1.Effect of concanavalin A on mouse islet transplantation

To test whether Con A has antirejection effect on mouse islet allograft, 600-700 handpicked islets of C57BL/6 mice was transplanted beneath the kidney capsule of the streptozotocin induced diabetic BALB/C recipients. In control group the mean survival time (MST) of the grafts was 14.1?2.4 days. Treatment with Con A significantly prolong the MST of the islet allografts. In group with Con A treatment of the donor islets prior to the transplantation the MST was 25?5.4 days. The MST increased sigificantly after ip injection of Con A to recipient mice. The combined treatment of islet and recipient with Con A resulted in a prolongation of MST to 25.2?4.4 days. The experiments concerning the mechanisms of the antirejection effect of Con A showed that Con A was able to combine with the islet cell membrane well masking perhaps the histocmpatibility antigen. Furthermore the lymphocytes of mice after i. p Con A injection suppressed the lymphocyte proliferation reaction of the svnergic mice to mitogen.
2.Observation of the curative effect of colonoscopy combined with laparoscope in the treatment of colonic polyps

Objective To explore the curative effect of colonoscopy combined with laparoscope in the treatment of colonic polyps.Methods The clinical data of 48 cases with colonic polyps were retrospectively analyzed.According to the operation pattern,the patients were divided into observation group(23 cases)and control group(25 cases).The observation group received colonoscopy combined with laparoscopy for radical surgery of colon polyps,the control group used the traditional open surgery to remove polyps.The operation time,bleeding volume, exhaust and defecation time,hospital days and cost as well as postoperative complications were observed.Results The two groups were successfully completed surgery.The operative time,bleeding volume of the observation group were (78.3 ±8.2)min and (1 3.1 ±4.5)mL respectively,which of the control group were (1 1 5.5 ±1 0.1 )min, (63.6 ±1 8.1 )mL,the differences between the two groups were statistically significant(t =1 3.93,1 3.01 ,all P <0.05).The anal exhaust time and defecation time of the observation group were (25.1 ±6.5)h and (42.5 ±7.6)h, which were significantly shorter than those of the control group[(36.6 ±8.1 )h,(82.3 ±1 1 .5)h],the differences were statistically significant(t =5.39,1 4.01 ,all P <0.05).The hospitalization time,hospital costs of the observation group were (5.5 ±0.6)d,(1 3 842.5 ±875.5)yuan,which of the control group were (8.2 ±1 .5)d,(1 8 992.5 ± 1 001 .5)yuan,the differences between the two groups were statistically significant(t =8.05,1 8.89,all P <0.05). Pulmonary infection in 1 case,anastomotic fistula in 2 cases,1 case of intestinal obstruction in the observation group, and in the control group,pulmonary infection occurred in 2 cases,2 cases of intestinal obstruction,the postoperative complications between the two groups had no statistically significant difference(P >0.05).Conclusion For simple colonoscopy treatment difficulties of colon polyps,colonoscopy,laparoscopy combined treatment is minimally invasive and can improve the safety,strictly control surgical indications,can give full play to the double mirror combined advantage.
3.Effect of ginseng-sanqi extract on the Ras associated signal proteins.

OBJECTIVETo explore the effect of ginseng-sanqi extract (GSE) on the vascular endothelium growth factor receptor-2 (VEGFR-2), Ras and mitogen-activated protein kinase (MAPK) in VEGFR-2-Ras-MAPK signal pathway of angiogenesis in cultured human umbilical vascular endothelial cells (HUVECs).
METHODSThe protein expressions of VEGFR-2, Ras and MAPK in HUVEC and the effects of GSE (at different doses) and basic fibrin growth factor (bFGF) on them were detected by Western-Blot test.
RESULTSGSE can enhance the expression of angiogenesis signaling proteins (VEGFR-2, Ras, MAPK) to different extents, especially in the groups treated by bFGF or higher dose GSE, the levels showed significant differences to those in the blank group (P<0.05).
CONCLUSIONGSE can promote the expressions of VEGFR-2, Ras and MAPK in the angiogenesis signaling pathway, which may be the foundation of Chinese medicine for tonifying qi and activating blood circulation in promoting endothelial proliferation and angiogenesis.

