2.Growth effects of deoxycholic acid on colorectal cancer cell by regulating the expression of cyclooxygenase-2

Objective The activity and cyclooxygenase (COX-2) protein expression of colorectal adenoc arcinoma cell treated by deoxycholic acids (DCA) were examined in order to find out the carcinogenesis mechanism of DCA. Methods The proliferation of colorectal cancer cells (SW 1116) was detected by MTT metho d . COX-2 protein expression was measured by immunohistochemistry and Western blo t. Results DCA lower than 100 ?mol/L concentration can stimulate the growth of the adenoca rcinoma cells with effect reversible, while higher concentration shows the long -acting effect of inhibition. DCA of 10,50 and 100 ?mol/L concentration can up -regulate the expression of COX-2 in cells, while only 10 ?mol/L can maintain this effect long than 72 hours. The level of COX-2 protein treated by the late r two concentration descends after 48 hours. Conclusion DCA affects the proliferation of SW1116 in two sides. D CA concentration lower than 100 ?mol/L can promote COX-2 protein expression, w hich may be the mechanism of its carcinogenesis.

Objective To study the effect of FSH on the secretion of gastrin in the rat stomach,offering experimental evidence for reproductive endocrine hormone regulating digestive function. 5.Effects of cyclooxygenase-2 inhibitor Piroxicam on the growth of colorectal cancer

Objective To study the effect of nonsteroidal anti-inflammatory drugs (NSAIDs) ,cy-clooxygenase-2 ( COX-2 ) inhibitor Piroxicam on the growth of colorectal cancer cells and to evaluate the preventive significance from COX-2 expression and apoptosis. Methods The cell growth of colorectal adenocar-cinoma cell line SW1116 was measured by MTT (3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyl tetrazolium bromide) assay, and COX-2 protein expression by Immunohistochemistry and Western Blot. Apoptosis was characterized by DNA fragmentation. Results The results showed that COX-2 inhibitor Piroxicam could restrain the proliferation of SW1116, which had positively related to its concentration. Concentration higher than 1. 0mmol/L showed cytotoxic effects. The inhibition of COX-2 by Piroxicam appeared within 12 hours, but COX-2 protein level recovered within 24 hours, its expression had negatively related to the concentration of Piroxicam. Apoptosis was induced in SW1116 culture with Piroxicam higher than 0. 1mmol/L. Conclusion It can be concluded that cell inhibition effect is associated with Piroxicam-mediated cell apoptosis and inhibition of COX-2 protein expression in SW1116 cell, because the effects of Piroxicam have the concentration and time dependence, in further clinical research its dosage and time of medication should be considered in preventing or treating colorectal cancer.
6.Effects of Indometacin on Apoptosis and Proliferation of Cervical Cancer Hela Cell

OBJECTIVE:To study the effects of indometacin on apootosis and proliferation of cervical cancer Hela cell. METH-ODS:Hela cell was cultured in vitro as study object,and cultured with 0(blank control),200,400,600,800 and 1 000 μmol/L indometacin for 24,48 and 72 h. The inhibitory rate of indometacin to the proliferation of Hela cells was detected by MTT assay. After treated with 0(blank control),400,600 and 800 μmol/L indometacin for 24 h,the change of cellular morphology was ob-served by invert microscope;cell cycle phase and apoptosis were analyzed by flow cytometry. RESUITS:Indometacin of 600, 800,1 000μmol/L could inhibit the proliferation of Hela cell,which was positively correlated to drug concentration and time. Com-pared with blank control,indometacin could induce that Hela cell transformed from polygonous to round in appearance,and result-ed in cell apoptosis and necrosis;the proportion of cells at G0/G1 phase increased,while the proportion of cells at S phase reduced;the apoptotic rate of cells raised. CONCLUSIONS:Indometacin could inhibit the proliferation of Hela cell,block cell cycle at G0/G1 phase and induce apoptosis.
8.Advanced in solubilization methods of water-insoluble natural drugs.

With the development of natural products, the research activities on the solubilization methods of water-insoluble natural products have been carried out worldwide. Big molecular weight and poor solubility of most natural active ingredients lead to a very poor oral absorption and low bioavailability, which has extremely limited their development in pharmaceutical fields and clinical application. As a result, it is necessary to find out a suitable technique to improve the solubility and enhance the oral bioavailability of insoluble natural drugs. Based on the related references published in these years, this review introduced some new techniques to improve the solubility and bioavailability of natural drugs, including prodrugs, inclusion complex, solid dispersion, cocrystals, osmotic pump, liquisolid compacts, micronization, self-microemulsifying, nanosuspensions, lipsomes, polymeric micelles and so on, and summarized the theory, characteristics, application range, application examples, problems and development direction of each technique.

9.The decrease of CD4~+ T cells of patients with SARS in early stage and its clinical significance

Objective:To observe the change of CD4 + T cells of SARS patients in early stage of disease and determine its clinical significance on the progress of disease and therapy selection.Methods:Detection the absolute counts of T cell subset from the peripheral blood samples of 52 SARS patients in initial 10 days' duration by Flow cytometry.Serial frontal chest radiographs had done and the progression of disease was observed during the treatment of all patients.Results:The 34 cases among the 52 patients were in normal range of CD4 + cell absolute count.At the same time,their pulmonary lesions were in limited degree and the states of an illness were stabilization and the pulmonary lesions were absorbed quickly.Therefore they reuired the treatment without corticosteroids.The other 18 patients were with low CD4 + cell absolute count, of which 13 cases showed progressive deterioation of radiographic change and were treated with corticosteroids additional.The other 5 cases with the mild low of absolute count of CD4 + cells were also static during the period of observation,and no treatment with coticosteroids.There was statistically significant relation between two groups(P=0 000).The results of Pearson correlate analysis between absolute count of CD4 + cells and pulmonary lesions were r=-0 737;P=0 000;The result between absolute count of CD4 + cells and corticosteroids treatment were r=-0 573;P=0 000.Conclusion:Peripheral blood CD4 + cell absolute counts of SARS patients on early stage of onset were negatively related to the degree of pulmonary lesions.The patients with remarkable decrease of CD4 + cell were in need of treatment with corticosteroids.
10.Thanssphenoidal mircrosurgery for the elderly patients aged over 70 years with pituitary adenoma

Objective To summarize the clinical features and transsphenoidal microsurgical experiences of pituitary adenomas in the elderly patients over 70 years old Methods Twenty four elderly patients accepted transsphenoidal microsurgery were conductded a retrospective review,whose clinical data including clinical main presentation,histological types,surgical conditions,perioperative treatment and outcome were also analyzed Results There were 16 non functional adenomas (66 7%) and 18 giant macroadenomas(75%) respectively Visual deterioration was the commonest complain in 20 (83 3%) Despite the majority of patients having coexisting medical conditions,we had no mortality and no postoperative adjunctive morbidity with an average hospital stay of 17 8 days For an average follow up of 3 5 years,we did observe major patients(79 2%) vision had improved Conclusions Pituitary adenomas in the elderly are most frequently non functional macroadenomas that present with visual deterioration and hypopituitarism If special care is devoted to the perioperative treatment,in particular to fluid and electrolyte balances,transsphenoidal microsurgery is safe and effective in hands of an experienced team
