3.Activation effect of natural flavonoid compound baicalein on human wild cystic fibrosis transmembrane conductance regulator chloride channel

Objective To study activation effect of a natural compound baicalein on cystic fibrosis transmembrane conductance regulator(CFTR) chloride channel.Methods A cell-based fluorescence assay was used to determine CFTR-mediate iodide influx rate/dt(mmol?L-1?s-1)] activated by baicalein(the concentrations were 0.18,0.55,1.65,5,15,44,133 and 400 ?mol?L-1).Results The Ka of flavonoid baicalein stimulating CFTR was about 16 ?mol?L-1.The half of maximal activity was reached in ten minutes and the activation disappeared in 20 min after baicalein was washed out.The activation of baicalein was not affected obviously under different concentrations of Forsklin(0,20,50 and 100 nmol?L-1)and the activation could be totally inhibited by CFTRinh-172.Conclusion Baicalein can stimulate CFTR-mediated iodide influx in a dose-dependent way and its activity manifests a rapid and reversible characteristic.It might work in both elevating CFTR protein phosphorylation and direct binding way.
4.The role of peer education for rehabilitative patients with schizophrenia

Objective To discuss peer education on the rehabilitation in schizophrenia recovery.Methods According to the order of admission,108 cases of hospitalized recovery of schizophrenia patients were divided into observation group and control group,each group in 54cases.The observation group received psychiatric conventional treatment and care at the same time add peer educations,the control group only accepted psychiatric conventional treatment and care.The observation group was given for a period of 20 weeks peer education.The nurse brief mental scale (N -BPRS),self -knowledge and treatment attitude questionnaire scale (ITAQ)were assessed in the two groups at the beginning,10 weeks,20 weeks.Results The difference of N -BPRS score between the two groups was not statistically significant (t =0.191,P =0.850).In the observation group,the scores had significant differences between 10 weeks and 20 weeks peer education (t =2.757,P =0.069;t =4.000,P =0.000).The ITAQ scores of the observation group at the beginning,20 weeks peer education were (11.22 ±3.55)points,(17.22 ±4.31)points, the difference was statistically significant (P <0.01).In control group,ITAQ scores before and after treatment were (10.84 ±5.19)points,(13.12 ±6.12)points,the difference was not statistically significant (P >0.05).Conclusion Schizophrenia convalescence patients accept peer education is helpful to rehabilitation.
6.Conjunctive Use of Various Adjuvant and Fusion Protein Which Composed of M2e and N P Genes of Avian Influenza Virus,and the Influence on Immunogenicity

Based on the gene sequence of AIV matrix protein 2(M2) and two cytotoxic T-lymphocyte epitopes derived from nucleoprotein,the prokaryotic expression vector pET-3M2e-NP1-NP2 was constructed.The target gene was expressed in the solvable form in BL21(DE3) when induced with 1.0 mmol/L IPTG and Western blot analysis showed that the expression product had good immunogencity.Purified fusion protein was mixed with various amount of adjuvant,including Freund's,Vash oil and chitosan,and then immunized 20-day-old chicken by intramuscular injection and boosted 3weeks later.Blood samples were collected weekly following the primary vaccination.The anti-M2e antibody was detected with ELISA method with the synthesized peptide as coated antigen;the neutralizing ability of anti-serum was evaluated on MDCK cell line and chick embryo,the CD4+ and CD8+ T lymphocyte amounts in peripheral blood of immunized chicken was measured by flow cytometry.Results showed that the fusion protein can induce immunological reaction,and the antibody can bind with the viral M2 protein that expressed on the surface of MDCK cells.Flow cytometry result showed that CD4+ and CD8+ T lymphocyte in peripheral blood increased obviously following immunization(P
7.Study on the Immune Efficiency for General Vaccine Against Avian Influenza Virus Using Human Mycobacterium Tuberculosis hsp70 as the Carrier for Peptide Epitopes

M2e gene of three copies for H5N1 subtype AIV was synthesized and fused with human mycobacterium tuberculosis hsp70 gene.The fused gene was cloned into the prokaryotic expression vector to get pET-3M2e and pET-3M2e-hsp70.Recombinant protein r3M2e and r3M2e-hsp70 were successfully expressed induced with IPTG and purified with Ni2+-NTA collumn.Following that,the immunity of the recombinant protein was analysized with Western blot.20-day-old AIV non-immunized chickens were vaccined with r3M2e and r3M2e hsp70,at the same time,Trx and KLH-M2e inoculated chickens were served as vector and positive controls.Two weeks after the primary vaccination,every group was boosted with the same vaccine as in the primary vaccination.The humoral immunity of the vaccined chickens was evaluated with antibody detection against M2e,cytopathic suppression test,and indirect fluorescence assay.The cellular immunity was estimated according to lymphocyte subtype analysis with flow cytometry and M2e specific cytokine detection.Four weeks after the boost vaccine,all groups were challenged with 100EID50 AIV of H9N2 subtype,and the virus from swabs was detected with Real-time PCR.Results indicated that r3M2e hsp70 vaccined chicken developed the better humoral and cellular immune response,also,made a better performance compared with r3M2e vaccined group in virus challenge.
8.Risk factors of return visit for hospitalization in children with hand-foot-mouth disease

