1.Pharmacokinetics of gatifloxacin mesylate and gatifloxacin in rats in vivo

Aim To investigate the difference in pharmacokinetics between gatifloxacin mesylate(GATM) and gatifloxacin(GAT) in SD rats in vivo.Method The plasma concentration-time courses of GAT in rats were measured by HPLC method after a single oral dose of GATM and GAT,and the pharmacokinetic parameters were estimated by program 3p97 software.Results The results shown that both of GATM and GAT plasma concentration-time courses were best fitted to two-compartment models after a single oral dose(GATM 23.43 mg?kg~(-1)and GAT 20 mg?kg~(-1),respectively).The major average pharmacokinetic parameters of GATM were as follwing: T_(2)?: 9.13 h,AUC: 15.05 mg?L~(-1)?h,C_(max): 4.41 mg?L~(-1),and T_(max): 0.5 h,respectively.On the other hand,the pharmacokinetic parameters of GAT were as follwing: T_(2)?:10.70 h,AUC:13.84 mg?L~(-1)?h,C_(max):4.25 mg?L~(-1),T_(max):0.5 h,respectively.Conclusion Under the equi-mole dose condition treated as above,both of pharmacokinetic characters between GATM and GAT were not significantly differences in rats
2.Effect of Family Support Training on Quality of Life for Patients with Post-stroke Depression

Objective To investigate the effect of family support training on quality of life of patients with post-stroke depression(PSD).Methods68 PSD patients were divided into the observation group and control group with 34 cases in each group.All cases of tow groups received regular diagnosis,treatment,and nursing care.And the same time,the family members of the patients of the observation group were given support training.The therapeutic effect of two groups was compared.ResultsThe scores of Hamilton Depression Scale(HAMD)of the patients of the observation group were significantly lower than that of the control group after treatment(P<0.01).ConclusionFamily support training can improve quality of life of stroke patients.
3.Study of rpoB Mutation in Rifampin Resistants Mycobacterium Tuberculosis

Objective To understand the rpoB gene mutation in M.tuberculosis isolates,and to evaluate their clinical value.Method 335 clinical isolates of mycobacterium tuberculosis(109 isolates drug susceptible,246 isolates rifampin-resistance or multidrug resistance including rifampin) were detected using polymerase chain reaction-single strand conformation polymorphism(PCR-SSCP).Results SSCP pattern of reference mycobacterium tuberculosis H37Rv as control,no mutation was found in rifampin-suscepitible 109 strains.SSCP patterns of 225/246 rifampin resistant clinical isolates were different from the normal control.The sensitivity was 91 5%.31 resistant isolates,included 25 abnormal isolates and 6 normal isolates of SCCP were identified.Sequencing showed 29 isolates had rpoB gene mutations and 3 isolates were not found rpoB gene mutations.The 3 most frequent rpoB gene mutation situs were Leu-531(19 isolates.TCGTTG) and His-526(7 isolates,CACTAC) and Asp-516(3 isolates,GACGTC).Conclusions The results confirm that rpoB gene mutation is the most important mechanism in rifampin resistant tuberculosis mutation situs are 531 tryptophan and 526 histidine;respectively.It is feasible that using PCR-SSCP to detect drug resistance in mycobacterium tuberculosis.
4.Study on the distribution of lymphocyte subsets in the eutopic and ectopic endometrium of women with endometriosis

Objective To investigate the abnormal distribution of lymphocytes in eutopic and ectopic endometrium of patients with endometriosis and its significance. Methods In 43 cases, biopsies of ectopic tissues were taken by laparoscopy and laparotomy from patients with endometriosis and eutopic endometrium by curettage at the same time. In 19 cases, eutopic endometrium was taken from hysterectomy for myomatous uterus. Immunohistochemical techniques were employed to demonstrate the difference in the number and ratio of the lymphocyte subsets between the patients with endometriosis and the controls. Results In the patients with endometriosis, in the proliferative phase, ectopic endometrium contained respectively CD+3、CD+8T cells and CD+68 macrophages (67.2±13.5)/5HP, (45.0±14.0)/5 HP and (37.2±10.6)/5 HP, significanly higher then that in the eutopic endometriun (52.4±11.3)/5HP (P<0.01), (32.5±10.0)/5HP (P<0.05), and (30.7±10.3)/5HP, and also higher as compared with the control group (52.1±14.9)/5HP (P<0.05), (28.9±12.7)/5HP (P<0.01), (26.3±9.3)/5HP (P<0.05); in the secretory phade, CD+8/CD+4, and CD+68 content was respectively 3.5±1.2,(40.3±12.2)/5HP, higher than that in the control group, 3.2±0.8 (P<0.05), (28.6±10.6)/5HP (P<0.01). The number of macrophages was also significantly increased. No cyclic changes in the number of lymphocytes in each subpopulation in ectopic endometrium were found. Conclusions In the patients with endometriosis, the changes in T lymphocytes and macrophages are mainly on the endometriotic sites. The infiltration of many lymphocytes and macrophages into the ectopic endometrium formed a chronic inflammatory process. The lymphocytes are not able to clear the ectopic endometrium in the late stages of endometrium,on the contrary, they stimulate the further growth of the endometrium.
5.Evaluation on the clinical effects for asynchronous cerebral arterial thrombosis with treatment of cerebral circulation therapeutic equipment

