1.Study on Estrogenic Effects of DDT-derivative Pesticides

Objective To study the estrogenic effects of DDT-derivative pesticides. Methods Using a recombinant human estrogen receptor gene yeast cell to estrogenic activity of DDT pesticides. The different concentrations of 17?-estradiol and individual DDT isomers and homologue such as p,p′-DDT, o,p′-DDT, p,p′-DDD and p,p′-DDE and their mixtures were added to yeast cultures, then estrogenic activity was measured by quantification of ?-galactosidase after 4 hours culture. Results DDT showed the ability to play the estrogenic effects like a ligand to bind hER and the dose dependent model existed between the estrogenic effects and the DDT concentration. The relative activity of o,p′-DDT, EC50=5.30?10-7mol/L, was higher and the estrogenic effects of isomers exhibited synergism, AI=1.5. The relative activity of p,p′-DDE , EC50=9.28?10-5mol/L, was lower and the antagonistical action between the homologues was found, AI=-1.38. DDT pesticides presented a synergism, the regression equation was =-2.600 6x2-28.079x-21.757,EC50 was 2.57?10-8mol/L, AI=0.83. Conclusion DDTs are ER mediated environmental estrogens, p,p′-DDT, o,p′-DDT, p,p′-DDD and p,p′-DDE show a synergic effect.
2.Application of Gene Recombinant Yeast Cell in Screening to Environmental Estrogen

Objective To research the estrogen-like activity of the DDTs and the extract of vehicle exhaust by recombinant yeast system.Methods Being added into the yeast cell culture fluid,the objects reached the final concentration of 10-9~10-15 mol/L for 17?-estradiol,10-4~10-9 mol/L for DDTs(p,p'-DDT,o,p'-DDT,p,p'-DDD,p,p'-DDE),0.25~8.00 ml/ml for the extract of vehicle exhaust,then the cultivation was performed for 4 hours,the quantitative assay of ?-galactosidase activity accounts for the estrogenlike effect of the samples was carried out.Results DDTs were a group of environmental estrogens hormones,which could integrate estrogen receptors as the stimulators do and express the estrogen-like effect.Being tested separately,the DDTs showed the estrogen-like activity,which was stronger for o,p'-DDT and weaker for p,p'-DDE.The?-galactosidase activity of gasoline-fueled vehicle exhaust at the final concentration of 2.00~4.00 ml/ml was significantly different from that of the DMSO(P
3.mRNA expression of ciliary neurotrophic factor during injury and repair of optic nerves in rats

Objective To investigate the mRNA expression of ciliary n eurotrophic factor on the retina during injury and repair of optic nerves in rat s. Methods Thirty-five healthy SD rats were randomly divided into 3 groups: 5 in the control group, 15 in the simply transected optic nerve g roup and 15 in the optic nerve-sciatic nerve anastomosis group. The simply tran sected and optic nerve-sciatic nerve anastomosed models were set up, and the re tinal tissues of all of the rats were taken out after 3, 7 and 14 days, respecti vely; and the mRNA expression of CNTF in the 3 groups were observed by semi-qua ntitative reversal transcription-polymerase chain reaction method. Res ults A minimum expression of CNTF mRNA was found in the reti nae of the control group, and the increased rates of expression were found in th e retinae of the simple transection of optic nerve group with the increase rate of 100%, 594%, and 485% on the 3rd, 7th, and 14th day respectively after the ope ration, while in optic nerve-sciatic nerve anastomosis group, the increase rate s were found to be 258%, 752% and 515% on the 3rd, 7th, and 14th day respectivel y after the operation. Conclusion Retinal neurons can respond to axonal reaction of retinal ganglion cells by up-regulate endogenous CNTF aft er the injury of the optic nerves, which may provide a theoretic base for the application of the exogenous CNTF.
4.Pioglitazone, a peroxisome proliferators-activated receptor gamma ligand, inhibits dimethylhydrazine (DMH) induced aberrant crypt foci in rats

