3.Inhibitive Effect of Interleukin-4,-10 on Cell Apoptosis in Rats with Renal Ischemic-Reperfusion Injury
Journal of Applied Clinical Pediatrics 1992;0(05):-
Objective To study the inhibitive effect of interleukin-4(IL-4) and (or) IL-10 on cell apoptosis of renal ischemic-reperfusion injury(IRI) in rats and its mechanism. Methods Thirty female Sprague-Dawley rats were divided into five groups(n=6): sham-operated group,operation+9 g/L NaCl control (NaCl group),operation+IL-4(IL-4 group),operation+IL-10(IL-10 group),operation+ IL-4+IL-10 (IL-4+IL-10 group). The animal model of IRI was set up by clamping bilateral renal pedicles for 45 min. When the clamp was loosed, relevant agents was injected in every group. Apoptosis of renal epithelial cells (TUNEL method) was obserued. Results IL-4 could not reduce level of apoptosis of renal epithelial cells. To some extent,IL-10 could reduce level of renal epithelial cells. The combination of IL-4 and IL-10 was more effective in inhibiting apoptosis of renal epithelial cells. Conclusion The combination of IL-4 and IL-10 is effective in inhibiting apoptosis of renal epithelial cells, indicating that it has a relationship in preventing renal histological and functional damage.

Objective To explore the regulatory effect of Nervilia fordii Injection ( NFI ) on inflammatory cytokines in rats with lipopolysaccharide ( LPS) -induced acute lung injury ( ALI) , and to explore its possible interfering mechanism . Methods The rats were randomly divided into normal group , model group , Shenmai Injection group, and NFI group. J774 macrophages were stimulated by LPS to establish the cell model in vitro, and in vivo ALI rat model was established by injection of LPS through the sublingual veins. Electronic microscope and enzyme-linked immunosorbent assay (ELISA) were used for observing the proliferation of J774 macrophages, the levels of supernatant inflammatory cytokines secreted by J774 cells, and the levels of serum inflammatory cytokines . Results The proliferation of LPS-induced J774 macrophages was increased , and the secretion of inflammatory cytokines was disordered. Uncontrolled inflammatory reaction occurred in the lung after the rats were administrated with intravenous injection of LPS . Both NFI and Shenmai Injection could inhibit the proliferation of J774 macrophages. NFI could also significantly inhibit the levels of supernatant and serum tumor necrosis factor (TNF) -α and interleukin 6 (IL-6) expression (P<0.05 or P<0.01), and it could increase the level of supernatant IL-10 (P<0.01) and decrease the level of serum interleukin 10 (IL-10) in rats (P<0.05), but couldn’t regulate the secretion of monocyte chemoattractant protein 1 (MCP-1) (P>0.05). Conclusion NFI has better preventive and therapeutic effect for ALI than Shenmai Injection, and its possible mechanism is related with the inflammatory regulation and lung injury relief through the suppression of excessive expression of TNF-α and IL-6 .

Objective To screen the liver cell proteins interacting with the novel protein encoded by the 2.2 kb singly spliced variant of hepatitis B virus genome. Methods The splicing-specific gene TPss generated by the 2.2 kb singly spliced variant of HBV genome was amplified by PCR and cloned into the bait vector pGBKT7. After exclusion of self-activatian capacities of TPss protein, a two-hybrid library screening was performed using a pre-transformed human liver cDNA library to screen the liver cell proteins interacting with TPss. Mammalian two-hybrid assay was also done to further confirm the interactions between the bait and prey proteins in Huh7 and HepG2 hepatacytes. Results TPss gene with the size of 336 bp was successfully amplified and cloned, and the TPss protein expressed well in AHI09 yeast cells. Four liver cell proteins interacting with TPss, i. e. , cathepsin B, epoxide hydrolase 1, cathepsin D and fibrinogen gamma chain, were screened by yeast two-hybrid assay and further confirmed by mammalian two-hybrid assay. Conclusion TPss could interact with liver cell proteins.

Objective To evaluate the diagnostic value of Achilles tendon rupture by using X-ray plain films and MR images. Methods The plain films and MRI findings in 21 patients with operation proved acute Achilles tendon rupture were analyzed, and every manifestation on MR and operation were compared. Results The Achilles tendon rupture was diagnosed on MRI in all 21 cases. Complete rupture of Achilles tendon was showed in 7 and partial rupture in 14. MRI appearance of the rupture represented as thickening of Achilles tendon, hyperintensity in the tendon, and completely or partly discontinuance of the fibrous bundle. X-ray showed Achilles tendon rupture in 5, suspicious rupture in 12, and normal in 4. Operation showed Achilles tendon complete rupture in 9 and partial rupture in 12.Conclusion MRI can accurately show the degree and location of acute Achilles tendon rupture, which is very helpful for treatment. X-ray is the adminicular method in detecting Achilles tendon rupture and the diagnosis should be depended on MRI to confirm the the acute Achilles tendon rupture.
