1.The Empirical Study on mtDNA4834 Deletion in Mimetic Aging Model Rat’s Cochlear

Objective To study the effect of Shenqiwan and vitamin E on mtDNA 4834 deletion in a mimetic aging rat model .Methods Forty rats three months old ,were randomly divided into four groups :group A ,which was treated with hypodermic 5 % D -galactose for 8 weeks ,and group B (shenqiwan group ,n=10) which was treated daily with Shenqiwan for 2 weeks after hypodermic 5 % D -galactose for 6 weeks ,group C (vitamin E group ,n = 10) which were treated daily with Vitamin E for 2 weeks after hypodermic 5 % D -galactose for 6 weeks ,group D were given saline only .Auditory brainstem response (ABR) was used to detect the hearing thresh-old of rats before and after the treatment .The inner ear tissue was harvested and mitochondrial DNA was amplified to identify the 4834 bp deletion mutation by nested primer polymerase chain reaction (nested PCR) technique .Re-sults The threshold of ABR in the group A ,B and C were significant higher than that in group D .However the difference in shift of ABR threshold between group A ,B ,C and group D was not significant .The mtDNA4834 de-letion rate in three modeled groups (group A ,B ,C) was significant high than that of in group D .There was statis-tically significant difference between the treated group (group B ,C) and the model group (group A) on mtDNA4834 deletion in cochlea ,but the difference between the treated groups on mtDNA 4834 deletion in cochlea was not statistically significant .Conclusion Shenqiwan and vitamin E can reduce the aging induced mtDNA 4834 deletion in aging rats inner ear .
2.Investigation on body composition among children and adolescents in Dalian

Objective To explore the development regularities and characteristics of child and adoles-cent body composition in Dalian,to provide scientific basis for their healthy growth and basic evidence for nutri-tional status and healthcare,and to accumulate dynamic date for evaluation of children's growth and development in the future.Methods Two primary schools、two secondary schools and one high school in Dalian were en-rolled,where four classes were taken from each grade,and 2 027students aged from 6-18 years were systematical-ly collected.The collected students were given height and weight tests,and body composition was examined with bioelectrical impedance analysis.Overweight and obesity were determined by body fat percent.Results (1 ) Height,weight,body fat percent,fat mass,lean body mass,muscles,bone mass,BMI and basal metabolism rate of the students increased from 6-18 years in Dalian,and the growth curves of boys and girls appear rising tenden-cy,while the percentage of total body water decreased.(2)The muscles,lean body mass and basal metabolism rate were obvious sex differences among all the age groups,and the others were sex difference in part of age. Body fat percent and fat mass of girls were greater than those of boys,while the other indexes of boys were grea-ter.(3)The obesity rate of girls in Dalian was 35.7%,while the boys'was 16.7%.Conclusion (1 )The find-ings were consistent with the growth characteristics of adolescent boys and girls aged 6-18 years in Dalian.(2) The muscles,lean body mass and basal metabolism rate were obvious sex differences among all the age groups, and the others were sex difference in part of age.(3)The obesity rate of students in Dalian was higher than the government standard.
3.Expression of androgen receptor in breast cancer and its prognostic significance

Objective · To investigate the expression of androgen receptor (AR) in human breast cancer and its prognostic significance. Methods · A total of 183 female patients with breast cancer were selected. The expression of AR in breast cancer tissues was detected by immunohistochemical staining. The relationship of AR expression with the clinicopathological characteristics was analyzed. Survival analysis of AR gene expression by an online database (Kmplot) was also performed. Results · The positive rate of AR expression was 47.5% and was significantly higher in grade 1 and grade 2 group than that in grade 3 group (57.6% vs 25.0%, P<0.05). Breast cancer patients with estrogen receptor, progesterone receptor and P53 positive expression had significantly increased AR levels (P<0.05). AR expression were significantly higher in patients with luminal type as compared withthose in patients with human epidermal growth factor receptor-2 (HER2) positive and patients with triple negative breast cancer (51.7% vs 31.6%,P<0.05). Kaplan-Meier curves identified that AR gene expression was positively correlated with relapse-free survival, overall survival and distant metastasis-free survivalof breast cancer patients (P<0.05). Conclusion · The breast cancer patients with AR positive expression have a better prognosis, which suggests that inhibitors of AR pathway may be a treatment option for breast cancer.
4.Nationwide intercomparison of 226Ra,232Th and 40K for soil and building material by γ-spectrometry analyses in 2008

