Objective To examine changes infibrinogen (FIB),D-dimer and homocysteine (HCY) levels and their significance in patients with cerebral small vessel disease (CSVD).Methods This was a prospective study consecutively recruiting 116 patients with CSVD from January 2015 to August 2016 at the Second Affiliated Hospital of Bengbu Medical University, including 50 patients with lacunar infarction and 66 patients with leukoaraiosis. As a control group,89 age-matched healthy elderly individuals were enrolled in the study.FIB,D-dimer and HCY levels in the groups were measured and compared. Results Serum FIB,D-dimer and HCY levels in CSVD patients, including the lacunar infarction group and the leukoaraiosis group, were all significantly higher than in the control group[(3.3±0.9)g/L vs.(3.3±1.0)g/L vs.(3.3±0.8)g/L vs.(2.6±0.5)g/L,(631.1± 487.1)μg/L vs.(479.2 ± 277.6)μg/L vs.(750.2 ± 577.4)μg/L vs.(202.8±81.7)μg/L,(20.6 ± 12.8)μmol/L vs.(14.4 ± 4.8) μmol/L vs.(25.4 ± 15.0)μxmol/L vs.(12.8 ± 4.6) μmol/L,all P < 0.05)].Multiple Logistic regression analysis showed that FIB,D-dimer and HCY were independent risk factors for CSVD (P<0.01).Additionally,multiple regression analysis of lacunar infarction and leukoaraiosis showed that HCY had no significant correlation with lacunar infarction (P > 0.05).Conclusions FIB,D-dimer,HCY may be risk factors for cerebral small vessel disease, which may not carry the same weight for lacunar infarction and leukoaraiosis.
4.Immunotherapy in melanoma patients with autoimmune disease
Journal of International Oncology 2017;44(4):310-312
Immunotherapy has become an important treatment for melanoma. The anti-cytotoxic T lymphocyte-associated antigen-4 (CTLA-4) antibody or programmed death-1 (PD-1)/programmed death-ligand 1 (PD-L1) inhibitor is safe and effective for melanoma patients with autoimmune disease (AD).However, when such patients are treated with anti-CTLA-4 antibody or PD-1/PD-L1 inhibitors, their condition changes should be closely monitored to prevent and avoid possible AD progression and immune-related adverse events.
5.Effect of compound preparation of Astragalus mongholicus on subsets of lymphocytes in patients with myasthenia gravis
International Journal of Laboratory Medicine 2008;29(9):787-788,791
Objective To investigate the effect of compound preparation of Astragalus mongho-licus on subsets of lymphocytes in patients with myasthenia gravis (MG), and to explore its regulatory mechanism. Methods 60 MG patients were randomly divided into test group (n=30, given compoundpreparation of Astragalus mongholicus for 12 wks) and control group (n=30, given prednisone tablets for 12 wks). Flow cytometry was applied to examining the distribution change of the subsets of lym-phocytes in patients before and after treatment. Results After treatment, the percentages of CD4+ T cells and the ratio of CD4+/CD8+ in both groups decreased compared with those before treatment. The ratio of CD4+/CD8+ in test group was significantly lower than that in control group (P<0.05).The percentage of CD8+ T cells in test group after treatment increased significantly (P<0.05) com-pared with that before treatment, but had no significant difference compared with that in control group(P>0. 05). Conclusion The result from the current study suggests that one of the mechanisms of the compound preparation of Astragalus monghoticus regulating immune response may be achieved through its modulating effect on the distribution of subsets of lymphoeytes and humoral immune func-tion.
6.A fifty-year history of lung transplantation
Chinese Journal of Tissue Engineering Research 2007;0(31):-
Lung transplantation is the unique method to cure pulmonary tuberculosis at final stage. This paper reviewed the history, indication and contraindication, method of donor and recipient selection and donor preservation, operation methods, postoperative complications and protocol for complications to well understand issues in lung transplantation. Moreover, lacking of donors and increasing mortality of patients waiting for transplantation are urgent problems. With technique development, prognosis amendment, and improvement of survival rate and quality of life, donor source becomes important. Many studies have been conducted to explore the application of cadaveric lung, and several animal trials have been successful. In addition, scope of in vivo pulmonary lobes transplantation is increasing. Based on split liver transplantation, split double-lung transplantation is developed. This method utilizes existed donor sources, and provides options for patients waiting for transplantation.

