3.Preparation of novel bulk carbon nano tubes dental materials for implant
Chinese Journal of Tissue Engineering Research 2008;12(1):180-184
BACKGROUND: Carbon nanotubes (CNTs) is about 1 mm in diameter and about 1-10 μm in length cylinder-like material, and it only contains the spiral of carbon atoms arranged on the surface. Because of its unique tubular structure, lower density than that of graphite and with a high enough intensity, CNTs are expected to substitute bone.OBJECTIVE: To primarily study the influence of the content of polycarbosilane (PCS) and the sintering pressure on CNTs, look for the most optimum condition for the preparation of chunk CNTs, analyze the physical properties and biocompatibility of the material, and provide a choice for candidate implant denture material.DESIGN: The content of cement and the sintering pressure are adjusted in the experiment to make sure the most optimum condition for the preparation, and the capabilities of the material are observed and analyzed to make sure if that the material is suitable for implant denture.SETTING: Department of Stomatology, Xiangya Hospital, Central South University; Prosthodontics Staff Room, Oral Medical College of Central South University. MATERIALS: 80% pure molecules of CNT (Nano Lab Company USA), 20-40 nm in diameter, 5-20 mm in length, was provided by Nacheng Technology Development Corporation of Beijing. PCS was compounded by 502 Development Room of Defense Technology University, and the molecular weight is 1 300; Ten male white Wistar rats with 8 weeks old were provided by Animal Experiment Center of Guangxi Medical University. The treatments with the animals accorded to animal ethical standard in the experiment.METHODS: The experiment was carried out in the Experimental Center of Material College, Central South University from January to August 2005. By using the spark plasma sintering (SPS), at the sintering temperature of 1 200 ℃, the pressure from 20 MPa to 60 MPa, PCS was used as the cement to produce the new chunk multi-wall CNTs.MAIN OUTCOME MEASURES: By using Archimedes method and adsorption of gases method, Japanese MitutoyoHM-101 little burden Vickers hardness meter was used to test the apparent bulk density, specific surface area, Vickers hardness at the different contents of PCS and sintering pressures. Scanning electron microscope (SEM) and transmission electron microscope (TEM) were used to observe the microstructure of the material.RESULTS: ① Microstructure: SEM and TEM indicated that the nanotube structure of the material was retained after sintered. ② Conforming phase of material: X-ray diffractometer indicated that carbon nanotubes were felt by PCS pyrolysis product of SiC; along with the PCS content and sintering pressure increased, the specific surface area was reduced, and Vickers hardness and density increased. Specific surface area decreased. ③ Histological compatibility: The inflammatory reactions of the CNTs/PCS were little more serious along with the increase of the content of PCS. The physical property of carbon nanotubes was prepared in this way, it was closed to bone tissues, it had the better biocompatibility, and it was the suitable candidate implant denture material.CONCLUSION: Appropriate to PCS for the bonding agent, through SPS, at 1 200 ℃, 20-60 MPa sintering pressure, CNTs/PCS which the material property is closed to bone tissues can be prepared, and CNTs/PCS subcutaneous implantation in rats does not show significant rejection; therefore, it is suitable candidate for dental implants material.

Journal of Chinese Physician 2001;0(08):-
Objective To survey the significance of specific immune response in the relationship between Sagnation the Liver-qi syndrome and the spleen Deficiency syndrome. Methods The rats with the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome were respectively corresponding and crossely treated with Radix Bupleuric dispersing the stagnated Liver-qi powder Decolion of Four Noble Drugs. Their T, B lymphocyte activity and specific immune response indexes changes (such as IgG, IgM in the serum) were respectively observed by lymphocyte transformation test and immunoturbidimetry. Results There were no notable changes about the IgG level among all kinds of groups. The IgM levels in the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome decreased, especially in the Liver-qi Stagnation syndrome (P0 05), Decolion of Four Noble Drugs could increase the IgM level of the patients with the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome (P0 05); The T lymphocyte transformation rate of the Spleen Deficiency syndrome obviously decreased, and Decolion of Four Noble Drugs could recover it to the normal level (P0 05). The rate of B lymphocyte transformation the Liver-qi Stagnation syndrome's also showed the increasing tenderly (P0 05). Conclusions The relationship between the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome could occur in the phase of antigen presentation, especially in the phase of the T H cell activation stimulated by antigen. Radix Bupleuric dispersing the stagnated Liver-qi powder could enhance T, B lymphocyte activities by reverse the Liver-qi Stagnation syndrome, and Decolion of Four Noble Drugs could enhance T lymphocyte by improving the Spleen Deficiency syndrome.

