1.GC Fingerprint analysis for quality control of volatile oil in Radix Bupleuri

Objective To establish the GC fingerprint analysis for the quality control of volatile oil in Radix Bupleuri. Methods The GC with capillary column DB-1 (30 m?0.25 mm?0.25 ?m) was used. The column was maintained at 50 ℃ for 5 min after injection then programmed at 3 ℃/min to 170 ℃ and at 5 ℃/min to 230 ℃ which was maintained for 5 min. Gasification temperature: 250 ℃, carrier gas: N_2, flow rate: 1.03 mL/min, inlet volumn: 0.5 ?L, spliting ratio: 10∶1, FID Injector temperature: 250 ℃. The internal standard was [WTBX]n-nonane used to determine 25 batches of Radix Bupleuri from different habitats by GC fingerprint. Results The 25 batches of Radix Bupleuri are classified to be the qualified and unqualified based on the results of cluster and similarity analyses. Conclusion The method is simple and reliable and it is capable of effectively controlling the quality of Radix Bupleuri.
2.Application of Circulation Water to Induce Hypothermia in the Operation of Treating Upper end of Shinbone Tumor

It had some advantages when microwave induced hyperthermia were used to treat inferior femora and superior shinbone tumor.The device of circulation water,which could induce hypothermia during the treatment procedure needed to be used.Large amount of normal saline at the room temperature was perfused through this device,so the temperature of surrounding normal tissues and organs was reducd and the functions of nerve,vessel and knee were protected.It is safe and convenient measure that keep the operation go on smoothly.Summarized 54 cases of superior shinbone tumor that were used this device in the operation and experienced that it provided an effectiveness support for the operation.
3.Determination of Chlorinated Anilines Residues in Soil by Gas Chromatography-Triple Quadrupole Mass Spectrometry Coupled with Solid Phase Microextraction

A gas chromatography-triple quadrupole mass spectrometry coupled with solid phase microextraction method ( SPME-GC/QQQ) was developed for the determination of chlorinated anilines ( CAs) residues in soil. The multi-reaction monitoring ( MRM ) parameters were optimized. The SPME conditions such as amount of soil sample, volume of added water, extraction temperature and absorption time were investigated. Optimum SPME conditions including 8 g of soil sample, 4 mL of water, 80℃ of extraction temperature and 40 min of absorption time were employed in the experiment. Method detection limits were within the range of 1-10 pg/g, while linearities were within the range of 5-1000 pg/g. Under the optimized conditions, the soil samples spiked with 200 pg/g targets were determined with recoveries of 96. 8%-144. 2%and relative standard deviations of 13 . 4%-27 . 8% ( n=5 ) . The proposed method combines simplicity and rapidity of SPME and high selectivity of MRM. It is especially suitable for the quick screening and determination of trace amounts of chlorinated anilines residues in soil.
4.Clinical and Pathological Analysis of Acute Renal Failure in Children

Objective To study the etiology, clinical and pathological characteristics, therapy and prognosis of acute renal failure (ARF). Methods The literature of 80 treated children with ARF from 1988. 1 to 2003.10 was reviewed. Results Sixty three cases (78.8%)suffered from mtrarenal ARF;47 glomerulonephritls (58. 8%); 15(18. 8%)prerenal ARF;2(2 5%)postrenal obstructive ARF. Twenty three cases with renal biopsy,9(39. 1%)MsPGN;4 (17. 4% ) MPGN; 4 (17. 4%) crescentic glomerulonephntis; 3 (13% ) endocapillary glomerulonephritls;2(8 7% )proliferative and sclerosing glomerulonephritis; 1(1. 3% )minimal changes. According to the different etiologic and pathological characteristics,different therapies were applied,infusion,oral prednisone ,and intravenous pulses of methylprednisone and cyclosphosphamide ,dialysis, or diuretic therapy and lowering blood. Thirty one (38. 8% )cases recov-ered;27(33 8% )cases improved; 10( 12.5% ) cases gave up treatments;21 (26 3% )died. 6.Study on the inhibitory effect of decorin on rat mesangial cell growth and its signal transduction pathway

Objective To explore the inhibitory effect of decorin(DCN) on rat mesangial cell (MsC) growth and on the expression of mitogen-activated protein kinases (MAPKs) and p21 protein. Methods Lipofectin-mediated method was used to transfect DCN vector into MsC. After the screen and identification of transfected MsC, DCN-containing supernatant was collected and added into the culture medium of normal MsC, then flow cytometer was used to detect the cell cycle. Objective To explore whether the antagonistic effect of decorin (DCN)on progression of glomeruloselerosis is associated with the inhibition of the expression of type I and type II transforming growth factor-? receptors (TGF-?R I and TGF-?R II )in mesangial cells (MsC). Methods RT-RCR and Western blot analysis were used to detect the expression of TGF-?R I and TGF-?R II mRNA and their proteins on cultured rat MsC stimulated by exogenous TGF-?1. Lipofectin-mediated method was used to transfect DCN vector into MsC. After screening and identifying of transfected MsC, RT-PCR and Western blot analysis were adapted to detect the changes of TGF-?R I and TGF-?R II expression respectively. Results The expression of TGF-?R I and TGF-?R II mRNA and their proteins on normal MsC stimulated by exogenous TGF-?1 increased in time-dependent manner and reached the peak at 24th hour. Compared with normal and untransfected MsC (1P-1), the mRNA and protein expression of TGF-?R I and TGF-?R II on MsC (3D-5, 7D-1) transfecled by DCN gene decreased significantly, and DCN gene transfection could antagonize the increase of mRNA and protein expression of both receptors caused by exogenous TGF-?1. Conclusions The expression of both TGF-?R I and TGF-?R II decreases obviously in MsC overexpressing DCN gene, which may be one of the importan antagonistic mechanisms of decorin involved in the development of glomeruloselerosis mediated by TGF-?.
8.Observation on the Effect of Lidocaine Treated in Three Different Ways on the Prevention of General Anesthesia in the Elderly Patients

