3.Epigenetic regulation in spermatogenesis.

Spermatogenesis is a process consisting of spermatogonial proliferation, spermatocytic meiosis, and spermiogenesis, and is also considered to be a process in which heterochromatins gradually aggregate and finally reach a highly condensed formation in the sperm head. Recent studies show that epigenetic regulation plays a key role in spermatogenesis. This review discusses the mechanisms of epigenetic regulation in spermatogenesis in three aspects, DNA methylation, histone modification, and noncoding RNAs. These factors are essential for spermatogenesis, fertilization, and embryogenesis by mutual regulation as well as by gene expression regulation, transposon activation, sex chromosome inactivation, and genome imprinting.

4.Research development of prophylactic central lymph node dissection for papillary thyroid cancer

Papillary thyroid cancer (PTC) is the most common malignant neoplasm originating from the thyroid gland and has an excellent prognosis.But cervical lymph node metastases are common and the most common sites of metastases are the central lymph nodes of the neck (level Ⅵ).While therapeutic central lymph node dissection for involved lymph nodes is needed in our nation and others,the controversial topic is whether routine prophylactic central lymph node dissection in patients without evidence of lymph node metastasis should be performed in patients with PTC.The author reviewed the research development of prophylactic central lymph node dissection for papillary thyroid cancer in the article.
5.Comparison of mineral trioxide aggregate and Vitapex paste in the treatment of permanent teeth apexification

BACKGROUND: Vitapex is an injectable root canal filling material. It possesses effects of antibiosis, analgesia, as well as promotes formation of alveolar bone, dentin and cementum, which had been widely used in apexification. However, it also has weakness of long induction time and need reduplicated subsequent visit.OBJECTIVE: To explore the therapeutic effect of mineral trioxide aggregate (MTA) and Vitapex paste on apexification.DESIGN, TIME AND SETTING: The contrast observation experiment was performed at the Department of Stomatology,Shenyang Children's Hospital from February 2006 to November 2008.PARTICIPANTS: A total of 42 patients with 49 teeth were included in this study, including 28 anterior maxilla and 21 premolars.The X-ray film showed that the root tip of teeth was immature.METHODS: The teeth were randomly divided into the experimental group (n=25) and control group (n=24), which was apexificated by MTA and Vitapex paste, respectively.MAIN OUTCOME MEASURES: The effective rate of two groups was compared at months 3, 6 and 12 after operation.RESULTS: All teeth were included in the final analysis. The effective rate of experimental group was greater than the control group at 3 months after operation, and the difference had significant (P < 0.05), while, the effective rate was similar in two groups at 6 and 12 months after operation (P > 0.05).CONCLUSION: Both MTA and Vitapex paste are ideal filling materials in apexfication. Nevertheless, MTA may be a substitute in apexfication for its short therapy duration and precisely therapeutic effect.
6.Retinal toxicity of repeated intravitreal injection with bevacizumab in diabetic rats

Objective To observe the retinal toxicity of repeated intravitreal injection with bevacizumab(Avastin)in diabetic rats.Methods Forty male Sprague Dawley(SD)rats were randomly divided into normal group(Group A,10 rats)and diabetes mellitus group(30 rats).The rats in diabetes mellitus group were induced with streptozotocin injection for diabetic retinopathy model.And then randomly divided into diabetic retinopathy(DR)group(Group B,10 rats),the rats were not intervened;the left eyes of the other 20 rats were intravitreal injected with bevaeizumab 3 μ1(25 mg/m1)for 3 times as experimental group(Group C);the right eyes of the 20 rats were not intervened as experimental control group(Group D),20 days after last intravitreal injection,retinal function was measured by Flicker Electroretinogram (F-ERG);retinal vascular pattern was determined by fluorescence microscopy of ethidium bromide(EB)stained retinal flat mounts;retinal morphological changes were determined by light microscope on hematoxylin-eosin (HE) stained sections;Thy-1 and VEGF expression was measured by immunohistochemistry staining.Results F-ERG showed that-the differences of a-and b-waves-the b-wave amplitude and the Ops-wave amplitude in the implicit time between group A,B,C and D were significant (F=33.165,36.162,19.955,23.243;P=0.000);the differences of a-wave amplitude between group A,B,C and D was not significant(F=0.097,P=0v961).Retinal blood vessel pattern was normalin Group A;retinal vascular vessels were tortuous and irregularly expanded in Group B:retinal vascular vessels of Group C were regular and thinner than Group A;microaneurysm were showed in Group D.Light microscope displayed that the layers of the rat retina of Group A were regular,the retinal architectures of Group B were irregular,the retinal layers were regular in Group C,the retinal layers were irregular in Group D.Immunohistochemistry staining discovered that Thy-1 and VEGF were mainly expressed in ganglion cell layer(GCL).Conclusion Repeated intravitreal injection of bevacizumab iS toxic tO retina of diabetes mellitus rats.
7.Study on nuchal skin thickness of normal fetal by ultrasonography

