2.SELECTION OF ADULT HAIR FOLLICLE STEM CELLS AND THE EFFECTS OF ?-CATENIN

Objective To establish a convenient method for the selection of hair follicle stem cells and study the effects of ?-catenin on the regulation of proliferation and differentiation of hair follicle stem cells.Methods Based on the strong specific adherent ability of stem cells,hair follicle cells were incubated in the plates coated with human collagen before use.Hair follicle stem cell's marker K19 and clone forming efficency were used to identify the stem cells.The expression of ?-catenin was tested by immunocytochemistry and in situ hybridization.Results After adherence to collagen for 20min,the number of Kl9 positive cells among quickly-adhered cells increased 1.6 times more than unselected cells and clone forming efficiency increased 3.5 times.The expression of ?-catenin protein increased in the nuclear of quickly-adhered cells.Conclution The selection method of human hair follicle stem cells by quick-adherence to collagen was established for the first time.?-catenin was involved in the regulation of hair follicle stem cells'proliferation and differentiation.
3.Prevention for occurrence of sclerotomy-related complications during vitrectomy

Pars plana vitrectomy (PPV) is a common vitreoretinal surgical procedure. With its rapid advances many vitreoretinal diseases that have the potential to blind can now be treated. Although surgical complications are decreasing due to the improvement of surgical tools and techniques, these are not uncommon in our daily clinical practice. The sclerotomy-related complications, for example, are of great concern because they can result in retinal detachment and loss of vision. This article will review the common sclerotomy-re-lated complications including the sclerotomy-related retinal breaks and detachment, fibrovaacular ingrowth into the sclerotomy site, and vitreous incarceration. 4.Establish Centralized Sterilization and Supply Center to Control Hospital Infection

OBJECTIVE To discuss the effect of the central sterilization and supply center in controlling the infection in hospital.METHODS Eliminated the old supply pattern,to implement centralized management for the supply office.The whole sterilization and supply center was rebuilded according to the standard,cleared the function,standardized the procedure and strengthened the management.RESULTS The centralized sterilization and supply center could form effective and standard circulatory system for cleaneiness,sterilization and antiseptic,and ensure the quality of disinfectants.CONCLUSIONS The centralized sterilization and supply center is better to the management and control of quality, it can reduce the pollution,simplify operational procedures,reduce hospital infection,and realize the zero flaw of aseptic goods.

6.Clinical significance of high sensitive C-reactive protein and immune function in children with Mycoplasma pneu-moniae pneumonia

Objectives To detect the clinical significance of high sensitive C-reactive protein (hs-CRP) and immune function in children with Mycoplasma pneumoniae pneumonia (MPP). Methods 103 children with MPP, 47 cases of systemic inflammatory response syndrome (SIRS group), 56 cases of non-systemic inflammatory response syndrome (non-SIRS group) were recruited. 26 healthy children served as the control group. ELISA was used to detect the level of serum hs-CRP, immune in-dexes, IgG, IgA, and IgM, Cellular immune CD3+, CD4+, CD8+, CD4+/CD8+. Results The level of serum hs-CRP、IgG、IgM and CD8+in control group were significantly lower than those in non-SIRS group and SIRS group (P<0.05). The level of IgA、CD3+, CD4+, CD4+/CD8+in control group were significantly higher than those in non-SIRS group and SIRS group (P<0.05). The level of serum hs-CRP、IgG in SIRS group were significantly higher than those in non-SIRS group (P<0.05). The level of IgA、CD3+、CD4+、CD4+/CD8+in SIRS group were significantly lower than those in non-SIRS group. There was no significant difference in non-SIRS group and SIRS group of IgM and CD8+(P>0.05). The level change of serum hs-CRP were positively related with IgG (r=0.66,P=0.001) and were negatively related with IgA、CD4+、CD4+/CD8+(r=0.79, 0.67, 0.82, P all were<0.05) in chil-dren with MPP. Conclusion Children with MPP have Immunity function( including humoral immunity and cellular immunity) disorder which is related to the disease status. The level of hs-CRP could be an anpation index for the severity and immune func-tion of the children with MPP.
7.Practice of Clinical Pharmacists Participating in One Case of Multiple Organ Cryptococcus Infection during Perinatal Period

