1.Effection of immune memory of tuberculosis vaccine on memory T cell in mice

Objective Exploring different strengthening immune strategy on long-term immune memory effects for tuberculosis vaccine lays the theoretical foundation.Methods The fusion protein AMM,adjuvant DDA and BCG-PSN mixed built AMM subunit vaccine.Choose only 50 C57BL/6 mice (SPF),according to the table of random number method was divided into 5 groups and each group 10 only,the experiment 1 group injected mice to phosphate buffer solution (PBS) as control; Experiment 2 mice only injected BCG vaccine (BCG) from early; Experiment 3 mice BCG initial free,in 10 weeks with AMM subunit vaccine strengthen immunity; The four mice from early after BCG,respectively on 8 weeks,10 weeks with AMM subunit vaccine strengthen immune mice one time.Experimental 5 groups of mice after BCG initial free,respectively on 6 weeks,8 weeks,10 weeks with AMM subunit vaccine strengthen immune mice a.28 weeks to remove spleen lymphocytes in mice,add 2.5μg/ml Ag85B stimulus 72 h after collecting spleen cell culture supernatant,with ELISA test Ag85B specificity splenic lymphocyte caused IFN ppar-gamma level.Results Spleen cells by antigen Ag85B stimulation,BCG initial free,AMM subunit vaccine strengthen immune once,twice,three times group and PBS compared all had difference (P <0.01 orP <0.05) ;BCG initial free-AMM subunit vaccine strengthen immune once,twice,three times group and BCG compared all had difference (P <0.01 or P <0.05) ; BCG initial free-AMM subunit vaccine strengthen immune once and twice or three times group compared with difference (P <0.01 orP <0.05) ; Initial free-AMM subunit vaccine strengthen immune two group and three times than group difference was statistically significant (P >0.05).Conclusion Mice strong immune strengthened the number is not the more the better,the appropriate number of strengthen the immune to induce the most lasting immunity memory,subunit vaccine strengthen immunization strategy may affect the immune memory is one of the important factors.
2.Study on the Thermic Effect of Food of Patients With Chronic Obstructive Pulmonary Disease

Objective To study on the thermic effect of food (TEF) of patients with chronic obstructive pulmonary disease(COPD).Methods Basal energy expenditure (BEE) of 12 male cases and resting energy expenditure (REE) of 11 male cases of COPD were detected by indirect calorimetry with metabolic cart. There were no significant difference in sex, age, height and body weight between two groups' e.g:TEF=REE-BEE.Results BEE and REE were 1114?208kcal/d,1433?136kcal/d respectively, the latter was higher 28% than BEE.Conclusion TEF increases significantly which may contributes to malnutrition and non-response to nutritional support in patients with COPD.
3.Effects of rosiglitazone on the atherosclerosis and oxidative stress in diabetic rabbits

Rosiglitazone was used for intervention of atherosclerosis in diabetic rabbits.The results showed that the intima/medium thickness ratio,cross section area of plaque,and the expressions of NADPH oxidase p22phox,gp91 phox were decreased;while the total anti-oxidative ability was increased after administration of rosiglitazone as compared with the non-intervention group(all P<0.05).Compared with rosiglitazone treatment group,serum hepatocuprein leveI in rosiglitazone prevention group was increased,while serum malonaldehyde level decreased(both P<0.05).This study suggests that rosiglitazone may have the effect of reducing the oxidative stress and the formation of atherosclerotic plaque in diabetics.
4.Influence of health education on Hying quality and medical compliance of patients suffering from gynecologic malignant tumor

Objective To explore the influence of health education on living quality and medical compliance of patients suffering from gynecologic malignant tumor. Methods 69 cases of patients suffer-ing from gynecologic malignant tumor were divided into the observation group (35 cases) and the control group(34 cases). The control group was conducted with routine instructions of treatment and nursing, while individualized health education was given to the patients of the observation group and their relatives based on routine treatment and nursing. The living quality and medical compliance were observed in two groups separately on admission and half year after treatment. Results The living quality of the observation group was better than that of the control group as well as the medical compliance. Conclusions Individualized health education can improve the living quality and medical compliance of patients suffering from gyneco-logic malignant tumor to ensure the treatment effect of disease and is feasible as well.
5.Screening and identification of anomously expressed proteins in colon mucosa in diarrhea-predominant irritable bowel syndrome

