1.Analysis of allergen skin prick test in children with bronchial asthma

Objective To investigate the outcome of allergen skin prick test in children with bronchial asthma,and propose prevention strategy.Methods Sensitinogen prick test on the skin was applied on 200 children who were in asthma clinic of our hospital between Mar 2007 and Oct 2010.Then,the distribution of sensitinogen and their correlation with age and sex were analysed by SPSS 17.0.Results (1) The positive rate of sensitinogen prick test in 200 children with asthma was 72.5%(145/200).The positive rate of inhalant allergen and ingestion allergen were 62.5%(125/200),14.0%(28/200),respectively,of which dermatophagoides farinae,dermatophagoides pteronyssinus,outdoor mould,mugwort,cat epidermis,tree Ⅰ,egg,milk were 36.5%,40.5%,19.0%,17.0%,17.0%,13.0%,8.0%,5.5% respectively.(2) The frequency of sensitivity to cat epidermis,mugwort,outdoor mold and tree Ⅰ increased with age and the frequency of sensitivity to egg and milk decreases with age.The sensitivity rate to all other allergens had no relation with sex.(3) The severity of sensitivity to dermatophagoides farinae,dermatophagoides,ragweed and pigweed increased with age,while severity of sensitivity had no significant relation with sex among the 18 kinds of allergens.Conclusion Dermatophagoides farinae,dermatophagoides pteronyssinus,outside mould,mugwort,tree Ⅰ,cat epidermis,egg and milk are the major allergens in children with bronchial asthma,who should avoid contact with these facters.
3.Research progress on hepatic oval cells

Hepatic oval cells (OCs) are the stem cells of the liver. They can differentiate into liver cells, bile duct epithelial cells and other cells, and also are closely related to liver regeneration and liver diseases. In this article, we give a brief summary focused on the source, location, proliferation, differentiation, apoptosis, transplantation of hepatic oval cells, and its role in liver regeneration and liver diseases.
4.Comparison of consumption of drugs for patient-controlled intravenous analgesia in patients with different blood groups

Objective To compare the consumption of the drugs for patient-controlled intravenous analgesia (PCIA) in patients with different blood groups. Methods One hundred female ASA Ⅰ or Ⅱ patients, aged 30-50 yr, body mass index < 30 kg/m2 , scheduled for elective total hysterectomy or myomectomy, were enrolled in this study. The patients were divided into 4 groups according to the blood group: group A ( n= 25), group B ( n = 30), group AB ( n = 11 ) and group O ( n = 34). Patients received iv injection of flurbiprofen 30 min before the end of surgery and sufentanil at the end of surgery. PCIA with sufentanil 1.2 μg/ml ( bolus dose 2.0 ml, background infusion 0.5 ml/h, lockout interval 5 min) was performed when the VAS score ≤4. The loading dose and consumption on 1 st and 2nd day after operation for sufentanil were recorded. The adverse reactions were observed.Results The loading dose and consumption on 1st day after operation for sufentanil were the highest in group A and B, the second in group AB, and the lowest in group O among the 4 groups. Conclusion The consumption of the drugs for PCIA is different in patients with different blood groups.
5.Gd-polybutylcyanoacrylate nanoparticles as a novel magnetic resonance imaging specific contrast agent Synthesis and liver targeting

