1.Development and application of anti-adherent evaluation model of H pylori adherent to MKN45 cell

Aim To establish the adherence of H pylori to MKN45 cell model for screening and evaluating anti-adhesive drugs.Methods H pylori suspension was incubated with FITC solution in different conditions(FITC concentration and incubation time)to optimize the labeling conditions.At the same time,Gram staining was used to determine the optimal adhesion time of H pylori to MKN45 cells.Results H pylori suspension(1?106 cuf?ml-1)incubating with FITC(5 mg?L-1)for 1 h could achieve the optimal effect for FITC-labeling H pylori.Besides,Gram staining indicated that the optimal adhesion time for H pylori to MKN45 cells was 1 h.Based on the model,the crude Aloe polysaccharide was evaluated for its anti-adhesive activity and the results showed it could significantly reduce the adherence of H pylori to MKN45 cells at the concentration of 1 g?L-1(P
2.CpG-ODN Enhances Anti-Tumor Effects of Non-Replicating Vaccinia Virus-Modeified Oncolysates

Objective:To investigate the antitumor effect of CpG-ODN combining non-replicating recombinant vaccinia virus in tumor immunotherapy.Methods: CpG-ODN and constructed vaccinia virus v△11?75 were combined to study the antitumor effects in the walker′s rat tumor model.Results: Recombinant vaccinia virus v△11?75 lost the replicating capacity on human cell line,143TK - cell.The genes between HindⅢ C & K fragment of vaccinia virus TianTan strain were deleted,verified with Southern-blot. In the Walker′s tumor model of Wistar rats,Combinational immunotherapy with CpG-ODN and non-replicating vaccinia virus-modified WRC256 oncolysates resulted in prolonged life span and reduced tumor hyperplasia. Conclusion: CpG-ODN can enhance the antitumor effects of non-replicating vaccinia virus-modified oncolysates,providing a new route of tumor therapy.
3.The protective effect of propofol on pulmonary ischemia/reperfusion injury

Objective To study the protective effects of propofol against ischemia/reperfusion injury in rat lung. Methods Rat model of pulmonary ischemia/reperfusion injury was used in this study. Rats were randomly divided into three groups, including sham opera-tion group (group A), iachemia/reperfusion group (group B) and propofol group (group C), 15 rats in each group. The concentration of tumor necrosis factor -α and interleukin-6 in bronchoalveolar lavage fluid (BALF) were measured by enzyme linked immunosorbent assay (ELISA). Then blood gas analysis, lung wet/dry (W/D) weight ratio were detected in each group. Results Propofol could significantly improve PaO2, reduce the W/D value and the contents of TNF-α and IL-6 in BALF. Conclusion Propofol effectively suppressed the pro-duction and release of inflammatory cytokine, therefore it can protect the lung from isehemia/reperfusion injury.
4.Visualization for Three-dimensional Data

7.Studies on Nephrotoxicity Induced by Triptolide on MDCK Cell Model in vitro

Objective:To study the nephrotoxicity induced by triptolide ( TP) on MDCK cell model and investigate its effect on oxidative stress. Methods:Aristolochic acid was chosen as the positive control. After the MDCK cells were incubated with 0. 5, 5, 50 and 500 nmol·L-1 TP for 24h, MTT method was used to observe the cell inhibiting rate and lactate dehydrogenase (LDH) release test was used to detect the cell membrane damage caused by TP. The cell morphology was observed under an inverted microscope. After the MDCK cells were incubated with 500 nmol·L-1 TP respectively for 30min, 1h, 2h, 4h and 6h, the level of reactive oxygen species ( ROS) was detected using 2′,7′-dichlorodihydro-fluorescein diacetate ( DCFH-DA) as the fluorescent probe. Results:Compared with those of the negative control group, the cell inhibiting rates and the relative LDH release rates in TP-treated group were increased sig-nificantly(P<0. 01). The cells in TP-treated group were creased, turned into the round shape and began to shed off. After the MDCK cells were incubated with TP for 30min, the level of ROS reached the highest value, and then began to decrease (P<0. 01). Conclu-sion:TP can induce the toxic effects on MDCK cells and the mechanism may be related to oxidative stress.
8.Impact of accelerator′s multi-leaf collimator leaves on the intensity modulated radiation therapy dose at different gantry angles

