1.The comparison of diagnostic value of FNAB and CNB in the treatment of patients with invasive breast cancer

Objective To investigate and compare the diagnostic value of fine needle aspiration biopsy(FNAB) and core needle biopsy(CNB) in the treatment of patients with invasive breast cancer.Methods Given 100 cases of patients with newly diagnosed breast cancer biopsy in our hospital from November 2015 to November 2016,50 cases of patients with FNAB into the observation group,50 cases of patients with CNB into the control group.Taken the pathologic results as a diagnostic gold standard and compared the diagnostic value of two diagnostic methods in invasive breast cancer.Detected the human epidermal growth factor receptor 2(Her-2),estrogen receptor(ER) and progesterone receptor(PR) preoperatively and compared the adverse reactions of puncture of patients in two groups.Results 30 cases of patients in the observation group showed axillary lymph node metastasis and 20 cases of patients had no axillary lymph node metastasis,21 cases of patients were positive cases of axillary lymph node metastasis accepted FNAB examination,the sensibility was 70.00%.29 cases of patients in the control group showed axillary lymph node metastasis while 21 cases of patients didn′t have axillary lymph node metastasis,23 cases of patients were positive cases of axillary lymph node metastasis accepted FNAB examination,the sensibility was 74.19%.Compared the ER、PR、Her-2 antibody with the testing result of primary tumor after operation.The diagnostic accordance rates in the observation group were 93.55%,91.67%,92.86% respectively.The diagnostic accordance rates in the control group were 94.12%,92.31%,86.67%,the differences in two groups were not statistical significance(P>0.05).The adverse reaction rate was 4.00% in the observation group and 0.00% in the control group after puncture.Conclusion The diagnostic sensibility of FNAB is poorer than CNB in the diagnostic procedure of invasive breast cancer.ER,PR,Her-2 in two diagnostic methods keep a high consistency with the testing result of primary tumor after operation.
3.Effects of He-Ne laser irradiation on the periodontal blood vessel remodeling during orthodontic movement in rabbits

BACKGROUND: Blood vessel remodeling is the basis of the remodeling of orthodontic periodontal tissues. Can He-Ne laser irradiation promote blood vessel remodeling and then accelerate the process of the remodeling of orthodontic periodontal tissues or not?OBJECTIVE: To investigate the effects and action mechanisms of He-Ne laser irradiation on the blood vessel remodeling of orthodontic periodontal tissues of rabbits.DESIGN: Randomized controlled study.SETTING: Animal experimental center of Jilin University.MATERIALS: Thirty-five flap-eared rabbits weighting 2.0 kg were provided by Animal Experimental Center of Jilin University, license No. SYXK(Ji)2002-0002, production license No. SCXK2003-0003. Animal intervention met animal ethical standard. HNGSQ-1 type of He-Ne laser therapeutic instrument was produced in Shanghai. HPIAS-1000 image analytical system was produced by Tongji Medical University. CD34 monoclonal antibody with the product number of MAB-0034 and vascular endothelial growth factor monoclonal antibody with the product number of MS-350 were offered by Fuzhou Maixin Company.METHODS: Experiments were performed at the Provincial Laboratory of Animal Experimental Center of Jilin University from May to December 2003. ①Thirty-five flap-eared rabbits were randomly divided into seven groups, an unforced group and forced 1-, 3-, 5-, 7-, 14- and 21-day groups with 5 rabbits in each group. A stainless steel closing coil spring was deposited between the maxillary first molar and the corresponding maxillary incisor bilaterally, delivering 0.08 N force. The right maxillary of rabbits of each forced group was irradiated by He-Ne laser, and the left side was as control.The cheek corresponding to the right maxillary first molar was selected as irradiation spot and then received He-Ne laser local irradiation. He-Ne laser's wavelength is 632.8 nm, power output of 20.0 mW and energy density of 2.50 J/cm2.Animals of the unforced group did not proceed with anything, sacrificed after raising for a week. Animals of the forced groups were respectively sacrificed on days 1, 3, 5, 7, 14 and 21. ②The premaxillary bones, including the upper first molar, were cut into sagittal sections. Sections were preceded with CD34 and vascular endothelial growth factor immunohistochemical straining. Microvessel density and microvessel area were measured in the tension zone and the pressure zone of orthodontic periodontal tissues. The staining intensity for vascular endothelial growth factor was evaluated by the measurement of gray value using the Computer Image Analyzing System.MAIN OUTCOME MEASURES: ①Gray integral of vascular endothelial growth factor after immunohistochemical staining. ②Microvessel density and microvessel area.RESULTS: Thirty-five rabbits were involved in the result analysis. ①Microvessel density and microvessel area in He-Ne laser irradiated sides were all obviously larger than in the control sides in every forced group in the tension zone and the pressure zone (P < 0.05-0.01), except for the 1-day group. ②In the 1-day group, the vascular endothelial growth factor expressions in periodontal tissues in pressure zone of He-Ne laser irradiated sides were significantly lower than that of control sides (P < 0.05). In the 3- and 5-day groups, vascular endothelial growth factor expressions in pressure zone of irradiated sides were significantly higher than that of control sides (P < 0.05). The vascular endothelial growth factor expressions in pressure zone of both irradiated sides and control sides reached a peak at 5-day group. The vascular endothelial growth factor expressions of irradiated sides were significantly higher than the corresponding control sides in tension zone at 3-, 5- and 7-day group (P < 0.05). The vascular endothelial growth factor expressions in tension zone of both irradiated sides and control sides reached a peak at 14-day group.CONCLUSION: He-Ne laser irradiation promotes blood vessel remodeling of periodontal tissues, increases the vascular endothelial growth factor concentrations in periodontal tissues and makes the angiogenesis of periodontal tissues more active, followed by the remodeling of orthodontic periodontal tissues.
4.STUDY ON PROLIFERATION AND APOPTOSIS IN DEVELOPING RENAL CORPUSCLES OF MOUSE