BACKGROUND: An effective method to enhance the accuracy and sensitivity of magnetic resonance imaging (MRI) is to select a targeted contrast agent. OBJECTIVE: To prepare gadopentetic acid -polybutylcyanoacrylate nanoparticle (Gd-PBCA-NP) and to investigate its liver-targeted values in animal experiments.DESIGN, TIME AND SETTING: A comparative observation was performed at the Laboratory of Department of RadiologicelCenter, Nanfang Hospital between September 2004 and January 2005. MATERIALS: Gd-PBCA-NPs were prepared by anionic emulsion polymerization. METHODS: A total of 18 Wistar rats were randomly divided into 3 groups, with 6 rats per group: Gd-DTPA, PBCA-NP, and Gd-PBCA-NP. Following caudal venepuncture (catheter remained), rats received 0.1 mmol/kg Gd-DTPA, 10 mL/kg PBCA-NP, or 10 mL/kg Gd-PBCA-NP. MRI scans (coronal and transverse) were performed prior to and after different enhancement protocols using a head coil for transmission and reception of the signal. Immediately after contrast medium injection, dynamic data of each group were acquired at 5, 15, 30, 60, and 120 minutes using imaging parameters the same as for the pre-contrast images. MAIN OUTCOME MEASURES: The morphology of Gd-PBCA-NPs was observed using transmission electron microscope. The size and the index of size distribution were measured through the use of laser granulometer (Malvern-3000HS). The Gd ion concentration in the dialysate was determined by inductively coupled plasma atomic emission spectroscopy for calculation ofencapsulation efficiency and loading capacity of Gd-PBCA-NP. The enhanced rate of the liver at each time point was calculated. RESULTS: The morphology of Gd-PBCA-NPs was spherical shape with core-shell structure and without adherence between particles. The average size of Gd-PBCA-NPs was 65.7 nm, and the index of size distribution was 0.09 measured by dynamic light scattering. The average encapsulation efficiency and loading capacity of Gd-PBCA-NPs were 81.97% and 51.23%, respectively. After intravenous injection of Gd-DTPA, the enhanced rate of the liver was 35.2% and 48.6% at 5 and 15 minutes respectively and the kidney was obviously enhanced; at 30 minutes, the enhancement of the liver recovered to normal level. After intravenous injection of Gd-PBCA-NPs, the average enhanced rate of the liver was 3.4%, 22.7%, 36.1%, 56.4%, and 24.8% at 5, 15, 30, 60, and 120 minutes accordingly. After intravenous injection of PBCA-NP, the enhancement of the liver was not found. CONCLUSION: Gd-PBCA-NPs can be taken as liver-targeted MRI contrast agents and have potential application value in MRI.
6.Analysis of Drugs Used in 10 Hospitals of Hunan Province From 1996 to 2000

OBJECTIVE: To analyze the current situation and developing trend of drugs used in 10 hospitals of Hunan Province from 1996 to 2000, and to serve as reference for drug production, marketing and clinical application .METHODS:The sum of money and percentages of different kinds of drugs used each year were added up separately.RESULTS: The sum of money of drugs consumed was increasing at a rate of 18% each year.About 40% of the money was spent on anti - infective drugs and 20% on imported drugs.CONCLUSION: The sum of money of drugs consumed will grow year by year .The clinical use of drugs will be more rational and of more choices,but the sum of money spent on imported drugs will decrease.
7.Pathogenesis of diseases with antineutrophil cytoplasmic antibody and its clinical significance

Objective To investigate the pathogenesis of diseases with antineutrophil cytoplasmic antibody (ANCA)and its diagnostic value in primary small vessel vasculitis.Methods 57 patients with serum ANCA positive were involved in this study,and ELISA was employed to assay anti-MPO.Patients with ANCA positive,anti-MPO positive and/or anti-PR3 positive were involved in group A.Patients with ANCA positive,anti-MPO negative and anti-PR3 negative were involved in group B.X2 was used to analyze the differences between the two groups.Results The etiology of 57 ANCA positive patients included primary small vessel vasculitis (20 cases,35.1%),non inflam-matory connective tissue disease(19 cases,33.3%),non connective tissue disease(18 cases,31.6%).A group of primary small vessel vasculitis accounted for 58.6%,which was significantly higher than 10.7% of the B group (χ2 =14.354,P<0.01);while the B group of non inflammatory connective tissue disease accounted for 50%,which was significantly higher than 17.2%of the A group (χ2 =6.879,P<0.01).Conclusion ANCA should be found in many kinds of diseases,so combined detection of anti-MPO and anti-PR3 should be employed to improve the diag-nosis specificity in primary small vessel vasculitis.Furthermore,non vessel vasculitis connective tissue disease should be excluded in patients with ANCA positive,anti-MPO and anti-PR3 negative.