Objective:To compare the three effects of lidocaine in the prevention of general anesthesia in elderly patients.Methods:A total of 120 elderly patients (65-85 years old) underwent anesthesia with general anesthesia (ASA) Ⅰ ~ Ⅱ were randomly divided into thyrocricocentesis group (group H),throat surface anesthesia group (Group Y),intravenous injection group (group J) and control group (group D).Group H was injected with lidocaine for surface anesthesia;group Y used laryngeal spray for laryngeal sprayed lidocaine for surface anesthesia;group J was anesthetized induction of intravenous lidocaine to prevent intubation reaction;group D the control group was not treated with lidocaine.(SBP) and heart rate (HR) were measured before and after induction (T0),tracheal intubation (T1) and 1 (T2),3 (T3) and 5 min (T4),the changes of hemodynamics related indexes were compared.Results:Compared with the same group of T0,the SBP and HR of four groups of T1 moments were significantly decreased (P <0.05);In the other three groups,SBP was significantly increased at T2 and T3 (P <0.05),HR ofT2 was significantly increased (P <0.05);Compared with group D,SBP and HR in group H,Y and J were significantly different at T2,T3 and T4,he difference was statistically significant (P <0.05);but there was no statistically significant difference between the three groups (P> 0.05).Conclusion:The three ways of lidocaine can be used to prevent the general anesthesia reaction in elderly patients,the effect of three ways is parallel.However,cricothyroid membrane puncture increased the patient's pain,throat spray method increased the cumbersome operation and enhanced the cost of the patient,and intravenous injection method is simple and worthy of popularization and application in clinic.
9.Effects of Acupuncture in Patients with Mild or Moderate Depression Treated with Paroxetine Hydrochloride

Objective To observe the clinical efficacy and reverse reactions of acupuncture combined with paroxetine hydrochloride in the treatment of mild or moderate depression. Methods The patients with mild or moderate depression (n=73) were randomly divided into control group (treated with paroxetine hydrochloride,n=33) and observation group (treated with acupuncture and paroxetine hydrochloride,n=40). The therapeutic course lasted for 6 weeks. The total score changes of Hamilton Depression Scale (HAMD) were observed before treatment and treated for 1, 2, 4 and 6 weeks. Rating Scale for Side Effects (SERS) was evaluated before treatment and treated for 2, 4, 6 weeks.Results The total effective rate of clinical efficacy in the observation group was 78.95% (30/38) and the control group was 68.75% (22/32), without significant difference between the two groups (P>0.05). The scores of HAMD decreased 4, 6 weeks after the treatment in the control group and 2, 4, 6 week after the treatment in the observation group compared with those in the same group before the treatment (P<0.05,P<0.01). There was a significant difference in HAMD scores between two groups after the treatment for 4 weeks (P<0.05). The scores of SERS showed a significant difference 4, 6 weeks after the treatment between the two groups (P<0.05).Conclusion Acupuncture can improve the curative effect of paroxetine hydrochloride and decrease its side effects in the treatment of depression.
10.Effect of rapamycin on apoptosis of mouse astrocytes in vitro

[ ABSTRACT ] AIM: To observe the effect of rapamycin on the apoptosis of mouse astrocytes in vitro.ME-THODS:The astrocytes from C57BL/6J newborn mouse pups were isolated and primarily cultured.The effect of rapamycin on the viability of astrocytes was assessed by MTT assay.The mean fluorescence intensity of SYTOX?Green stain in the astrocytes was detected by fluorescence microplate reader in order to analyze the effects of rapamycin on the cell death in-duced by H2 O2 , ionomycin and/or deferorxamin.DiOC6 (3) staining was used to analyze the mitochondrial membrane po-tential of the astrocytes induced by H2 O2 .Flow cytometry analysis was used to determine the production of ROS in the as-trocytes and mitochondria by staining with H2 DCFDA and MitoSOXTM Red reagent, respectively.RESULTS: Rapamycin at concentration of 0.5 μmol/L protected the astrocytes against cell death induced by H2 O2 or deferoxamine plus ionomy-cin.Rapamycin protected the mitochondrial membrane potential of astrocytes from the injury of H2 O2 .It also reduced the production of ROS in the astrocytes and decreased the level of ROS in the mitochondria.CONCLUSION:Rapamycin re-duces the ROS overload in the mitochondria, keeps mitochondrial membrane potential safety and protects the astrocytes a-gainst apoptosis in vitro.