Objective To study cell proliferation and apoptosis of renal corpuscle in normal developing mouse.Methods PCNA immunohistochemisty method,terminal deoxynucleotidyl transferase mediated dUTP-biotin nick endlabeling(TUNEL staining method)and light,electron microscopy were used to observe proliferation and apoptosis within corpuscle at different stages of development.Results Nearly all cells in Ⅰ and Ⅱ stages had nuclear PCNA immunolabeling.The number of PCNA immunolabeling cells during Ⅲ and Ⅳ stages was gradually decreased,and to less in Ⅴ stage.TUNEL positive cells could be easily observed before birth and identified in Ⅲ and Ⅳ stages.Electron microscope revealed that the mitotic activity was observed in all cells of Ⅰ and Ⅱ stages renal corpuscles.The mitotic phases of endothelial cells and podocytes could be seen in Ⅲ stage,but only endothelial mitotic activities in Ⅳ stage of renal corpuscle.Apoptotic activities of endothelial cell and Bowman's parietal cell were noted,predominantly in Ⅲ and Ⅳ stages.Conclusion Proliferation and apoptosis exist universally in the course of developing renal corpuscle,and supervise and control the regular development of renal corpuscle cooperatively.
5.The Persistent Secretion of TNF-? by60Co Irradiated TNF Gene Modified Human Hepatoma Cell

?-tumor necrosis factor can enhance tumor cells autolysis and the expression of HLA antigenes. So we infected Human Hepatoma Cell Line (HHCL)with recombinant retroviral vector containing an insert of human tumor necrosis factor-? cDNA. The transduced tumor cells were isolated by G418 selection. After being irradiated by different doses of 60Co, their expression of TNF in 24 hours were tested. Observations of the relationship between the doses of irradiation and the expression levels of TNF were done. We also preserved the irradiated HHCL-TNF cells into liquid nitrogen, after recovering, tested their expressions of TNF again. This paper shows that: (l)following irradiation by 60Co, the production of ?-TNF persisted for about four weeks, and there is no difference among the different irradiated doses(40G~100G); (2)After being irradiated and kept into liquid nitrogen, the recovering HHCL-TNF cells constitutively secrete ?-TNF for about three weeks.These findings provide a logical basis for designing protocols for active immunotherapy in humans.

