1.New ADR Reported in Our Hospital during 2013-2014:Analysis of 151 Cases

OBJECTIVE:To probe into the characteristics and general pattern of new ADR in our hospital. METHODS:A to-tal of 151 new ADR cases reported to Nationwide ADR Monitoring Network by our hospital during 2013 to 2014 were analyzed statistically. RESULTS:Of the 151 ADR cases,males(56.29%)showed higher proportion than females(43.71%);most of ADR cases were induced by anti-infective agents,including 45 cases(29.80%). ADR cases most easily induced by injection and intrave-nous dripping,accounting for 83.44% and 91.39 %,respectively. Lesions of skin and its appendants were most common clinical manifestations of ADR,which accounted for 28.82%(66 cases). There were 10 new and severe ADR cases(6.62%). All of ADR cases were cured or improved. CONCLUSIONS:More attention should be paid to ADR monitoring especially indication of anti-in-fective agent to reduce the occurrence of ADR.
2.An study of Tephroseris Kirilowii Turez.Houlub extract on cell cycle and apoptosis marker AnnexinV/PI of U266

Objective To study Tephroseris Kirilowii Turez.Houlub extract on cell cycle and apoptosis marker AnnexinV/PI of U266 in vitro..Methods U266 cells were cultured together with Tephroseris Kirilowii Turez.Houlub extract.Cell cycle and apoptosis marker AnnexinV/PI were detected by flow cytometry(FCM).Results After exposure of U266 cells to Tephroseris Kirilowii Turez.Houlub extract.the cell cycle distribution was changed.There Was a decrease of cells in the G0/G1 phase with an increase of cells in the S phase and G2/M phase and apoptosis.FCM with staining of Annexix V FITC/PI showed a dependence of apoptotic cells with the dosage of Tephroseris Kirilowii Turez.Houlub extract.Conclusion Tephroseris Kirilowii Turez extract has strong cell apoptosis effect on U266 cells.
3.Histological observation of the angiogenic effect of bFGF combined to freeze- dried tendon in ACL reconstruction at early stage

Objective To observe histologically the effect of exogenous application of bFGF (basic fibroblast growth factor) combined to freeze- dried tendon on angiogenic enhancement in early ACL (anterior cruciate ligament) reconstruction. Methods 14 dogs were used in the experiment. 100 ng/mL bFGF was combined to freeze- dried tendon and then transplanted into one side knee to substitute the original ACL, while only freeze- dried tendon was used in the transplantation at the other side as control. The ACLs were harvested each week from 1 to 6 weeks from the 14 dogs respectively. The histological observation with HE staining was done under microscope to mainly observe the angiogenesis in the transplanted ACL. Results Neovascularization occurred at 2nd to 3rd week and reached the peak at 4th to 5th week postoperatively at the experimental sides. By contrast, the neovascularization occurred at 4th to 5th week postoperatively at the control sides. Conclusion The time of neovascular formation and the depth of vascular penetration into the tendon in the group of bFGF combined to freeze- dried tendon are superior to those in the control group.
4.Arthroscopic internal fixation of avulsion fracture of the anterior cruciate ligament from the tibial eminence with stainless steel wire guided by meniscal stitching needle

Objective To investigate the technique and efficacy of arthroscopic internal fixation of avulsion fracture of the anterior cruciate ligament(ACL) from the tibial eminence with stainless steel wire guided by meniscal stitching needle.Methods Fifteen cases of avulsion fracture of the ACL from the tibial eminence were treated by arthroscopic internal fixation with stainless steel wire guided by meniscal stitching needle from December 1999 to August 2005.The Lysholm scoring scale system was used to evaluate knee functions before and after surgery.Regular X-ray plain films at AP and lateral views were conducted to detect the bony healing of avulsed fragments.Results The operation was completed within 30 min.No complications of intraarticular infection,iatrogenic injury,fibroarthritis,or non-union of fracture were found.The 15 cases were followed for 6~54 months(mean,20.9?15.0 months).X-ray films revealed good bone union in all the 15 cases.The Lysholm scores were improved from 19.1?15.2(range,10~56) preoperatively to 97.5?3.7(range,91~100) postoperatively,with significant statistical difference by the student-t test(t=18.483,P=0.000).Wire breakage was found in 2 cases at 8 and 14 months after operation,respectively.But the bone union in the 2 cases was not affected.Conclusions Arthroscopic internal fixation with stainless steel wire guided by meniscal stitching needle in the treatment of avulsion fracture of the anterior cruciate ligament from the tibial eminence has advantages of simplicity,broad indications(from type Ⅱ to Ⅳ injuries),minimal invasion,and rapid postoperative recovery.
5.Study of the Changes of Endothelin,Tumor Necrosis Factor in Different Stress-related Gastric Mucosal Disease

Objective To observe the changes of Endothelin,Tumor Necrosis Factor in different stress; and to explore the corelation between the changes of Endothelin,Tumor Necrosis Factor and the occurrence of stress-related gastric mucosal disease.Methods The models of the acute methamidophos poison and the acute myocardial infarction in rats were made.The gastric ulcer index was counted and the content of blood plasma Endothelin and blood serum Tumor Necrosis Factor were checked.Results Models of the acute methamidophos poison and the acute myocardial infarction in rats were set up successfully. The gastric ulcer index of rats in the stress groups increased.The blood plasma Endothelin and blood serum Tumor Necrosis Factor in the stress group were significantly higher than those in the control group.Conclusion The gastric mucosal disease under different stress was observed with the gastric mucosal ulcer index.The blood plasm Endothelin and blood serum Tumor Necrosis Factor under different stress were increased,suggesting they play an important role in this phenomenen.
6.Efficiency comparison of Esomeprazole and Omeprazole in the treatment of gastric ulcer

