1.The ouabain signal transduction and its role in cell growth and death

The ouabain is a member of the cardiotonic steroids (CT S) that specifically bind to Na+,K+-ATPase. Binding of ouabain to Na+,K +-ATPase causes rapid activation of Src family kinases, and forms a Na+,K +-ATPase-Src complex as a active binary receptor to phosphorylate and assem ble other proteins into different signaling modules.Oubain stimulates hypertroph ic growth in cardiac myocytes and proliferation in smooth muscle cells, it also induces apoptosis and a newly recognized "hybrid cell death" of concurrent apopt osis and necrosisin the same cells.
3.Treatment outcome of patients with idiopathic sudden sensorineural hearing loss and concomitant benign paroxysmal positional vertigo.

OBJECTIVE:
To evaluate the treatment outcome of patients with idiopathic sudden sensorineural hearing loss (ISSHL) with concomitant benign paroxysmal positional vertigo (BPPV).
METHOD:
Out of 252 ISSHL patients, 96 patients were diagnosed with complicating vertigo and examined using electronystagmography. All patients were divided into three groups, namely, ISSHL without vertigo group (n=156), ISSHL with non-BPPV vertigo group (n=70) and ISSHL with BPPV group (n=26). All patients received daily intravenous infusion of 200,000 U urokinase for 3 consecutive days and 100,000 U for 3 additional days. Concomitant medications included intravenous Ginkgo biloba leave compound and vitamin B6 and intramuscular vitamins B1 and B12 for 2 weeks. Twenty-six patients diagnosed with BPPV on electronystagmography positioning test also received canalith repositioning maneuver.
RESULT:
Vertigo-associated symptoms completely resolved after canalith repositioning maneuver in ISSHL patients with complicating BPPV. With respect to post-treatment hearing, ISSHL patients without vertigo exhibited a similar improvement as compared to those with BPPV, while those with non-BPPV vertigo had a significantly less improvement than those without vertigo and with BPPV.
CONCLUSION
ISSHL patients with complicating BPPV exhibit a relatively favorable outcome with respect to hearing and vertigo-associated symptoms after medical and repositioning treatment.

4.The diagnostic value of 18F-FDG PET/CT in the localization of ectopic adrenocorticotrophic hormone-secreting tumors

Objective To investigate the diagnostic value of 18F-fluorodeoxyglucose (FDG) PET/CT in the localization of ectopic adrenocorticotrophic hormone ( ACTH)-secreting tumors. Methods Seven patients with ectopic ACTH syndrome (confirmed by clinical symptom, biochemical data or bilateral simultaneous inferior petrosal sinus sampling) were included, 4 males and 3 females with age ranged 27-63 years old.All patients were examined with head MR, chest and abdomen CT, abdomen ultrasonography, and whole body 18F-FDG PET/CT. Location, size, and the maximum standardized uptake value (SUVmax) of the tumor were measured. Results 18 F-FDG PET/CT detected three ectopic ACTH-secreting tumors with the sizes 1. 1 cm×1.0 cm (SUVmax =9.4), 1.5 cm×1.1cm (SUVmax =6.4), and 1.6 cm ×1.5 cm (SUVmax =12.0). The pathological findings of the three tumors were thymoma, lung carcinoid, and mediastinal carcinoid respectively. Conclusion 18 F-FDG PET/CT may help the localization of ecto pic ACTH-secreting tumors .
5.An ultrastructural observation of the effect of persistent stress on cancellous bone healing WAN

Objective To explore the effect of persistent stress on the ultrastructure of cancellous bone in the process of fracture healing.Method A transverse osteotomy on both sides of the femoral tuberositas was performed in 20 adult dogs. One side was fixed with a self-designed tuberositas memofy-pressure connector (TMC) which was made of NiTi shape memory alloy, and the other with compressive steel-wire (SW) cerclage.Five animals in each group were sacrificed at 2, 4, 8, 12 weeks after operation and observed under transmission electron microscope.Results The fracture healing was accelerated in the TMC group.The osteoblasts, chondroblasts and collagen fibers at the fracture sites showed significantly different morphology between the 2 groups at every observation time.Most cells and collagen fibers in the TMC group grew along the same direction as the stress, but those in the SW group grew in disorder.Both the number and functional activity of the osteoblasts and chondroblasts were higher in the TMC group than in the SW group.Conclusion Persistent stress can enhance healing and bone reconstruction following a cancellous bone fracture.
6.The control and prognostic factors analysis of acute gastroesophageal variceal hemorrhage

