1.Effects of selective psychological intervention on cytokine and unhealthy emotion in patients with breast cancer

Objective:To study the role of selective psychological intervention on peripheral blood cytokine(interleukin-2,interleukin-4,tumor necrosis factor-?)(IL-2,IL-4,TNF-?)and unhealthy emotion in patients with chemotherapeutical breast cance.Methods:Randomized-control study method was adopted.Eighty chemotherapeutical inpatients with breast cancer were divided into intervention group and control group(40 patients in each group).Patients in both groups received mutine chemotherapy and health education,while the intervention group also received selective psychological intervention.The psychological and behavior rating were observated with Zung anxiety scale(SAS)/Zung depression scale(SDS)and symptom checklist(SCL-90)pre-and three month post-chemotherapy.Psychological intervention methods were selected independently and completed by patients.The effect of selective psychological intervention on unhealthy emotions were observed in breast cancer inpatients undergoing chemotherapeutics.The changes of IL-2/IL-4/TNF-?in peripheral blood were detemined by radioimmunity.Results:The breast cancer inpatients in both groups were noticeably depressed after chemotherapy(P0.05).Conclusion:The selective psychological intervention could inhibit the unhealthy emotion of breast cancer patients undergoing chemotherapy and increase the content of some cytokines in peripheral blood,which may be related with immunity enhancement.
2.Features and applications of acid-base changes in cardiopulmonary resuscitation

The cases of cardiac arrest are increasing because of the increased incidence of cardiovascular diseases. Cardiopulmonary resuscitation (CPR) is the necessary measure to rescue these patients. But the success rate of recovery is still low. Acid-base disturbance is one of the important factors affecting the recovery effect. Overseas and domestic studies in recent years focused on the acid-base changes and mechanism in CPR have founded that it exits extraordinary hypercapnia and CO2 spike, which have important guidance to the clinical work. The research for the arterial blood gas analysis during CPR, changes in the exact time, and treatment of hypercapnia and acid-basic change in different etiology and different populations were very few, therefore the a lot of clinical data is needed to accumulate for further study.
3.Reduction of fat embolic risks in total hip arthroplasty using cannulated rasps for preparation of femoral canal

Objective To describe the effect of cannualted rasps in reduc in g the fat embolic risks during preparation for femoral canal in experimental tot al hip arthroplasty. Methods Twelve mongrels were used to perform resection of b ilateral femoral heads, and randomly assigned to one of two groups with six each . In experiment group, impacting cannulated rasp was used in preparing femoral c anal; while, in control group, impacting traditional rasp was used. Both the ras ps were of same shape; however, there was a 4 mm diameter canal inside the cannu lated rasp connecting its two ends. A bone hole was made through the lateral fem oral cortex to the femoral canal at a point 12 cm distal to the greater trochant er in order to measure the intramedullary pressure of femoral canal when the fem oral canal was prepared with either rasp. The pathology of the postmortem pulmon ary tissue was analyzed. Using quantitative morphometry, the size of fat embolus in the lung tissues and the volume proportion of lung tissues occluded by fat w ere measured respectively. The statistical differences between the two groups we re analyzed using t test. Results In control group, the intramedullary pressure during preparation of femoral canal with traditional rasp increased with a peak of (34.5?10.2)kPa, pulmonary fat emboli were found in all mongrels, the volum e proportion of postmortem lung tissue occluded by fat was 0.64%, and pulmonary arterial pressure increased to (2.2?0.4)kPa[the normal value was (0.8?0.3 )kPa]; while in the experiment group, the intramedullary pressure peak increas ed to (22.4?7.7)kPa, significantly lower than that of the control group; the volume proportion of lung tissue occluded by fat was 0.21% which was also signif icantly lower than that of the control group, and pulmonary arterial pressure wa s (1.7?0.4)kPa. The differences between the two groups were significant in al l measured values. Conclusion The present study suggests that conventional prepa ration of canal can cause an increase of intramedullary pressure, which may lead to pulmonary fat embolism and cardiopulmonary impairment. However, the cannulat ed rasp can decrease the volume proportion of lung tissue occluded by fat and al leviate the influence of preparation of canal on cardiopulmonary function.
4.Study on apoptosis of the HCT116 cells mediated by cisplatin and its mechanisms

Objective To study the apoptosis mediated by cisplatin on human colorectal tumors (HCT116) cell line and its mechanisms in vitro. Methods The apoptosis levels of HCT116 cells mediated by cisplatin at vari-ous time and in different concentration were measured by M30-ApoptosisTM -ELISA-kits and flow cytometry assay. The expressions of the protein p53,p21 and Bcl-2 were assessed through Western-Blotting. Results Cisplatin in-hibited the proliferation in a time-and dose -dependant manner ( F = 1129. 383, P = 0. 000 and F = 125. 267, P = 0. 000, respectively). The sub-G1 peak detected by flow cytometry at 24,48,72 hours of the apoptosis rates showed a significant difference between the experimental group and the control group (χ2= 5. 669,14.110,12. 221, P = 0. 010,0.003,0. 000,respectively). We found significant differences on the HCT116 cell growth between different time under cisplatin effect (χ2 = 14.008 ,P =0. 003 ). There were significant difference on the CK18-Asp237-Asp396 re-leased by HCT116 cell between different casplatin concentration (F =48. 667 ,P =0.000) as well as different times ( F = 1194. 394, P = 0.000). The Western-Blotting results indicated that the expression level of the p53 ( t = 9.873, -2.906,7. 229,2.776,P =0.000,0. 007,0. 000,0. 011 ) and p21 (t = - 10. 692, - 8. 867, - 15. 063, - 16.281, P = 0. 000,0.001,0.000,0.000 ,respectively) increased gradually, while there are no effect on the expression of the protein Bcl-2(t=1.429,2.011,2.247,2.001,P=0. 178,0.069,0.053,0.062,respectively). Conclusions Cis-platin can induce the apoptosis on HCT116 cells and thus inhibit the reproduction of tumor cells by recovering the function of p.53.

