzed the characteristics of the American response mode, such as net-worked laboratories, special corps,budgets and national animal identification systems.
4.Clinical Observation of Locally Advanced Gastric Cancer Treated by Compound Matrine Injections Combined with Concurrent Chemoradiotherapy

Objective To study the recent clinical efficacy of compound matrine injections combined with concurrent chemoradiotherapy in the treatment of locally advanced gastric cancer. Methods Totally 65 cases of untreated locally advanced gastric cancer were randomly divided into treatment group (33 cases) and control group (32 cases). The control group was treated by the conformal intensity modulated radiotherapy combined with chemotherapy. On the basis of the control group, the treatment group was given compound matrine injections 20 mL/time, once a day for 5 weeks. The recent curative effect, the quality of life and adverse reaction of the two groups were observed. Results The recent curative effect rate of the treatment group was 66.67% (22/33), the control group was 40.63% (13/32), the difference was statistically significant (P=0.031). On hematologic toxicity, leukopenia occurred in the treatment group was 30.30% (10/33), the control group was 65.63% (21/32), the difference was statistically significant (P=0.004). KPS of 26 cases (78.79%) improved in the treatment, and 18 cases (56.25%) improved in the control group. Conclusion The effects of compound matrine injections combined with concurrent chemoradiotherapy for locally advanced gastric cancer are good, they can improve short-term efficacy, reduce side effects, and improve the quality of life.
5.Laparoscopic Cholecystectomy Combined with Endoscopic Retrograde Cholangiopancreaticography for Gallbladder and Bile Duct Stones

Objective To investigate the efficacy of laparoscopic cholecystectomy(LC) followed by endoscopic retrograde cholangiopancreaticography(ERCP) for gallbladder and bile duct stones.Methods A total of 58 patients with both gallbladder and bile duct stones received endoscopic sphincterotomy(EST) in our hospital.After the stones were removed,LC was performed on the patients.Results Of the 58 patients,56 were successfully treated with EST and LC.Two cases were unsuccessful owing to difficulty in intubation during ERCP in one and failure to remove all the stones from the bile duct in the other.These two patients were then cured by LC combined with electronic choledochoscopy.Perioperative complications included bleeding during EST in 1,acute edematous pancreatitis in 2,and mild infection of the puncture site in 1.The overall complication rate was 7.1%(4/56).Retained common bile duct stone was observed in 1 patient,and was successfully removed by subsequent ERCP.Conclusions LC combined with ERCP is a safe and effective minimal invasive method for the treatment of concurrent gallstones and bile duct stones.

8.Effect of Splenectomy & Different Splenic Preservation Operations on Immune Functions in Rats

It is still controversial whether total splenectomy on adult animals would affect their immune function. In this study, total splenectomy and different kinds of splenic preservation operations including splenorrhaphy, partial splenectomy and spleen autotransplantation were performed on adult rats after the porta of the spleen of the rats was artificially ruptured and the splenic artery ligated.In the 4th month after the operations, the hemolysin antibody and the de-layed-type hypersensitivity reaction were determined in the rats. It was found that no significant difference occurred in the hemolysin antibody and the delayed type hypersensitivity reaction among the rats of the control group, the group with total splenectomy, and the groups with different kinds of splenic preservation operations.
9.The biologic behavior and gene expression of malignant choroid melanoma cell strain OCM-1-gfp modified with green fluorescent protein

Objective To investigate the development and metastasis of malignant choroidal melanoma cell strain OCM-1-gfp modified with green fluorescent protein(GFP) and the factors which affected the tumor biological behaviors. Methods GFP was transfected into malignant melanoma cell strain OCM-1.Melanoma cells with high and stable expression of GFP were injected into subretinal space and the subcutaneous space of hind leg of Balb/c nude mouse respectively in order to establish orthotopic and heterotopic transplanted tumor models.The development and metastasis process of orthotopic tumor models was observed directly by fluorescence microscope,and the size of the hypodermal tumor was measured by vernier.The expressions of 13 genes in melanoma were detected by means of immunohistochemistry staining. Results Malignant choroidal melanoma cell strain OCM-1 stably expressed GFP and preserved the characteristics of parental generation,OCM-1-gfp may develop melanoma and continue to metastasize in nude mouse.Positive expression of most of the antibodies,including Rb,p53,p21,E_2F,NF?B,cyclin D1,proliferation cellular nuclear antigen(PCNA),bcl-2、bcl-XL/S,bax,and epithelial growth factor(EGF)and its receptor(EGFR),was found.While the staining of inhibition gene p16 was negative. Conclusions GFP is the marker for observing the development and metastasis of malignant choroidal melanoma in vivo.The rate of tumor formation and development process in orthotopic models does not differs much from which in heterotopic models of malignant choroidal melanoma.The expressions of lots of genes in malignant choroidal melanoma developed from OCM-1-gfp including p16、p53、NF?B,cyclin D,PCNA,EGF,and EGFR are abnormal.
