1.Clinical Observation of Qianggu Capsule Plus Alendronate on Postmenopausal Osteoporosis

[Objective] To investigate the effects of Qianggu capsule plus alendronate on postmenopausal osteoporosis.[Methods]By random number generating method,120 patients were randomly divided into a treatment group with Qianggu capsule plus alendronate,control group A only with Qianggu capsule,and control group B only with alendronate.The pain of the patient before treatment,3 m and 6 m after treatment was assessed.At the same time,the bone mineral density (BMD) before and 6 months after treatment was recorded.[Results] There was significant difference before and after 3 m in the score of pain in three groups,but the difference in treatment group was higher than other two control groups.After 6 m,all the scores of pain in 3 groups had changed significantly compared to the ones before treatment,but the scores of pain in treatment group were lower than 2 control groups.The BMD of both vertebral and ward in treatment group after 6 months treatment was higher than before.[Conclusions]Qianggu capsule plus alendronate has definite therapeutic effects on postmenopausal osteoporosis.
2.Anterior cervical plate fixation and graft fusion in the repair of adult cervical tuberculosis:good biocompatibility

BACKGROUND:Spinal tuberculosis seldom involves cervical vertebra. The application of anti-tuberculosis drug has slight effects on patients combined with nerve dysfunction and severe vertebral destruction, which results in unstable cervical vertebra. OBJECTIVE: To evaluate biocompatibility of graft and host after one-stage anterior debridement graft fusion and internal fixation in the repair of adult cervical tuberculosis. METHODS: A total of 14 patients who suffered from cervical tuberculosis were admitted into Department of Spinal Surgery, First Affiliated Hospital, Xinjiang Medical University between May 2010 and June 2012. They underwent Zephir anterior cervical plate for one-stage anterior debridement graft fusion and internal fixation. RESULTS AND CONCLUSION:Compared with pre-fixation, erythrocyte sedimentation rate, C-reactive protein and visual analog scale score were lower in final folow-up (P < 0.05), and Japanese Orthopaedic Association score increased (P < 0.05). Except that Frankel grade recovered to grade D from grade C in one case, Frankel grade did not alter in the remaining patients. Compared with pre-fixation, Cobb angle was apparently shortened in seven patients with kyphosis. Folowing internal fixation, bone trabecula was visible between the vertebral body and graft region after fixation. No displacement, bone resorption, nonunion or pseudoarthrosis occurred. Neck pain and limited function relieved or disappeared to different degrees after fixation. These findings suggest this method can effectively treat cervical tuberculosis. Moreover, the biocompatibility of the plate and host is good.
3.Continuous veno-venous hemodiafiltration on acute kidney injury in children

ObjectiveTo investigate the effect and outcome of critically illness with acute kidney injury (AKI) treated with continuous veno-venous hemodiafiltration (CVVHDF) in children.Methods Twenty-four cases of critically illness with AKI were treated with CVVHDF in our pediatric intensive care unit from Jan 2008 to Dec 2010.The levels of creatinine (Cr),blood urea nitrogen (BUN),K +,Na + and HCO3- were observed before CVVHDF and 6,12,24,48,72 h after CVVHDF.ResultsCatheter was successfully established for CVVHDF in 24 cases of AKI.The average duration of CVVHDF was 46 h ( 16 ~142 h).The blood levels of Cr and BUN were significantly decreased at 6 h after CVVHDF [ ( 196.3 ±112.4) μmol/L,( 13.3 ± 8.5 ) mmol/L] and 12 h after CVVHDF [ ( 106.1 ± 84.2) μ mol/L,( 10.2 ± 9.7 )mmol/L] as compared to those before treatment [ (340.6 ±298.2) μmol/L,(31.6 ± 11.3) mmol/L] (P <0.05,P < 0.01 ).After 48 h of CVVHDF,the Cr,BUN returned to normal range.The imbalance of blood K +,Na +,and HCO3- improved at 6 h after CVVHDF and returned to nomal levels at 24 h.Total 28 d fatality rate was 29.2% (7/24),and all death cases were complicated with multiple organ dysfunction syndrome.ConclusionCVVHDF therapy for AKI can quickly clear Cr,BUN and excess water,correct electrolyte disorders,improve kidney function in children.

5.Protective effect of COPP on hypoxia/reoxygenation injury of H9c2 myocytes

Aim To study the effect and molecular mechanism of pretreatment with COPP on hypoxia/reoxygenation injury of H9c2 myocytes.Methods H9c2 myocytes model of hypoxia/reoxygenation injury was established and H9c2 myocytes were given COPP pretreatment before hypoxia/reoxygenation.Treatment with Znpp and all-trans retinoic acid(ATRA)inhibited HO-1 and Nrf2-ARE respectively.The level of LDH and CK in cell supernatants were measured.HO-1mRNA expression was analyzed by RT-PCR.HO-1 and Nrf2 protein expressions were analyzed by Western blot.Results Compared with hypoxia/reoxygenation group,the level of LDH and CK in COPP pretreatment groups decreased significantly and the level of HO-1mRNA,HO-1 protein expression and Nrf2 protein expression in the nucleus significantly increased.Znpp abolished protective effect of COPP pretreatment.ATRA blocked the nuclear accumulation of Nrf2 and decreased HO-1 protein expression that COPP pretreatment induced.Conclusions COPP can induce HO-1 overexpression which has protective effect on hypoxia/reoxygenation injury of H9c2 myocytes.Its mechanism is related to Nrf2-ARE signaling pathway.
6.Expression of the estrogen receptor ? and ? in mouse thymus

