1. Therapeutic effect of 153sm-clodronate on nude mice orthotopically transplanted with human osteosarcoma

Objective: To observe the therapeutic effect of 153Sm-clodronate on nude mice orthotopically transplanted with human osteosarcoma. Methods: Severe combined immune deficiency (SCID) mice were subcutaneously inoculated with human osteosarcoma cell line HOS-8603. The formed tumors were then transplanted into BALB/c nude mice at the tibia for passage; the tumors were harvested for establishing orthotopical mice model of human osteosarcoma. Mice treated with 153Sm-EDTMP and clodronate group served as controls. The distribution of 153Sm-clodronate was determined 24 h later. The appearance, tumor weights, X-ray examination, histological observation, and survival of mice were compared between the 3 groups. Results: It showed that the distribution of 153Sm-clodronate in the lesion bone was much higher than that in other organs, with the radioactive ratio of lesion bone to blood being 4 627.1 : 1, lesion bone to normal bone being 10.8 : 1, and normal bone to liver being 8. 4 : 1. 153Sm clodronate treated mice had a slighter osteolytic reaction and a longer survival period, but the tumor mass was not significantly different compared with those of the control group. Conclusion: 153Sm-clodronate gathers at the lesion bone and it has good therapeutic effect on the mice model of human oscosarcoma, making it a promising agent for treatment of bone tumor.
2.Combined fibrinogen concentration and neutrophil-lymphocyte ratio as a pro-gnostic marker of colorectal cancer

Objective:To explore the significance of combined fibrinogen concentration and neutrophil-lymphocyte ratio as a prognostic marker of colorectal cancer.Methods:A retrospective analysis of the hospital in June 2010-June 2011 of received and accepted the surgical treatment of colorectal cancer patients,respectively analysis the relationship NLR and FIB with the pathological characteristics of colorectal cancer,neutrophils and lymphocytes ratio (NLR) and the concentration of fibrinogen (FIB) were combined to form a index (FIB-NLR).250 patients with colorectal cancer recording the scores were divided into three groups,patients NLR acuity 2.95 and FIB≥348 mg/dl as FIB-NLR 2 groups,NLR acuity 2.95 and FIB < 348 mg/dl or NLR < 2.95 and FIB acuity 348 mg/dl for 1 group,NLR < 2.95 and FIB < 348 mg/dl of 0 group,and analyzed three groups with invasion depth,clinical staging and lymph node metastasis of colorectal cancer,nerve invasion,distant metastasis and histological grades.And the three groups respectively for survival analysis,and carried on the comparison to the survival rates of three groups.Results:Moderate and advanced and with lymph node metastasis of colorectal cancer patients was significantly higher than the earlier stage and no lymph node metastasis in patients with NLR,statistically significant difference (P<0.001);tumor infiltration depth deeper,nerve invasion and distant metastasis of the NLR value was significantly higher in patients with infiltrating depth shallow,without nerve invasion and distant metastasis in patients with NLR values,the difference was statistically significant (P=0.006,P=0.002,P=0.007).Moderate and advanced stage,lymph node metastasis and distant metastasis of colorectal cancer patients with the FIB values were significantly higher than the early and without lymph node metastasis and distant metastasis of colorectal cancer patients FIB values,the difference was statistically significant (P<0.001),the deeper the infiltration depth and colorectal cancer patients with neural infiltrates FIB values significantly higher than the infiltration and shallow depth,of FIB values for patients with nerve infiltration,the difference was statistically significant (P=0.015,P=0.012).NLR and FIB in tumor histologic stage,age,gender,tumor location,had no significant association (P>0.05). The patients with later clinical stage of colorectal cancer,the deeper the infiltration depth,lymph node metastasis,distant metastasis had a high FIB-NLR score than infiltration and shallow depth,without lymph node metastasis and distant metastasis,no nerve infiltration patients was high,the difference was statistically significant (P<0.001).Survival analysis found that the higher the score of patients the 5-year survival rate was lower,the difference had statistical significance (P=0.001).Conclusion:Neutrophil-lymphocytes ratio (NLR) and fibrinogen (FIB) joint form a index FIB-NLR may be a potential progression effective index which estimate the progression of colorectal cancer.
3.Epidemiological Analysis of Leprosy in Zhejiang Province