Objective To investigate the risk factors in children with hand-foot-mouth disease(HFMD)hospitalized after the second visit to outpatient department(OPD),and to evaluate their predictive value.Method In May 2008,180 of 343 pediatric patients with HFMD were repatriated to family or community after preliminary diagnosis in Bering Ditan Hospital.The ill children hospitalized after revisit(RVH)were compared with the remaining children(control group)for seeking the differences in age,HFMD contagion,temperattLre,interval between onset and visit,WBC count,skin lesion and comphcating risk symptoms by respective analysis.Logistic regression analysis was performed to find the risk factors involved in patients with RVH and diagnostic evaluation was introducted to predict the probability of RVH.Results Of 180 ill children,17(9.44%)ones returned to family or community requested admission into hospital for further consultation because who symptoms urtremitted or complications developed.The average duration between the preliminary visit and second visit was 3.26 days(0.5~14 days).No significant differences were found between RVH group and control group in age(P=0.669),ratio of gender(P=0.101),conttagion with HFMD(P=0.104),and typing of vires(P=0.475).Compared with control groups,the significant prolongation of interval between onset and visist(3.71±1.10)d,significant eleva-tion of temperature(38.74±0.57)℃ and WBC counls(10.99±3.67)×109 were noticed in ill children of RVH(P<0.05).According to logistic regression,interval between onset and visit nlore than 3 days,body tem-perature at the first visit higher than 38.5℃,WBC count over 10.0×109,and the accompaniment of serious symptoms were confirmed to be independent risk factors involved in RVH.Of them,67.85%(11/16)ill children with three or more risk factors of RVH showed diagnosis sensitivity and specificity reach to 64.53%and 97.14%,respectively.Conclusions Risk factors confirmed are the interval between onset and the visist more than 3 days,bodv temperature higher than 38.5℃ at the first visit,WBC count more than 10.0×109,and the accompaniment of severe symptoms.The ill children with three or more risk factors are in great request of more closely monitoring and should not be the candidates for repatriation to family or community after preliminary diagnosis.
9.DR bedside and CR bedside photographic quality comparative analysis in chest

Objective To explore the advantages of the bedside DR,taking the quality analysis of the chest image taken from the bedside digital radiography systems (DR) and computed radiography system (CR).Method All of the 900 pieces chest image taken by bedside DR,compared with the 900 pieces CR image randomly chosen.Results Bedside DR could further reduce the rate of the remake and the radiation dose,and provided more excellent image information.Conclusion The chest image taken by bedside DR has obviously advantage than CR.It can play a positive role in the diagnosis and treatment of the critical patient and surgical patient.
10.Expression of B1a cells and IgA1 positive cells in tonsil of IgA nephropathy patients and analysis of associated clinicopathological factors

Objective To examine the expression of IgA1 and B1a positive cells in palatine tonsils of IgA nephropathy (IgAN) patients, and to analyze the association between B1a cells and clinicopathological changes. Methods Eight patients diagnosed as IgAN by renal biopsy and 8 chronic tonsillitis patients without nephritis as control were enrolled in the study.Immunofluorescence and laser scanning confocal microscope (LSCM) were applied to observe the localization and quantitative calculation of Bla and IgA1 positive cells. Statistic analysis of the association of B1a cells with proteinuria and pathological Lee's grading was performed. Results Bla cells were mainly localized in germinal center of tonsil, and IgA1 positive cells were mainly localized in subepithelium of tonsil. Compared to control group, the percent of B1a cells and IgA1 positive cells was significantly higher in IgAN (P<0.01). There was a positive correlation between Bla cells and IgA1 cells (P<0.05). In IgAN, the percent of B1a cells in patients with hematuria and proteinuria was obviously higher than that of patients with hematuria only (P<0.05). The number of Bla cells in IgAN patients with≥Lee's grade Ⅲ was significantly higher than that of those < grade Ⅲ (P<0.05). Conclusions IgA1 may be secreted by Bla cells in the tonsil of IgAN patients. The number of B1a cells is correlated with exacerbation of proteinuria and pathological severity, which may play an important role in pathogenesis of IgAN.
