Lgr5 is one of the G protein-coupled receptors families, the newest research displays that it has the potency as the special marker of stem cell, it also has a great relationship with tumor formation and development. Lgr5 may be a marker of tumor stem cells, and can be the new treatment target.

Objective This study aims to investigate the correlation of CD4+ CD25+ FOXP3+ Tregs and PD‐1+ CD4+ T cells located at cervical lesions and the prognosis of CIN .PD‐1 expression on T cells induces Tregs .Methods Cervical lymphocytes were collected from CIN patients and analyzed by FACS .Comparisons were made between populations of cervical Tregs and PD‐1+CD4+T cells in CIN regressors and non‐regressors .Results A median of 11 .8% of cervical CD4+ T cells were CD4+ CD25+ FOXP3+Tregs ,while a median of 30 .3% were PD‐1+ CD4+ T cells .The proportions of CD4+ CD25+ FOXP3+ Tregs and PD‐1+ cells were significantly lower in CIN regressors when compared with non‐regressors .Conclusion The prevalence of CD4+ CD25+ FOXP3+Tregs negative correlates with spontaneous regression of CIN ,suggesting that cervical Tregs may play an important role in HPV‐related neoplastic immunoevasion .
4.Effect of high epidural anesthesia on interleukin-6 and soluble interleukin-2 receptor in patients with dilated cardiomyopathy

BACKGROUND: Serum cytokines in patients with dilated cardiomyopathy are increased obviously, and the expression of interleukin-6mRNA is also observed in myocardial tissues. High epidural anesthesia can block the vicious cycle involving cytokines and improve cardiac function. OBJECTIVE: To observe the changes of interleukin-6 and soluble interleukin-2 receptor in patients with dilated cardiomyopathy after high epidural anesthesia treatment. DESIGN: A case-controlled analysis. PARTICIPANTS: Thirty-five inpatients with dilated cardiomyopathy were selected from the Department of Cardiology, First Hospital Affiliated to Harbin Medical University, from October 2001 to May 2002. All the patients were randomly divided into high epidural anesthesia group and conventianal treatment group. High epidural anesthesia group consisted of 22patients, 15 males and 7 females, whose cardiac function was classified into grade Ⅱ in 4 patients, grade Ⅲ in 9 and grade Ⅳ in 9. Conventional treatment group consisted of 13 patients, 11 males and 2 females, whose cardiac function was grade Ⅱ in 1 patient, grade Ⅲ in 5 and grade Ⅳ in 7. Healthy control group comprised 21 people, 13 males and 8 females,who received physical examination at the same period. INTERVENTIONS: Patients with dilated cardiomyopathy were treated with high epidural anesthesia and conventional method, whereas those in conventional treatment group were treated with conventional method only.Elbow venous blood of 3 Ml was sampled from all patients on empty stomach in the morning before treatment and after 4-week treatment. The level of serum interleukin-6 and soluble interleukin-2 receptor was measured with enzyme-linked immunoadsorbent assay. MAIN OUTCOME MEASURES: The level of serum interleukin-6 and soluble interleukin-2 receptor of patients in eachgroup. RESULTS: Blood samples of the 56 subjects were qualified and entered the final analysis.①The level of serum interleukin-6 was higher in dilated car diomyopathy group (13.9ng/L) than in healthy control group (11.22 ng/L) (z= -3.072, P < 0.05). ② The level of serum interleukin-6 in high epidural anesthesia group was obviously decreased aftertreatment (11.42 ng/L) as compared with that before treatment (20.42ng/L) (z =2.582 9, P < 0.05).The le vel of serum interleukin-6 in conventional treatment group was similar before and after treatmant(12.16 ng/L and 12.80 mg/L, z = -1.89,P > 0.05). The difference of interleukin-6 level in high epidural anesthesiagroup (-2.04 ng/L)was obviously higher than that in conventional treat ment group (0.28 ng/L) (P < 0.01) before and after treatment.③The level of serum soluble interleukin-2 receptor was higher in dilated car diomyopathy group[(1 306.17±1.46)ng/L]than in healthy control group [(1078.95±1.23) ng/L] (t =2.51, P < 0.05). ④ The level of soluble intedeukin-2 receptor in high epidural anesthesia group was decreased after treatment [(1 086.68±1.34)ng/L]as compared with thatbeforetreatment [(1 328.01±1.51) ng/L], (t =2.145, P < 0.05). The level of soluble interleukin-2 receptor in conventional treatment group was similar before and after treatment [(1 473.33±1.66) ng/L and (1 331.07±1.52) ng/L,t=-1.06, P > 0.05]. CONCLUSION:The level of serum interleukin-6 and soluble inter leukin-2 receptor is significantly decreased after high epidural anesthesia, suggesting that high epidural anesthesia can regulate cytokines better than conventional treatment.The regulation of high epidural anesthesia is related to inhibition of sympathetic nervous system and humoral-im mune system.