Objective: To investigate the chemopreventive effects of pioglitazone (exogenous PPAR? ligand) on rat colon aberrant crypt foci, a rat carcinogenesis model induced by dimethylhydrazine (DMH),and to compare pioglitazone with sulindac (a NSAID). Methods: Thirty two, 8 week old, female Sprague Dawley rats were randomly divided into four groups ( n =8 each). Group 1 rats were injected with DMH alone (120 mg?kg -1 , single subcutaneous injection). Group 2 rats were injected with saline alone. Group 3 rats were pre treated with sulindac (320 mg?kg -1 ) for 7 days before DMH initiation. Group 4 rats were treated with pioglitazone (100 mg?kg -1 ). The animals were killed at the end of the experiment (week 5) and the colons were stained with methylene blue. The aberrant crypt foci (ACF) of the colonic mucosa were assessed. Results: In Group 1 rats (DMH only), the average numbers of ACF/colon and AC/colon were (182?93) and (263?198), respectively. In Group 2 (saline group) rats, no ACF were found. In Group 3 (sulindac group) rats, the average numbers of ACF/colon and AC/colon were (91?49) and (140?69), respectively. 5.E-Flow imaging on detection of the fingertip's microvascular changes of hemodynamics in patients with essential hypertension

objective To assess relationship between microvascular changes of hemodynamics of the fingertip and evolution of hypertension in patients with essential hypertension(EH)by enhanced power Doppler imaging(E-Flow).Methods The right ring fingertip'S blood flow peak systolic velocity(PSV),end-diastolic velocity(EDV),resistance index(RI)of finger ventral arteriole and nail bed arteriole in thirty-nine patients with EH were separately observed and were made contrast with 35 age-matched healthy subjects by E-Flow imaging.Results Compared with traditional colour Doppler flow imaging and power Doppler imaging,E-Flow imaging was clearer to demonstrate the distribution and movement of the fingertip'S microvascular.It was shown that the PSV and EDV in the ring finger ventral arteriole and nail bed arteriole of EH were decreased(P<0.05),but the RI increased(P<0.05),along with the evolution of EH.Conclusions Fingertip'S microvascular changes of hemodynamics could be a criterion to detect obstruction alternations of essential hypertension.E-Flow imaging iS superior to traditional color Doppler flow imaging and power Doppler imaging in differentiating microvascular and could provide a new method in assessing microvacular changes of patients with EH for clinic.
6.Application of predictive and high- quality nursing in partients with pulmonary embolism or having risk factors of pulmonary embolism

Objective To summarize the effect of predictive nursing combined with high-quality nursing in patients with pulmonary embolism or having risk factors of pulmonary embolism (PE).Methods A retrospective analysis of nursing in 57 PE patients was camied out,Nursing flow chart was established.For patients with high risk factors of PE,predictive nursingwas administered,including analysis of risk factors,health education,general nursing and pertinent nursing.For PE patients,holistic nursing was administered,including emergeney nursing,basic nursing,observation of illness,nursing of thrombolytic therapy,psychological nursing and rehabilitation nursing, Results All patients accorded with thrombolysis indication and received thrombolysis treatment. Conclusions Predictive and high-quality nursing should be used in PE patients in order to improve treatment effect.
7.Incidence of deep vein thrombosis in patients with obstructive sleep apnea-hypopnea syndrome

Twenty five patients with obstructive sleep apnea hypopnea syndrome (OSAHS) were classified as mild, moderate and severe grade according to apnea hyponea index and lowest oxyhemoglobin saturation.Lower limbs were examined with ultrasonography, blood routine and D-dimer levels were measured in all patients.The results revealed an increased incidence of deep vein thrombosis with the extent of obstructive sleep apnea.There was no deep vein thrombosis in patients with mild-grade OSAHS, while there were 2 in moderate and 3 in severe patients.The blood D-dimer levels were (498 ± 22) pg/L, (659 ±43 ) μg/L, ( 1528 ± 181 ) μg/L in mild, moderate and severe patients, respectively.The hemoglobin levels were (150 ± 8) g/L, ( 183 ± 15) g/L and (261 ± 26) g/L in mild, moderate and severe patients,respectively.There may be some association of obstructive sleep apnea hypopnea syndrome with deep vein thrombosis.
8.Effect of Small Interfering RNA Silencing the Fatty Acid Synthase Gene on Lipid Metabolism in Human Hepatic Cell Line HepG2