Objective To screen and identify the anomously expressed proteins at protein level in colon mucosa of diarrhea-predominant irritable bowel syndrome(D-IBS)patients and healthy individuals.Methods Four D-IBS patients and four healthy individuals were included in the study.Biopsies of mucosa at the ileocecal junction and sigmoid colon were performed via colonoscopy.The specimens were cleaned with ice cold saline which contained 0.1% PMSF,and were snap frozen in liquid nitrogen.The two-dimensional gel electrophoresis(2-DE)was used to screen the anomously expressed proteins,and the two proteins which presented the greatest changes in each group were identified by mass spectrometry method.Results Proteomics maps of human colon mucosa were obtained successfully with 2-DE.Average protein spots were 336 in normal control group,and the matching rate among them was 92%,while in D-IBS group,the average protein spots were 426,and the matching rate was 95%.The average matching rate between the test group and the control group was 74%.It was found that the volume value of 24 proteins spots changed more than two folds.The protein expression was found to decrease in 3 spots and increase in 21 spots.Four proteins were chosen for analysis with mass spectrometry analysis.The two up-regulated proteins were identified as Immunoglobulin J chain and heat shock protein 27,while the other two down-regulated proteins were identified as hemoglobin beta subunit and fructose-bisphosphate aldolase A.Conclusion With 2-DE,proteomics maps of human colon mucosa have been obtained successfully.The proteome expressions of colon mucosa in D-IBS patients are remarkably different from that of the healthy individuals.The four identified proteins may play a certain role in the pathogenesis of D-IBS.
6.Correlation of Immune Infertility with Chronic Genital Tract Inflammation and Intervention of Chinese Medicine

【Objective】To explore the correlation of antisperm immune infertility(AII) in females with chronic genital tract inflammation and to observe the therapeutic effect of Chinese medicine of Qingshi Huayu Mixture(QHM) and Yishen Huoxie Prescription Ⅰ(YHPⅠ) for AII.【Methods】Fifty-eight AII patients were screened from 200 infertility patients,and were randomized into three groups.Group A(N=18) was treated with YHPⅠ,group B(N=20) with QHM and group C(N=20) with western medicine.Three months constituted one treatment course.The incidence of positive antisperm antibody(AsAb+)combined with chronic genital tract inflammation,therapeutic effect,pregnancy rate,the content of serum AsAb,as well as the changes of lymphocyte subtypes,natural killer(NK) and interleukin 6(IL-6) were observed.The correlation of the above indexes with serum AsAb content was also analyzed.【Results】(1) The incidence of positive antisperm antibody(AsAb+)combined with chronic genital tract inflammation: There were 96(48%)with AsAb+ in 200 patients,and 62(64.58%) with AsAb+ in 122 combined with chronic genital tract inflammation;there were 34(35.42%)AsAb+ patients in 78 without inflammation,the difference of AsAb+ incidence was significant between the inflammation patients and the non-inflammation patients(P0.05).There were 2 in 5 cured patients of group A,2 in 7 of group B and 1 in 7 of group C being pregnancy.(3) Content of serum AsAb: serum AsAb content was decreased in the three groups after treatment,the difference being significant as compared with that before treatment(P0.05).(4) The changes of lymphocyte subtypes,NK and IL-6: CD8+ was increased,CD4+/CD8+ and NK were decreased in groups A and B,the differences being significant as compared with those before treatment and those in group C after treatment(P
7.Local decompression and selective interbody fusion with pedicle screw fixation technique for the treatment of degenerative lumbar scoliosis

BACKGROUND:Conservative treatment is mostly invalid in treatment of severe degenerative lumbar scoliosis-induced low back pain, so surgical intervention is general y required. Repair methods are decompression, rigid fixation, and selective interbody fusion for the“responsible vertebra”to re-establish normal lumbar curvature, spinal balance and good stability. OBJECTIVE:To investigate clinical curative effect of local decompression and selective interbody fusion with pedicle screw fixation technique for the treatment of degenerative lumbar scoliosis. METHODS:Data of 57 cases of degenerative lumbar scoliosis from Armed Police Corps Hospital of Hunan Province from January 2011 to January 2014 were retrospectively analyzed. According to clinical characteristics of the patients, selective fusion, local decompression and pedicle screw fixation were used for repair. The imaging analysis and Suk criteria were used to evaluate fixation, decompression and bone fusion. Japanese Orthopaedic Association scores were utilized to assess therapeutic effect. RESULTS AND CONCLUSION:(1) Operation time was 157-255 minutes, averagely 176 minutes. Amount of bleeding was 480-1 700 mL, averagely 835 mL. Postoperative drainage volume was 140-210 mL, averagely 155 mL. (2) 57 patients were fol owed up for 12-38 months. Postoperative X-ray revealed that Cobb’s angle was (3.8±2.1)° on the coronal plane after treatment, and the correction rate of lordosis was 54%. The sagittal Cobb’s angle was (6.3±10.5)°, and the correction rate of lordosis was 35%. According to Suk standard, the fusion rate of vertebra was 95%. According to low back pain score system recommended by Japanese Orthopaedic Association scores, the effective rate was 91%at the end of fol ow-up. (3) There were 2 cases of cerebrospinal fluid leakage, 1 case of nerve injury, and 1 case of pedicle screw failure, no infection, no blood vessel injury. (4) These findings suggest that according to imaging data and self clinical features, local decompression and selective interbody fusion with pedicle screw fixation technique for the treatment of degenerative lumbar scoliosis can rebuild and stabilize degenerative lumbar scoliosis in the coronal and sagittal planes to different degrees, with satisfactory clinical curative effect.