Three nuclear type segeragants were isolated from a heterokaryon of Fusarium oxysporum f.sp.vasinfectum strain Ag149.Their cultural character and pathogenicity were different.The differences of their GC content for total DNA and nuclear DNA were between 0 1% to 0 4% and 0 4% to 0 8% respectively,as measured by thermal denaturation method.The 18S rDNA partial sequences of 1477 base pairs in these segregants were identical.
6.CT anatomic study and 3D reconstruction of lumbar nerve roots and main adjacent structures

Objective To observe the positional relationship of lumbar nerve roots and its main adjacent structures with CT data, and to conduct 3D construction and measurement,so as to provide anatomical basis for percutaneous lumbar diskectomy(PLD).Methods CT serial tomography dataset of 30 healthy adults' lumbosacral portions were imported into Mimics 17.0 software to segment related structures and obtain the layers of different structures.The structures were 3D reconstructed and virtually demonstrated.The lumbar nerve roots and their main surrounding structures were observed and measured.Results The minimum distances between the lumbar nerve roots and the superior articular processes increased gradually from L3 to L4 intervertebral disc layer to L5 to S1 intervertebral disc layer.The 3D model clearly displayed the morphology and spatial relationship of the lumbar nerve roots and their adjacent structures in lumbosacral portion.The lower the lumbar intervertebral discs were,the thicker they were.The distances from the middle point of the back of lumbar intervertebral disc to the end of the superior spinous process of lumbar vertebra increased at first and decreased then,and the distance in L3 to L4 intervertebral disc layer was the farthest.Conclusion The relationship between lumbar nerve roots and superior articular processes,the intervertebral disc thickness variance and the change of the distances from lumbar intervertebral disc to the end of the superior spinous process have been analyzed in this study,and the clinic basis has been laid for percutaneous lumbar diskectomy.
7.Effect of hypoxic preconditioning on levels of cPKC ?I and ?II membrane translocation in the mouse brain

AIM: To explore the role of cPKCs in the development of cerebral hypoxic preconditioning, the effect of repetitive hypoxia on the level of protein kinases C ?I and ?II (cPKC ?I and ?II ) membrane translocation in the brain of mice was observed. METHODS: The hypoxic preconditioned mouse model was adapted with minor modification from our previous report. The biochemistry techniques of SDS-PAGE and Western blotting were applied to determine the level of cPKC ?I and ?II membrane translocation in cortex and hippocampus of mice. RESULTS: cPKC ?II translocated from the cytosolic fraction to the particulate fraction in response to the repetitive hypoxic exposure (H1-H4) both in hippocampus and cortex of mice, which was regarded as membrane translocation. The significant membrane translocation of cPKC ?II was found in hippocampus of H4 group (173.3%?21.3% vs H0: 100% or H1: 79.5%?10.7%, P0.05, n=8). CONCLUSION: These results suggest that cPKC ?II may play an important role in the development of cerebral hypoxic preconditioning, but the changes of novel and typical PKC isoenzymes are still under investigation.
8.A case-control study about the preventive effect of magnesium sulfate on hypertention caused by ACTH in the treatment of infantile spasms

Objective To examine the preventive effect of magnesium sulfate on hypertension caused by ACTH in the treatment of infantile spasms (IS). Methods 46 children diagnosed as IS were recruited from two hospitals during May, 2011 to October, 2013.23 patients in group A (treatment group) were treated with magnesium sulfate and ACTH in hospital A;another 23 cases in group B (control group) were treated with ACTH only in hospital B. The therapy course was 2 weeks. Results Hyperten-sion was not observed in the treatment group, while 6 children were observed with hypertension in the control group. There was signiifcant difference between the two groups (P<0.05). Conclusions Magnesium sulfate could prevent the incidence of hyper-tension in the treatment of IS with ACTH, and beneift the completion of treatment course.

Objective: To establish a quality control standard for YurongXiaocuo granule.Methods: TLC was used to identify Radix Puerariae,Fructus Forsythiae,Bulbus Fritillariae Thunbergii,while the content of tanshinoneⅡA was determined by HPLC.Result: The methods of qualitation and quantitate was simple,accurate and with strong specifi city,the good linear rang of tanshinoneⅡA was 3.2-32?g and the average recovery was 99.51%(RSD=2.74%).Conclusion: This method can control effectually the quality of YurongXiaocuo granule.
10.Analysis of distribution and drug resistance of pathogens in premature infants with ventilator-associated pneumonia

Objective To investigate the distribution and characteristics of drug resistance of pathogens in premature infants with ventilator‐associated pneumonia(VAP) ,and explore the therapeutic measures for premature infants with VAP ,so as to provide references for clinically reasonable administration of antibacterial agents .Methods A total of 54 cases of premature infants diag‐nosed with VAP in the First Affiliated Hospital of Xiamen University from January 2013 to June 2014 were enrolled in this study . Specimens of respiratory tract secretion were collected ,and species identification of pathogens and drug sensitivity test were per‐formed by VITEK 2 Compact system .Results A total of 69 strains were isolated .Gram negative bacteria was accounted for 94 .20% ,and gram positive bacteria was accounted for 5 .80% .Four of the most common pathogenic bacteria were Klebsiella pneu‐monia (29 strains ,42 .03% ) ,Enterobacter aerogenes (12 strains ,17 .39% ) ,Pseudomonas aeruginosa (10 strains ,14 .49% ) and Stenotrophomonas maltophilia(7 strains ,10 .14% ) .The results of antibiotics‐sensitivity tests showed that these strains were multi‐drug resistant to most commonly used antibiotics ,but sensitive to only a few antibiotics .Conclusion Gram‐negative bacteria are the major pathogens for premature infants with VAP in this hospital .Mixed infection and multidrug resistance are common .Therefore , the rational use of antibiotics according to drug sensitivity tests′results is critical in treating VAP .