Objective To investigate the impact of accelerator′s multi-leaf collimator (MLC) on the radiotherapy dose with different gantry angles.Methods Measured with Mapcheck 2D diode array and 30 cm×30 cm×3 cm solid water, Pre-selecting the 30 appropriate single fields and 0°,45°,90°,270°,315° gantry angles of static and dynamic intensity modulated radiation therapy (IMRT), quantification analysis of the passing rate with MapCheck γ(3%/3 mm) and (5%/3 mm) analysis methods, and the same method to 30 examples static and dynamic IMRT plans.Results When the accelerator collimator angle is 0°,the 30 appropriate single fields′ passing rate of between 0°gantry angle and 45°,90°,270°,315°gantry angles of static and dynamic IMRT accordingly is 97.71% and 96.25%(t=1.70, P=0.389), 96.34% and 93.72%(t=2.95, P=0.002), 96.65% and 92.98%(t=2.87, P=0.005), 95.87% and 93.15%(t=2.71, P=0.006), 96.09% and 93.51%(t=2.89, P=0.004) with MapCheck γ(3%/3 mm) analysis methods, however, the passing rate also does not have the difference, respectively is 99.31%-99.73% and 98.89-99.68%(t=0.57-1.90, P=0.913-0.725) with MapCheck γ(5%/3 mm) analysis methods;the passing rate of 30 examples static and dynamic IMRT plans accordingly is 96.11%-96.76% and 94.88%-95.78%(t=1.02-1.61, P=0.317-0.235).Conclusions When the accelerator collimator angle is 0°, at different gantry angles, MLC leaves due to gravity, friction, inertia, etc caused by errors in place, the physical penumbra and leakage radiation will indeed affect the IMRT dose, however, the deviation of dose distribution is within acceptable 5%.
9.Patterns of Frequency Domain Indexes of Surface Electronic Signals of Adolescent Idiopathic Scoliosis Patients' Paraspinal Muscles

Objective To investigate the patterns of frequency domain indexes of surface electronic signals of adolescent idiopathic scoliosis (AIS) patients' paraspinal muscles by using surface electromyographic (sEMG) techniques .Methods 25 AIS patients enrolled, 7 males, 18 females, ages range from 11 to 21 years old. All of the enrolled patients undertook the Biering Sorensen test(BST) and the object-lifting test, a Finland made type ME3000P sEMG instrument was applied to record the electronic activities of paraspinal muscles(convex /concave) of all subjects, and the frequency domain indexes: median frequency(MF),mean power frequency(MPF), zero crossing rate(ZCR)were analyzed.Results The wave amplitudes and scales of paraspinal muscles electronic frequency domain indexes(MF, MPF, ZCR) were lower than the other tested positions when recorded at the zone of apex vertebrae, and MFslope, MPFslope and ZCRslope all showed a linear degressive tendency as the exercise time was extended.Conclusion Paraspinal muscles at the zone of apex vertebrae have low fatigue durabilities and more likely to be exhausted. sEMG ought to be one of the objective examinations used to evaluate the differences of electronic activities of paraspinal muscles(convex /concave) of AIS patients, and may have a promising value in clinical practice.
10.Interstitialcells of liver cancer influence the biological function of malignant through the HGF/cMET system

Objective To explore the effects of interstitial cells of liver cancer and normal liver cells co‐cultured on the biological function of liver cancer malignancy so as to understand the signal pathway involved by the interaction between these cells and confirm the role of interstitial cells in cancer progression in tumor microenvironment .Methods We co‐cultured interstitial cells or hepatocyte growth factor (HGF ) and human normal liver cell L‐02 ,and then detected the expressions of the tumor‐suppressing gene PTEN and the oncogene K‐RAS and changes of cell proliferation .The downstream signaling pathways were detected by Real‐time PCR and Western blot .Results The expression of PTEN was downregulated at the transcription level by 1 .15 times and translation level by 10 times (P<0 .05) ,while the transcription level and translation level of K‐RAS increased by 1 .4 times and more than 9 times , respectively ( P< 0 .05 ) in normal liver cells co‐cultured with liver cancer mesenchymal cells .The proliferation ability was increased by more than 2 times .ELISA experiment results showed that the medium from co‐culture contained HGF over 3 times more than the control group ( P<0 .05 ,1 085+108 vs .387+23) .At the same time ,cells in the experimental group expressed more than four times of cMET than the control group cells (P< 0 .05) .Exogenous HGF consistently promoted liver cell proliferation and viability (P<0 .05) .Conclusion Our study shows that liver cancer interstitial cells activate the HGF/cMET signaling pathway by secreting HGF and promote the proliferation of normal liver cells ,suggesting a new way to explore the molecular mechanism of tumor microenvironment in tumor development and treatment of hepatocellular carcinoma .