Objective To analyze the epidemiological trends of leprosy and evaluate the effectiveness of control in Zhejiang Province. Methods A retrospective study was conducted. Results By the end of 1999, 16 262 leprosy patients had been registered, of them 11 935 had been cured and only 97 were still remained. In 1999, active cases decreased by 99.02% over 1973. The epidemiological trends of leprosy showed the increase of mean age at onset and type ratio, and the decrease of incidence in children. Conclusion The epidemicity of leprosy has been continuously dropping in Zhejiang Province due to the sustained effort and effective control.
4.Effects of hyperventilation on jugular bulb pressure and optic nerve sheath diameter during robotic laparoscopic radical prostatectomy

Objective To observe the effects of hyperventilation on jugular bulb pressure and optic nerve sheath diameter during RALRP. Methods Twenty patients undergoing elective RALRP were enrolled in this study. After hyperventilation of General anesthesia,we monitored and recorded changes of Heart rate(HR),mean arterial pressure (MAP),end-tidal carbon dioxide (ETCO2),arterial oxygen tension (PaO2),arterial carbon dioxide tension(PaCO2),peak airway pressure(PIP),JBP and ONSD in 5 minutes after intubation(T0),Trendelenberg posture in 15 minutes (T1),the established pneumoperitoneum in 30,60,90 minutes (T2,T3,T4) and termination of pneumoperitoneum in 15 minutes in supine position(T5)respectively. Results Compared with T0, JBP increased at T1 ~ T5(P < 0.05). ETCO2 and PaCO2 of T1 ~ T5 were lower than those of T0(P < 0.05)during hyperventilation. After establishment of Trendelenburg posture and pneumoperitoneum,PIP values increased at T1~T4 than that at T0 (P < 0.05),However,ONSD increased from T3 to T5 compared with T1 to T2 (P < 0.05). Conclusion Through over-ventilation,pneumoperitoneum and Trendelenburg posture,JBP and PIP can increase, and ONSD becomes wider evendually.
5.Effects of shenning on rats with membranous glomerulonephritis

AIM To investigate effects of shenning on nephrotic rats. METHODS Adult male wistar rats were randomly devided into normal control group, pathological pattern group, high-dosage shenning group,middle-dosage group,low-dosage group,shenfukang group and prednisone group. Rats with membranous glomerulonephritis were induced by injection of cation bovine serum albumin into tail vein. The rats were treated respectively for 32 days by gavage. Urinary protein was observed once a week after 2 weeks. After 7weeks ,plasma albumin,renal histology,parameters of renal function and prostaglandins were examined. In addition TXB 2/6-Keto-PGF 1? in serum were also examined. RESULTS The urinary protein,serum cretinine level,BUN and TXB 2/6-Keto-PGF 1? can be reduced in shenning groups compared with pathological pattern group. Plasma albumin was significantly increased in high-dosage shenning group. CONCLUSION High-dosage and middle-dosage shenning can significantly decrease urinary protein in rats with membranous glomerulonephritis. High-dosage shenning can increase the plasma albumin level in rats with membranous glomerulonephritis.
6.Effects of trehalose on bcl-2 and bax mRNA expression in the cryopreserved sternum

BACKGROUND: Studies have demonstrated that trehalose possesses protective effects on cyropreserved sternum. But the mechanism of action remains poorly understood. OBJECTIVE: To investigate the effects of trehalose on bcl-2 and bax mRNA expression in cryopreserved sternum. METHODS: Four groups of freshly prepared solution were used: low-potassium dextran (LPD), LPD + dimethyl sulfoxide (DMSO), LPD + trehalose, LPD + DMSO + trehalose. Rat sternum was cut and then immediately cryopreserved in the tubes containing each group of solution. Fresh rat sternum tissue and 4 groups of samples cryopreserved for 120 days were taken and bcl-2 and bax mRNA expression in fresh and cryopreserved sternum was detected using reverse transcription-polymerase chain reaction.RESULTS AND CONCLUSION: bcl-2 mRNA expression in the LPD + trehalose group was significantly higher, but bax mRNA expression was significantly lower, than in the LPD, LPD + DMSO groups (both P < 0.01). LPD + DMSO + trehalose group showed highest bcl-2 mRNA expression and lowest bax mRNA expression, which were basically similar to fresh bone tissue (P > 0.05). These findings indicate that trehalose may protect cell activity in cryopreserved sternum by enhancing bcl-2 mRNA expression and inhibiting bax mRNA expression, and trehalose together with DMSO shows better protective effects.
7.Immunocytochemistry analysis of human neutrophil defensins