Objective To analyze the correlation between the bleeding control time,treatment and prognosis in acute gastroesophageal variceal hemorrhage and to explore the predictive factors in treatment failure.Methods The clinical data of 309 patients with acute gsstroesophageal variceal hemorrhage was collected.The bleeding control was retrospectively analyzed within and at two hours after treatment,over two hours but less than six hours,over six hours but less than five days,at and after five days.The correlation between bleeding control time and mortality rate was analyzed.The differences in success rate of bleeding control and mortality rate between medication treatment alone and medication combined with endoscopic treatment were compared.Logistic regression mode was used to analyze the predictive factors of treatment failure.Results Of the 309 acute gastroesophageal variceal hemorrhage hospitalized patients,the failure of bleeding control within and at two hours,over two hours but less than six hours and over six hours but less than five days were 79 cases,25 cases and 26 cases respectively and there were one,four and 16 cases who died respectively.The success rate of bleeding control by medication treatment alone was 57.93% (179/309) and the mortality was 10.36% (32/309). The success rate of bleeding control by medication combined with treatment under endoscope was 82.35% (28/34) and the mortality rate was 0.The results of Logistic regression mode analysis indicated that the admission systolic blood pressure (≤90 mm Hg,1 mm Hg=0.133 kPa),increase of total bilirubin,increase of Child-Pugh grade and ascites were the predictive factors of treatment failure.Conclusions The time of bleeding control failure mostly was within two hours.With time extended,the mortality increased. The success rate of bleeding control by medication combined with endoscopic treatment was higher and the mortality was lower.The admission systolic blood pressure lower than 90 mm Hg,increase of total bilirubin,increase of Child-Pugh grade and ascites may be the predictive factors of treatment failure.

8.Expressions and roles of long non-coding RNA UCA1 in tumors

Urothelial carcinoma associated antigen 1(UCA1)is a highly bladder cancer-specific long non-coding RNA(lncRNA),and it does not have the function of encoding protein. UCA1 differentially expre-sses in various fetal tissues,but its expression is turned off in most adult tissues. It reactivates during tumori-genesis. Researches indicate that UCA1 may regulate cell proliferation,apoptosis,metastasis and chemoresis-tance of tumors,such as bladder cancer,breast cancer and hepatocellular cancer.
9.The effects of compressive stress on TGF-?1 expression in condylar cartilage in vitro

Objective:To investigate the expression of transforming growth factor-?1 (TGF-?1) in mandibular condylar chondrocytes after the condylar cartilage was stimulated by compressive stress.Methods:20 condylar cartilage samples were obtained from 10 new borne SD rats and cultured for 24 h.Then,10 samples were treated by a static compressive stress with a magnitude of 10 kPa for 1 h.Another 10 samples without stress treatment were used as the controls. 10.The role of immune complex(IgG) and complement(C_(3c))in immunopathogenesis of ulcerative colitis

Objective To explore new insight into the immunopathogenesis of ulcerative colitis.Methods Immunohistochemistry for the immunoglobulin(IgG)and complement(C 3c )was performed in biopsy specimens of the mucous membrane from 50 patients with ulcerative colitis(32 in active state and 18 in inactive state)and 5 controls.Results The immunoglobulin and activated complement were found in the epithelium mucosae and basement membrane of capillary walls in ulcerative colitis,with highest density in patients with active ulcerative colitis as compared with inactive ones.Conclusions The activation of immunoglobulin(IgG)and complement(C 3c )are closely related to active ulcerative colitis which plays an important role in the tissue damage of active ulcerative colits.Immunoglobulin(IgG)and complement(C 3c )are important components of immunoregulatory network of ulcerative colitis
