Objective To verify the targeting regulatory relationship between microRNA (miR)-21-5p and mitogen-activated protein kinase kinase 3 (MKK3).Methods miR-21-5p and MKK3 targeted relationship was analyzed by bioinformatics database predict.Constructing MKK3 3'UTR reporter vector (MKK3-3U) and mutation reporter vector (MKK3-3 U-M) was constructed by synthesizing miR-21-5p mimics (mimic),miR-2 1-5p inhibitor (inhibitor),nonsense miR (NC).NC (NC1 group),mimics (mimic1 group),inhibitor (inhibitor1 group) together with MKK3-3U cotransfected human embryonic kidney (HEK293) cells,respectively.Empty vector-pYr-MirTarget (NC2 group),MKK33U (Mt group),M KK3-3U-M (Mut group) together with mimic co-transfected HEK293,respectively.The fluorescence ratio was detected by dual luciferase assay.NC,mimics,inhibitor intervened human renal tubular epithelial (HK-2) cells,respectively.RT-PCR and Western blot were used to test cell MKK3 expression level in HK-2.Results 1.The fluorescence ratios of NC1 group,mimicsl group,and inhibitor1 group were 100.00％,67.99％,133.17％,and there was difference between mimic1 group and NC1 group(t =19.93,P ＜ 0.05).2.The fluorescence ratios of NC2 group,Mt group and Mut group were 100.00％,65.30％,98.48％,and there was difference between Mt group and NC2 group (t =14.39,P ＜ 0.05).There was no significant difference between NC2 and Mut group (t =0.63,P ＞ 0.05).3.MKK3 mRNA relative expression levels of the control group mimic2 group and inhibitor2 group were 1.36 ± 0.02,1.01 ± 0.04,1.43 ± 0.06 ; relative protein expression levels were 0.97 ± 0.05,0.62 ± 0.06,1.36 ± 0.32.MKK3 mRNA and protein levels of mimic2 group were significantly lower compared with the control group (F =85.98,118.26,all P ＜ 0.01).Conclusions MKK3 is the target gene of miR-21-5p.miR-21-5p can regulate the expression of MKK3 both in mRNA and protein levels.miR-21-5p may be an important regulatory molecule in p38 mitogen-activated protein kinase signaling pathway.

Background and purpose:Nasopharyngeal carcinoma is sensitive with radiotherapy, and it is of great signiifcance to study the resistance mechanism. This study aimed to analyze the expression of TNFAIP3 in the radiotherapy of nasopharyngeal carcinoma, the difference between radiosensitivity and radioresistance, the relationship with the occurrence and development of radioresistance.Methods:Detection of TNFAIP3 mRNA expression was carried out by TNFAIP3 multi-point labeled DIG probein situ hybridization.Results:In radiosensitivity and radioresistance of nasopharyngeal carcinoma, the moderately and strongly positive TNFAIP3 mRNA expression rates were 15% and 45%, (χ2=5.274,P=0.021 6), there were statistical significance. In the radioresistat nasopharyngeal carcinoma, TNFAIP3 mRNA moderately and strongly positive expression was positively correlated with TNM stage. In the degree ofⅢ-Ⅳ tumor, it had moderately and strongly positive expression rate of 48.28%.In the degree ofⅠ-Ⅱtumor, it had moderately and strongly positive expression rate of 36.36% (χ2=33.240,P<0.005); The expression rates in distant metastasis and no distant metastasis were 81.81% and 31.03% (χ2=6.385,P=0.011 5), the difference had statistical signiifcance.Conclusion:TNFAIP3 has antitumor effect, and it is correlated with the occurrence and development of radioresistant in nasopharyngeal carcinoma.

Objective To investigate the expression of visfatin in the livers of rats suffering from non-alcoholic fatty liver disease (NAFLD) and to expect the development of new diagnosis and treatment for NAFLD.Methods A total of 40 Sprague-Dawley (SD) rats was randomly divided into control and NAFLD model groups.For control group,the rats were given basic diet and subcutaneous injection of saline(0.2 ml/100 g weight)twice a week.However,rats in the model group were given high fat diet and were given subcutaneous injection of 40 ％ carbon tetrachloride (CCl4) salad oil (0.2 ml/100 g) twice a week.The levels of alanine aminotransferase (ALT),aspartate aminotransferase (AST),triglyeride (TG),total-cholesterol (TC) in serum together with the visfatin expression in hepatic tissue were analyzed at the 2nd,4th,6th,and 8th weeks after operation,respectively.Results The levels of ALT,AST,TG,and TC in model group were increased gradually as time going(P ＜ 0.05).Immunohistochemistry and reverse transcription polymerase chain reaction(RT-PCR) experiments showed the level of visfatin in model group was significantly higher than control group in each period,with a increasing trends with the modeling time (P ＜ 0.05).Conclusions (1)After high-fat feeding and intraperitoneal injection of CCL4,a model of adult SD rats suffering from NAFLD was constructed.(2)In livers of rats suffering from NAFLD,the expression of visfatin was increased while the level of liver steatosis was enhanced.Visfatin would be a new target to observe the severity of NAFLD.

Good nursing care can improve patients' com-fort degree and satisfaction to the treatment. It can also effectively relieve abdominal distention and reduce ad-verse reactions.

