1.Cyclooxygenase-2 expression and its localization in the mature rat testis

Objective To confirm the expression and localization of COX-2 gene in mature rat testis,as COX-2 not only plays a central role in inflammation by up-regulating the synthesis of prostaglandins,but also mediates critical reproductive functions by other mechanisms. Methods In the mature Wistar rats,COX-2 expression and its localization were examined by using immunohistochemistry,RT-PCR and Western-blot. Results The results of RT-PCR and Western-blot showed that COX-2 mRNA and its protein exist in the testis. Expression of COX-2 was localized in spermatogenic cells on immunohistochemical study.No COX-2 expression in sertoli cells and leydig cells was found. Conclusions The present study confirmed the expression of COX-2 in rat testis and its localization in spermatogenic cells.These suggest that COX-2 may play a role in spermatogenesis.
2.Genotyping and resistant mutants analysis of P gene in HBV fragment sequencing

Objective To establish a method for efficient,accurate genotyping and nucleoside drug-resistant mutation analysis for hepatitis B virus ( HBV ).Methods The 48 HBV serum samples were collected from the Third People's Hospital of Taiyuan from July to August 2011,and HBV DNA were extracted using the commercial kit.The HBV whole genome and P gene were amplified and sequenced.Each HBV sample was genotyped by both constructing phylogenetic trees and genotyping software analysis.The results from two strategies were compared for every sample.Results A total of 48 HBV full genome sequences were identified into 12 B and 36 C genotype's by both constructing phylogenetic trees and genotyping software analysis,which was exactly the same as the analysis using P gene fragment sequencing.Seven forms of nucleoside drug-resistant mutation were found in the P gene for all the samples,with the ratio of 27.1% ( 13/48 ),in which all the mutation forms were associated with lamivudine or adefovir,and no other nucleotide drugs-related resistance mutations existed.In addition,there were 11 B and 35 C genotype and 2 B/C hybrid type with the analysis using Real-time PCR genotyping for the 48 samples.Conclusion P gene sequencing can be used as a new clinical method for efficient,accurate HBV genotyping and resistant mutation analysis,which provides guidance for hepatitis B treatment.
3.Comparation of magnetic activated cell sorting and chemotherapy agent DDP in enriching cancer stem cell of laryngeal carcinoma cell line Hep-2

Objective To compare effect of chemotherapy agent DDP to MACS in sorting cancer stemcells (CSC) of laryngeal carcinoma cell line Hep-2. Methods CD133 magnetic beads were applied to sort Hep-2 cells. Different dosages of DDP were used to treat Hep-2 cells for 48 hours. Enrichment rate of CD133+ cells by MACS and after DDP treatment was detected by Flow Cytometer (FCM). Morphologic change was observed under inverse-phase microscope. Results FCM showed that the sorting rate of CD133+ cells through MACS was 64.33 %, while after DDP treatment for 48 hours, the rate of CD133+ cells was enriched significantly in each dosage of DDP, with the maximal rate was 50.7 %, in the dosage of 4 μg/ml. There was a significantly difference between MACS and each of DDP group (P <0.01). Cells treated with DDP were abnormal in morphology. Conclusion MACS and DDP sorting has respective advantages in enriching CSC in Hep-2 cell lines.
4.Technology Study of Separation and Purification of Active Constituents from Morus alba L

OBJECTIVE:To optimize the technology of separation and purification of active constituents from Morus alba L. METHODS:With the adsorption rate and desorption rate as the indexes,static and dynamic macroporous resin adsorption-desorp-tion methods were adopted to optimize the type of macroporous resin,the loading amount of fluid,the volume fraction and amount of eluent in the separation and purification technology of total flavonids. With the yield and content of polysaccharide as the index-es,water extraction and alcohol precipitation method was used to optimize the volume fraction of alcohol for polysaccharide precipi-tation in the separation and purification technology of total polysaccharides. With the absorbance of total alkaloids as the index,ion exchange method was employed to optimize the loading amount of fluid and the amount of eluent in the separation and purification technology of total alkaloids. Then the verification was conducted. RESULTS:The optimal technology of total flavonids from Morus alba L. was with D101 macroporous resin as the adsorbent,loading fluid mass concentration of 0.1 g(material medicinal)/ml,loading fluid amount 9 times as the column volume(BV),70% alcohol of 5 BV for elution. The optimal volume fraction of alcohol for polysaccharide precipitation in the technology of total polysaccharides was 80%. The optimal technology of total alka-loids was with loading fluid amount of 3 BV,0.5 mol/L ammonia water of 8 BV for elution. After separation and purification,the purities of total flavonids,total polysaccharides and total alkaloids from Morus alba L. were 35.68%,47.14% and 55.79%,and the RSDs were 1.28%,1.61% and 1.14%(n=3). CONCLUSIONS:The optimal technology is stable and suitable for the separa-tion and purification of active constituents from M. alba L.
5.Postoperative radiotherapy combined with intravesical chemotherapy for T2/T3 bladder cancer