ObjectiveTo explore ultrasonographic featurs of nuchal skin of normal fetus in its thickness and changes.MethodsThe thickness of nuchal skin of 900 normal fetal was measured by abdominal ultrasound with 7.5 MHz probe.ResultsThe nuchal skin only displayed whole layer before pregnant 27 weeks. After pregnant 27 weeks, the derma and subderma layer was distinguishable, the whole thickness of nuchal skin was ( 1.18 ? 0.05 )mm (10 weeks) to ( 6.01 ? 0.46 )mm (40 weeks) in normal fetus. All layers of nuchal skin were increased proportionally with gestation [r= 0.973 (whole layer), P 0.05 ).Conclusions The abnomal chromosome of fetus could be sifted by the thickness of nuchal skin and the character of sonographic image. It′s a useful diagnostic method for eugenics.
8.THE EFFECTS OF ACTINIDIA CHINENSIS PLANCH IN THE PREVENTION OF CANCERⅤ. INHIBITORY EFFECTS ON THE FORMATIONOF N-NITROSOPROLINE IN VIVO INRATS AND MEN

This paper deals with the inhibitory effects of Actinidia chinensis Planch (AC) on the formation of N-nitrosoproline (NPRO). In self-control studies, 15 male rats and 10 healthy men were the subjects. It was found that AC juice inhibited formation of NPRO in vivo in rats (inhibitory rate, 59.6%), and the effect was better than the same amount of a vitamin C (VC) solution (41.8%); and 150g AC fruit containing 75mg VC could completely inhibit the NPRO formation in vivo in men ingesting 300mg NaNO3 and 500mg L-proline, but 75mg VC only partially inhibited.

This paper deals with the inhibitory effect of Actinidia chinensis Planch (AC) juice on the formation of N-nitrosoproline (NPRO) in pregnant rats and women. In 21 Wistar pregnant rats and 27 pregnant women studies, it was found that NPRO formed in vivo might be transferred into the fetus, and AC juice might block it by inhibiting the NPRO formation in vivo. This was the first time to report that N-nitrosamino acid could be transferred into the fetus
10.Down-regulation of Gucocorticoid Receptor mRNA by Glucocorti-coids in Rats

The effect of glucocorticoids on the down-regulation of glucocorticoid receptor (GR) mRNA was studied in intact rats. GR mRNA was characterized by Northern blot hybridization and quantitated by dot blot hybridization using a human GR cDNA fragment as a probe. Administration of hydrocortisone in polyvinyl alcohol resulted in a rapid increase in plasma glucocorticoids which maintained at stress levels (20 to 40 ?g/dl) for about 3 d. Hepatic GR mRNA decreased significantly to 73.5?63% of control values 6 h following hydrocortisone treatment, after which the decline of GR mRNA was gradual, reaching a minimum of 44.0?5.0% of control levels 3 d after the treatment The effect of hydrocortisone on the down-regulation of hepatic GR mRNA lasted up to 11 d. In contrast, hydrocortisone treatment had no effect on GR mRNA in rat brain. These results are consistent with the changes in GR in rats as reported previously, except that even though the hepatic cytosol GR decreased markedly, no significant changes in hepatic GR mRNA were found 1 h after hydrocortisone treatment, strongly suggesting that the down-regulation of GR by its ligands in vivo occurs at both transcriptional and posttranscriptional levels and is of tissue-specific fashion.