Journal of International Pharmaceutical Research 2017;44(5):402-408
Colorectal cancer(CRC)ranks the third in incidence and mortality rate among human tumors. Tumor relapse,me?tastasis and drug resistance remain the main obstacle to the success of CRC treatments. Compared to traditional chemotherapy ,target therapy seems to treat tumors in more precise and rational fashion with good response and often less toxic side-effect. Although target agents provide hope for more effective therapy, recent clinical studies have shown only modest benefit from target therapy similar to tra?ditional chemotherapy. Primary and secondary resistance to target agents is still observed and contributes to CRC treatment failure. The review summarizes research progress in the mechanism of drug resistance to target therapy in CRC treatment.

[Objective]To determine the effect of Oxfenmino hydrochloric acid on delayed allergic reaction in mouse. [ Method ] 120 ICR mice weighing 18~22g, half female and half male, were randomly and averagely divided into 6 groups according to weight:control group and model group (treated with distilled water at a dosage of 10ml/kg), three Oxfenmino hydrochloric acid groups (treated with Oxfenmino hydrochloric acid at a dosage of 10.0mg/kg, 5.0mg/kg and 2.5mg/kg respectively) and positive drug group (treated with CsA at a dosage of 15mg/kg); they were orally administered one time every day. After a continuous week of administration, all the groups were smeared evenly with 1%DNFB solution on the abdomen for allergy except the control group. Five days after allergy, 1%DNFB solution was smeared to right ear of all mice to stimulate the allergic reaction except that the control group was smeared with non-allergenic substance. 24 hours after attack, the auricle swelling, spleen index and thymus index in right mouse were determined. [ Result] Different dosages of Oxfenmino hydrochloric acid benefited to reduce auricle swelling, spleen index and thymus index in mouse with allergic reaction induced by DNFB at different degree. [ Conclusion] Oxfenmino hydrochloric acid can inhibit the delayed allergic reaction in mouse induced by DNFB and depress the ability of specific cell-mediated immunity.

Objective To study the effect of nonsteroidal anti-inflammatory drugs (NSAIDs) ,cy-clooxygenase-2 ( COX-2 ) inhibitor Piroxicam on the growth of colorectal cancer cells and to evaluate the preventive significance from COX-2 expression and apoptosis. Methods The cell growth of colorectal adenocar-cinoma cell line SW1116 was measured by MTT (3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyl tetrazolium bromide) assay, and COX-2 protein expression by Immunohistochemistry and Western Blot. Apoptosis was characterized by DNA fragmentation. Results The results showed that COX-2 inhibitor Piroxicam could restrain the proliferation of SW1116, which had positively related to its concentration. Concentration higher than 1. 0mmol/L showed cytotoxic effects. The inhibition of COX-2 by Piroxicam appeared within 12 hours, but COX-2 protein level recovered within 24 hours, its expression had negatively related to the concentration of Piroxicam. Apoptosis was induced in SW1116 culture with Piroxicam higher than 0. 1mmol/L. Conclusion It can be concluded that cell inhibition effect is associated with Piroxicam-mediated cell apoptosis and inhibition of COX-2 protein expression in SW1116 cell, because the effects of Piroxicam have the concentration and time dependence, in further clinical research its dosage and time of medication should be considered in preventing or treating colorectal cancer.

Chinese Journal of Digestion 2001;0(01):-
Objective The activity and cyclooxygenase (COX-2) protein expression of colorectal adenoc arcinoma cell treated by deoxycholic acids (DCA) were examined in order to find out the carcinogenesis mechanism of DCA. Methods The proliferation of colorectal cancer cells (SW 1116) was detected by MTT metho d . COX-2 protein expression was measured by immunohistochemistry and Western blo t. Results DCA lower than 100 ?mol/L concentration can stimulate the growth of the adenoca rcinoma cells with effect reversible, while higher concentration shows the long -acting effect of inhibition. DCA of 10,50 and 100 ?mol/L concentration can up -regulate the expression of COX-2 in cells, while only 10 ?mol/L can maintain this effect long than 72 hours. The level of COX-2 protein treated by the late r two concentration descends after 48 hours. Conclusion DCA affects the proliferation of SW1116 in two sides. D CA concentration lower than 100 ?mol/L can promote COX-2 protein expression, w hich may be the mechanism of its carcinogenesis.