5.Effect of preoperative sleep disturbance on efficacy of postoperative analgesia and comfort in patients undergoing colorectal cancer resection surgery

Objective To evaluate the effect of preoperative sleep disturbance on efficacy of postoperative analgesia and comfort in patients undergoing colorectal cancer resection surgery.Methods 60 colorectal cancer patients with elective anesthesia laparoscopic radical resection were selected.According to the presence or absence of preoperative sleep disorders,they were divided into the sleep disorder group (group A) and non-sleep disorders group (group B),30 patients in each group.Two groups of patients used the same method to maintain anesthesia induction.Intraoperative intravenous nicardipine controlled hypotension in parallel to maintain MAP 60-75mmHg,HR 65-95times/min.The patients received PCIA pump at the end of surgery.Postoperative 24h VAS scores were maintained VAS score ≤3 points,when VAS score > 3 points,given the pressing PCA analgesia remedy.The remedy situation within 24 hours after surgery and comfort analgesic score were recorded.Results Compared with group B,postoperative PCIA pressing number of group A significantly increased [A group:(11 ± 5),group B:(6 ± 5),t =2.44,P < 0.05].At the end of surgery,postoperative 2h,4h,8h pain scores of group A were (2.8 ± 0.4),(2.5 ± 0.7),(3.2 ± 0.6),(3.5 ± 0.5),respectively,which were significantly higher than those of group B [(1.8 ± 0.5) points,(1.8 ± 0.4) points,(1.9 ± 0.3) points,(2.9 ± 0.3) points; at the end of surgery t =5.78,postoperative 2h t =4.56,postoperative 4h t =4.17,postoperative 8h t =2.09,all P < 0.05],but postoperative 12h,24h had no significant change in pain scores (P > 0.05).Compared with group B,the comfort scores of A group at the end of surgery,postoperative 2h,4h,8h,12h[group A:(2.8 ±0.3) points,(2.5 ±0.9)points,(2.6 ±0.9) points,(2.5 ±0.5) points,(2.9 ± 0.6) points ; group B:(3.8 ± 0.4) points,(3.3 ± 0.5) points,(3.6 ± 0.8) points,(3.3 ± 0.3) points,(3.3 ± 0.6) points] were significantly lower (at the end of surgery t =9.87 ; postoperative 2h t =4.94 ; postoperative 4h t =6.87 ; postoperative 8h t =7.61 ; postoperative 12h t--2.79,all P < 0.05),after 24h comfort score had no significant change (P > 0.05).Conclusion Preoperative sleep disorders can affect laparoscopic colorectal cancer after radical surgery analgesia and comfort of patients.
6.The effect of small doses of dexmedetomidine on postoperative analgesia and comfort after colorectal cancer radical operation in elderly patients

Objective To observe the effect of small doses of dexmedetomidine on postoperative analgesia and comfort after colorectal cancer radical operation in elderly patients .Methods 60 cases of elderly patients for elective colorectal cancer resection ,ASAⅠ~Ⅱgrade anesthesia were selected .According to randomly digital table , they were divided into the dexmedetomidine given group ( group D) and control group ( group C),30 cases in each group.The patients of group D was given to 0.5μg/kg dexmedetomidine for induction of anesthesia infusion , 15minutes bolus injection was completed ,intraoperative maintain 0.2μg· kg-1 · h-1 dexmedetomidine fixed to the front end of surgery 10 min.And group C was given to infusion of 0.9%sodium chloride injection in the same capacity . The operation time, amount of fentanyl using , comfort and VAS scores were recorded within 24h, maintain VAS score≤3 points.When VAS score>3 points,the pressing PCA was given ,and the number of times of pressing the PCIA was recorded.The adverse reactions occurred within 24h such as nausea and vomiting were also recorded . Results In the two groups,the results of postoperative analgesia were good ,but the VAS score of group D at each time of PCA point were less than those of group C (tend of surgery =5.99,tafter operation 2h =4.76,tafter operation 4h =4.08,all P<0.01).The Bruggrmann comfort score of group D were greater than those of group C (tend of surgery =9.86,tafter operation 2h =4.91,tafter operation 4h =6.84,all P<0.01).The incidence of adverse reactions of group D was 13.30%,which was less than 26.67%of group C(χ2 =4.32,P<0.05).Conclusion Small doses of dexmedetomidine given to improve the analgesic effect in elderly patients with colorectal cancer cure can improve comfort and reduce the incidence of adverse reactions ,and has a role in reducing postoperative nausea and vomiting .
7.Retrospective analysis on Chinese medicinal treatment on 302 chronic heart failure patients of coronary heart disease and diabetes.