Objective To assess the accuracy and precision of γ-spectrometry analysis, and to obtain accurate and valid measurement results in the middle term and long term. Methods A nationwide intercomparison on gamma-ray spectrometry measurement of activity concentration of 226Ra, 232Th and 40K in soil and building material was organized by National Institute for Radiological Protection( NIRP) , China CDC. Results 15 laboratories participated in this intercomparison, with 13 laboratories produced acceptable results. Only 2 laboratories were classified as " not acceptable" , including one for inappropriate accuracy in determination of 40K and another for inappropriate precision determination of 226Ra in both kinds of the samples. Through comment and discussion, the second round intercomparison got satisfactory results. Conclusions The overall measurement results of samples for intercomparison are in close agreement with the reference values. Most of the laboratories involved in the intercomparison have good ability in γ-spectrometry analysis.
5.Long-term clinical observation of anterior allogenic iliac graft in treatment of thoracolumbar fractures

Objective To evaluate the long-term clinical efficacy of iliac bone block allograft in anterior reconstruction of thoracolumbar fractures.Methods A follow-up study was carried out on 93 patients with thoracolumbar fractures treated by anterior decompression and interbody fusion with iliac bone block allograft from February 2004 to March 2007.The radiographic and clinical outcomes were retrospectively assessed.Results Sixty-six patients were followed up for 60-97 months (mean,78 months).The effective interbody fusion rate was 100%,with the fusion time of 6-12 months (mean,10 months).The Cobb' s angle corrected from pre-operative (21.6 ± 8.3)° to post-operative (5.8 ±5.2)°,but when the infusion became effective,the angle showed average loss of 2.4°.All the 66 patients were associated with various degrees of neurological deficiency,but the patients with incomplete nerve dysfunction obtained different degree of recovery.Conclusions Iliac bone block allograft is effective in reconstruction of spinal anterior-middle column stability with the aid of internal fixation instruments.Thereby,it may be a potential alternative to autograft in the treatment of thoracolumbar fractures and can attain early and late stage persistent spinal stability.
6.The protective effect of sufentanil postconditioning on myocardial ischemia-reperfusion injury in rats

Objective To evaluate the effect of sufentanil postconditioning on myocardial ischemia-reperfusion injury in rats, and to investigate the possible mechanism. Methods Thirty male Sprague-Dawley ratswere randomly divided into 5 groups (n = 6 each): sham group (group S), I/R group (group I/R), low dose of sufentanil postconditioning group (group SL), high dose of sufentanil postconditioning group (group SH) andsufentanil postconditioning plus wortmannin group (group WM). Different drugs were injected before reperfusion: normal saline (NS) and sufentanil 1 μg/kg in group SL, NS and sufentanil 3 μg /kg in group SH, wortmannin 15 μg/kg andsufentanil 1 μg/kgin group WM, and NS in group S and I/R. At the end of reperfusion, artery blood was collected for assessment of plasma cTnI concentration; And the heart was harvested for determination of infarct size (IS) and area at risk (AAR). Results Compared with group S, cTnI concentration was increasedin the rest groups (P< 0.01); Compared with group I/R, cTnI concentration was decreased in group SL, SH and WM, while IS/AAR reduced in group SL and SH (P < 0.01); Compared with group SL, cTnI concentration and IS/AAR were decreased in group SH while increased in group WM (P <0.01). Conclusion Sufentanil postconditioning could attenuate myocardial ischemia-reperfusion injury in rats, and the mechanism involved PI3K related pathway.
7.Construction of mouse interleukin-10 recombinant adenovirus vector and gene modification for dendritic cells

BACKGROUND: Few reports concern the effects of dendritic cells-a kind of antigen presenting cells, and interleukin-10 (IL-10) on airway hyperreactivity or inflammation. OBJECTIVE: To construct mice IL-10 recombinant adenovirus vector Ad-mIL-10 to acquire the dendritic cells modified by mIL-10, which can provide a foundation for the further study. METHODS: Mouse IL-10 (mIL-10) gene comprise of enzyme cutting spot was synthesized according to the mIL-10 gene sequence and multiclone spot of adenovirus vector, connected to pMD18-T vector and sequenced. MIL-10 was subcloned to BD Adeno-X~(TM) vector, packed and augmented in HEK 293 cells, following determine the protein expression, and the vector was transfected to mice bone marrow-derived dendritic cells. RESULTS AND CONCLUSION: Recombinant adenovirus vector Ad-mIL-10 was successfully synthesized, packed and augmented, which could highly express protein IL-10. Bone marrow-derived dendritic cells were successfully cultured and transduced in vitro. It suggested that it is feasible to transfect mice dendritic cells by Ad-mIL-10 adenovirus vector. The study can provide more sufficient theoretic evidence for the possibility of correlative gene therapy.
8.Curative effect of L-carnitine on neonatal abnormal cardiac troponin I caused by asphyxia