Objective To observe the expressions of Clara cell secretary protein (CC16) mRNA and protein in senile rats. Methods The histological changes, the level of CC16 mRNA in lung tissue and CC16 protein in terminal bronchioles and respiratory bronchioles were measured in 10 senile rats (at the age of 26 months) and compared with 7 young rats (aged 3 months). Objective: To study the effect of Interleukin(IL)-17 on neutrophil apoptosis and try to explain the possible mechanism involved. Methods: Neutrophils isolated from healthy donors were incubated in enriched RPMI 1640 cell culture medium at 37 ℃ in 5% carbon dioxide. Subgroups were incubated with IL-17, heat-denatured IL-17 (X-IL-17), dexamethasone (DEX), or buffer alone. Apoptosis was assessed by morphologic changes, by detecting DNA strand breaks. Production of proapoptotic protein Bax by neutrophils was evaluated by immunocytochemistry. Results: At the time of neutrophil incubation, neutrophils in the control subsets exhibited morphologic evidence of apoptosis. A steady rise in apoptosis index (AI) was noted, with (1.54?0.08)% for 0 h, (11.48?1.80)% (compared with 0 h, P0.05) for 0 h, (20.47?6.22)% ( compared with control 12 h, P0.05).Neutrophils apoptosis was accompanied by DNA fragmentation. Objective:To investigate the significance of 25 ( OH )-vitamin D in its diagnostic value of primary biliary cirrhosis. Methods:44 cases with PBC,41 cases with Hepatitis B cirrhosis and 110 normal people as control group were all analyzed bi-ochemical results and significance of serum 25 ( OH )-vitamin D. We Used receiver operator characteristic curve ( ROC ) to analyze serum 25(OH)-vitamin D in its diagnostic value of primary biliary cirrhosis. Results:Biochemical results of TBIL,DBIL,IBIL,GLB, AST,ALT,ALP,γ-GT were higher in PBC than that in normal people and Hepatitis B cirrhosis. While the serum 25(OH)-vitamin D of PBC was much more lower than that in normal and people and Hepatitis B cirrhosis. The difference between them were statistical difference(P<0. 05). The area under the ROC curve of PBC was 0. 926,it showed serum 25(OH)-vitamin D diagnosis for PBC had quite value. When the cut-off level of serum 25(OH)-vitamin D was 17. 275 ng/ml,the sensitivity for the diagnosis was 92. 8%,the re-spectively was 84. 1%. Conclusion:Serum 25(OH)-vitamin D diagnosis for PBC has quite value,the cut-off level of serum 25(OH)-vitamin D is 17. 275 ng/ml.
9.Causes of false-positive anti-HCV in non-syndromic cleft lip and palate with serum chemiluminescence test

Objective:To investigate the causes of false-positive anti-HCV in non-syndromic cleft lip and palate with serum chemiluminescence test. Methods:We performed a hospital-based,case-control study of 4 050 patients with non-syndromic cleft lip and palate and 8 547 control patients without cleft lip and palate to evaluate associations between positive anti-HCV and non-syndromic cleft lip and palate. CLIA test were used for detect anti-HCV in control group. RIBA and FQ-PCR were used for confirming the result of the positive samples of CLIA test. CLIA test were used for detect anti-HCV in case group until CLIA test was negative. IgG,RF and ANA were used for case-control comparisons. Results:The detection rate of anti-HCV were higher in the case group than that of control group (P<0. 05). The false-positive rate was 1 percent based on testing of 4 050 patients. These samples were negative after opration 1-19 months. There were no change among IgG, RF and ANA in the false-positivepatients. Conclusion: False-positive anti-HCV in non-syndromic cleft lip was possible associated with abnormal plasma metabolic product. It can be excluded by track monitoring.

Objective This study was designed to investigate the importance of H2S in rats with adriamycin-induced dilated cardiomyopathy.Methods Weight-matched adult male Wistar rats were randomly divided into 5 groups as follows:(1)the ADR group(n=12)where adriamycin(2.5 mg/kg)was given once a week;(2)ADR + small-dose NaHS group(n=12)where NaHS solution [2.8 ?mol/(kg?d)] was given;(3)ADR+large-dose NaHS group(n=12)where NaHS solution [14 ?mol/(kg?d)] was given;(4)the control group(n=9)where physiological saline was administered instead of adriamycin.(5)NaHS group(n=9)where NaHS solution [14 ?mol/(kg?d)]was given.Hemodynamic and echocardiographic measurements were carried out after treatment.Histological examination was implemented at the 10th week after the rats were sacrificed.Meanwhile,H2S concentrations in serum and myocardial tissues were evaluated by modified sulfide electrode method.Results The heart function of ADR rats decreased obviously.Pathological examination demonstrated that characteristics consistentwith cardiomyopathy were found in ADR group.H2S concentrations in serum and myocardial tissues of the rats in ADR group were both significantly decreased as compared with those in the control group(both P