Objective:To detect estrogen receptor ? and ?(ER-?,ER-?) protein expression in different age of mouse thymus.Methods:Protein expression of ER-? and ER-? in thymus was analyzed via immunohistochemistry.Moreover,the relationship between ER-? and cytokeratin 18(epithelial cell marker) was further tested through fluorescence double-staining.Results:Immunohistochemical analysis revealed that both ER-? and ? protein was found in nuclei of some thymocytes of 3 month mice.However,expression of ER-? was absence while ER-? was still positive in aged mice,such as 12 months and 16 months old.Double staining further confirmed that lots of ER-?/? positive cells were Foxp3 negative cells.Conclusion:Expression of ER-? is absent while ER-? is still positive in thymus of aged mice,which indicates ER-? is the critical estrogen receptor that involves in thymic involution.Moreover,ER-?/? do not participate in Treg development within thymus.
7.DEVELOPMENT OF A NOVEL SUSPENSION CONCENTRATE OF NICLOSAMIDE AND ITS MOLLUSCICIDAL EFFECT AGAINST ONCOMELANIA SNAILS

Objective To develop a novel suspension concentrate of niclosamide (SCN) and evaluate its characteristics and molluscicidal effect against Oncomelania snails. Methods Niclosamide was milled by a sand granule mill and mixed with different amounts of wetting agent, dispersant agent, thickener, and water etc., to develop suspension concentrates, and its dispersion, suspensibility and stability were evaluated. According to the results of evaluation the best recipe and quality indexes for producing SCN were selected. The molluscicidal effects against Oncomelania snails were tested under lab condition and in field. Results The novel SCN contained 25% niclosamide (w/w), 1.5%-2.0% wetting agent (RS 3), 4% dispersant (FS 2), 0.10% thickener, a litte of other agent and water. The quality indexes which the SCN reached were as following: the content of niclosamide was more than 25%(w/w); the suspensibal rate was more than 90%; the pH was from 4 to 7; the size of more than 98% granules of niclosamide was smaller than 44?m; the thickener was smaller than 600 mpa.s. The SCN was very stable when it was stored in high or in low temperature. Under lab condition the LC 50 concentrations of SCN by the immersion method for 24, 48 and 72 hours were 0.0474 mg/L, 0.0412 mg/L and 0.0412 mg/L respectively while the LC 50 concentrations of 50% wettable powder of niclosamide (WPN) were 0.0947 mg/L, 0.0583 mg/L and 0.0442 mg/L. In the field death rates of the snails sprayed with 2.0 g/(L?m 2 ) of 25% SCN after 3, 7 and 15 days were 95.77%, 99.07%, 97.09% while the death rates of the snails sprayed with 2.0 g/(L?m 2 )of 50% WPN were 97.37%, 95.17% and 97.41%. Conclusion SCN had stable quality and high molluscicidal effect against Oncomelania snails, and it was suitable to be used in the field. The molluscicidal effect using 2.0 g/(L? m 2 ) of 25% SCN was similar with that using 2.0 g/(L?m 2) of 50% WPN.
8.Association between polymorphisms in HLA-Ⅱ genes and multiple sclerosis in Guizhou Province

Objective To discussed the relationship between multiple sclerosis(MS)and the genepolymorphism of HLA hoping that these results would be useful for further pathogeny studies,diagnoses,therapy and prognosis estimation of MS.Methods HLA-Ⅱ alleles in 32 patients with MS,36 nonimmunological neurological disease controls and 30 healthy controls,were identified by polymerase chain reaction-specific sequence primers(PCR-SSP)methods.Results The gene frequency of HLA-DR16 (7/32,1/36,0/30),DR11(7/32,3/36,1/30)and DQB1*0502(10/32,6/36,4/30)in patients with MS were higher than those in the 2 control groups.The gene frequency of DQB1*0601(8/32,12/36,17/30)in the patients with MS was lower than the controls.However,only the HLA-DR16 had significant difference (χ2=7.398,RR=17.94,P=0.011;χ2=5.52,RR=9.8,P=0.022).Conclusion HLA-DR16 alleles may be associated with the susceptibility to MS in Guizhou Province.

10.Effects of isoquercitrin from Craibiodendron yunnanense on osteogenic differentiation of MC3T3-E1 cells.

Natural products especially flavonoids are being explored for their therapeutic potentials in reducing bone loss and maintaining bone health. The present study is to investigate the effects of isoquercitrin from Craibiodendron yunnanense with different concentrations at 1 x 10(-4), 1 x 10(-5), 1 x 10(-6), 1 x 10(-7) mol x L(-1) on proliferation, differentiation and mineralization of MC3T3-E1. Cell proliferation was assessed by CCK-8 kit at 1, 3, 5 and 7 days of culture. Alkaline phosphatase (ALP) activity were performed qualitatively and quantitatively on day 7, and alizarin red S staining was employed to access the mineralization of cells on day 21. The osteogenic markers ALP, collagen type I (COL 1A1), runt-related transcription factor 2 (Runx-2) and Osterix were detected to analysis early osteogenic differentiation of cells on day 3 by RT-PCR. The results showed that isoquercitrin had a dose-dependent effect on the proliferation, osteogenic differentiation, mineralization and gene expression of MC3T3-E1 in the range from 1 x 10(-7) to 1 x 10(-5) mol x L(-1). At concentrations above 1 x 10(-4) mol x L(-1) isoquercitrin showed cytotoxicity, while 1 x 10(-6) mol x L(-1) is the optimal concentration of isoquercitrin to improve the osteoblastic activity. All these results implied that isoquercitrin might be the major composition of traditional Chinese medicine C. yunnanense to treat bone fractures.