Objective: To investigate the effect about the treatment in asynchronous cerebral arterial thrombosis of cerebral circulation therapeutic equipment with disease function rehabilitation and the trigger factor of stroke and cerebral blood flow in 108 cases. Methods:Treatment group on the basis of conventional drug treatment plus brain circulation function therapeutic apparatus (KJ-3000-a) treatment in patients with stroke in different disease period. Using the neural function defect score and controllable risk factors of stroke and TCD determination of internal carotid artery and vertebral artery blood flow changes in evaluation of cerebral circulation function therapeutic equipment therapy in treatment group and control group is effective and feasible. Results:Different stages of stroke treatment group after treatment of acute phase of the neural function defect score in the control group (t=-13.26, P<0.05), and factors caused troke such as blood pressure and cholesterol leels also declined significantly than control group (t=7.39, t=7.75, P<0.05), cerebral blood flow between treatment group and the control group after treatment mainly reflected in the increased blood flow of part of left and right internal carotid artery and Vertebral Artery ,as Vd, Vs and Vm. Conclusion:Against the illness severity stroke with drug therapy on the basis of brain circulation function therapeutic instrument to stimulate rehabilitation treatment is convenient, economic, safe, effective, can be applied to stroke periods with high blood pressure, cholesterol levels, but acute stage used the best, after post-stroke patients vital center smoothly, can take early used cerebral circulation function therapeutic equipment for rehabilitation.
6.Clinical research of relationship between silent brain infarction and coronary heart disease

Objective:To explore the relationship between coronary heart disease (CHD)and silent brain infarction (SBI).Methods:A total of 430 patients who had received coronary angiography were enrolled,including 330 CHD patients and 100 non-CHD patients.According to complicated with SBI or not scanned by cranial MRI,CHD pa-tients were divided into CHD+SBI group (n=208)and CHD without SBI group (pure CHD group,n=122).Levels of high sensitive C reactive protein (hsCRP)and homocysteine (Hcy)were measured,and coronary artery score (CAS)and Gensini score (GS)were calculated in all patients.Results:Incidence rate of complicated SBI in CHD patients was significantly higher than that of non-CHD patients (63% vs.31%,P <0.01);compared with pure CHD group,there were significant rise in age [(60.79±9.65)years vs.(67.14±9.77)years],percentages of hy-pertension (41.0% vs.66.8%),hyperlipidemia (37.7% vs.55.8%)and levels of hsCRP [(3.92±2.17)g/L vs. (5.85±6.27)g/L],Hcy [(9.20±3.20)μmol/L vs.(10.54±4.25)μmol/L],CAS [(0.88±0.88)scores vs. (1.94±0.94)scores]and GS [(18.62±16.23)scores vs.(53.04±39.61)scores],and percentage of left anterior descending artery stenosis (25.4% vs.69.2%)in CHD+SBI group,P <0.01;Logistic regression analysis indicated that risk factors of SBI were age (OR=1.07,95%CI:1.032~1.108),hypertension history (OR=2.195,95%CI:1.104~4.366),Hcy (OR=1.088,95%CI:1.001~1.182),CAS (OR=1.776,95%CI:1.049~3.007)and GS (OR =1.039,95%CI:1.013~ 1.066),P <0.05 or <0.01. Conclusion:Incidence rate of SBI in CHD pa-tients is significantly higher than that of non-CHD patients;the more severe coronary artery disease is,the higher percentage of complicated SBI is.CAS and Gensini score abnormities suggest there may be SBI existence simultaneously.
7.Changes of haemorheology during induction and maintenance of anesthesia

