Objective To evaluate the relationship between fragile histidine triad(FHIT) transcription abnormalities and HPV16 infection and human cervical tumorigenesis.Methods Total RNA from 5 cervical carcinoma cell lines(SiHa,HeLa,RJC-1,CS1213 and C4-1),58 primary cervical cancer specimens and 18 normal cervical epithelial tissues were extracted and FHIT transcripts were characterized by a two-step(nested) reverse transcription(RT)-PCR.The seven of the PCR products with different size were purified and sequenced.HPV16 infection was assessed by PCR.Results ① There were altered FHIT transcripts in SiHa,HeLa and C4-1 cells.Aberrant FHIT transcripts were detected in 39 out of the 58 cervical cancer samples(67.2%),but none out of 18 in the normal cervix tissue specimens(0%);HPV16 infections were identified in 37 of the 58 cervical cancer tissues(63.8%),but 1 in the 18 normal cervical epithelial tissues(5%),which showed a significant difference between these two groups(P0.05).③ The exon 5 and exon 6 were mainly deleted in the altered FHIT transcripts and no insertion and point mutation were found by DNA sequencing.Conclusion Aberrant FHIT expression was significantly common in cervical cancers and was correlated with HPV16 infection.These findings suggest that the tumor suppressor gene FHIT and high risk HPV16 may play a very important role in human cervical carcinogenesis.

Objective To investigate the expression of Matrix Metalloproteinase 9 (MMP 9) in bronchial biopy mucosal tissue and its relation to airway eosinophilic infiltration,lung function and airway hyperresponsiveness.Method 10 subjects with asthma were enrolled,meanwhile,10 healthy subjects and 6 subjects with COPD were as controls.At the same time,immunohistochemistry was employed.Results Immunohistochemistry for MMP 9 was positive in all asthmaticus,and just as such in 2/6 subjects with COPD.On the contrary,immunohistochemistry for MMP 9 was negative in all healthy subjects.The amount of MMP 9 expression was positively correlated with the number of eosinophil infiltrated to airway (r=0 62,P

Objective To explore the application value of uric indicant monitoring on patients with impaired glucose tolerance. Methods 9 6 health examination people were selected who with glucose tolerance impairment diagnosis standard population in Jan to Feb 2015 in LaiZhou,There were no significant differences in age and sex composition between the two groups in 48 cases of experimental group and 48 cases in control group.There were no other diseases,and no drugs were taken to con-trol blood glucose.IGT patients in the experimental group using uric indican monitoring to establish personal diet guide, while the control group used normal health education guidance,the two groups respectively before experiment and at the end of detecting urinary indican,fasting blood glucose,2h glucose tolerance levels,and were compared.Results According to the findings of the two groups follow up and the experimental data collected during one year,at the end of the experiment the IGT patients in the experimental group levels of urinary indicant was (50.7±6.2 mg/L vs 78.5±7.9 mg/L),fasting blood glucose was (6.34±0.11 mmol/L vs 6.91±0.29 mmol/L),2h glucose tolerance was (7.81 ± 0.52 mmol/L vs 9.57 ± 1.13 mmol/L)which was lower than the control group.There was significant difference between two groups (t=2.0,2.6, 3.06,P<0.05).Compared with IGT in the experimental group at the end of the experiment,the level of uric indican was (50.7±6.2 mg/L vs 78.3±8.1 mg/L),fasting glucose was (6.34±0.11 mmol/L vs 6.78±0.19 mmol/L),2h glucose tol-erance was (7.81±0.52 mmol/L vs 9.33±0.93 mmol/L)was lower than before the start of the level (t=2.01,2.28, 2.83,P<0.05),and the difference was statistically significant.In terms of the outcome of the various stages of diabetes, IGT patients in the experimental group was significantly better than the control group (χ2=10.85,P<0.05),the difference was statistically significant.Conclusion Through the determination of uric indican to guide the glucose tolerance decrease in patients with a reasonable diet,is conducive to glucose tolerance in the patients with impaired on blood glucose control,can effectively reduce the glucose tolerance decrease (IGT)conversion to diabetes mellitus (DM),change to a positive direction.