Objective To analysis pudendal nerve block anesthesia combined with noninvasive delivery on maternal vaginal delivery quality.Methods 60 primipara who were received vaginal delivery in our hospital were collected.All primipara were randomly divided into anesthesia group and no anesthesia group by the order of entry, 30 cases in each group.The no anesthesia group was given noninvasive delivery operation, and the anesthesia group received unilateral or bilateral nerve block anesthesia.Perineal laceration degree and incidence rate, time of the second stage of labor, neonatal asphyxia and two groups of maternal postpartum recovery were compared after the treatment.Results After the birth of a child, Compared with no anesthesia group, the incidence of maternal perineal laceration was lower in the anesthesia group, and the difference was statistically significant(P<0.05);the second stage of labor time of primipara was shorter in the anesthesia group(P<0.05); the pain score,getting out of bed activity time,average length of hospital stay,and incidence of postpartum complications of primipara were lower in the anesthesia group(P<0.05).Conclusions Pudendal nerve block anesthesia combined with noninvasive delivery operation can improve vaginal delivery quality, and have a guiding significance to clinical.

Objective To investigate the 6 short tandem repeats(D2S1338,D8S1179,D16S539,D18S51,D19S433,D21S11) polymorphism in Maonan and Mulao minority and their genetic relationships with other 5 minorities in Guangxi Province and to enrich the genetic database of Maonan and Mulao populations. Methods The allelic frequencies and the genotype of six STR loci were generated from 383 unrelated Maonan and Mulao individuals by PCR_STR and ABI3100. Results Sixty_one and 65 STR alleles were found in the six STR loci of Maonan and Mulao populations,with their frequencies ranging from 0.002 5-0.320 0 and 0.002 7-0.284 2 respectively.Two hundred and sixteen and 218 genotypes were found in Maonan and Mulao populations,with their frequencies ranging from 0.005 0-0.150 0 and 0.005 5-0.158 5 respectively.Their average heterozygosities were above 0.8;polymorphism information content were above 0.8 their accumulative discrimination power were over 0.999 999 98,and the probability of paternity exclusion were over 0.998 3.The results of genetic distance and phylogenetic tree showed that there were closest genetic relationship between Maonan and Mulao minorities,and farthest genetic relationship was existed between Shui and Yi minorities.Seven minorities were clustered to 2 groups on phylogenetic tree.Yi minority was clustered to one group,whereas the other 6 minorities(Maonan,Mulao,Hui,Miao,Shu and Jing nationalities) were clustered to the other group.Conclusion Six STR loci of Maonan and Mulao minorities possesse the characteristic of high genetic diversities which has great practical value.Therefore,the obtained data can be used in human population genetics investigation,individual indentification and paternity test.The genetic variation of STR among 7 populations is basically consistent with their historical culture and geographic distribution.

Objective To explore the clinical features, diagnosis, treatment, and prognosis of pertussis in neonates. Methods Clinical data from 17 neonates with pertussis conifrmed by real-time polymerase chain reaction (PCR) of Bordetella pertussis, were retrospectively analyzed. Results In 17 neonates ( 8 males and 9 females), 2 neonates were premature. The age at onset was from 15 to 27 days. Thirteen cases had a history of close contact with coughing patients, 4 cases had nasal congestion and runny nose in the early stage, 5 cases had typical spastic cough, one case had cough with inspiratory“whoop”, 6 cases had lfushing in face after severe cough, 7 cases had severe cough with cyanosis, 2 cases had systemic cyanosis before hospitalized, 7 cases had regurgitation after cough, and 3 cases had fever. Fifteen cases had increased peripheral blood white blood cell count, ( 12 . 57-79 . 63 )× 109/L, with lymphocyte at 45 . 1% -75 . 2%. The PCR copy number of pertussis was 7 . 12 × 102-1 . 04 × 108/mL. Seventeen neonates were all treated with intravenous infusion of erythromycin, and all of them had a signiifcantly improvement, and no death case. Conclusions Neonates with a history of contact with coughing patients, paroxysmal cough and increased of peripheral blood leucocyte presented as lymphocyte increase, need to be alert to pertussis, and relative examination should be taken as early as possible.

To analyze the clinical features and prognosis of fulminant Wilson's disease (FWD) in patients with hepatitis B virus (HBV) infection.Twenty-seven patients were enrolled in Guangzhou Eighth People's Hospital from 2005 to 2015,including 13 FWD patients with HBV infection and 14 FWD patients without HBV infection.Clinical efficacy and survival rate were evaluated.Baseline biochemical data in two groups were comparable(P ＞ 0.05),including total bilirubin,prothrombin activity,serum albumin,alpha fetal protein,alanine transaminase,ceruloplasmin and 24 hours urine copper.Treatment in FWD group with HBV infection was ineffective,including 9(9/13) deaths and 4(4/13) patients receiveing liver transplants.However,7 (7/14) cases in the other group did not response to the treatment,including 6 (6/14) deaths and 1 (1/14)patient receiving liver transplant.The prognosis in the two groups is significantly different(P =0.006),which is much worse in FWD patients with HBV infection.

Objective:To evaluate the methods of nano-dispersion systems and the stability of samples in medical devices.Methods: By analyzing the cause of agglomeration and comparing kinds of methods to orient the elements of nano-dispersion Systems and the evaluation means. Results: To obtain the repetitive results, the foundation methods of nano-dispersion systems should be recorded in detail and all the following tests should be based on it.Conclusion: The constitution and evaluation of a dispersion system in obtaining representative and reproducible results in biological evaluation is very important and should be studied case by case.

Angiogenesis is one of the effective strategies for the treatment of ischemic cerebrovascular disease, and a variety of angiogenic factors and antiangiogenic factors involved in the process of regulation. Apelin is a newly discovered peptide, ari it is the endogenous ligand of G protein-coupled receptor APJ (Angiotensin Ⅱ Type 1 Receptor-Associated Protein). Apelin and APJ express widely in rat and human hearts and brains, etc. Apelin-APJ system has a role in promoting angiogenesis. It has an important significance in the treatment of ischemic diseases. This article review the mechanisms of action and advances in angiogenesis of apelin-APJ system.