9.Diagnostic value of magnetic resonance diffusion weighted imaging (DWI)in T staging of rectal cancer

Objective To evaluate the clinical value of magnetic resonance diffusion weighted imaging (DWI)in T staging of rectal cancer.Methods Rectal cancer patients were accepted in our hospital from January 2015 to December 2015.Rectal cancer patients were examined with T2 weighted imaging (T2 WI)and magnetic resonance DWI and given preoperative T staging.The sensitivity,specificity and accuracy of T2 WI and T2 WI combined with DWI in the diagnosis of DWI staging of rectal cancer were compared,and the relationship between the apparent diffusion coefficient (ADC value)and T staging of rectal cancer was analyzed.Results The accurate rate of T2 WI in diagnosis was 65.22%(60 /92),T2 WI and DWI diagnosis accurate rate was 76.09% (70 /92).The two diagnostic methods of colorectal cancer T staging diagnosis accurate rate had no significant difference (χ2 =1.18,P >0.05). Patients with good prognosis (T2 and T3 a)ADC value were significantly higher than the patients with poor prognosis in patients with ADC (T3 b,T3 c and T4 ),and the differences were statistically significant (all P <0.05).Conclusion DWI can be used as a complementary sequence for the diagnosis of T staging of rectal cancer by T2 WI.It had better consistency with pathologic stage.The ADC value of the tumor showed a decreasing trend along with the progression of T staging of rectal cancer,and provided reliable reference information for the preoperative treatment plan.
10.Effects of IGF-1R gene silencing on the expression of Caspase12 in L02 cells with peroxidation damage

Objective To observe the effects of silencing IGF-1 receptor on the expression of Caspase12 , a endoplasmic reticulum specificity apoptosis protein of the peroxidation damage in L02 cells. Methods Four the recombined plasmids expressing short hairpin RNA (shRNA) specific for sileincing IGF-1R gene were constructed and these shRNAs were potentially targeted on different mRNA sequences of the IGF-1 receptor gene (Sh-H-IG1R- 1108, Sh-H-IGF1R-2797, Sh-H-IGF1R-3215 and Sh-H-IGF1R-3787). These constructs were classed as the experimental group. Another two constructs containing an irrespective sequence and no sequence were defined as a positive control group a negative control group , respectively. All these constructs were transfected into the L02 cells by Lipefectamine 2000. The interference effect was detected by RT-PCR after 24 h and the plasmid stably silencing the expression of IGF-1 receptor was selected for further analysis which was divided into four groups: the blank control group(Group A), the peroxidation damage group(Group B), the negative plasmid transfection group (Group C) and the positive plasmid transfection group (Group D). The expression of Caspase12 was detected by Westen Blot after 12 h. Results The amplification folds of experimental groups were statistically significantly lower than those of the Sh-H-IGF1R-V1 group and Sh-H-IGF1R-V2 group (P < 0.05). The silence effect of the Sh-H-IGF1R-3215 group (0.02 ± 0.01) and the Sh-H-IGF1R-3787 group (0.05 ± 0.05) were most stable and the repeatability of the later was better. The expressions of Caspase12 in Group B (2.41 ± 0.03), Group C (2.45 ± 0.02) and Group D (3.60 ± 0.06) were higher than Group A (0.34 ± 0.02) (P < 0.01). The Group Cexpression was higher than that of the Group B (P > 0.05). The Caspase12 expression in Group D was higher than that in Group B and C (P < 0.01). Conclusion (1) Of all the four constructs in this study, Sh-H-IGF1R-3787 is the most suitable for silencing IGF-1 receptor. (2) Silencing IGF 1R expression using shRNA plasmids can aggravate the L02 cells apoptosis, suggesting that the IGF-1/IGF 1R signal pathway might be involved in regulating the activation of apoptosis in L02 cells with peroxidation damag via endoplasmic reticulum stress.