9.Construction of Rat Extracellular Signal-regulated Kinase 1 Gene 3' Untranslated Regions Dual-luciferase Reporter Plasmids and Effect of rno-miR-15b-5p on Its Activitiy

Objective To construct dual-luciferase reporter plasmids containing the wild type and mutant rat extracellular signal-regulat-ed kinase 1 (ERK1) gene 3' untranslated regions (UTR) which were used to detect rno-miR-15b-5p's putative target gene. Methods The rat ERK1 gene 3' UTR fragment was amplified by polymerase chain reaction (PCR) from PC12 cell cDNA and cloned into pmiR-RB-ReportTM vector. The mutant rat ERK1 gene 3' UTR fragment was obtained by overlap PCR and inserted into pmiR-RB-ReportTM vector. Successful wild type and mutant recombinant plasmids were confirmed by DNA sequencing. PC12 cells were co-transfected with rno-miR-15b-5p mim-ic and pmiR-ERK13' UTR or pmiR-ERK1-mut 3' UTR and then analyzed by dual-luciferase reporter assay system. The achieved mutation sequence of the target site TGCTGCT was mutated to CGAACGT and GTACACG, respectively. Results The wild-type reporter vector pmiR-ERK13' UTR and the mutant reporter vector pmiR-ERK1-mut 3' UTR were successfully constructed. The rno-miR-15b-5p mimic de-creased the activity of pmiR-ERK13' UTR plasmid (P<0.001) but did not decrease the activity of pmiR-ERK1-mut 3' UTR plasmid. Conclu-sion The recombinant pmiR-ERK13' UTR and pmiR-ERK1-mut 3' UTR plasmids were constructed successfully, and luciferase activities demonstrated that the 3' UTR of ERK1 gene might be a potential target of rno-miR-15b-5p.
10.Surface labeling of bone marrow mesenchymal stem cells by biotin-streptavidin

BACKGROUND:Currently, there is a lack of efficient, non-invasive way to transplant stem cels to the target organ or tissue. Exploring a way to guide targeting transplantation of stem cels and to improve the efficiency of stem cel homing is now one of focuses in the field of stem cels research.
OBJECTIVE: To establish a simple and feasible method to chemicaly modify the cel surface using biotin-streptavidin reaction system, and to evaluate the efficiency of this method to label bone marrow mesenchymal stem cels (BMSCs) and its effects on cel biological functions.
METHODS: Passage 3 BMSCs were obtained by whole bone marrow culture method and verified by flow cytometry. Biotin, streptavidin, sulfonated biotin-N-hydroxy-succinimide were used to equip the adhesion molecule ligand, sialyated LewisX (SLeX), to the BMSCs surface. The labeling rate of BMSCs was assessed using fluorescence microscope, the vitality of BMSCs was evaluated by trypan blue staining, and the proliferation of BMSCs was evaluated by cel counting kit-8 assay. Adipogenic and osteogenic inductions were used to evaluate the effect of the method on the multi-differentiation function of BMSCs.
RESULTS AND CONCLUSION: After culture for 2 weeks, passage 3 BMSCs were obtained and confirmed by expressing CD90, CD29 and lack of CD34, CD45. Biotin, streptavidin, sulfonated biotin-N-hydroxy-succinimide were successfuly used to equip sialyated LewisX (SLeX) to the BMSCs surface and had minor effects on the vitality, proliferation, and differentiation of BMSCs. This method was simple for surface modification and had a high modification rate of 88%. The homing of BMSCs modified by this method to the target organ or tissue could be greatly enhanced. Therefore, this method potentialy could have extensive and important applications.