Our laboratory isolated three defensin molecules from the circulating neutrophils of healthydonors.The three molecule mixture was conjugated with BSA and immunized New Zealand rab-bit.ELISA showed that the rabbit antisera was of a very high titters,which were specific for hu-man defensins.Immunoperoxidase staining with this rabbit antisera indicated that normal humanneutrophils,acute myeloid leukemia cell lines KG1 and HL-60 were highly positive.In con-trast,normal mononuclear leukocytes,erythrocytes,erythroleukemia cell line K562 were all neg-ative,even rabbit and guinea pig neutrophils had no response to this rabbit anti-human de-fensins antisera.Our preliminary experiments suggested that the rabbit antisera was highly spe-cific for human defensins.Probably,defensin is a differentiation antigen of granunocyte lineage,and antibody against human defensins would be a candidate immunological reagent specific for a-cute myeloid leukemia.Further work for this should be done.
8.Establishment and functional analysis of a cell model expressing the C-type lectin receptor Langerin

Objective To establish a cell model expressing the Langerhans cell-specific C type lectin receptor Langerin in vitro.Methods The cDNA of Langerin was obtained by PCR and cloned into a eukauotic green fluorescent protein (EGFP) expression vector pEGFP-C 1 with EGFP located in the N terminal region of the Langerin gene.Then,the recombinant plasmid was transfected into a human embryonic kidney carcinoma cell line HEK293T.Subsequently,laser confocal microscopy was performed to observe the expression of EGFP-Langerin fusion protein,and flow cytometry to measure the expression of Langerin.Laser confocal microscopy was also conducted to visualize the recognition and endocytosis of dust mite antigen (nDer p 2) by Langerin.Results PCR and Western blot confirmed the successful transfection of HEK293T cells with the recombinant plasmid as well as the expression of Langerin in the transfected cells.As flow cytometry revealed,the expression level of Langerin in transfected HEK293T cells was increased by 43% compared with untransfected cells.Laser confocal microscopy showed that green fluorescein-labeled Langerin was successfully expressed,and could bind to and endocytose the red fluorescein-labeled antigen nDer p 2.Conclusions The fusion protein EGFP-Langerin expressed in HEK293T cells showed the distribution characteristic of cell surface receptors,and could bind to and endocytose allergens.

Objective:To promote clinical innovation and patent transformation, Ningbo First Hospital built a medical device innovation and transformation management information platform to optimize the supply of clinical innovation and transformation resources and explore a new path of clinical innovation and transformation management.Methods:A series of processes of medical device innovation research, patent declaration, and transformation were managed by the cloud data platform based on Springboot micro-service architecture. The functions and practices of the medical device innovation and transformation management information platform were elaborated on in detail, and the problems in the platform construction were discussed.Results:The medical device innovation and transformation management information platform has been accepted and recognized by clinical care, enterprises, and third-party agents for shortening the research and development and transformation time of innovative projects.Conclusions:The medical device innovation and transformation management information platform can promote the output and achievement transformation of clinical innovative projects.
10.In vitro binding ability of transglutaminase 3 to dendritic cell-specific intercellular adhesion molecule 3-grabbing nonintegrin

Objective To evaluate the recognition and uptake of transglutaminase 3 (TG3) by dendritic cell-specific intercellular adhesion molecule 3-grabbing nonintegrin (DC-SIGN) receptors on the membrane surface of DC-SIGN-transfected human embryonic kidney (HEK) 293T cells and monocytederived dendritic cells (MDDCs).Methods The eukaryotic expression vector pGCMV-enhanced green fluorescent protein (EGFP) containing DC-SIGN gene fragments was transfected into HEK293T cells to prepare DC-SIGN-EGFP-HEK293T cells by using liposome transfection method.CD14+ monocytes were isolated from peripheral blood samples by magnetic bead-based negative selection,and then were induced by granulocyte-macrophage colony stimulating factor (GM-CSF) and interleukin-4 (IL-4) to prepare MDDCs.Laser confocal microscopy and flow cytometry were performed to evaluate the recognition and uptake of TG3 protein by DC-SIGN receptors on the surface of HEK293T cells and MDDCs.MDDCs treated without Alexa Fluor 647 dye-tagged TG3 served as blank control group,and those treated with Alexa Fluor 647 dye alone served as negative control group.Results After co-culture with TG3 for 3 hours,laser confocal microscopy and flow cytometry both showed that TG3 could be recognized by and uptaken through DC-SIGN receptors into HEK293T cells and MDDCs.Flow cytometry also revealed that the binding of TG3 to MDDCs could be partially blocked by DC-SIGN blocking antibodies.Neither the negative control group nor the blank control group showed the recognition and binding of TG3 to HEK293T cells and MDDCs.Conclusion TG3 can serve as a kind of autoantigen to be recognized and bound by DC-SIGN receptors,followed by uptake by dendritic cells.