Objective To explore the protective effect of L-carnitine on neonates with myocardial injury caused by as?phyxia. Methods Forty-four neonates with myocardial injury caused by asphyxia were randomly divided into L-carnitine treatment group (21 cases) and control group (23 cases). Patients in control group were received routine treatment and pa?tients in treatment group were given L-carnitine 0. 1 g/(kg · d) on the basis of routine treatment for 7 days. Symptoms and physical signs were observed before therapy and during the treatment in two groups. Before and after the treatment, plasma levels of free L-carnitine and cardiac troponin I (cTnI) were detected with the method of colorimetric assay and chemilumi?nescent, respectively. Results The clinical effective rate was significantly higher in treatment group than that of control group (90.48%vs 60.87%, P<0. 05). Compared with the control group, there was a significantly higher plasma concentra?tion of free L-carnitine in treatment group after treatment [(27.00±5.69)μmol/L vs (13.20±3.04)μmol/L, P<0.05]. In treat?ment group, plasma concentration of free L-carnitine was significantly higher after treatment than that of pre-therapy [(14.87 ± 3.95)μmol/L,P<0.05]. Compared with the control group, there was a significantly lower plasma concentration of cTnI after treatment in treatment group [(0.025±0.006)μg/L vs (0.046±0.010)μg/L, P<0.05]. In the treatment group, there was a significant correlation between decreased plasma concentration of cTnI and increased plasma concentration of free L-carnitine (r=0.899, P<0.05). Conclusion Administration of L-carnitine can effectively decrease the abnormal plasma lev?el of cTnI in neonates with myocardial injury caused by asphyxia, and thereby protect the myocardium.
9.Analysis of nosocomial infection in patients with multiple trauma in intensive care unit

Objective To discuss the features of nosocomial infection in patients with multiple trauma in intensive care unit and the preventive measures. Method The nosocomial infections occurred in 137 patients with multiple trauma were analyzed including site of infection, pathogenic organisms and bacterial strains. Results The rate of nosocomial infection was 62.8%, mainly the lower respiratory tract infection and wound infection, the G-negative bacteria infection accounted for 67.8%, mostly Escherichia Coli, Pseudomonas aeruginosa and A cinctobacter baumanni I. The G-positive bacteria infection accounted for 32.2%, mainly Staphylococcus aureus and Coagulase negative staphylococcus. The positive detection rates of Klebsiella and pseudomonas aeruginosa were increasing year afer year. Conclusions The incidence of nosocomial infection in patients with multiple trauma is high, suggesting some appropriate preventive measures should be taken to reduce the rate of infection.
10.The changes of serum bilirubin level in elderly patients with acute cerebral infarction and its significance

Objective To investigate the changes of serum bilirubin level in elderly patients with acute cerebral infarction and its significance.Methods 164 hospitalized elderly patients,who suffered from acute cerebral infarction within 1 week after onset,were divided into 2 groups according to age:group A aged over 60 years(n=85) and group B aged 40-60 years(n=79),and 66 healthy subjects aged over 40 years were collected as controls(group C).Serum bilirubin levels in all subjects were determined.The ratio of pulse pressure over mean arterial pressure(PP/MAP) in group A and B was calculated.Nerve function scores in the three groups were detected before and after 2 weeks of treatment.Meanwhile,the data of risk factors including blood glucose,blood pressure,blood lipids,smoking and drinking in group A and B were collected.Results Compared with group C,serum total,direct,indirect bilirubin levels were increased in group A and B(both P<0.01),and the change was smaller in group A than in group B(P<0.05).The nerve function scores was lower in group A than in group B before and after treatment [(35.2±12.6) vs.(44.3±7.9),(40.7±9.1) vs.(51.3± 4.1),t=5.58,9.73,both P<0.01],but PP/MAP and the numbers of risk factors were higher in group A than in group B [(0.46±0.06) vs.(0.38±0.06),93.01 vs.71.20,both P<0.01].There were no significant correlations of serum total,direct and indirect bilirubin levels with nerve function scores in group A or B(all P>0.05).Conclusions Serum bilirubin level is increased in patients with acute cerebral infarction,but the endogenous antioxidant capacity is decreased because of aging,multiple risk factors and more serious atherosclerosis in elderly patients,and the increment of bilirubin level is relatively smaller in acute cerebral ischemia,leading to the reduced protective effect against stress.Serum bilirubin level may influence the prognosis in patients with acute cerebral infarction.