Objective To study the effect of berberine on the lipid metabolism and the expression of ox-LDL,CD36 and PPAR? in the peritoneal macrophage(PM) and alveolar macrophage(AM) from the type 2 diabetic rats. Total hip arthroplasty (THA) has developed rapidly in recent years, which reconstructs the joint function of some terminal hip joint diseases and obtains favorable efficacy. However, the stress load alters in the proximal femur following THA, leading to "adaptable bone reconstruction", which is characterized by thin cortical bone, decreased bone density, and bone loss surrounding prosthesis, in particular calcar femorale. This paper analyzed the effects of cement and cementless prosthesis, coated prosthesis, and THA approaches on stress shielding in the proximal femur to reduce stress shielding effect by improving prosthesis design, low elastic modulus materials, decreased section area of the prosthesis, cement prosthesis, appropriate coated materials and improved THA approaches.
9.Relationship of autoantibodies against angiotensin Ⅱ-1 receptor, α1-and 1-adrenergic receptors with thyrotoxicosis heart disease

Objective To explore the relationship of autoantibodies against G protein coupled angiotensin Ⅱ-1 receptor (AT1 R),α1-adrenergic and β1-adrenergic receptors (α1 R and β1 R) with thyrotoxicosis heart disease (THD).Methods The epitopes of the second extracellular loop ofAT1 R (165-191),α1 R (192-218),and β1 R(197-222) were synthesized for screening autoantibodies from 277 participants by ELISA.237 patients with thyrotoxicosis were subdivided into thyrotoxicosis treatment group (n =148) and thyrotoxicosis recovery group (n =89),or into THD group (n =46) and simple hyperthyroidism group (n =191).40 healthy subjects were served as control group.Results (1) The positive rates of autoantibodies against AT1 R,α1 R and β1 R in thyrotoxicosis patients were higher than those in the control subjects (31.6% vs 12.5%,27.8% vs 10.0%,and 23.6% vs 7.5%,all P<0.05).The positive rates of the three autoantibodies in the patients with Graves' disease were higher compared with thyrotoxicosis caused by other reasons (36.3% vs 19.7%,32.2% vs 16.7%,and 28.1% vs 12.1%,all P<0.05).(2) In thyrotoxicosis treatment group,the positive rates of autoantibodies against AT1 R and α1 R were higher than those in the hyperthyroidism recovery group (40.5% vs 16.9% and 33.1% vs 19.1%,both P<0.05).(3) The incidence of autoantibodies against AT1 R and α1 R in THD were significantly higher compared with simple hyperthyroidism (52.2% vs 26.7% and 43.5% vs 24.1%,both P<0.05).Conclusions Autoantibodies against AT1 R,α1 R,and β1 R may play important roles in the pathogenesis of hyperthyroidism,which may be involved in the progression of THD.
10.Effects of paeonol on lipopolysaccharide/adenosine 5′-triphosphate induced NLRP3 inflammasome activation in primary rat microglia

Aim To investigate the effects of paeonol on lipopolysaccharide ( LPS) and adenosine 5′-triphos-phate ( ATP) induced NLRP3 inflammasome activation in primary rat microglia and the mechanisms responsi-ble for this anti-inflammatory effects. Methods Pri-mary rat microglia were identified immunohistochemi-cally using the cluster of differentiation 11 b ( CD11 b ) antibody. Proinflammatory cytokine IL-1β was deter-mined by ELISA. Western blot was performed to ob-serve the protein expression of NLRP3 , ASC and caspase-1 in cultured primary rat microglia. The level of intracellular reactive oxygen species ( ROS) was mo-nitored by using the fluorescent probe 2′, 7′-dichlo-rofluorescein diacetate ( DCFH-DA ) . Results LPS (0. 5 mg · L-1 )/ATP ( 5 mmol · L-1 ) significantly increased intracellular ROS level and IL-1β secretion and upregulated NLRP3 , ASC and caspase-1 protein expression in primary rat microglia. Paeonol signifi-cantly decreased intracellular ROS level and IL-1β se-cretion, and inhibited LPS/ATP induced overexpres-sion of NLRP3 , ASC and caspase-1 in cultured primary rat microglia. Conclusion Paeonol can inhibit LPS/ATP induced NLRP3 inflammasome activation in pri-mary rat microglia, and this inhibitory effect may be through the suppression of intracellular ROS.