9.Construction of pEGFP-N1-TGF-β1 recombinant plasmid and transfected it into primary cultured AEC-Ⅱ of neonatal piglet using lipofectamine 2000
Chinese Journal of Emergency Medicine 2016;25(1):50-56
Objective To construct pEGFP-N1-TGF-β1 recombinant plasmid and transfect it into primary cultured neonatal piglet type Ⅱ alveolar epithelium cell (AEC-Ⅱ) by using lipofectamine 2000,in order to provide basis of methodology for producing recombinant plasmids for transplantation of transfected AEC-Ⅱ into ALI/ARDS animal model lungs. Methods PCR primers were designed to amplify the human TGF-β1 cDNA fragment from plasmid.XhoI and EcoRI were used for double digesting the empty plasmid pEGFP-N1 and cDNA fragment of human TGF-β1.Then the products of double enzyme digestion by using T4 DNA ligase were connected and transformed into DH5α and cultured over night for 16 hours. The structure of recombinant plasmid was identified by using PCR and base sequencing to verify the correctness of pEGFP-N1-TGF-β1 recombinant plasmid. It was then transfected into primarily cultured AEC-Ⅱ by lipofectamine2000 mediated transfection and cultured for another 48 hour.Plasmid DNA (pEGFP-N1-TGF-β1 recombinant plasmid) and lipofectamine 2000 were added into serum-free DMEM respectively, then DNA suspension and Lipofectamine 2000 suspension were blended together and added into cells. After 24-48 hours later, the expression level of enhanced green fluorescent protein (EGFP) was evaluated under fluorescence microscope. Results The structure of vector was verified as pEGFP-N1-TGF-β1 recombinant plasmid by using PCR and base sequencing.Green fluorescence found in some cells showed that the pEGFP-N1-TGF-β1 recombinant plasmids had been successfully transfected into primary cultured AEC-Ⅱ,however,the transfection efficiency still need tobe further improved such as repeating the transfection procedure once again or using adenovirus mediated transfection method to improve the efficiceny and to transplant the cells into animal lungs eventually. Conclusions pEGFP-N1-TGF-β1 recombinant plasmid was successfully constructed and,for the first time, transfected into primarily cultured AEC-Ⅱ of newborn piglets. This established method should be useful for investigation of therapeutic effect and outcomes of lung with experimental acute lung injury.
10.Risk factors of vagus nerve invasion in patients with advanced adenocarcinoma of esophagogastric junction
Chinese Journal of Digestive Surgery 2013;(4):298-301
Objective To investigate the risk factors of vagus nerve invasion of advanced adenocarcinoma of esophagogastric junction (AEG).Methods The specimens from 98 patients with advanced AEG who underwent radical total gastrectomy and esophagogastric Roux-en-Y anastomosis from January 2011 to August 2012 at the First Affiliated Hospital of Anhui Medical University were collected.Serial section cutting was done and the upper and bottom incisional edges of every section were marked. The specimens were stained by hematoxylin-eosin solution,S-100 and CK immunohistochemistry.Vagus nerve invasion could be affirmed if cancer cells were observed in the peripheral space of vagus nerve and vagus nerve parenchyma or cancer cells infiltrated along the peripheral space of vagus nerve.Relationship between nerve invasion and clinicopathological factors was analyzed using chi-square test or Fisher exact probability.Factors related to vagus nerve invasion were analyzed using one-way analysis of variance and multi-factor logistic regression analysis. Results The incidence of vagus nerve invasion was 28.6% (28/98),and the tumor only invaded the vagus nerve which had the same altitude as the upper incisional edge. The results of one-way analysis of variance showed that Siewert classification, intravascular cancer emboli, lymph node metastasis, the degree of lymph node metastasis, clinical staging, the degree of tumor differentiation were correlated with the vagus nerve invasion (x2 =14.156,14.552,5.454,10.706,6.919,14.767,P < 0.05).The results of multi-factor logistic regression analysis showed that Siewert classification, intravascular cancer emboli and degree of tumor differentiation were the independent influencing factors of vague nerve invasion (OR =3.667,10.368,0.249,P < 0.05).Conclusion Vagus nerve invasion is correlated with Siewert classification, intravascular cancer emboli and degree of tumor differentiation. The range of vagus nerve invasion is restricted under the upper incisional edge of tumor section.