Haemorheological changes, including blood viscosity (?b) and plasma viscosity (?p) measured on a cone-plate viscometer, erythrocyte aggregation calculated, erytkrocyte deformability (DI) determined by the nuclear pore microfiltration technique and red blood cell shape using scanning electromicroscopy, were observed in patients during anesthesia There acs a significant reduction in ?b (at shear rates range 7. 68- 307. 20 s-l ), ?p and haemotocrit after induction (preintubation ) and 30 min after enflurane in the presence of iv fentanyl, compared with pre-aneathesia, accompanying remarkable increased DI. No changes of erythrocyte shape, however, was found add, nor was the difference of the index between induction period and maintenance of anesthesia. These findings suggest that general anesthesia leads to haemorheological alterations which may improve the microcirculation of the patients.

Objective To evaluate effects of pulmonary surfactant (PS) containing ketamine on respiratory failure induced with lung-lavage in rats Methods Twenty Wistar rats were anesthetized with intraperitoneal pentobarbital sodium and were ventilated with the peak inspiratory pressure(PIP) being 1 47 kPa and E:I at 1:1 following intubation through tracheotomy In all rats whole double lung lavage were performed with normal saline (37℃, 40 ml?kg -1 ) 8 10 times after PIP and PEEP were adjusted to 1 96 kPa and 0 49 kPa respectively The rats were randomly divided into A and B groups after PaO 2 decreased to less than 12 kPa In group A (n=10): PS (25mg, 0 5ml) was injected intratracheally to lungs; in group B (n=10): PS (25mg, 0 5ml) containing ketamine 2 5 mg was injected At 120th min after PS injection, PIP and PEEP were gradually adjusted to 1 47 kPa and 0 kpa (ZPEEP), respectively Results PaO 2 in both groups decreased significantly from 59 3 kPa to 10 3 kPa after lung lavage(P0 05 vs pre-lavage) All rats in group B and 4 of 10 rats in group A survived at the end of experiment Conclusions Intratracheal administration with PS containing ketamine not only reverses hypoxemia , but also maintains the respiratory function under low airway pressure without PEEP after respiratory failure induced with lung lavage,
9.Effects and mechanism of celecoxib on rat colitis induced by trinitrobenzene sulfonic acid

Objective An investigation was conducted to assess the effects and mechanism of celecoxib [a selective cyclooxygenase(COX) 2 inhibitor] on a rat colitis model induced by trinitrobenzene sulfonic acid (TNBS). Methods The rats were divided into four groups. Group 1 and group 2 were experimental groups. Group 3 and group 4 were control groups. Colitis was induced by intracolonic administration of TNBS (25 mg/ml) in a vehicle of 50% ethanol (0.25 ml) in rats of experiment groups. Three hours before induction of colitis ,the rats were beginning and continuing to treat orally with celecoxib (1.25 mg/kg, group 1) and distilled water (1 ml/0.3 kg, group 2) twice per day for 7 days , respectively. The rats in group 4 were treated orally with celecoxib (1.25 mg/kg) twice per day for 7 days. Group 3 served as healthy control. All rats that survived until the end of the experiment (7 d) were killed and the severity of damage were assessed. The prostaglandin E2 (PGE2) concentrations of colonic mucosa were tested by radioimmunoassay. Results The colonic damage scores were 11.15?3.3 in group 1 and 8.50?2.82 in group 2. 10.Study on the Preparation of Euphol Reference Substance in Euphorbia pekinensis

OBJECTIVE:To establish a preparative separation method for euphol reference substance in Euphorbia pekinensis. METHODS:Parts of petroleum ether extraction from E. pekinensis ethanol extract were separated by silica gel column chromatogra-phy with petroleum ether-ethyl acetate(95∶5-70∶30,V/V)by gradient elution. The enriched fractions of euphol were collected. With the methanol repeated recrystallization,nuclear magnetic resonance spectroscopy(NMR)method,mass spectrometry(MS)meth-od and other spectroscopic methods were used to identify the chemical structures,and thin layer chromatography (TLC),UV, HPLC-UV,HPLC-MS were combined to detect the mass fraction. RESULTS:The mass fraction of euphol reference substance sepa-rated from E. pekinensis was>99%. CONCLUSIONS:The reference substance of euphol acquired by this method meet the relative requirements of the chemical reference substance in the content dertermination of TCM new drug quality standard. It provides chemi-cal reference substance for the quality control of E. pekinensis and prescription preparations containing E. pekinensis and the basic research of effective substances.