Objective To select a safe and feasible way by comparing the two main styles of splitting liver transplantation classical splitting and modified splitting.Methods In order to compare the distribution of artery, vein and bile duct of all segments of liver, 58 formaldehyde fixed livers and 8 fresh livers were divided into two groups. One group was subjected to classical splitting and the other group to modified splitting. Results Among 66 livers, 66.7?% of the segment VIII and 100?% of the segment V (all or some) drained to middle hepatic vein. In classical splitting group, the vein drainage of 5th and 8th segments of right graft was destroyed.Conclusion Modified splitting is safer and more reliable than classical splitting, so the former is the first choice when the size of donor liver is suitable.

Objective To analyze the clinicopathological characteristics and the related factors influencing the prognosis of pancreatic neuroendocrine carcinoma.Methods The clinicpathological and follow-up data of 21 patients with pancreatic neuroendocrine carcinoma admitted between April 2000 and August 2013 in Zhongshan Hospital of Fudan University were analyzed retrospectively.The influence facts on patients' prognosis were analyzed statistically.Results Univariate analysis showed that age,stage,tumor size,degree of tumor differentiation and lymph node metastasis were factors separately influenced patients' survival.Multivariate analysis (Cox regression) revealed that age (P =0.019) was the only independent factor affecting the prognosis.Conclusions Patients with pancreatic neuroendocrine carcinoma were mostly non-functional and presented no specific clinical features.Tumors tended to metastasize and the prognosis was poor.Age was the independent factor affecting the prognosis.

Objective To investigate whether antisense integrin αV and β3 gene therapy has antitumor activity for hepatocellular carcinoma.Methods In this study,120 male nude mice at the age of 4 weeks old were devided into 4 groups randomly.The antisense integrin αV and β3 expression vectors were iniected into HepG2 hepatomas established in nude mice to monitor tumor growth after 6 weeks.Tumot inhibit rate was calculated.Tunor microvessel density(MVD)was determined by immunohistochemical staining,and tumor apoptosis by TUNEL. Results The tumor weight of the control group,αV group,β3 group and αVβ3 group was(1.38±0.92)g、(1.28±0.27)g、(1.30±0.34)g、(1.08±0.16)g respectively.The difference between each two groups was significant(q12=4.76,q13=3.73,q14=14.28,P＜0.05);The MVD was(17.53±1.88)、(16.06±1.92)、(15.83±2.00)、(14.86±1.69)respectively,(q12=3.91,q13=4.55,q14=7.13,P＜0.01);The AI of the control group,αV group,β3 group and αVβ3 group was 10.53%±3.29%,19.80%±4.06%,21.93%±3.26%,24.03%±4.45%respectively,with the difference being significant among groups(q12=29.41,q13=33.52,q14=39.7,P＜0.01).Conclusions Antisense gene therapy targeting αV and β3 integrins exerts dramatic inhibition on the growth of the tumor.

Objective: To develop new kinds of stationary phases suitable for applications in capillary electrochromatography (CEC), as well as in LC, which can be tailored to realize selective separations of solutes difficult to separate with conventional stationary phases. Methods:Purchased spherical silica particles (5 ?m) were refluxed in anhydrous toluene with vinyl triethoxysilane for 18 h, to modify the surface with vinylsilyl groups. The silylated silica particles were subsequently stirred in an n henanol acetonitril mixture (1∶4, volume fraction) for 18 h. Styrene and divinylbenzene in the mixture co polymerized to form a crosslinked encapsulation layer on the silica surface, and bonded on the surface vinyl groups at the same time, both reactions were initiated by azo iso butyronitrile. The poly(Styrene divinylbenzene) encapsulated silica(PS DES) produced can be used as a non conventional stationary phase for CEC itself. By sulphonation of the PS DES phase with chlorosulphonic acid, strong cation exchange stationary phase of sulphonic type was obtained. Results: The manufactured PS DES as well as the sulphonated phases were respectively packed into capillaries,and the columns thus prepared were tested for their chromatographic characteristics. It was found that the PS DES phase showed reversed phase characteristics. Due to the phenyl groups in the encapsulated polymer, it introduced ?-? electronic interaction between the solutes molecules and stationary phase during the chromatographic separation process, therefore it showed unique selectivity on separating aromatic compounds, also polar as well as some alkaline drugs was analyzed on the column packed with the phase. It was demonstrated that the sulphonated phase could be used to prepare columns for the separation of alkaline drugs, symmetric peaks were obtained for them and base line separation was realized. Conclusion: It is possible that these stationary phases prepared can be used for solving the analytical problems in which non conventional selectivity are needed ( the analyses of alkaline drugs are examples) In some cases, the analysts can realize the anticipated separation results based on the different separation mechanism from that of the conventional stationary phases.