Objective To compare the efficacy of esomeprazole and omeprazole in the treatment of gastric ulcer, and to provide a reference for the clinical treatment of gastric ulcer.Methods80 cases of gastric ulcer patients in our hospital from February to April 2014 were randomly divided into observation group and control group, with 40 cases in each group.The patients in the control group were treated with omeprazole,the patients in the observation group were treated with Esomeprazole, patients received three courses of treatment,the changes of body after treatment in two groups were observed, the degree of stomach pain and the changes of the condition were analyzed, the healing degree of gastric ulcer was observed, the clinical therapeutic effect of two groups was analyzed.ResultsAfter treatment, the recovery rate of the observation group was 100%, significantly higher than the control group 90%, the difference was statistically significant (P<0.05).The scores of pain in the observation group and the drug use score were significantly lower than those in the control group (P<0.05).The adverse reaction rate of the observation group was significantly lower than that of the control group, the difference was statistically significant (P<0.05).ConclusionThe efficiency of Esomeprazole in the treatment of gastric ulcer is better than omeprazole, patients with high satisfaction, can effectively shorten the wound healing time, reduce patient treatment time, reduce medical costs, and adverse reactions light.
7.Prostaglandin E_2 and omeprazole in treating poisoning related acute mucosal lesion

Objective To determine the effects of prostaglandin E_2 alone and the combination with omeprazole for poisoning related acute mucosal lesion. Methods Forty-two healthy SD rats were randomly divided into 7 groups: control group (control), methamidophos poisoning model group (intox), 10 mg/kg omeprazole group (ome1), 50 mg/kg omeprazole group (ome2), 10 μg/kg prostaglandin E_2 group (PG1), 50 μg/kg prostaglandin E_2 group (PG2), 10 μg/kg prostaglandin E_2 combined with 10 mg/ kg omeprazole group (union). The gastric ulcer index was counted and the content of serum cholinesterase, endothelin, and tumor necrosis factor were measured, and dependability was analyzed. Results There was no obvious difference between the prostaglandin group and the poisoning model group in gastric ulcer index, endothelin, and tumor necrosis factor. The omeprazole alone and the combination treatment could improve the acute gastric mucosa lesion induced by methamidophos poisoning. Conclusion Prostaglandin E_2 alone can not obviously improve the acute gastric mucosa lesion by methamidophos poisoning. The combination with omeprazole may be a promising treatment.
8.Effect of BQ123 on the voltage-gated K~+ current of pulmonary artery smooth muscle cells of rats exposed to chronic hypoxia

AIM: To study the effect of BQ123 on voltage-gated K+ current in pulmonary artery smooth muscle cells(PASMCs) from chronic hypoxic rats.METHODS: Twelve age and body weight matched Wistar rats were randomly divided into control and chronic hypoxic group.Single PASMCs were obtained with acute enzyme(collagnaseⅠ plus papain) dispersing method.Using the whole cell patch-clamp technique in freshly isolated PASMCs from normorxic and hypoxic rats,the effects of ET-1 and BQ123,a selective ETA receptor antagonist,on voltage-gated K+ current were recorded.RESULTS:(1) ET-1(10-8 mol?L-1) caused inhibition of K+ current in PASMCs from normoxic and hypoxic rats.The effect of ET-1 on K+ current in PASMCs from hypoxic rats was greater than that from normoxic rats [+50 mV,percent inhibition were(71.04?6.58)% and(60.21?5.32)%,respectively,P0.05,n=5),nor ETA receptor blockade had change of ET-1 mediated IKV inhibition.(3) In chronic hypoxic PASMCs,BQ123 significantly reduced the effect of ET-1 mediated IKV inhibition,from(28.49?6.69) pA/pF to(74.19?9.74) pA/pF at +50 mV(P

AIM: To elucidate the effect of ET-1 on the expression of voltage-gated K~+ channel ? subunits Kv2.1, Kv9.3 in pulmonary artery smooth muscle cells in rats. METHODS: The mRNA expression of Kv2.1, Kv9.3 in the 2nd, 3rd, 4th subculture of intrapulmonary artery smooth muscle cells isolated from Wistar rats, which exposed to either normoxia or chronic hypoxia, were detected with reverse transcription-PCR (RT-PCR). At the same time, PASMCs were treated with BQ123, an ET_A receptor antagonist. RESULTS: The expression of Kv2.1 and Kv9.3 gene were found in the subculture PASMCs of rats exposed either to normoxia or chronic hypoxia. AIM: To investigate the role of potassium channels in the regulation of intracellular free calcium concentration (_i) of pulmonary artery smooth muscle cells (PASMCs) in rats. METHODS: The fluorescence Ca 2+ indicator Fura-2/AM was used to observe _i of rat PASMCs in normal and chronic hypoxic condition. The influences of potassium channels on PASMCs proliferation were assessed by MTT assay. RESULTS: 1. In normoxic condition,_i was (156.91?8.60) nmol/L,and in hypoxic condition,_i was (294.01?16.81) nmol/L. 2. In normoxic condition,the voltage-dependent K +-channel antagonist 4-aminopyridine (4AP),but not the Ca 2+ -activated K +-channel antagonist tetraethylammonium (TEA) and the ATP-sensitive K +-channel antagonist glibenclamide (Glib) increased _i. 3.
