Objective To study the expression of MAGE-3 in colorectal cancer (CRC) with different differentiation grades. Method Immunohistochemical technique for MAGE-3 antigen was performed in paraffin embedded sections of 101 CRC specimens. The relationship between differentiation and Dukes classification of CRC and the expression of MAGE-3 antigen was analyzed. Result MAGE-3 was only expressed in tumor tissues with the total positive rate of 31.7%. The rate and intensity of MAGE-3 in poorly differentiated group were higher than that of the well differentiated group (P0.05). Conclusion MAGE-3 may be a specific molecular differentiation related marker of CRC. Its antigen may have potential use as a new peptide vaccine for immunotherapy for CRC.

OBJECTIVE: To study the changes of serum vascular endothelial growth factor (VEGF) and beta 2-microglobulin levels before and after radiotherapy in 58 patients with nasopharyngeal carcinoma (NPC), and to elucidate the clinical significance of VEGF and beta 2-microglobulin test before radiotherapy. METHODS: Serum VEGF level was measured by sandwich ELISA in 58 patients with NPC and 24 healthy individuals, and the serum beta 2-microglobulin was assayed with time-resolved fluoroimmunoassay. RESULTS: The serum VEGF level in patients with NPC was (174.0+/-130.0) ng/L, higher than (134.1+/-66.6) ng/L of the healthy subjects, but the difference was not significant (P>0.05). The serum VEGF level was higher in T4 and IV a patients with NPC. No significant differences in the levels of serum VEGF were found among various N classifications in the patients with NPC. Patients with the serum VEGF level higher than 267.3 ng/L (mean+2 s of the serum VEGF level in the healthy individuals) had a shorter metastasis-free survival time (P<0.05). Although the patients with high beta 2-microglobulin level had a shorter survival time, the difference was not significant. CONCLUSION: The NPC patients with high serum VEGF level have a poor prognosis.

Total hip arthroplasty (THA) has developed rapidly in recent years, which reconstructs the joint function of some terminal hip joint diseases and obtains favorable efficacy. However, the stress load alters in the proximal femur following THA, leading to "adaptable bone reconstruction", which is characterized by thin cortical bone, decreased bone density, and bone loss surrounding prosthesis, in particular calcar femorale. This paper analyzed the effects of cement and cementless prosthesis, coated prosthesis, and THA approaches on stress shielding in the proximal femur to reduce stress shielding effect by improving prosthesis design, low elastic modulus materials, decreased section area of the prosthesis, cement prosthesis, appropriate coated materials and improved THA approaches.

Objective To study the effect of matrine on the cell cycle and DNA content of fibroblasts derived from hypertrophic scarring tissues. Methods The fibroblasts were derived from 3~5 generations of the cultured hypertrophic scarring tissues. After the cells were treated with different concentration of matrine, the population doubling time was calculated and the cell cycle and DNA content were measured with flow cytometry. The fibroblasts treated with normal saline were as control. Results Compared with the control group, the distributions in cell cycle showed that the percentage of G 2 M and S phase cells decreased, and the percentage of G 0 G 1 phase cells increased significantly ( P

Journal of Environment and Health 1989;0(06):-
Asthma is a chronic respiratory disease with various etiologic factors.Asthma is believed as a result of interaction between gene variation and environmental factors.Asthma increased rapidly worldwide in recent 40 years, air pollutants are believed to be the important triggers, crossover design and multi-pollutant models were used in the further studies.Although some efforts had been made in the mechanism of asthma triggers, they were limited to the lab studies.Studies in asthma related SNP and its interaction with air pollutants will be a revised method in mechanism researches.Based on epidemiological studies, in vivo and in vitro studies, the biological effects and mechanisms of air pollutants on asthma were discussed and evaluated in this paper.

A remarkabale inhibitory activity was exhibited by the aqueous extract from Rhizoma smilacisglabrae(RSG) against both picryl chloride(PC)- induced contact dermatitis and sheep red bloodcells (SRBC)- induced footpad reaction. The effect of RSG was displayed more distinctlywheng given after than before the 2nd antigen challenge.RSG also showed a marked anti-in-flammatory activity against xylene- induced ear and egg whiteinduced footpad edema.Addi-tionally,RSG did not show a potable influence on IgM- and IgG-PFC counts against SRBC inmice. An increase but not decrease in serum hemolysin level, however, was observed in both groupsof RSG,peralleled with the finding that hemolytic plaques in PFC test were obviously biggerthan those in control. These results suggest that RSG has a selective activity to suppress the cellularimmune response without inhibiting the humoral immune response. The suppression to cellular im-munity by RSG may be presented mainly through affecting the inflammatory process after lym-phokine release.

Objective To study the effect of neuroepithelial cell transforming gene 1 (Net1) on the cellular radiosensitivity and underlying mechanism. Methods Real-time quantitative PCR was used to measure the variations in Net1 expression level upon irradiation.Radiosensitivity was analyzed by colonyforming assay after Net1-siRNAs.Net1-associated proteins were identified by co-immunoprecipitation.Results The Net1 mRNA level in the cells was increased significantly (t =-10.52,P < 0.05) after irradiation.Compared to the control group,siRNA-mediated silencing of Net1 enhanced cell radiosensitivity (t =15.31,11.65,P <0.05).Net1 was found to interact with Ku70,Ku80 and DNA-PKcs under either normal conditions or after irradiation. Conclusions Net1 could protect cells from irradiation by interaction with DNA repair proteins in non-homologous end joining pathway.

Objective To identify the types, frequency and risk factors of major complications using microwave ablation (MWA) to treat liver malignancies in a single center. Methods Using the electronic medical record system, patients with liver cancer who were treated with MWA from January 2010 to November 2015 were retrospectively studied, and the risk factors of severe complications were analyzed. Results Of the 7 403 patients who were treated with MWA (12 558 ablations).96 cases experienced severe complications (1.3%) and 5 patients died (0.07%).The complications included liver abscess (n = 34,0.46%),pleural effusion (n = 20,0.27%),bile duct injury (n = 15,0.20%),hemorrhage (n = 6,0.08%),liver failure (n = 6,0.08%),extrahepa-tic organ injuries (n = 5,0.07%),cancerous cells implantation of abdominal walls (n = 2),cardiac arrest (n = 1),vascular thrombosis (n = 1),renal insufficiency (n = 1),and patients with other associated diseases which developed after the MWA therapy (n =2).Metastatic liver cancer and earlier treatment of MWA sessions were associated with a higher major complication rate (P < 0.05).Conclnsions MWA is a well-tolerated local treatment for liver cancer, with an acceptably low incidence of severe complications.Liver abscess was the most common postoperative severe complication. The surgeons' experience and tumor type were associated with the incidence of severe complications, however, not relavant with the frequency of preoperative ablations.Appropriate measures can be taken to effectively reduce the risk of serious complications.