Objective: To investigate the effect of the gene interfering technology on fatty acid synthase (FAS) gene silencing for lipid contents in human hepatic cell line HepG2 and to study the lipid metabolism related gene expression in HepG2 cells. Methods: A total of 3 pairs of small interfering RNA (siRNA) targeting different sequences of FAS mRNA were synthesized as FAS-siRNA-1, FAS-siRNA-2 and FAS-siRNA-3, meanwhile, 2 controls were established as Blank control group, in which HepG2 cells were not treated, and Negative control group, in which HepG2 cells were transfected by non-effective siRNA. The mRNA, and protein expression levels of FAS in HepG2 cells were examined by real-time lfuorescence quantitative RCR and Western blot analysis to screen the most effective pair of siRNA for FAS gene silencing; and that speciifc siRNA was transtected to HepG2 cells for 48 hours to detect the intra-/extra-cellular TG, TC levels and the mRNA expression related to lipid metabolism in HepG2 cells. Results: The screening experiment indicated that FAS-siRNA-3 was most effective for FAS gene silencing. Compared with Blank control group, the mRNA and protein expressions in FAS-siRNA-3 transfected HepG2 cells (Transfected group)decreased to (52.33 ± 3.07) % and (51.57 ± 3.14) % respectively. Compared with Blank control group, Transfected group had the reduced intra-/extra-cellular TG levels and reduced extracellular TC level; while increased mRNA expression of hepatic lipase,P<0.0001 and decreased mRNA expression of TG transfer protein in HepG2 microsome,P<0.05. Conclusion: FAS gene silencing could signiifcantly decrease the intra-/extra- cellular TG level and extracellular TC level in HepG2 cells, those ifndings need to be conifrmed by furtherin vivo andin vitro studies.
9.Distribution of Helicobacter pylori in stomach before and after the administration of Lansoprazole

Objective: To investigate the effect of the acid inhibitor-Lansoprazole on the distribution of Helicobacter pylori (H.pylori) in stomach. Methods: Biopsy specimens were taken from the duodenal ulcer patients who underwent gastroscopy before and after the treatment of Lansoprazole. The biopsy specimens were taken from the lesser curvature of the antrum and the greater curvature of the corpus respectively. H&E and Warthin-Starry staining were used for detecting the changing of active gastritis and the positive rate of H.pylori. Results: (1)The positive rates of H.pylori before treatment, 4 weeks after treatment and 3 months after treatment, in the lesser curvature of the antrum were 93.02%, 58.14%, and 86.05%, respectively. Objective To investigate the cellular immunologic response of TH 17/Treg cells in the peripheral blood of pelvic tuberculosis patients and explore their roles in the pathogenesis of pelvic tuberculosis.Methods The intracellular flow cytometry was performed to evaluate the expressions of TH 17 and Treg cells in 46 pelvic tuberculosis patients and 25 healthy controls in childbearing age.Twenty-eight of the 46 pelvic tuberculosis patients were followed up to monitor the variation of the TH17/Treg cells after 3 months and 6 months of anti-tuberculosis treatment.Results The percentage of TH 17 cells in the peripheral blood of pelvic tuberculosis patients was (3.26 ± 1.30) % which was significantly lower than that of healthy controls [(4.92 ± 1.71) %,P < 0.01].The percentage of Treg cells in the patients was (5.18 ± 1.53) % which was significantly higher than that of healthy controls [(3.26 ± 1.10) %,P < 0.01].The percentage of TH17 cells in the pelvic tuberculosis patients after 6 months of treatment was (4.67 ± 1.75) % which was significantly higher than that in the patients before treatment and after 3 months treatment [(3.26 ± 1.30) %,P < 0.01 and (3.70 ± 1.06) %,P <0.01,respectively].The percentage of Treg cells in pelvic tuberculosis patient after 6 months of treatment was (3.93 ±0.94)% which was significantly lower than that in the patients before treatment and after 3 months of treatment [(5.18 ± 1.53)%,P <0.01 and (4.94 ± 1.51) %,P < 0.01,respectively].The percentage of Treg cells in the patients after 6 months of treatment was still significantly higher than that of controls (P < 0.05).The TH 17/Treg ratio before treatment was significantly lower than that of healthy controls (P < 0.01),and the TH 17/Treg ratio was increased after 3 months of treatment but it did not show significant difference compared with that before treatment.The TH 17/Treg ratio after 6 months of treatment (1.18 ± 0.34) % was significantly increased in contrast to those after 3 months of treatment and before treatment [(0.77 ± 0.21) %,P < 0.01 and (0.55 ± 0.13) %,P < 0.01,respectively].The TH 17/Treg ratio could not rise to the normal level even after 6 months of treatment.Conclusion Both the TH 17 and Treg cells may involve in the immunologic responses of pelvic tuberculosis patients and the imbalance of TH1T/Treg cells may remain persistently.