Objective To investigate the influence of very late antigen-4 (VLA-4) antibody and VLA-4 antibody combined with VP_(16) in vitro on childhood leukemic cells' apoptosis and explore the protection of bone marrow stromal cells (BMSC) upon leukemic cells and its related mechanisms. Methods Leukemic bone marrow stromal cells were isolated by human lymphocyte separation medium and in vitro culture of BMSC (adherent) and leukemia cells (suspended) BMSC+leukemic cells group were as control. Then VLA-4 antibody and/or VP_(16) were added respectively to VLA-4 antibody group, VP_(16) group and VLA-4 antibody combined with VP_(16) group to detect the apoptosis of leukemic cells in different groups through Annexin V-FITC double-labeled flow cytometry and the expression of Survivin, bcl-2 genes in each group of leukemic cells detected by RT-PCR. Results The results showed by flow cytometry that compared with the control groups, for 12 h or 24 h, the early and total apoptosis rates of leukemic cells of the three experimental groups were significantly increased(P <0.05); the early and total apoptosis rates of leukemic cells treated with VLA-4 antibody combined with VP_(16) group was markedly increased, compared with the control group (P <0.05); the comparison of the early and total apoptosis rates for the three experimental groups between 12 h and 24 h was significantly different (P<0.01). Moreover, RT-PCR results showed that the expression of Survivin and bcl-2 genes of leukemic cells in three experimental groups was reduced in varying degrees and the reduction of VLA-4 antibody combined with VP_(16) group was the most obvious. Conclusion BMSC plays a protective role on leukemic cells, and VLA-4 antibody can block the adhesion between BMSC and leukemic cells promoting leukemic cells apoptosis and enhance the sensibility of apoptosis of leukemic cells induced by chemotherapeutics.

Objective To study the expression of S100A8 and the relationship between S100A8 and Toll-like receptor 4 (TLR4) in focal cerebral ischemia reperfusion (I/R) injury.Methods C3H/HeJ mice with TLR4 gene mutation (n =30) and C3H/HeN with normal TLR4 gene mice (n =30) were divided into 4 groups at random (random number),namely C3H/HeJ model group (n =18),C3H/HeJ control group (n =12),and C3H/HeN model group (n =18).C3H/HeN control group (n =12).Middle cerebral artery was occluded to make I/R model in mice by using thread embolism method.Brain tissues were collected after ischemia for one hour and reperfusion for 12 hours.Stroke outcome was evaluated by determination of infarct volume of brain tissue and assessment of neurological scores.And brain injury after cerebral I/R was observed by optical microscope after TTC and HE staining.The immunofluorescence technique and RT-PCR were used to determine the protein level and expression of S100A8 mRNA in damaged brain tissues.Results Compared with C3H/HeN model mice,TLR4-deficient model mice (C3H/ He J) had lower infarct volumes and better outcomes of neurological function after resuscitation for 12 hours.Compared with control groups,the expression of S100A8 mRNA and level of S100A8 protein increased greatly in damaged brain tissues of model mice after I/R injury.In addition,model mice with lacked TLR4 (C3H/HeJ) had lower expression of I/R-induced S100A8 mRNA than C3H/HeN mice in model group,indicating that the close relationship between the levels of S100A8 and TLR4.Conclusions S100A8 interaction with TLR4 might be involved in brain damage and in inflammation triggered by I/R injury.