OBJECTIVETo explore the characteristics of Chinese medicinal treatment on 302 chronic heart failure (CHF) patients of coronary heart disease and diabetes.
METHODData of CHF patients of coronary heart disease and diabetes hospitalized in 9 hospitals of Beijing and Tianjin from Jan. 2003 to Sep. 2006 were collected and their clinical syndromes, condition of treatment (both western and Chinese medicine) were investigated and analyzed using the data system of Chinese medicine clinical research for coronary heart disease.
RESULTIn the 302 patients, the intravenous perfusion were mainly injections of puerarin, Salvia mitrorrhiza, sanchi total saponin and fleabane. The Chinese medicinal treatment applied was mainly the methods of supplementing Qi, activating blood circulation, dissolving phlegm. In the apozem, hoelen is the highest frequency in all drugs and the frequency in order is hoelen, glycyrrhizae, Salvia mitrorrhiza, astragali, Rhizoma Pinelliae, etc.
CONCLUSIONThe patients of these 9 hospitals were mainly treated by integrative medicine and the Chinese medicinal treatment applied was mainly the methods of supplementing Qi, activating blood circulation, dissolving phlegm.

8.Application of Single-cell RNA Sequencing Technology to Exploring Cardioprotective Mechanism of Chinese Medicine After Myocardial Infarction: A Rewiew

The pathological changes of the heart after myocardial infarction (MI) are complex, involving multiple molecular mechanisms and various cells, such as myocardial cells, endothelial cells, fibroblasts, and macrophages. With multiple targets, Chinese medicine demonstrates ideal cardioprotective effect. However, the complex mechanism of multi-component Chinese medicine formulas has not been elucidated, thus limiting the further application. The high-throughput single-cell RNA sequencing (scRNA-seq) technology offers single-cell transcriptome analysis of hundreds of drugs under different processing conditions in a single experiment and identifies the differences in the response of different cells and cell subtypes to drug treatment. scRNA-seq technology helps us to understand the exact cellular and molecular mechanisms of cardiac remodeling from acute ischemic events to chronic cardiac scarring. The application of scRNA-seq to studying the cardioprotective mechanism of Chinese medicine after MI can boost the development of Chinese medicine, help obtain richer, more accurate and comprehensive information. It can give us a clear insight into the mechanism of Chinese medicine based on complex network. In this study, we summarized the research on cardioprotective mechanism of Chinese medicine and introduced the development of scRNA-seq technology and the application to MI research. Finally, we explored the possible application prospects of scRNA-seq in the research on cardioprotective mechanism of Chinese medicine after MI, hoping to provide ideas for the modernization of Chinese medicine.

Coronavirus disease-2019 (COVID-19) is highly contagious. In the early stages of the disease, the symptoms of coldness, dampness and depression in the lungs often appear, including fever, fatigue, soreness, dry cough, poor appetite, and white greasy tongue coating. During the development of the disease, the damp toxin epidemic often enters the inner and generates heat, and the damp heat epidemic toxin blocks pleurodiaphragmatic sites, the triple energizer and lungs, with manifestations of chest tightness, shortness of breath, exacerbation after exercise, or high fever without bringing down, poor appetite, bitterness in mouth, fatigue, diarrhea, loose stools, and yellow and thick tongue fur. As the pathogen can go outward or enter more deeply inside to cause death at the moment, it is the crucial time to treat the disease. In this study, Haoqin Qingdantang was used to clear dampness and heat, reconcile Shaoyang channel and recover the triple energizer, detoxify the dampness fever epidemic toxin, and block the toxin inside, with a good efficacy. This prescription focuses on smoothing the Shaoyang gallbladder channel and the triple energizer, and regards the spleen and stomach as the acquired essence. In the prescription, Erchentang reconciles the spleen and stomach, elevates clear Qi and lower turbid Qi. Bupleuri Radix is added to increase its detoxification function, and Paeoniae Rubra Radix is added to circulate the blood and prevent pathogen from the blood. 10.Screening, identification and analysis of platelet differential functional proteins in patients with coronary heart disease of blood-stasis pattern.

OBJECTIVETo seek the key platelet functional proteins (PFPs) for the occurrence of blood-stasis pattern (BP) in patients with coronary heart disease (CHD).
METHODSPeripheral blood platelet protein of 22 patients and 24 healthy person (for control) were extracted respectively in 4 batches for carrying 4 times of the test out. Differential PFPs in samples were screened out by two-dimensional fluorescence difference gel electrophoresis, and identified with matrix-assisted laser desorption/ionization-time of flight mass spectrometry; then the identified proteins were further authenticated by Western-blotting.
RESULTSThirteen differential PFPs were screened out, and among them the 7 identified by spectrometry were: isoform 1 of integrin alpha- II b, isoform 2 of integrin alpha- II b, actin-cytoplasmic 1, actin-cytoplasmic 2, cDNA FLJ52842, cDNA FLJ55253, and cDNA FLJ43573 fis. Among them isoform 2 of integrin alpha- II b (CD41) and actin-cytoplasmic 2 (Acting) were authenticated successfully.
CONCLUSIONCD41 and acting are the possible marker proteins, and the other PFPs might play crucial roles in the occurrence and development of BSS in CHD.