Objective To compare the result of T2/T3 transitional cell carcinoma (TCC) of the urinary bladder after segmental cystectomy, treated by postoperative radiation plus intravesical chemotherapy and postoperative intravesical chemotherapy alone.Methods From 1985 to Dec.1995 patients with T2/T3 TCC bladder cancer who had been treated by segmental cystectomy were eligible for this retrospective analysis. Fifty-eight patients received postoperative radiotherapy plus intravesical chemotherapy (RT+IVC) and 35 patients were given postoperative intravesical chemotherapy (IVC) with thio-TEPA or calmette-Gue'rin bacilli (BCG). For radiation,8 or 18 ?MV X-ray was given with total dose of 50-60 ?Gy. Vesicoclysis was performed on 50-60 mg thio-TEPA twice per week and 0.5 mg BCG per week.Results The 3-year local control rates of RT+IVC and IVC groups were 68.6% and 48.2% showing a difference statistically significant (? 2=4.08,P=0.044).The 3- and 5-year survival rates of RT+IVC and IVC groups were 70.7%,49.5% and 59.9%,35.7% ,showing no significant difference (? 2=1.77,P=0.184). Among the 5 year survivors of the RT+IVC patients, 78.6% had their bladder preserved. Though untoward radiation reactions were severer, they were tolerated well.Conclusions Combined radiation therapy plus intravesical chemotherapy is indicated for T2/T3 bladder cancer after segmental cystectomy. Multimodality therapy is more favored to improve both the local control and the possibility of preserving the bladder.
6.Research of urinary incontinence and depression among aged female patients

Objective To explore the prevalence and relationship of urinary incontinence and depression among elderly female patients.Methods A total of 118 female patients older than 60 years old were interviewed face to face.The prevalence of urinary incontinence and depression were estimated using the International Consultation on Incontinence Questionnaire Urinary Incontinence Short Form-Chinese (ICIQ-SF-Chinese) and Geriatric Depression Scale-15 (GDS-15).Linear regression and Spearman correlation were used to identify the relationship between urinary incontinence and depression.Results Urinary incontinence was found in 27.1% (32/118),and depression was in 39.0% (46/118) of the survey population.Urinary incontinence with depression was found in 13.6% (16/118) of the overall population.Urinary incontinence was positively correlated with depression,r=0.211,P<0.05.The frequency of entertainment and urinary incontinence were significantly related to depression symptoms,P<0.05.Conclusions Urinary incontinence appears to possess an additive effect on incidence of depression,which seriously affects both physical and mental health of patients.Clinicians should give patients health education and intervention measures at early stage.
7.The Effects of Gantaikang on Immunological Hepatic Fibrosis Induced by Porcine Serum in Rats

Objective To explore the effects of Gantaikang on porcine serum-induced liver fibrosis in rats. Methods Hepatic fibrosis was induced in rats by intraperitoneal injection of 0.5ml porcine serum twice a week. Gantaikang was simultaneously administered intragastrically to rats in the intervention group at dose of 0.27g/kg?bw for 7 weeks. Then blood was collected, and serum levels of hyaluronic acid(HA), laminin(LN) and precollagen III(PCIII) were measured. Liver tissues was removed, and the pathologic changes of liver were observed by routine, Masson and James staining. The semi-quantitive analysis of collagen and reticulin in the liver tissues were performed. Results Gantaikang could effectively counteract the increase of HA, LN and PCIII level in serum, and attenuate hepatic collagen and reticulin proliferation induced by porcine serum in rats. Conclusion The results suggested that Gantaikang could, to a certain extent, prevent and attenuate immunologic hepatic fibrosis induced by porcine serum in rats.
8.Detection for fetal DNA in maternal plasma by fluorescence quantitative polymerase chain reaction

Objective To investigate the fluorescence quantitative PCR (FQ-PCR) based on TaqMan-MGB( Minor Groove Binder) technique for quantification of fetal DNA in maternal plasma and its variation during pregnancy. Methods Maternal DNA extracted from 237 plasma samples obtained from 30 pregnant women (5-40 gestational weeks and post delivery). The TaqMan-MGB probe and SRY primers were designed to amplify the SRY gene sequence of Y chromosome in maternal plasma by FQ-PCR. Results This system was sensitive enough to detect a male DNA among 20 000 female DNA. Fetal DNA can be detected in maternal plasma as early as 6+6 weeks of gestation and increased with the pregnant progress with the peak level at the third trimester. Between 24- 48 h after delivery, the SRY gene was negative in maternal plasma. The percentage of fetal DNA concentration in maternal total plasma DNA was 4. 88% in the first trimester, 6. 10% in the second and 4. 77% in the third trimester. The SRY positive signal was obtained from samples of 13 women bearing male fetuses and no signal was detected for all of the 17 women bearing female fetuses. Conclusions The FQ-PCR for quantification of fetal DNA in maternal plasma is highly sensitive, specific and reliable. Fetal DNA does present in maternal plasma at a higher concentraton. FQ-PCR may be useful in nonin-vasive prenatal diagnosis.