Objective: To evaluate the clinical pharmacists’ role in the drug treatment of perinatal women with multiple organ cryptococcus infection. Methods:Clinical pharmacists participated in the treatment of one case of perinatal women with multiple organ cryptococcus infection and provided comprehensive pharmaceutical service, including antifungal drug selection, adverse drug reactions prevention and monitoring, glucocorticoid selection in fetal lung maturity and recommendation in lactation. Results: Clinical pharma-cists provided suggestions on the drug treatment in many ways to enhance the safety, efficiency and economic of the drug treatment, and improve the patient's compliance as well. Conclusion:Clinical pharmacists can optimize the treatment regimen and play an active role in the drug treatment of perinatal patients with cryptococcus infection.
8.The diagnostic value of ultrasound-guided transperineal systematic prostate biopsy and cognitive fusion multi-parameter magnetic resonance imaging transperineal suspicious targeted biopsy in the prostate cancer in different serum prostate specific antigen levels

Objective:To evaluate the value of ultrasound-guided transperineal systematic prostate biopsy(SPB)and cognitive fusion multi-parameter magnetic resonance imaging(mpMRI) suspicious transperineal targeted biopsy(CFTB) in the prostate cancer with different serum prostate specific antigen(PSA) levels.Methods:A retrospective analyses were performed in 527 patients with suspected prostate cancer who underwent ultrasound-guided SPB from January 2018 to December 2019 in Shanghai Jiaotong University Affiliated 6th People′s Hospital. According to the PSA levels, they were divided into group A(PSA 4-10 μg/L) and group B(PSA>10 μg/L). All the patients underwent ultrasound-guided SPB, 376 patients with suspicious mpMRI had two additional targeted biopsies. The detection rates of ultrasound-guided SPB and CFTB in prostate cancer were tested by χ 2 test. Compared with pathological results, the sensitivity, specificity, accuracy of two methods were calculated and tested by χ 2 test, and a P<0.05 was defined as statistically significant difference. Results:Prostate cancer was detected in 319 of 527 patients(60.5%). One hundred and three cases of 198 patients in group A were diagnosed as prostate cancer, with an overall detection rate was 52.0%. Among them, ultrasound-guided SPB detected 72 cases of prostate cancer, the detection rate was 36.4%, sensitivity was 67.9%, specificity was 17.7%, accuracy was 26.5%, the detection rate, sensitivity, specificity and accuracy of CFTB were 39.9%, 75.6%, 91.6% and 88.8%, respectively. In this group, there were no statistically significant differences in the detection rate and sensitivity of the two methods in the diagnosis of prostate cancer (χ 2=0.525, 0.005, both P>0.05), and the differences in specificity and accuracy were statistically significant (χ 2=108.340, 79.829, respectively, both P<0.05). Two hundred and sixteen cases of 329 patients in group B were diagnosed as prostate cancer, with an overall detection rate was 65.7%. Among them, 160 cases of perineal prostate cancer were detected by ultrasound-guided SPB, with the detection rate was 48.6%, sensitivity was 78.2%, specificity was 37.6% and accuracy was 49.5%. A total of 189 cases of prostate cancer detected by CFTB, the detection rate was 57.4%, the sensitivity was 89.3%, the specificity was 90.6%, and the accuracy was 90.2%. All the differences were statistically significant in group B(χ 2=5.131, 4.391, 61.339, 38.982, all P<0.05). Conclusions:When PSA is greater than 10 μg/L, CFTB has a higher diagnostic efficiency than SPB.When PSA is 4-10 μg/L, there are no significant differences between the two methods in the detection rate and sensitivity of prostate cancer.

10.Combined surgery or two-step procedures for the management of primary retinal detachment accompanied with cataract

Postoperative cataract appeared 6 months after operation in the patients without IOL implantation, while it appeared 14 months after operation in the patients with IOL implantation. There was no significant difference in the postoperative complications between group A and group B. Conclusions Combined surgery is effective and safe for the selected patients of primary retinal detachment accompanied with cataract.
